Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von Jod-substituierten Benzodiazepinderivaten der Formel

Die Verbindungen der obigen Formel I bzw. ihre Säureadditionssalze werden erfindungsgemäss hergestellt, indem man das Diazoniumsalz einer Verbindung der Formel

worin R1 obige Bedeutung besitzt, mit einem Jodidsalz behandelt.
Erwünschtenfalls wird eine erhaltene Verbindung der Formel I in ein Säureadditionssalz übergeführt.
Bei der Reaktion werden die Verbindungen der obigen Formel I, ausgehend von den entsprechenden 7-Amino-1,4benzodiazepin-2-onen, einer Austauschreaktion vom Sandmeyer-Typus unterworfen. Bei der Durchführung dieses Verfahrens wird zunächst das Diazoniumsalz der Verbindung der obigen Formel II hergestellt.
Die Aminogruppe der Verbindung der Formel II kann nach üblichen Methoden diazotiert werden, beispielsweise durch Behandlung mit irgend einer Kombination von Reagenzien, welche in situ salpetrige Säure erzeugt, wie beispielsweise Natriumnitrit in Mineralsäure, z. B. Salzsäure oder Schwefelsäure. Hierauf behandelt man das Diazoniumsalz der Verbindung der Formel II mit einem Jodidsalz, vorzugsweise einem Alkalimetalljodid, wie Natriumjodid oder Kaliumjodid, oder mit Kupfer(I)-jodid, wobei man die Verbindung der Formel 1 erhält.
Wie oben erwähnt, kann die Diazotierung der Verbindung der Formel II nach allgemein gebräuchlichen Methoden durchgeführt werden. Beispielsweise löst man das Amin in einer wässrigen Lösung von Mineralsäure und kühlt das Gemisch auf eine Temperatur zwischen 0 und - 100 C. Hierauf gibt man langsam eine wässrige Natriumnitritlösung zu, worauf sich das Diazoniumsalz bildet. Um eine Zersetzung zu vermeiden, wird das Salz dann sofort mit dem Jodidsalzzur Reak- tion gebracht, wobei die Austauschreaktion und die Bildung der Jodverbindung erfolgt.
Die Ausgangsprodukte des erfindungsgemässen Verfahrens können durch Reduktion der entsprechenden 7-Nitroverbindung der Formel

worin Rl obige Bedeutung besitzt, hergestellt werden, beispielsweise mittels Zinn(II)-chlorid in wässriger Salzsäure.
Die erfindungsgemäss erhältlichen Verbindungen der obigen Formel I sowie ihre pharmazeutisch verträglichen Salze sind nützlich als Antikonvulsiva, Muskelrelaxantien und Sedativa. Somit können diese Verbindungen und ihre pharmazeutisch verträglichen Salze als Medikamente verwendet werden.
Sie können in Form pharmazeutischer Präparate Verwendung finden, welche sie oder ihre Salze in Mischung mit einem für die enterale oder parenterale Verabreichung geeigneten pharmazeutischen, organischen oder anorganischen inerten Trägermaterial, wie z. B. Wasser, Gelatine, Milchzucker, Stärke, Magnesiumstearat, Talk, pflanzliche Öle, Gummi arabicum, Polyalkylenglykole, Vaseline usw., enthalten. Die pharmazeutischen Zubereitungen können in fester Form oder in flüssiger Form vorliegen. Gegebenenfalls enthalten sie Zusatzstoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Salze zur Veränderung des osmotischen Druckes oder Puffer. Sie können auch noch andere therapeutisch wertvolle Substanzen enthalten.
Bei der Verabreichung der Verbindungen der Formel I oder ihrer pharmazeutisch verträglichen Salze wird die Do sierung den individuellen Erfordernissen und dem pharma zeutischen Bedürfnis der Situation angepasst. Zweckmässiger weise liegt die Dosierung im Bereich von etwa 0,05 bis etwa
1 mg pro Tag.
Wie oben erwähnt, sind die erfindungsgemäss erhältlichen Verbindungen der Formel I und ihre pharmazeutisch ver wendbaren Salze nützlich als Sedativa, Muskelrelaxantien und
Antikonvulsiva. Die Nützlichkeit der erfindungsgemäss er hältlichen Verbindungen kann in zahlreichen Versuchen be obachtet werden. In den nachfolgend beschriebenen
Versuchen verwendete man als Untersuchungssubstanzen die
Verbindungen der obigen Formel I, d. h. 5 -(2-Fluorphenyl) - 7-jod-1,3-dihydro-l -methyl-2H-1,4-benzodiazepin-2-on (Verbindung A) und 5-(2-Fluorphenyl)-7 jod-1,3-dihydro-
2H-1,4-benzodiazepin-2-on (Verbindung B).
Die sedative und muskelrelaxierende Aktivität der erfin dungsgemäss erhältlichen Verbindungen kann in folgenden
Versuchs anordnungen beobachtet werden:
Versuch an der geneigten Ebene: Man verabreicht die
Versuchssubstanz in Dosen-bis zu maximal 500 mg/kg p. o.
an männliche Mäuse, bringt die Tiere auf eine geneigte Ebene und beobachtet während mindestens 4 h, ob-sie aufgrund von
Lähmungserscheinungen an der Ebene abgleiten. Man ermit telt die PDso nach der Methode von Behrens [Arch.- Exp.
Pathol. Pharmakol. 140, 237 (1929)1 auf der Basis von 6
Mäusen pro Dosis und mindestens 2 Punkten zwischen 100 und 0%. In diesem Versuch zeigt die Verbindung A eine PDso von 1 mg/kg und Verbindung B eine PDso von 3,5 mg/kg.
Versuch an der wachen Katze: Man behandelt normale Katzen auf oralem Wege und beobachtet, ob wesentliche Ver änderungen in ihrem Verhalten auftreten. Die minimale wirksame Dosis (M. E. D.) ist die niedrigste Dosis, bei welcher Symptome beobachtet werden. In diesem Versuch zeigt Verbindung A eine M. E. D. von 0,05 mg/kg und Verbindung B eine M. E. D. von 0,1 mg/kg.
Kampf-Test: Man bringt 2 Mäuse unter ein umgekehrtes
1 -Liter-Becherglas auf ein Gitter, durch welches ihnen Schläge auf die Füsse versetzt werden. Man wählt Mäusepaare, welche unter diesen Umständen einer 2minütigen Versuchsperiode mindestens fünfmal gekämpft haben, verabreicht ihnen die Versuchssubstanz auf oralem Weg und wiederholt den Versuch nach einer Stunde. Hierbei verwendet man logarithmisch ansteigende Dosenbis zu maximal 100 mg/kg. Man definiert als 100teige hemmende Dosis (PDXoo) diejenige Dosis, welche bei 3 von 3 Mäusepaaren während einer Stunde jeglichen Kampf verhindert. In diesem Versuch zeigt Verbindung A eine PD100 von 1,0 mg/kg und Verbindung B eine PD1oo von 2 mg/kg.
Die antikonvulsive Aktivität der-erfindungsgemäss erhältlichen Verbindungen kann in folgendem Versuch demonstriert werden:
Maximaler Elektroschock: Dieser Versuch erfolgt nach der Methode von Swinyard, Goodman und Brown P, Pharmacol. Exp. Ther. 106, 319 (1952)1, jedoch mit folgenden Modifikationen. Man verwendet männliche Mäuse und versetzt ihnen den Schock in gleicher Weise und Dauer wie in der oben beschriebenen Methode; der Stromstoss beträgt jedoch nicht 50 mA, sondern 30 mA. Eine Stunde nach Verabreichung der Versuchssubstanz erhalten-die Tiere den erwähnten Elektroschock. Kriterium für die Wirksamkeit der Versuchssubstanz ist das Ausbleiben des Streckkrampfes der Hinterpfote.
In diesem Test zeigt Verbindung A eine EDso von 1,6 mg/kg und Verbindung B eine EDso von 1,3 mg/kg.
7-Halogen-1,4-benzodiazepin-2-one gehören einer bekannten Verbindungsklasse an. Die ganz besonders ausgeprägten sedativen, muskelrelaxierenden und antikonvulsiven Eigenschaften der 5-(2-Fluorphenyl)-7 -jod-1 ,4-benzodi- azepin-2-one der Formel I, aufgrund welcher diese Verbindungen ausserordentlich nützlich als Tranquillizer siadi sind jedoch bisher noch nicht erkannt worden. Die Entdeckung dieser bisher nicht beschriebenen Verbindungen innerhalb einer bekannten Verbindungsklasse und ihre ungewöhnliche Fähigkeit, als'besonders wirksame Sedativa zu dienen, machen den Hauptteil dervorliegenden Erfindung: aus
In den folgenden Beispielen sind alle Temperaturen in Celciusgraden angegeben.
Beispiel 1
Zu einer auf 0 gekühlten Lösung von 1,2 g 7-Amino-5 (2 -Fluorphenyl) -1,3 -dihydro-1 -methyl-2H-1 ,4-benzodi- azepin-2-on in 3,5 ml 6n Salzsäure gibt man eine Lösung von 0,33 g Natriumnitrit in 5 ml Wasser. Die gelbe Lösung wird während 10 min bei 0 bis +5 gerührt. Hierauf gibt man eine Lösung von 1,2 g Kaliumjodid in 5 ml Wasser zu und rührt während weiterer 30 min bei Raumtemperatur. Man filtriert, stellt das Filtrat mit Ammoniak alkalisch und extrahiert es mit Methylenchlorid. Die vereinigten Extrakte werden mit Wasser gewaschen, über Natriumsulfat getrocknet und eingedampft, wobei ein orangefarbiges Öl zurückbleibt.
Kristallisation aus Methylenchlorid/Äther/Petroläther liefert 5 -(2-Fluor- phenyl) -1,3 -dihydro-7 -jod-1 -methyl-2H-1 ,4-benzodiazepin- 2-on vom Schmelzpunkt 102-107 .
Das Ausgangsmaterial kann wie folgt hergestellt werden:
Zu einer Lösung von 12,5 g 5-(2-Fluorphenyl)-1,3-di- hydro-1 -methyl-7 -nitro-2H-1,4-benzodiazepin-2-on in 80 ml 6n Salzsäure gibt man eine Lösung von 30 g Zinn(II)-chloriddihydrat in 80 ml 6n Salzsäure. Man rührt das Gemisch über Nacht und stellt die erhaltene Suspension durch Zugabe von 50%igem wässrigem Kaliumhydroxyd alkalisch. Der ausgefallene gelbe Festkörper wird gesammelt, mit Wasser gewaschen, getrocknet und aus Tetrahydrofuran/Hexan umkristallisiert, wobei man 7-Amino-5-(2-Fluorphenyl)-1+3-dihydro-1- methyl-2H-1,4-benzodiazepin-2-on vom Schmelzpunkt 206-208 (Zers.) erhält.
Beispiel 2
In analoger Weise, wie in Beispiel 1 beschrieben, kann 5 (2-Fluorphenyl) -1,3 -dihydro-7-jod-2H-1 ,4-benzodiazepin- 2-on, Schmelzpunkt 222-224", hergestellt werden.

In the reaction, the compounds of the above formula I, starting from the corresponding 7-amino-1,4-benzodiazepin-2-ones, are subjected to an exchange reaction of the Sandmeyer type. In carrying out this process, the diazonium salt of the compound of the above formula II is first prepared.
The amino group of the compound of formula II can be diazotized by conventional methods, for example by treatment with any combination of reagents which generate nitrous acid in situ, such as sodium nitrite in mineral acid, e.g. B. hydrochloric acid or sulfuric acid. The diazonium salt of the compound of the formula II is then treated with an iodide salt, preferably an alkali metal iodide such as sodium iodide or potassium iodide, or with copper (I) iodide, the compound of the formula 1 being obtained.

The compounds of the above formula I obtainable according to the invention and their pharmaceutically acceptable salts are useful as anticonvulsants, muscle relaxants and sedatives. Thus, these compounds and their pharmaceutically acceptable salts can be used as medicaments.

To a solution, cooled to 0, of 1.2 g of 7-amino-5 (2-fluorophenyl) -1,3-dihydro-1-methyl-2H-1,4-benzodiazepin-2-one in 3.5 ml A solution of 0.33 g of sodium nitrite in 5 ml of water is added to 6N hydrochloric acid. The yellow solution is stirred at 0 to +5 for 10 min. A solution of 1.2 g of potassium iodide in 5 ml of water is then added and the mixture is stirred for a further 30 minutes at room temperature. To a solution of 12.5 g of 5- (2-fluorophenyl) -1,3-di-hydro-1-methyl-7-nitro-2H-1,4-benzodiazepin-2-one in 80 ml of 6N hydrochloric acid is added a solution of 30 g of tin (II) chloride dihydrate in 80 ml of 6N hydrochloric acid. The mixture is stirred overnight and the suspension obtained is made alkaline by adding 50% strength aqueous potassium hydroxide. The precipitated yellow solid is collected, washed with water, dried and recrystallized from tetrahydrofuran / hexane, 7-amino-5- (2-fluorophenyl) -1 + 3-dihydro-1-methyl-2H-1,4-benzodiazepine being obtained -2-one of melting point 206-208 (decomp.)
