Die vorliegende Erfindung bezieht sich auf ein neues Verfahren zur Herstellung von Benzodiazepinderivaten der allgemeinen Formel

worin R1 Wasserstoff, Halogen, nieder Alkyl oder Nitro, R2 Wasserstoff, nieder Alkyl oder Di-nieder-alkylaminonieder Alkyl, R3 Pyridyl Phenyl oder Halophenyl und B Methylen oder Carbonyl bedeuten.
Die erfindungsgemäss herstellbaren Verfahrensprodukte der Formel I sind bekannte Verbindungen und eignen sich als Sedativa, Tranquilizers, Anticonvulsiva und Muskelrelaxantien.
Der in dieser Beschreibung verwendete Ausdruck nieder Alkyl bezieht sich auf geradkettige und verzweigte Kohlenwasserstoffgruppen mit 1-7, vorzugsweise 1-4 Kohlenstoffatomen, wie Methyl, Äthyl, Propyl, Isopropyl u. dgl. Der Ausdruck Halogen umfasst alle vier Halogene, d. h.
Chlor, Fluor, Brom und Jod, wenn nicht anders angegeben.
Die Verbindungen der Formel I werden erfindungsgemäss erhalten, indem man eine Verbindung der allgemeinen Formel

In einer bevorzugten Ausführungsform der vorliegenden Erfindung bedeuten R1 Nitro oder Halogen, vorzugsweise Chlor. welches in 7- Stellung des Benzodiazepinrings sitzt, R2 Wasserstoff oder nieder Alkyl, vorzugsweise Methyl, und R3 Phenyl.
In einer anderen bevorzugten Ausführungsform der vorliegenden Erfindung bedeuten R1 Halogen, vorzugsweise Brom, welches in 7- Stellung des Benzodiazepinrings sitzt, R2 Wasserstoff oder nieder Alkyl, R3 Pyridyl, vorzugsweise 2Pyridyl, und B eine Carbonylgruppe.
In einer weiteren bevorzugten Ausführungsform bedeuten Rl Nitro oder Halogen, vorzugsweise Chlor oder Jod, welches in 7-Stellung des Benzodiazepinrings sitzt, R2 Wasserstoff, Methyl oder Diäthylaminoäthyl, R3 o-Chlorphenyl oder o-Fluorphenyl und B eine Carbonylgruppe.
Gemäss einer ganz besonders bevorzugten Ausführungsform werden die folgenden Verbindungen hergestellt:
7-Chlor-1 .3-dihydro-1 -methyl-5-phenyl-2H-1 ,4-benzo diazepin-2-on
1,3 -Dihydro-7-nitro-5-phenyl -2H- 1 ,4-benzodiazepin- 7-on.
7-Chlor-2.3-dihydro-1 -methyl-5-phenyl-1H-1,4-benzodiazepin.
7-Chlor-1 .3-dihydro-5-phenyl-2H-1 ,4-benzodiazepin2-on.
7-Brom-1 .3 -dihydro-5-(2-pyridyl) -H-i 4-benzo- diazepin-2-on.
7-Chlor-1 -(2-diäthylaminoäthyl)-5-(2-fluorphenyl)- 1.3-dihydro-2H-l.j--benzodiazepin-2-on,
5-(2-Fluorphenyl)-1,3 -dihydro-1 -methyl-7-nitro-2H 1 .4-benzodiazepin-2-on, 5-(2-Chlorphenyl)-l -1,3 -dihydro-7 -nitro-2H-1 ,4-benzo- diazepin-2-on, 5 -(2 -Fluorphenyl) -1,3 -dihydro -7 -jod-1 -methyl-2H 1,4-benzodiazepin-2-on.
Die Reduktion einer 4,5-Epoxyverbindung der Formel II zu einer entsprechenden Verbindung der Formel I wird so durchgeführt, dass man das Ausgangsmaterial der Formel II mit einem Reduktionsmittel behandelt. Für diesen Zweck geeignete Reduktionsmittel sind schwache Reduktionsmittel, wie Jodwasserstoffsäure und Hydroxylamin und stärkere Reduktionsmittel wie schweflige Säure (H2SO3), Natriumbisulfit (NaHSO3) in Essigsäure, Wasserstoff in Gegenwart eines Edelmetallkatalysators, wie Platin oder Palladium, Triphenylphosphin, Lithiumaluminiumhydrid oder andere gemischte Aluminiumhydride. Es versteht sich von selbst, dass die angewendeten Reaktionsbedingungen in erster Linie vom verwendeten Reduktionsmittel abhängen. Im allgemeinen wird die Reaktion vorteilhafterweise unter neutralen oder sauren Bedingungen durchgeführt.
Falls Jodwasserstoffsäure als Reduktionsmittel verwendet wird, so wird diese vorzugsweise in situ unter Verwendung einer neutralen oder sauren Lösung eines Alkalimetalljodids, vorzugsweise Natrium- oder Kaliumjodid, gebildet. Für die Bildung von Jodwasserstoffsäure aus einem Alkalimetalljodid geeignete Säuren sind niedere Alkansäuren, vorzugsweise Essigsäure, und wässrige Säuren wie beispielsweise wässrige Schwefelsäure und wässrige Chlorwasserstoffsäure, wobei die wässrige Schwefelsäure bevorzugt ist. Verwendet man Hydroxylamin als Reduktionsmittel, so wird dieses vorzugsweise in Gegenwart eines Überschusses eines Mineralsäuresalzes von Hydroxylamin, wie beispielsweise Hydroxylaminhydrochlorid, eingesetzt.
Die Reduktion einer Verbindung der Formel II wird vorteilhafterweise in Gegenwart eines inerten organischen Lösungsmittels durchgeführt. Repräsentative Beispiele solcher inerter Lösungsmittel, welche für den vorliegenden Zweck benützt werden können, sind Alkohole wie Methanol, Äthanol u. dgl., Äther wie Tetrahydrofuran, Dioxan und Polyäther wie Polyalkylenglycole, aromatische Kohlenwasserstoffe wie Benzol, Toluol u. dgl., Alkansäuren mit Ameisensäure, Essigsäure, Propionsäure u. dgl., wobei Tetrahydrofuran und Essigsäure die bevorzugten Lösungsmittel sind.
Die Reduktionsreaktion wird vorzugsweise bei einer Temperatur zwischen dem Gefrierpunkt des Reaktionsgemisches und ungefähr 80 , ganz besonders bevorzugt bei ungefähr Raumtemperatur durchgeführt.

Die oben beschriebene Reaktion erfolgt durch Bestrahlung mit einer Lichtenergie mit einem spezifischen Wellenlängenbereich, erzeugt mittels einer Quecksilberlampe. Eine besonders bevorzugte Lichtquelle für die Zwecke der vorliegenden Erfindung ist eine Quecksilber-Mitteldrucklampe, welche mit einem Pyrexfilter versehen ist, um Licht mit Wellenlängen unterhalb von etwa 350 m,u zu eliminieren. Der so erhaltene Lichtstrahl ist weitestgehend auf den Wellenlängenbereich oberhalb von etwa 350 m,u beschränkt, und dieses Licht ist genügend energiereich, um die erwünschte Bestrahlungsreaktion zu bewerkstelligen; es erreicht jedoch nicht diejenigen Energiewerte, welche einen Abbau der Verbindungen der Formel III zur Folge haben.
Die Bestrahlungsreaktion wird mit Vorteil in einem inerten organischen, für solche Reaktionen üblicherweise verwendeten Lösungsmittel durchgeführt, beispielsweise in Äthern wie Tetrahydrofuran, Estern, Ketonen, Kohlenwasserstoffen oder halogenierten Kohlenwasserstoffen. Ein besonders bevorzugtes Lösungsmittel ist Tetrahydrofuran.
Die Reaktion erfolgt in einer inerten Atmosphäre beispielsweise unter Stickstoff, Argon, Helium usw. Die Reaktionstemperatur ist nicht kritisch, weshalb die Reaktion bei Temperaturen ausgeführt werden kann, welche üblicherweise für Bestrahlungsreaktionen verwendet werden, beispielsweise in einem Temperaturbereich von etwa -70 bis 1500, vorzugsweise zwischen etwa 10 und 300.
Die Ausgangsmaterialien der Formel III sind bekannte Verbindungen oder können leicht nach bekannten Methoden hergestellt werden.
Die nachfolgenden Beispiele illustrieren die vorliegende Erfindung. Alle Temperaturen sind in Celcius Graden angegeben.
Beispiel 1 Methode A:
Zu einer Lösung von 287 mg (1,0 mMol) 7-Chlor 4,5-epoxy-1,3,4,5-tetrahydro-5-phenyl-2H-1,4-benzo- diazepin-2-on in 20 ml Tetrahydrofuran wird eine Lösung von 2,0 g (12 mMol) Kaliumjodid in 20 ml 2n wässriger Schwefelsäure bei Raumtemperatur auf einmal zugegeben, wobei sofortige Freisetzung von Jod beobachtet werden kann.
Titration dieses Jods, nach 15minütiger Reaktionsdauer, verbraucht 19,4 ml (97%) einer 0,10n Natriumthiosulfatlösung.
Die titrierte Reaktionsmischung wird mit 250 ml Eiswasser verdünnt und danach mit 10 %iger wässriger Kaliumcarbonatlösung auf pH 7 neutralisiert. Die Hauptmenge des Tetrahydrofurans wird unter vermindertem Druck abgedampft.
Durch Stehenlassen der wässrigen Reaktionsmischung bei 5 erhält man 210 mg (78%) eines Rohproduktes vom Schmelzpunkt 212-214". Umkristallisation aus Aceton liefert 7-Chlor-1,3-dihydro-5-phenyl-2H-1,4-benzodiazepin- 2-on, Schmelzpunkt 216-218 .
Methode B:
Zu einer Lösung von 2,4 g (35 mMol) Hydroxylaminhydrochlorid und 1,06 g (27 mMol) Natriumhydroxyd in 4 ml Wasser werden 70 ml Tetrahydrofuran zugegeben. Die ausgefallenen Salze werden abfiltriert. Zu der filtrierten Lösung werden 1,0 g 7-Chlor-4,5-epoxy-1,3,4,5-tetrahydro5 -phenyl-2H-1,4 -benzodiazepin-2-on zugegeben. Die Reaktionsmischung wird bei Raumtemperatur während 20 Stunden gerührt. Danach wird das Tetrahydrofuran abgedampft und der Rückstand zwischen Methylenchlorid und Wasser verteilt. Die Methylenchloridphasen werden mit Wasser gewaschen, über wasserfreiem Natriumsulfat getrocknet und zur Trockene eingedampft.
Kristallisation des öligen Rückstandes aus Äther liefert 7-Chlor-1,3-dihydro-5-phenyl-2H-1,4benzodiazepin-2-on, Schmelzpunkt 210-213 . Nach Umkristallisation aus Essigester steigt der Schmelzpunkt auf 213 bis 215 an.
Das Ausgangsmaterial kann wie folgt hergestellt werden:
Eine Lösung von 26,8 g (0,0935 Mol) 7-Chlor-1,3-di hydro-5-phenyl-2H-1,4-benzodiazepin-2-on-4-oxyd in 1,4 Liter Tetrahydrofuran wird in einer Stickstoffatmosphäre bei 20 während 22 Stunden durch ein Pyrexfilter mit einer 250 Watt Quecksilber-Mitteldrucklampe bestrahlt. Man dampft die Lösung im Vakuum auf ein kleines Volumen ein, gibt Hexan zu, kühlt und erhält 7-Chlor-4,5-epoxy-1,3,4,5 tetrahydro-5 -phenyl-2H-1 ,4-benzodiazepin-2-on in Form farbloser Prismen, welche unter plötzlicher Zersetzung im Temperaturbereich von 136-150 schmelzen. Durch Kristallisation aus Mischungen von Tetrahydrofuran und Methanol erhält man eine analysenreine Probe vom Schmelzpunkt 136 (Zers.).
Beispiel 2
In zu Beispiel 1 analoger Weise können die folgenden Verbindungen hergestellt werden:
7-Chlor-1 -(2-diäthylaminoäthyl) -5 -(2-fluorphenyl) 1,3 -dihydro-2H-1,4-benzodiazepin-2-on, 5 -(2-Fluorphenyl) - 1,3 -dihydro-1 -methyl-7 -nitro-2H1,4-benzodiazepin-2-on,
5 -(2-Chlorphenyl) -1,3 -dihydro-7 -nitro-2H- 1,4-benzodiazepin-2-on,
5 -(2 -Fluorphenyl) -1,3 -dihydro-7-jod-1 -methyl-2H1,4 -benzodiazepin-2 -on.
Beispiel 3
Zu einer Lösung von 1,0 g (3,3 mMol) 7-Chlor-4,5epoxy-1,3,4,5 -tetrahydro-1 -methyl-5 -phenyl-2H- 1 ,4-benzo- diazepin-2-on in 66 ml Tetrahydrofuran wird bei Raumtemperatur eine Lösung von 6,57 g (39,6 mMol) Kaliumjodid in 66 ml 2n wässriger Schwefelsäure auf einmal zugegeben.
Titration des freigesetzten Jods nach 15 Minuten verbraucht 55,8 ml (85%) einer 0,ln Natriumthiosulfatlösung. Das titrierte Reaktionsgemisch wird mit Eiswasser auf 600 ml verdünnt. Das pH der Reaktionsmischung wird mit 10%iger wässriger Kaliumcarbonatlösung auf 7 eingestellt. Die Hauptmenge des Tetrahydrofurans wird abgedampft. Die verbleibende wässrige Mischung wird zweimal mit Methylenchlorid extrahiert. Die vereinigten Methylenchloridphasen werden über wasserfreiem Natriumsulfat getrocknet und zur Trokkene eingedampft. Kristallisation des zurückbleibenden Öls aus n-Heptan liefert 7-Chlor-1,3-dihydro-1 -methyl5-phenyl- 2H-1,4-benzodiazepin-2-on, Schmelzpunkt 129-131".
Das Ausgangsmaterial kann wie folgt hergestellt werden:
Eine Lösung von 30 g (0,10 Mol) 7-Chlor-1,3-dihydro1-methyl-5-phenyl-2H-1,4-benzodiazepin-2-on-4-oxyd in 1.4 Liter Tetrahydrofuran wird, wie in Beispiel 1 beschrieben, bestrahlt. Die bestrahlte Lösung wird zur Trockene eingedampft. Behandlung des gummiartigen Rückstandes mit einer kleinen Menge Äthanol bewirkt Kristallisation von 7 -Chlor-4,5 -epoxy-1,3,4,5 -tetrahydro -1 -methyl-5 -phenyl- 2H-1,4-benzodiazepin-2-on. Einmalige Umkristallisation aus Äthanol liefert farblose Prismen vom Schmelzpunkt 99-100
Beispiel 4
Eine Lösung von 1,0 g (3,7 mMol) 1,3-Dihydro-7-nitro5-phenyl-2H-1,4-benzodiazepin-2-on-4-oxyd in 150 ml Tetrahydrofuran wird in einer Stickstoffatmosphäre durch ein Pyrexfilter mit einer 250 Watt Quecksilber-Mitteldrucklampe bestrahlt.
Die Reaktionsmischung gibt eine positive Stärke-Jodidreaktion, was die Anwesenheit von 4,5-Epoxy 1 ,3,4,5-tetrahydro -7-nitro-5-phenyl-2H-1,4-benzodiazepin- 2-on im Reaktionsgemisch anzeigt. (Der Test wird so durchgeführt, dass ungefähr 0,2 ml der Reaktionslösung zur Trokkene eingedampft werden. Ein feuchter Streifen eines Stärke Jodidpapiers wird in die Lösung des Rückstandes in 2 Tropfen Eisessig eingetaucht). Die Lösung wird auf ungefähr 60 ml eingeengt. Zu dieser Lösung wird bei Raumtemperatur eine Lösung von 6,57 g (39,6 mMol) Kaliumjodid in 60 ml 2n Schwefelsäure auf einmal zugegeben. Nach 10 Minuten wird das freigesetzte Jod durch Zugabe eines Überschusses von 0,ln Natriumthiosulfatlösung (70 ml) reduziert. Das Reaktionsgemisch wird mit 10%der Kaliumcarbonatlösung auf pH 7 neutralisiert.
Extraktionen mit Methylenchlorid liefern 1,1 g des rohen Produktegemisches in Form eines festgewordenen Schaumes. Das Gemisch wird an einer Säule von 100 g Kieselgel (gestopft mit Essigester) chromatographiert, wobei als Eluierungsmittel Essigester verwendet wird. Die Fraktionen, welche das erwünschte Produkt enthalten, werden vereinigt und zur Trockene eingedampft. Kristallisation aus Mischungen von Aceton und Hexan liefert 1,3-Dihydro 7-nitro-5 -phenyl-2H-1,4-benzodiazepin-2-on, Schmelzpunkt 223-225.
Das Ausgangsmaterial kann wie folgt hergestellt werden:
Zu einer Suspension von 6,3 g (0,02 Mol) 2-Chlormethyl 6-nitro-4-phenylchinazolin-3-oxyd in einer Mischung von 50 ml Äthanol und 20 ml Aceton werden 24 ml einer 1n Natriumhydroxydlösung tropfenweise zugegeben. Die Reaktionsmischung wird auf 40 erwärmt und dann über Nacht bei Raumtemperatur gerührt. Danach wird die Mischung mit verdünnter Salzsäure auf pH 5 eingestellt und im Vakuum zur Trockene eingedampft. Der Rückstand wird mit einer Mischung von 125 ml Äthanol und 30 ml Aceton digeriert und das Produkt aus dem Filtrat mittels Petroläther ausgefällt.
Umkristallisation des Produktes aus einer Mischung von Äthanol/Petroläther liefert 1,3 -Dihydro-7 -nitro-5 -phenyl- 2H-1,4 -benzodiazepin -2 -on-4-oxyd als gelbe Prismen, Schmelzpunkt 218-220 (Zers.).
Beispiel 5
Eine Lösung von 2,0 g (12 mMol) Kaliumjodid in 10 ml 2n Schwefelsäure wird bei Raumtemperatur auf einmal zu einer Lösung von 286 mg (1,0 mMol) 7-Chlor-4,5-epoxy2,3,4,5 -tetrahydro- 1 -methyl-5 -phenyl-1H-1 ,4-benzodiazepin in 10 ml Tetrahydrofuran zugegeben. Nach 5 Minuten wird das freigesetzte Jod durch Zugabe von 22 ml einer 0,ln Natriumthiosulfatlösung reduziert. Das Reaktionsgemisch wird mit einer 2n Kaliumhydroxydlösung basisch gestellt (pH 9-10). Durch Extraktionen mit Methylenchlorid wird das Produktegemisch isoliert. Das gelbe Öl, welches man nach Abdampfen des Methylenchlorids erhält, wird aus Hexan kristallisiert, wobei man 7-Chlor-2,3 -dihydro-1 -me- thyl-5-phenyl-1H-1,4-benzodiazepin als gelbe Prismen erhält, Schmelzpunkt 99-100 .
Das Ausgangsmaterial kann wie folgt hergestellt werden:
Eine Lösung von 5,4 g (0,02 Mol) 7-Chlor-2,3-dihydro 5-phenyl-1,4-benzodiazepin4-oxyd in 100 ml Dimethylformamid wird unter einer trockenen Stickstoffatmosphäre hergestellt. Zu dieser Lösung werden unter Rühren 3,18 g (0,066 Mol) einer 50%gen Suspension von Natriumhydrid in Mineralöl zugegeben und die Badtemperatur auf ungefähr 50 erhöht. Nach einer Stunde wird das Reaktionsgemisch mit einem Eisbad gekühlt und 6 ml (ungefähr 0,1 Mol) Methyljodid zugegeben. Die Lösung wird während 18 Stunden bei 25 gehalten und danach in Eiswasser gegossen.
Nach Extraktion mit Methylenchlorid wird ein Öl erhalten, welches nach Zugabe einer Mischung von Äther und Petroläther (Siedepunkt 30-60") kristallisiert, wobei man 3,9 g Kristalle vom Schmelzpunkt 133-139 erhält. Umkristallisation aus einer Mischung von Methylenchlorid und Petroläther liefert 3 g (53%) 7-Chlor-2,3 -dihydro-1 -methyl-5-phenyl 1H-1,4-benzodiazepin4-oxyd als gelbe Prismen, Schmelzpunkt 104-143 . Das Produkt kann auch in einer zweiten Kristallform mit dem Schmelzpunkt 169-172 erhalten werden.
Eine Lösung von 1,52 g 7-Chlor-2,3-dihydro-1-methyl5-phenyl-1H-1,4-benzodiazepin-4-oxyd in 150 ml Tetrahydrofuran wird in einer Stickstoffatmosphäre durch ein Pyrexfilter mit einer 250 Watt Quecksilber-Mitteldrucklampe bestrahlt. Die Reaktionslösung gibt eine stark positive Stärke-Jodidreaktion. Das Lösungsmittel wird danach abgedampft. Kristallisation des zurückbleibenden Öls aus Hexan liefert 7-Chlor-4,5 -epoxy-2,3,4,5-tetrahydro- 1 -me thyl-5-phenyl-1H-1,4-benzodiazepin als leicht gelbe Prismen, Schmelzpunkt 97-100 .
Beispiel 6
Zu einer Lösung von 33 mg (0,10 Mol) 7-Brom-4,5epoxy-1,3,4,5 -tetrahydro-5 -(2-pyridyl) -2H- ,4-benzo- diazepin-2-on in 2 ml Tetrahydrofuran wird eine Lösung von 100 mg (0,6 mMol) Kaliumjodid in 2 ml einer 2n wässrigen Schwefelsäure zugegeben, wobei sofort die Farbe von Jod erscheint. Nach 15 Minuten bei Raumtemperatur wird die Farbe des Jods durch Zugabe von 39 mg (0,3 mMol) Natriumsulfit zum Verschwinden gebracht. Die Mischung wird mit 20 ml Wasser verdünnt, mit Ammoniumhydroxyd auf pH 8 neutralisiert und zweimal mit gleichen Mengen Methylenchlorid extrahiert. Die vereinigten Methylenchloridphasen werden mit Wasser gewaschen, getrocknet und eingedampft.
Ein Dünnschichtchromatogramm zeigt an, dass die Überführung in 7 -Brom- 1,3 -dihydro-5 -(2-pyridyl)-2H- 1,4-benzodiazepin-2-on vollständig ist. Kristallisation des Rohproduktes aus Aceton/Hexan liefert 7-Brom-1,3-dihydro-5 -(2-pyridyl) -2H-1,4-benzodiazepin-2-on als gelbe Prismen, Schmelzpunkt 239-240 .
Das Ausgangsmaterial kann wie folgt hergestellt werden:
Eine Lösung von 360 mg (1,08 mMol) 7-Brom-1,3-dihydro -5 -(2-pyridyl) -2H-1,4-benzodiazepin-2-on-4-oxyd in 150 ml Tetrahydrofuran wird mit dem Licht einer 250 Watt Quecksilber-Mitteldrucklampe bestrahlt. Um Lichtanteile mit einer kürzeren Wellenlänge als 350 m,u zu entfernen, wird das Licht vorgängig durch ein Pyrexfilter filtriert. Sobald das ganze Ausgangsmaterial umgesetzt ist (Dünnschichtchromatographie) wird die Bestrahlung beendet (30 Minuten). Das Tetrahydrofuran wird im Vakuum bei 30" abgedampft. Kristallisation des Rückstandes aus Äther liefert 7 -Brom-4,5 -epoxy-1,3,4,5 -tetrahydro-5 -(2-pyridyl) -2H- 1,4-benzodiazepin-2-on als farblose Prismen, Schmelzpunkt 142-143" mit plötzlicher Zersetzung.

In a preferred embodiment of the present invention, R1 is nitro or halogen, preferably chlorine. which is in the 7-position of the benzodiazepine ring, R2 is hydrogen or lower alkyl, preferably methyl, and R3 is phenyl.
In another preferred embodiment of the present invention, R1 is halogen, preferably bromine, which is in the 7-position of the benzodiazepine ring, R2 is hydrogen or lower alkyl, R3 is pyridyl, preferably 2-pyridyl, and B is a carbonyl group.
In a further preferred embodiment, R1 is nitro or halogen, preferably chlorine or iodine, which is in the 7-position of the benzodiazepine ring, R2 is hydrogen, methyl or diethylaminoethyl, R3 is o-chlorophenyl or o-fluorophenyl and B is a carbonyl group.

The reduction of a 4,5-epoxy compound of the formula II to a corresponding compound of the formula I is carried out by treating the starting material of the formula II with a reducing agent. Suitable reducing agents for this purpose are weak reducing agents such as hydriodic acid and hydroxylamine and stronger reducing agents such as sulphurous acid (H2SO3), sodium bisulphite (NaHSO3) in acetic acid, hydrogen in the presence of a noble metal catalyst such as platinum or palladium, triphenylphosphine, lithium aluminum hydride or other mixed aluminum hydrides. It goes without saying that the reaction conditions used depend primarily on the reducing agent used. In general, the reaction is advantageously carried out under neutral or acidic conditions.
If hydriodic acid is used as the reducing agent, it is preferably formed in situ using a neutral or acidic solution of an alkali metal iodide, preferably sodium or potassium iodide. Acids suitable for the formation of hydriodic acid from an alkali metal iodide are lower alkanoic acids, preferably acetic acid, and aqueous acids such as, for example, aqueous sulfuric acid and aqueous hydrochloric acid, with aqueous sulfuric acid being preferred. If hydroxylamine is used as the reducing agent, this is preferably used in the presence of an excess of a mineral acid salt of hydroxylamine, such as, for example, hydroxylamine hydrochloride.
The reduction of a compound of the formula II is advantageously carried out in the presence of an inert organic solvent. Representative examples of such inert solvents which can be used for the present purpose are alcohols such as methanol, ethanol and the like. Like., ethers such as tetrahydrofuran, dioxane and polyethers such as polyalkylene glycols, aromatic hydrocarbons such as benzene, toluene and the like. Like., alkanoic acids with formic acid, acetic acid, propionic acid and the like. the like, with tetrahydrofuran and acetic acid being the preferred solvents.
The reduction reaction is preferably carried out at a temperature between the freezing point of the reaction mixture and about 80, very particularly preferably at about room temperature.

The reaction described above takes place by irradiation with light energy with a specific wavelength range, generated by means of a mercury lamp. A particularly preferred light source for the purposes of the present invention is a medium pressure mercury lamp which is provided with a Pyrex filter in order to eliminate light with wavelengths below about 350 μm. The light beam obtained in this way is largely restricted to the wavelength range above about 350 μm, and this light is sufficiently energetic to bring about the desired irradiation reaction; however, it does not reach those energy values which result in a degradation of the compounds of the formula III.
The irradiation reaction is advantageously carried out in an inert organic solvent usually used for such reactions, for example in ethers such as tetrahydrofuran, esters, ketones, hydrocarbons or halogenated hydrocarbons. A particularly preferred solvent is tetrahydrofuran.
The reaction takes place in an inert atmosphere, for example under nitrogen, argon, helium, etc. The reaction temperature is not critical, which is why the reaction can be carried out at temperatures which are usually used for irradiation reactions, for example in a temperature range from about -70 to 1500, preferably between about 10 and 300.
The starting materials of formula III are known compounds or can easily be prepared by known methods.
The following examples illustrate the present invention. All temperatures are given in degrees Celsius.
Example 1 method A:
To a solution of 287 mg (1.0 mmol) of 7-chloro 4,5-epoxy-1,3,4,5-tetrahydro-5-phenyl-2H-1,4-benzodiazepin-2-one in 20 ml of tetrahydrofuran, a solution of 2.0 g (12 mmol) of potassium iodide in 20 ml of 2N aqueous sulfuric acid is added all at once at room temperature, and immediate release of iodine can be observed.
Titration of this iodine, after a reaction time of 15 minutes, consumes 19.4 ml (97%) of a 0.10N sodium thiosulphate solution.
The titrated reaction mixture is diluted with 250 ml of ice water and then neutralized to pH 7 with 10% strength aqueous potassium carbonate solution. Most of the tetrahydrofuran is evaporated off under reduced pressure.
Leaving the aqueous reaction mixture to stand at 5 gives 210 mg (78%) of a crude product with a melting point of 212-214 ". Recrystallization from acetone gives 7-chloro-1,3-dihydro-5-phenyl-2H-1,4-benzodiazepine- 2-one, m.p. 216-218.
Method B:
70 ml of tetrahydrofuran are added to a solution of 2.4 g (35 mmol) of hydroxylamine hydrochloride and 1.06 g (27 mmol) of sodium hydroxide in 4 ml of water. The precipitated salts are filtered off. 1.0 g of 7-chloro-4,5-epoxy-1,3,4,5-tetrahydro5-phenyl-2H-1,4-benzodiazepin-2-one are added to the filtered solution. The reaction mixture is stirred at room temperature for 20 hours. The tetrahydrofuran is then evaporated and the residue is partitioned between methylene chloride and water. The methylene chloride phases are washed with water, dried over anhydrous sodium sulfate and evaporated to dryness.
Crystallization of the oily residue from ether gives 7-chloro-1,3-dihydro-5-phenyl-2H-1,4benzodiazepin-2-one, melting point 210-213. After recrystallization from ethyl acetate, the melting point rises to 213-215.
The starting material can be made as follows:
A solution of 26.8 g (0.0935 mol) of 7-chloro-1,3-dihydro-5-phenyl-2H-1,4-benzodiazepin-2-one-4-oxide in 1.4 liters of tetrahydrofuran becomes irradiated in a nitrogen atmosphere at 20 for 22 hours through a Pyrex filter with a 250 watt medium pressure mercury lamp. The solution is evaporated to a small volume in vacuo, hexane is added, the mixture is cooled and 7-chloro-4,5-epoxy-1,3,4,5 tetrahydro-5-phenyl-2H-1,4-benzodiazepine is obtained 2-on in the form of colorless prisms, which melt with sudden decomposition in the temperature range 136-150. An analytically pure sample with a melting point of 136 (decomp.) Is obtained by crystallization from mixtures of tetrahydrofuran and methanol.
Example 2
In a manner analogous to Example 1, the following compounds can be prepared:
7-chloro-1 - (2-diethylaminoethyl) -5 - (2-fluorophenyl) 1,3-dihydro-2H-1,4-benzodiazepin-2-one, 5 - (2-fluorophenyl) -1,3-dihydro -1 -methyl-7-nitro-2H1,4-benzodiazepin-2-one,
5 - (2-chlorophenyl) -1,3-dihydro-7-nitro-2H-1,4-benzodiazepin-2-one,
5 - (2 -Fluorophenyl) -1,3-dihydro-7-iodo-1 -methyl-2H1,4-benzodiazepin-2-one.
Example 3
To a solution of 1.0 g (3.3 mmol) of 7-chloro-4,5epoxy-1,3,4,5-tetrahydro-1-methyl-5-phenyl-2H-1, 4-benzodiazepine 2-one in 66 ml of tetrahydrofuran, a solution of 6.57 g (39.6 mmol) of potassium iodide in 66 ml of 2N aqueous sulfuric acid is added all at once at room temperature.
Titration of the released iodine after 15 minutes consumes 55.8 ml (85%) of a 0.1N sodium thiosulphate solution. The titrated reaction mixture is diluted to 600 ml with ice water. The pH of the reaction mixture is adjusted to 7 with 10% strength aqueous potassium carbonate solution. Most of the tetrahydrofuran is evaporated. The remaining aqueous mixture is extracted twice with methylene chloride. The combined methylene chloride phases are dried over anhydrous sodium sulfate and evaporated to dryness. Crystallization of the remaining oil from n-heptane gives 7-chloro-1,3-dihydro-1-methyl5-phenyl-2H-1,4-benzodiazepin-2-one, melting point 129-131 ".
The starting material can be made as follows:
A solution of 30 g (0.10 mol) of 7-chloro-1,3-dihydro1-methyl-5-phenyl-2H-1,4-benzodiazepin-2-one-4-oxide in 1.4 liters of tetrahydrofuran is, as in Example 1 described, irradiated. The irradiated solution is evaporated to dryness. Treatment of the gummy residue with a small amount of ethanol causes crystallization of 7-chloro-4,5-epoxy-1,3,4,5-tetrahydro -1 -methyl-5-phenyl-2H-1,4-benzodiazepine-2- on. One-time recrystallization from ethanol yields colorless prisms with a melting point of 99-100
Example 4
A solution of 1.0 g (3.7 mmol) of 1,3-dihydro-7-nitro5-phenyl-2H-1,4-benzodiazepin-2-one-4-oxide in 150 ml of tetrahydrofuran is in a nitrogen atmosphere through a Pyrex filter irradiated with a 250 watt medium pressure mercury lamp.
The reaction mixture gives a positive starch-iodide reaction, which indicates the presence of 4,5-epoxy 1, 3,4,5-tetrahydro-7-nitro-5-phenyl-2H-1,4-benzodiazepin-2-one in the reaction mixture . (The test is carried out so that approximately 0.2 ml of the reaction solution is evaporated to dryness. A damp strip of starch iodide paper is immersed in the solution of the residue in 2 drops of glacial acetic acid). The solution is concentrated to approximately 60 ml. A solution of 6.57 g (39.6 mmol) of potassium iodide in 60 ml of 2N sulfuric acid is added all at once to this solution at room temperature. After 10 minutes the released iodine is reduced by adding an excess of 0.1N sodium thiosulphate solution (70 ml). The reaction mixture is neutralized to pH 7 with 10% of the potassium carbonate solution.
Extractions with methylene chloride yield 1.1 g of the crude product mixture in the form of a solid foam. The mixture is chromatographed on a column of 100 g of silica gel (filled with ethyl acetate), ethyl acetate being used as the eluent. The fractions containing the desired product are combined and evaporated to dryness. Crystallization from mixtures of acetone and hexane gives 1,3-dihydro 7-nitro-5-phenyl-2H-1,4-benzodiazepin-2-one, melting point 223-225.
The starting material can be made as follows:
24 ml of a 1N sodium hydroxide solution are added dropwise to a suspension of 6.3 g (0.02 mol) of 2-chloromethyl 6-nitro-4-phenylquinazoline-3-oxide in a mixture of 50 ml of ethanol and 20 ml of acetone. The reaction mixture is warmed to 40 and then stirred at room temperature overnight. The mixture is then adjusted to pH 5 with dilute hydrochloric acid and evaporated to dryness in vacuo. The residue is digested with a mixture of 125 ml of ethanol and 30 ml of acetone and the product is precipitated from the filtrate using petroleum ether.
Recrystallization of the product from a mixture of ethanol / petroleum ether gives 1,3-dihydro-7-nitro-5-phenyl-2H-1,4-benzodiazepine -2-one-4-oxide as yellow prisms, melting point 218-220 (dec .).
Example 5
A solution of 2.0 g (12 mmol) of potassium iodide in 10 ml of 2N sulfuric acid is converted all at once to a solution of 286 mg (1.0 mmol) of 7-chloro-4,5-epoxy2,3,4,5 - tetrahydro-1-methyl-5-phenyl-1H-1,4-benzodiazepine in 10 ml of tetrahydrofuran was added. After 5 minutes, the released iodine is reduced by adding 22 ml of a 0.1N sodium thiosulphate solution. The reaction mixture is made basic (pH 9-10) with a 2N potassium hydroxide solution. The product mixture is isolated by extractions with methylene chloride. The yellow oil, which is obtained after evaporation of the methylene chloride, is crystallized from hexane, 7-chloro-2,3-dihydro-1-methyl-5-phenyl-1H-1,4-benzodiazepine being obtained as yellow prisms , Melting point 99-100.
The starting material can be made as follows:
A solution of 5.4 g (0.02 mol) of 7-chloro-2,3-dihydro 5-phenyl-1,4-benzodiazepine 4-oxide in 100 ml of dimethylformamide is prepared under a dry nitrogen atmosphere. 3.18 g (0.066 mol) of a 50% suspension of sodium hydride in mineral oil are added to this solution with stirring, and the bath temperature is increased to approximately 50%. After one hour, the reaction mixture is cooled with an ice bath and 6 ml (approximately 0.1 mol) of methyl iodide is added. The solution is kept at 25 for 18 hours and then poured into ice water.
Extraction with methylene chloride gives an oil which crystallizes after adding a mixture of ether and petroleum ether (boiling point 30-60 "), giving 3.9 g of crystals with a melting point of 133-139. Recrystallisation from a mixture of methylene chloride and petroleum ether gives 3 g (53%) 7-chloro-2,3-dihydro-1-methyl-5-phenyl 1H-1,4-benzodiazepine 4-oxide as yellow prisms, melting point 104-143. The product can also be in a second crystal form with the melting point 169-172 can be obtained.
A solution of 1.52 g of 7-chloro-2,3-dihydro-1-methyl5-phenyl-1H-1,4-benzodiazepine-4-oxide in 150 ml of tetrahydrofuran is passed through a pyrex filter with a 250 watt mercury in a nitrogen atmosphere -Medium pressure lamp irradiated. The reaction solution gives a strongly positive starch-iodide reaction. The solvent is then evaporated. Crystallization of the remaining oil from hexane gives 7-chloro-4,5-epoxy-2,3,4,5-tetrahydro-1-methyl-5-phenyl-1H-1,4-benzodiazepine as pale yellow prisms, melting point 97 -100.
Example 6
To a solution of 33 mg (0.10 mol) of 7-bromo-4,5epoxy-1,3,4,5-tetrahydro-5 - (2-pyridyl) -2H-, 4-benzodiazepin-2-one in 2 ml of tetrahydrofuran, a solution of 100 mg (0.6 mmol) of potassium iodide in 2 ml of 2N aqueous sulfuric acid is added, the color of iodine immediately appearing. After 15 minutes at room temperature, the color of the iodine is made to disappear by adding 39 mg (0.3 mmol) of sodium sulfite. The mixture is diluted with 20 ml of water, neutralized to pH 8 with ammonium hydroxide and extracted twice with equal amounts of methylene chloride. The combined methylene chloride phases are washed with water, dried and evaporated.
A thin-layer chromatogram indicates that the conversion into 7-bromo-1,3-dihydro-5 - (2-pyridyl) -2H-1,4-benzodiazepin-2-one is complete. Crystallization of the crude product from acetone / hexane gives 7-bromo-1,3-dihydro-5 - (2-pyridyl) -2H-1,4-benzodiazepin-2-one as yellow prisms, melting point 239-240.
The starting material can be made as follows:
A solution of 360 mg (1.08 mmol) of 7-bromo-1,3-dihydro -5 - (2-pyridyl) -2H-1,4-benzodiazepin-2-one-4-oxide in 150 ml of tetrahydrofuran is with irradiated with the light of a 250 watt medium pressure mercury lamp. In order to remove light components with a wavelength shorter than 350 m, the light is first filtered through a Pyrex filter. As soon as all of the starting material has reacted (thin layer chromatography), the irradiation is ended (30 minutes). The tetrahydrofuran is evaporated in vacuo at 30 ". Crystallization of the residue from ether gives 7 -Bromo-4,5 -epoxy-1,3,4,5-tetrahydro-5 - (2-pyridyl) -2H-1,4- benzodiazepin-2-one as colorless prisms, melting point 142-143 "with sudden decomposition.
