Die vorliegende Erfindung bezieht sich auf ein neues Verfahren für die Herstellung von Butyrophenonderivaten, und zwar von y-Piperidinobutyrophenonderivaten, welche in bezug auf das zentrale Nervensystem wirksam sind.
Ziel der vorliegenden Erfindung ist die Entwicklung eines vorteilhaften Verfahrens, gemäss welchem man substituierte y.Piperidinobutyrophenonderivate erhalten kann, welche in o-Stellung des Benzolkerns einen Halogensubstituenten aufweisen. Die als Ausgangsmaterial dienenden o-Amino-y-piperidinobutyrophenonderivate können durch Oxydation von 3y-Pipendinopropylindolderivaten zu o-Acylamino-y-pipefl- dinobutyrophenonderivaten, deren Acylaminogruppe sich durch eine gewöhnliche Hydrolysierungsreaktion in eine unsubstituierte Aminogruppe überführen lässt, erhalten werden.
Durch Diazotieren und Halogenieren wird die Aminogruppe erfindungsgemäss durch ein Halogenatom ersetzt. Somit kann man beim vorliegenden Verfahren verschiedene y-Piperidinobutyrophenonderivate in sehr vorteilhafter Weise herstellen.

Die 1 [fr(3-Indolyl).propionyfl-pipefldin-verbindungen der Formel (VIII), welche man als Zwischenprodukte verwendet, lassen sich herstellen, indem man eine Indolylpropionsäure der Formel (V) oder ein funktionelles aktives Derivat davon, z.B. ein Säurechlorid, Säurebromid, Säureanhydrid, gemischtes Säureanhydrid oder einen p-Nitrophenylester usw., mit einem Piperidin der Formel (VII) umsetzt. Die Umsetzung erfolgt vorzugsweise in Gegenwart eines basischen Mittels oder eines Kondensationsmittels, wie z.B. Pyridin, Triäthylamin, Natriumcarbonat, Natriumhydroxyd, Dicyclohexylcarbodiimid usw., in einem geeigneten inerten organischen Lösungsmittel, wie z.B.
Tetrahydrofuran, Äther, Dioxan, Benzol, Toluol, Chloroform, Dimethylformamid usw.
Die oben erwähnten gemischten Säureanhydride umfassen solche, welche man durch Behandeln mittels Äthylchloroformiat, Isobutylchloroformiat usw. erhält.
Die Zwischenprodukte gemäss Formel (VIII) lassen sich ebenfalls so herstellen, dass man eine Phenylhydrazonverbindung der Formel (VI) erhitzt.
Das Erhitzen erfolgt vorzugsweise in Gegenwart eines sauren Kondensationsmittels, wie z.B. Chlorwasserstoff, Schwefelsäure, Phosphorsäure, Zinkchlorid, Kupferchlorid, Borfluorid, Polyphosphorsäure usw., in Gegenwart eines geeigneten Lösungsmittels, wie z.B. Äthanol, Isopropanol, tert.-Butanol, Essigsäure, Benzol, Toluol, Wasser usw.

Die auf diese Weise erhaltenen Verbindungen der Formel (VIII) können in die entsprechenden 3-(y-Piperidinopro- pyl)-indolverbindungen der Formel (IX) übergeführt werden, indem man die ersteren mit einem Reduktionsmittel umsetzt.
Als Reduktionsmittel kommen beispielsweise Alkalimetalle in einem alkoholischen Lösungsmittel, Wasserstoff in Gegenwart eines Katalysators, Metallhydride und dgl. in Frage.
Eine elektrolytische Reduktion lässt sich für diesen Zweck gleichfalls anwenden.
Es ist insbesondere vorzuziehen, ein Metallhydrid, wie z.B. Lithium-aluminiumhydrid, Diisobutylaluminiumhydrid, Triisopropyl-aluminiumhydrid, Borhydrid oder dgl., in Gegenwart eines inerten organischen Lösungsmittels, wie z.B.
Äther, Tetrahydrofuran, Dioxan, N-Äthylmorpholin und dgl., zu verwenden.

Die Säureadditionssalze der 3 -(y-Piperidinopropyl)-indol- derivate lassen sich herstellen, indem man die freie Base mit einer Säure, wie z.B. Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Salpetersäure, Essigsäure, Oxalsäure, Citronensäure, Weinsäure, Glycolsäure, Benzoesäure, Sulfaminsäure, Mandelsäure usw., behandelt.
y-Piperidinobutyrophenone der Formel (II) lassen sich herstellen, indem man die oben erwähnten 3-y-Pipen-dinopro pylindole der Formel (IX) mit einem Oxydationsmittel behandelt. Die oxydative Aufspaltungsreaktion erfolgt vorzugsweise unter Verwendung eines Oxydationsmittels, wie z.B. Ozon, Wasserstoffperoxyd, Perameisensäure, Peressigsäure, Perbenzoesäure, Chromsäure oder Kaliumpermanganat.
Das Oxydationsmittel soll allerdings in keiner Weise auf die soeben genannten, rein beispielsweise angegebenen Oxydationsmittel eingeschränkt sein.
Im allgemeinen verläuft die Reaktion bei Zimmertemperatur, doch kann man auch höhere oder niedrigere Temperaturen anwenden, um den Reaktionsverlauf in erwünschtem Sinne zu regeln. Das Oxydationsmittel wird vorzugsweise Chromsäure oder Ozon sein. Die Umsetzung erfolgt vorzugsweise in Gegenwart eines Lösungsmittels. Die Wahl des Lösungsmittels hängt vom verwendeten Oxydationsmittel ab.
Als Lösungsmittel kommen Wasser, Aceton, Tetrachlorkohlenstoff, Essigsäure, Schwefelsäure usw. in Frage. Das Oxydationsmittel kann in stöchiometrischer Menge oder in einer grösseren Menge verwendet werden. Die Reaktionstemperatur schwankt je nach dem verwendeten Oxydationsmittel.
Erfolgt die Oxydation durch Verwendung von Chromsäure in Gegenwart von Essigsäure, so wird man die Chromsäure vorzugsweise in der doppelten bis 3fachen äquimolaren Menge verwenden und die Umsetzung bei Zimmertemperatur durchführen. Dabei wird man ein 3-y-Piperidinopropylindolderivat im Lösungsmittel auflösen oder suspendieren und das Oxydationsmittel der Lösung bzw. der Suspension hunter Rühren zugeben. Im allgemeinen ist die Umsetzung innerhalb von ungefähr 24 Stunden beendet.
In jenen Fällen, in denen die Oxydation unter Verwendung von Ozon erfolgt, wird man die Umsetzung bei Zimmertemperatur durchführen. Dabei wird man ein 3-y-Piperi- dinopropylindolderivat in einem Lösungsmittel, wie z.B. Ameisensäure, Essigsäure, Tetrachlorkohlenstoff oder dergleichen, auflösen oder suspendieren und ozonisierten Sauerstoff in die Lösung bzw. Suspension unter Rühren einblasen.
Die gewünschten y-Piperidinobutyrophenonderivate lassen sich aus dem Reaktionsgemisch in roher Form durch Extraktion, mit oder ohne vorherige Neutralisierung, und durch Eindampfen zur Trockne gewinnen. Das Produkt lässt sich gewünschtenfalls durch Umkristallisieren aus einem geeigneten Lösungsmittel, wie z.B. Äthanol, Isopropanol oder dergleichen, in üblicher Weise weiter reinigen.
Die erhaltenen Verbindungen der Formel (II) können hydrolysiert werden, wobei man eine entsprechende entacylierte Verbindung der Formel (III) erhält. Die Hydrolyse kann unter sauren oder alkalischen Bedingungen je nach der Hydrolysemethode durchgeführt werden.
y-Piperidinobutyrophenone der Formel (I) werden dadurch erhalten, dass man die oben erwähnten o-Aminoverbindungen der Formel (III) diazotiert und hierauf die Diazoniumgruppe durch ein Halogenatom ersetzt.
Die Diazotierung kann in an sich bekannter Weise durchgeführt und die Austauschreaktion durch Behandeln des Diazoniumsalzes mit Kupferpulver, Fluorborsäure oder einem Metallsalz, z.B. Cuprochlorid, Cuprobromid, Kaliumjodid, einem Mercurihalogenid usw., vorgenommen werden.
Die obigen Ausführungen sind eine allgemeine Beschreibung. Um die Diazotierung und die Austauschreaktion näher zu erläutern, seien nachstehend einige Reaktionsschemata wiedergegeben:

Diese Verbindungen bilden entsprechende Säureadditionssalze in an sich bekannter Weise, indem man beispielsweise die freie Base in einer wässrigen Lösung löst, welche eine geeignete Säure enthält, und hierauf das erhaltene Salz durch Eindampfen der Lösung isoliert. Man kann aber auch die freie Base mit einer Säure in einem organischen Lösungsmittel umsetzen.
Diese Salze umfassen pharmazeutisch annehmbare Säureadditionssalze, wie z.B. Chlorhydrate, Fumarate, Formiate, Acetate, Lactate, Citrate, Sulfonate, Maleinate, Tartrate, Methansulfonate, Salicylate und Hydrogensulfate.
Diese y- Piperidinobutyrophenonderivate der Formel (I), sowie deren pharmazeutisch annehmbare Säureadditionssalze besitzen eine Wirkung auf das zentrale Nervensystem. Sie eignen sich als anxiolytische und antipsychotonische Mittel, ferner als Mittel zur Besserung der emotionalen Stabilität und als krampfiösende oder antipsychotische Drogen.
Jede der pharmazeutisch wirksamen Verbindungen, welche man erfindungsgemäss erhält, lässt sich beispielsweise für die orale Verabreichung in Tabletten als einzige Wirksubstanz einarbeiten. Eine typische Tablette besteht aus 1 bis 2% Bindemittel, z.B. Tragacanth, 3 bis 10% Gleitmittel, z.B.
Talk, 0,25 bis 1,0% eines Gleitmittels, wie z.B. Magnesiumstearat, einer durchschnittlichen Dosis der Wirksubstanz und einer ausreichenden Menge eines Füllmittels, wie z.B. Lactose, um 100% zu ergeben. Die übliche tägliche Dosis liegt zwischen 1 und 100 mg per os.
Die folgenden Beispiele erläutern die vorliegende Erfindung.
Beispiel I Stufe I
Eine Mischung von 14,5 g Phenylhydrazinhydrochlorid, 19,2 g l-(y-Acetylbutyryl)-piperidin und 100 cm3 Essigsäure wird unter Rühren während 4 Stunden auf 75 bis 80"C erhitzt. Nach dem Kühlen wird das Reaktionsgemisch in 700 cm3 kaltes Wasser gegossen. Das feste Material, das sich ausscheidet, wird filtriert, mit Wasser gewaschen und aus Äthanol umkristallisiert, wobei man 1-[kS-(2-Methyl-3-indo- lyl)-propionyl]-piperidin mit einem Schmelzpunkt von 141 bis 143 C erhält.
Stufe 2
Zu einer Mischung von 2,0 g Lithium-aluminiumhydrid und 50 cm3 Äther gibt man tropfenweise unter Rühren eine Lösung von 4,9 g 1-[b-(2-Methyl-3-indolyl)-propionyl]-piperi- din in 130 cm3 Tetrahydrofuran innerhalb von 30 Minuten unter gelindem Erhitzen unter Rückfluss hinzu. Das Rühren und das Erhitzen unter Rückfluss wird während weiteren 3 Stunden fortgeführt, worauf man das Reaktionsgemisch tropfenweise mit einer Mischung von 10 cm3 Wasser und 40 cm3 Tetrahydrofuran unter Kühlen mit Eis versetzt Der erhaltene Niederschlag wird abfiltriert und das Filtrat zur Trockne eingedampft. Die Umkristallisierung des Rückstandes aus Benzol liefert 2-Methvl-3 -(y-piperidinopropyl).indol mit einem Schmelzpunkt von 140,5 bis 141 0C.
In ähnlicher Weise lassen sich die folgenden Verbindungen herstellen: 2-Methyl-3-[y.(4-phenylpiperidino)-propyU-indol > Smp. 119,5 bis l2l,00C, 2-Methyl-3 -y-(4-phenylpiperidino)-propyl] -5-fluorindol,
Smp. 153 bis 154"C, 2- Methyl-3 - [y-(4-hydroxy-4-phenylpiperidino)-propyl] -5- -fluorindol, Smp. 161 bis 162"C.
Stufe 3
Es wird Sauerstoff, enthaltend 3 bis 4% Ozon, in eine Lösung von 2,0 g 2-Methyl-3-(y-piperidinopropyl)-indol in 40 cm3 Essigsäurelösung eingeleitet, wobei das Produkt blassgelb wird (in der Praxis benötigt man hierfür ungefähr 35 Minuten), wobei man die Temperatur auf 16 bis 20"C hält.
Nachdem man das Reaktionsgemisch durch Zugabe einer 10% eigen wässrigen Natriumhydroxydlösung alkalisch gestellt hat, wird das Reaktionsprodukt mit Äthylacetat extrahiert.
Der Extrakt wird mit Wasser gewaschen und eingedampft, wobei man einen öBlgen Rückstand erhält. Der Rückstand wird aus einer Mischung von Äther und Petroläther auskristallisieren gelassen, wobei man y-Piperidino-2-acetaminobutyrophenon vom Smp. 45 bis 480C erhält.
Nach der obigen Methode lassen sich die folgenden Verbindungen herstellen: y-(4-Phenylpiperidino) -2-acetaminobutyrophenon-hydrochlo rid, Smp. 2400C (Zers.), y-(4-Phenylpiperidino)-2-acetamino-5-fluorbutyrophenon >
Smp. 130,0 bis 131,00C und sein Hydrochlorid, Smp.
2480C (Zers.), r-(4-Hydroxy-4-phenylpiperidino)-2-acetamino-5-fluorbutyro- phenon, Smp. 120,0 bis 121,00C.
Stufe 4
Eine Lösung von 3,5 g y-(4-Piperidino)-2-acetaminobutyrophenonhydrochlorid und 6 cm3 konzentrierter Salzsäure in 70 cm3 Äthanol wird unter Rückfluss während 3 Stunden erhitzt. Nach dem Kühlen wird das Reaktionsgemisch mit 100 cm3 Wasser verdünnt und dann mit 50%iger wässriger Natriumhydroxydlösung alkalisch gestellt. Die entstandene ölige Substanz wird mit Äthylacetat extrahiert. Der Extrakt wird mit Wasser gewaschen und zu einem Rückstand eingedampft, welcher aus wässrigem Äthanol umkristallisiert wird.
Auf diese Weise erhält man y-(4-Piperidino)-o-aminobutyro- phenon, Smp. bei 92 bis 93"C.
Stufe 5
Zu einer gekühlten Lösung von 3,6 g y-Piperidino-2-ami nobutyrophenon in 35 cm3 2n-Salzsäure gibt man tropfenweise eine Lösung von 1,1 g Natriumnitrit in 5 cm3 Wasser unter Rühren unterhalb 0 C hinzu. Die erhaltene Diazoniumsalzlösung wird einer kalten Suspension von 1,5 g Cuprochlorid in 5 cm3 konzentrierter Salzsäure unter kräftigem Rühren zugegeben. Das Gemisch wird dann unter Eiskühlung während weiteren 30 Minuten gerührt und hierauf während weiteren 2 Stunden bei Zimmertemperatur und danach während 1 Stunde bei 55 bis 60"C weitergerührt. Nach dem Kühlen wird das Reaktionsgemisch mittels Ammoniumhydroxyd alkalisch gestellt und mit Äther extrahiert.
Der ätherische Extrakt wird mit einer wässrigen, mit Natriumchlorid gesättigten Lösung gewaschen, über wasserfreiem Natriumsulfat getrocknet und eingedampft, wobei man einen öligen Rückstand erhält. Das erhaltene öl wird mit wasserfreiem Chlorwasserstoff in Äther behandelt, wobei man Kristalle von y-Piperidino-2-chlorbutyrophenonhydrochlorid erhält, die man aus Äthanol umkristallisiert. Smp. 166 bis 167"C.
In ähnlicher Weise, wie dies oben geschehen ist, kann man die folgende Verbindung herstellen: r-(4-Phenylpiperidino)-2-chlorbutyrophenonhydrochlorid,
Smp. 182 bis 184"C.
Beispiel 2
Zu einer gekühlten Lösung von 7,1 g y-(4-Hydroxy-4 -phenylpiperidino)-2-amino-5-fluorbutyrophenon in 200 cm3 2n-Salzsäure gibt man eine Lösung von 1,52 g Natriumnitrit in 10 cm3 Wasser unter Rühren bei einer Temperatur von weniger als 0 C hinzu. Die erhaltene Diazoniumsalzlösung wird einer gekühlten Suspension von 2,0 g Cuprochlorid in 20 cm3 konzentrierter Salzsäure unter kräftigem Rühren hinzugegeben. Das Rühren wird während weiteren 2 Stunden bei Zimmertemperatur und hierauf während weiteren 2 Stunden bei 55 bis 60"C fortgeführt. Nach dem Kühlen des Reaktionsgemisches wird es durch Zugabe einer wässrigen Ammoniumhydroxydlösung alkalisch gestellt und dann mit Chloroform extrahiert.
Der Extrakt wird mit Wasser gewaschen, über wasserfreiem Natriumsulfat getrocknet und eingedampft, wobei man r-(4-Hydroxy-4-phenyl)-2-chlor-5-fluor- butyrophenon erhält. Der Smp. des Chlorhydrates liegt bei 197,5 bis 2000C (Zers.).
Nach einer ähnlichen Methode wie die obige lassen sich die folgenden Verbindungen herstellen: r-(4-Phenylpiperidino)-2-chlor-5-fluorbutyrophenon, r-(4-Hydroxy-4-p-chlorphenylpiperidino)-2-chlor-5-fluor- butyrophenon, r-(4-Hydroxy-4-p-chlorphenylpiperidino)-2-brom-5-fluorbu- tyrophenon, y-(4-Hvdroxy-4-p-tolylpiperidino)-2-chlor-5-fluorbutywphe- non, y-(4-Hydroxy-4.m-trifluormethylphenylpiperidino)-2-chlor-5- -fluorbutyrophenon, y-(4-Hydroxy-4-p-methoxyphenylpiperidino).2-chlor-5-fluor- butyrophenon.
Beispiel 3 Stufe I
In eine unter Rühren gehaltene Lösung von 12,75 g p-(6- -Fluor-2-methyl-3-indolyl)-propionsäure und 5,83 g Triäthylamin in 70 cm3 Tetrahydrofuran gibt man tropfenweise eine Lösung von 6,25 g Chlorameisensäureäthylester in 20 cm3 Tetrahydrofuran bei einer Temperatur von weniger als 2"C hinzu. Dann wird während weiteren 15 Minuten bei weniger als 2"C gerührt und hierauf tropfenweise mit einer Lösung von 12,2 g 4-p-Chlorphenyl-4-hydroxypiperidin in 50 cm3 Tetrahydrofuran und 150 cm3 Chloroform versetzt Nach beendeter Zugabe wird das Reaktionsgemisch während 5 Std.
bei Zimmertemperatur gerührt und hierauf unter vermindertem Druck zur Trockne eingeengt. Der erhaltene Rückstand wird mit Wasser verdünnt und mit Äthylacetat extrahiert. Die Äthylacetatlösung wird nacheinander mit einer l0%igen wässrigen Salzsäurelösung, einer 10%igen wässrigen Natriumhydroxydlösung und schliesslich mit einer gesättigten wässrigen Natriumchloridlösung gewaschen, über wasserfreiem Natriumsulfat getrocknet und unter vermindertem Druck eingedampft, wobei man 1 -[p-(6-Fluor-2-methyl-3 - -indolyl)-propionyl]-4-p-chlorphenyl-4-hydroxypiperidin in Form eines rötlichen öligen Produktes erhält.
Stufe 2
In eine Mischung von 6,6 g Lithium-aluminium-hydrid und 50 cm3 Äther wird tropfenweise eine Lösung von 26,9 g 1 -[p-(6-Fluor-2-methyl-3 -indolyl)-propionyl] -4-p-chlorphenyl -4-hydroxypiperidin, erhalten nach der Stufe 1, in 250 cm3 Tetrahydrofuran unter gelindem Erhitzen unter Rückfluss eingerührt. Hierauf wird während weiteren 3 Stunden gerührt und unter Rückfluss erhitzt und das Reaktionsgemisch hierauf tropfenweise einer Mischung von Wasser und Tetrahydrofuran unter Kühlen mit Eis zugegeben. Der ausgefällte Niederschlag wird abfiltriert und das Filtrat zur Trockne eingedampft. Der Rückstand wird aus Toluol umkristallisiert, wobei man 2-Methyl-3 [(4-chlorphenyl-4-hydroxypiperidi- no)-propyl]-6-fluorindol vom Schmelzpunkt 167,0 bis 169,00C erhält.
Stufe 3
3 bis 4% Ozon enthaltender Sauerstoff wird in eine Lösung von 14,9 g 2-Methyl-3-[y-(4-p-chlorphenyl-4-hydrox piperidino)-propyl]-6-fluorindol in 150 cm3 Essigsäure bei einer Temperatur von 15 bis 20"C eingeführt. Während des Einleitens von ozonhaltigem Sauerstoff wird das Reaktionsgemisch dunkelrot und entfärbt sich allmählich. Das erhaltene Reaktionsgemisch wird mit Wasser verdünnt, mit einer 20%igen wässrigen Natriumhydroxydlösung alkalisch gestellt und mit Äthylacetat extrahiert. Der Extrakt wird mit Wasser gewaschen, über wasserfreiem Kaliumcarbonat getrocknet und zur Trockne eingedampft.
Das verbleibende öl wird aus wässrigem Äthanol umkristallisiert, wobei man Sy-(4-p-Chlor- phenyl-4-hydroxy-piperidino) -2-acetamino-4-fluorbutyrophe- non vom Schmelzpunkt 119,5 bis 121,00C erhält.
Stufe 4
Eine Lösung von 7,3 g -(4-p-Chlorphenyl-4-hydroxypiperi- dino)-2-acetamino-4-fluorbutyrophenon und 8 cm3 Salzsäure in 80 cm3 Äthanol wird während 3 Stunden unter Rückfluss erhitzt. Nach dem Verdampfen des Äthanols und dem Ein dampfen zur Trockne wird das verbleibende feste Material aus Methanol umkristallisiert, wobei man y-(4-p-Chlorphe- nyl-4-hydroxypiperidino)-2-amino-4-fluorbutyrophenon-mo- nohydrochlorid vom Schmelzpunkt 2360C (unter Zersetzung) erhält.
Das so erhaltene Hydrochlorid wird mit einer wässrigen Kaliumcarbonatlösung behandelt, wobei man die freie Base erhält, die man aus wässrigem Äthanol umkristallisiert. Auf diese Weise erhält man y-(4-p-Chlorphenyl-4-hydroxypiperidino)-2-amino-4-fluorbutyrophenon vom Schmelzpunkt 144,5 bis 146,5"C.
Stufe 5
Zu einer gekühlten Suspension von 4,7 g y-(4-p-Chlorphe nyl-4-hydroxypiperidino)-2-amino -4-fluorbutyrophenon-hydrochlorid in 100 cm8 In-Salzsäure gibt man portionenweise 0,7 g Natrium nitrit unter Rühren bei einer Temperatur von weniger als 50C hinzu. Nach dem Rühren dieses Reaktionsgemisches während weiteren 30 Minuten wird es in eine gekühlte Suspension von 1,5 g Cuprochlorid in 15 cm3 konzentrierter Salzsäure unter kräftigem Rühren zugegeben. Dann wird während weiteren 30 Minuten bei Zimmertemperatur und hierauf während 2 Stunden bei 50 bis 700C gerührt. Nach dem Kühlen wird das Reaktionsgemisch mittels konzentrierter Ammoniumhydroxydlösung alkalisch gestellt und anschliessend mit Chloroform extrahiert. Der Extrakt wird mit Wasser gewaschen, über wasserfreiem Kaliumcarbonat getrocknet und eingedampft.
Das verbleibende Ö1 wird mit Chlorwasserstoff in Äther behandelt, wobei man y-(4-p-Chlor- phenyl-4-hydroxypiperidino)-2-chlor-4-fluorbutyrophenon.hy.
drochlorid erhält, das man aus Isopropylalkohol umkristallisiert. Schmelzpunkt: 185,5 bis 188,0 C (unter Zersetzung).
Beispiel 4 Stufe 1
Eine Lösung von 29,8 g p-(6-Fluor-2-methyl-3-indolyl)- -propionsäure und 13,8 g Triäthylamin in 180 cm3 Tetrahydrofuran wurde bei einer Temperatur von 0 bis 5"C unter Rühren tropfenweise mit einer Lösung von 14,7 g Chlorameisensäureäthylester in 20 cm3 Tetrahydrofuran versetzt.
Man rührte weitere 15 Minuten lang bei einer Temperatur unter 5"C und gab tropfenweise eine Lösung von 33 g 4-m -Trifluormethylphenyl-4-hydroxypiperidin in 250 cm3 Tetrahydrofuran zu. Nachdem die Zugabe beendet war, wurde das Reaktionsgemisch 5 Stunden lang bei Raumtemperatur gerührt und dann unter vermindertem Druck zur Trockne eingeengt. Der erhaltene Rückstand wurde mit Wasser verdünnt und mit Äthylacetat extrahiert.
Die Äthylacetatlösung wurde nacheinander mit einer obigen wässrigen Salzsäurelösung, einer 10% eigen wässrigen Natriumhydroxydlösung und einer gesättigten wässrigen Natriumchloridlösung gewaschen, über wasserfreiem Natriumsulfat getrocknet und unter vermindertem Druck eingedampft, wobei 1-[f-(6-Fluor-2-methyl- -3 -indolyl).propionyl]-4-m-trifluormethylphenyl-4-hydrnxy piperidin als rötliches öliges Produkt erhalten wurde.
Stufe 2
Ein Gemisch aus 15,3 g Lithiumaluminiumhydrid und 100 cm3 Äther wurde unter gelindem Sieden am Rückfluss und unter Rühren tropfenweise mit einer Lösung von 50,1 g des in Stufe 1 erhaltenen rohen l-[p-(6-Fluor-2.methyl-3 -indolyl)-propionyl] -4-m-trifluormethylphenyl-4-hydroxypi- peridins versetzt. Man erhitzte weitere 4 Stunden lang unter Rühren zum Rückfluss und versetzte das Reaktionsgemisch dann unter Eiskühlung tropfenweise mit einem Gemisch aus Wasser und Tetrahydrofuran. Der resultierende Niederschlag wurde abfiltriert und das Filtrat über wasserfreiem Natriumsulfat getrocknet und unter vermindertem Druck eingedampft, wobei man 2-Methyl-3 -[y- (m4nfluormethylphenyI- -4-hydroxypiperidino)-propyl]-6-fluorinc mit einem Smp.
von 129 bis 133"C erhielt.
Stufe 3
3 bis 4% Ozon enthaltender Sauerstoff wurde bei einer Temperatur von 15 bis 20"C in eine Lösung von 10 g 2-Me t thyl-3-[Y-(4-m-trifluorrnethylphenyl-4-hydroxypiperidino)- -propyl]-6-fluorindol in 100 cm3 Essigsäure eingeleitet. Während 1 bis 5 Stunden lang ozonhaltiger Sauerstoff eingeleitet wurde, färbte sich das Reaktionsgemisch dunkelrot und entfärbte sich dann allmählich. Das erhaltene Reaktionsgemisch wurde mit Wasser verdünnt, durch Zugabe einer 20%igen wässrigen Natriumhydroxydlösung alkalisch gemacht und mit Äthylacetat extrahiert. Der Extrakt wurde mit Wasser gewaschen, über wasserfreiem Kaliumcarbonat getrocknet und zur Trockene eingedampft.
Das zurückbleibende Öl wurde aus wässrigem Äthanol kristallisiert und ergab y-(4-m-Tri fluormethylphenyl-4-hydroxypiperidino)-2-acetamino-4-fluor- butyrophenon mit einem Smp. von 134 bis 135,5"C.
Stufe 4
Eine Lösung von 3,8 g -(4-m-Trifluormethylphenyl-4- -hydroxypiperidino)-2-acetamino-4- fluorbutyrophenon und 3,6 cm3 konzentrierter Salzsäure in 36 cm3 Äthanol wurde 2 Stunden lang unter Rückfluss erhitzt. Nachdem das Äthanol vollständig verdampft worden war, wurde die zurückbleibende Festsubstanz in 10 cm3 Wasser suspendiert und durch Zugabe einer 10% eigen wässrigen Natriumhydroxydlösung alkalisch gemacht. Die unlösliche Substanz wurde mit Äthylacetat extrahiert. Die Äthylacetatschicht wurde dann mit einer gesättigten wässrigen Natriumchloridlösung gewaschen, über wasserfreiem Natriumsulfat getrocknet und unter vermindertem Druck eingedampft.
Die zurückbleibende Festsubstanz wurde aus wässrigem Äthanol umkristallisiert und ergab y-(4-m-Trifluormethylphenyl-4-hydroxypiperi dino)-2-amino-4-fluorbutyrophenor mit einem Smp. von 105,5 bis 1070C.
Stufe 5
Eine Lösung von 4,25 g r-(4-m-Trifluormethylphenyl-4- -hydroxypiperidino)-2-amino-4-fluorbutyrophenon in 25 cm3 konzentrierter Salzsäure und 10 cm3 Essigsäure, die gekühlt worden war, wurde bei einer Temperatur unter 50C unter Rühren portionenweise mit einer Lösung von 0,8 g Natriumnitrit in 4 cm3 Wasser versetzt. Nachdem das resultierende Gemisch 50 Minuten lang bei einer Temperatur unter 50C gerührt worden war, wurde das Gemisch unter kräftigem Rühren zu einer gekühlten Suspension von 1,5 g Cuprochlorid in 10 cm3 konzentrierter Salzsäure gegeben. Es wurde eine weitere Stunde lang bei Raumtemperatur und dann 30 Minuten lang bei 50 bis 550C gerührt. Nach dem Abkühlen wurde das Reaktionsgemisch mit Ammoniak alkalisch gemacht und mit Äthylacetat extrahiert.
Der Extrakt wurde mit Wasser gewaschen, über wasserfreiem Natriumsulfat getrocknet und zur Trockene eingedampft. Die zurückbleibende Festsubstanz wurde aus wässrigem Äthanol umkristallisiert, wobei y- (4-m-Trifluormethylphenyl-4-hydroxypiperidino)-2 -chlor-4-fluorbutyrophenon vom Smp. 70,5 bis 72,0 C erhalten wurde.
The present invention relates to a new process for the preparation of butyrophenone derivatives, namely γ-piperidinobutyrophenone derivatives, which are effective on the central nervous system.
The aim of the present invention is to develop an advantageous process according to which substituted y.piperidinobutyrophenone derivatives can be obtained which have a halogen substituent in the o-position of the benzene nucleus. The o-amino-γ-piperidinobutyrophenone derivatives used as starting material can be obtained by oxidation of 3y-pipendinopropylindole derivatives to o-acylamino-γ-pipefl-dinobutyrophenone derivatives, the acylamino group of which can be converted into an unsubstituted amino group by a customary hydrolysis reaction.
According to the invention, the amino group is replaced by a halogen atom by diazotization and halogenation. Thus, in the present process, various γ-piperidinobutyrophenone derivatives can be produced in a very advantageous manner.

The acid addition salts of the 3 - (γ-piperidinopropyl) indole derivatives can be prepared by treating the free base with an acid, e.g. Hydrochloric acid, hydrobromic acid, sulfuric acid, phosphoric acid, nitric acid, acetic acid, oxalic acid, citric acid, tartaric acid, glycolic acid, benzoic acid, sulfamic acid, mandelic acid, etc.
γ-Piperidinobutyrophenones of the formula (II) can be prepared by treating the above-mentioned 3-γ-pipen-dinopro pylindoles of the formula (IX) with an oxidizing agent. The oxidative cleavage reaction is preferably carried out using an oxidizing agent such as e.g. Ozone, hydrogen peroxide, performic acid, peracetic acid, perbenzoic acid, chromic acid or potassium permanganate.
The oxidizing agent should, however, in no way be restricted to the oxidizing agents just mentioned and given purely by way of example.
The reaction generally takes place at room temperature, but higher or lower temperatures can also be used in order to regulate the course of the reaction in the desired manner. The oxidizing agent will preferably be chromic acid or ozone. The reaction is preferably carried out in the presence of a solvent. The choice of solvent depends on the oxidizing agent used.
Water, acetone, carbon tetrachloride, acetic acid, sulfuric acid, etc. come into question as solvents. The oxidizing agent can be used in a stoichiometric amount or in a larger amount. The reaction temperature varies depending on the oxidizing agent used.
If the oxidation is carried out by using chromic acid in the presence of acetic acid, the chromic acid will preferably be used in twice to 3 times the equimolar amount and the reaction will be carried out at room temperature. A 3-γ-piperidinopropylindole derivative will be dissolved or suspended in the solvent and the oxidizing agent will be added to the solution or suspension while stirring. In general, the reaction is complete in about 24 hours.
In those cases in which the oxidation is carried out using ozone, the reaction will be carried out at room temperature. A 3-y-piperidinopropylindole derivative is used in a solvent such as e.g. Formic acid, acetic acid, carbon tetrachloride or the like, dissolve or suspend and blow ozonated oxygen into the solution or suspension with stirring.
The desired γ-piperidinobutyrophenone derivatives can be obtained from the reaction mixture in crude form by extraction, with or without prior neutralization, and by evaporation to dryness. If desired, the product can be purified by recrystallization from a suitable solvent, e.g. Ethanol, isopropanol or the like, further clean in the usual way.
The compounds of the formula (II) obtained can be hydrolyzed to give a corresponding deacylated compound of the formula (III). The hydrolysis can be carried out under acidic or alkaline conditions depending on the hydrolysis method.
γ-Piperidinobutyrophenones of the formula (I) are obtained by diazotizing the above-mentioned o-amino compounds of the formula (III) and then replacing the diazonium group with a halogen atom.
The diazotization can be carried out in a manner known per se and the exchange reaction can be carried out by treating the diazonium salt with copper powder, fluoroboric acid or a metal salt, e.g. Cuprous chloride, cupro bromide, potassium iodide, a mercury halide, etc. can be made.

These compounds form corresponding acid addition salts in a manner known per se, for example by dissolving the free base in an aqueous solution which contains a suitable acid and then isolating the salt obtained by evaporating the solution. These γ-piperidinobutyrophenone derivatives of the formula (I) and their pharmaceutically acceptable acid addition salts have an effect on the central nervous system. They are suitable as anxiolytic and antipsychotonic agents, furthermore as agents for improving emotional stability and as antispasmodic or antipsychotic drugs.
Each of the pharmaceutically active compounds which are obtained according to the invention can be incorporated into tablets as the only active substance, for example for oral administration. A typical tablet consists of 1 to 2% binder, e.g. Tragacanth, 3 to 10% lubricant, e.g.
Talc, 0.25 to 1.0% of a lubricant such as e.g. Magnesium stearate, an average dose of the active ingredient and a sufficient amount of a bulking agent, e.g. Lactose to make 100%. The usual daily dose is between 1 and 100 mg per os.
The following examples illustrate the present invention.
Example I Level I.
A mixture of 14.5 g of phenylhydrazine hydrochloride, 19.2 g of 1- (γ-acetylbutyryl) piperidine and 100 cm3 of acetic acid is heated to 75 to 80 ° C. for 4 hours with stirring. Recrystallization of the residue from benzene gives 2-methyl-3 - (γ-piperidinopropyl) indole with a melting point of 140.5 to 141 ° C.

Oxygen containing 3 to 4% ozone is passed into a solution of 2.0 g of 2-methyl-3- (γ-piperidinopropyl) indole in 40 cm3 of acetic acid solution, the product turning pale yellow (in practice, this is about 35 minutes), keeping the temperature at 16-20 ° C.
After the reaction mixture has been made alkaline by adding a 10% aqueous sodium hydroxide solution, the reaction product is extracted with ethyl acetate.
The extract is washed with water and evaporated, leaving a small residue. The residue is allowed to crystallize from a mixture of ether and petroleum ether, giving γ-piperidino-2-acetaminobutyrophenone with a melting point of 45 ° to 480 ° C.

Level 4
A solution of 3.5 g of y- (4-piperidino) -2-acetaminobutyrophenone hydrochloride and 6 cm3 of concentrated hydrochloric acid in 70 cm3 of ethanol is refluxed for 3 hours. After cooling, the reaction mixture is diluted with 100 cm3 of water and then made alkaline with 50% strength aqueous sodium hydroxide solution. The resulting oily substance is extracted with ethyl acetate. The extract is washed with water and evaporated to a residue which is recrystallized from aqueous ethanol.
In this way, y- (4-piperidino) -o-aminobutyrophenone, melting point at 92 to 93.degree. C., is obtained.
Level 5
To a cooled solution of 3.6 g of γ-piperidino-2-aminobutyrophenone in 35 cm3 of 2N hydrochloric acid, a solution of 1.1 g of sodium nitrite in 5 cm3 of water is added dropwise with stirring below 0 ° C. The diazonium salt solution obtained is added to a cold suspension of 1.5 g of cuprous chloride in 5 cm3 of concentrated hydrochloric acid with vigorous stirring. The mixture is then stirred with ice-cooling for a further 30 minutes and then stirred for a further 2 hours at room temperature and then for 1 hour at 55 to 60 ° C. After cooling, the reaction mixture is made alkaline with ammonium hydroxide and extracted with ether.
The ethereal extract is washed with an aqueous solution saturated with sodium chloride, dried over anhydrous sodium sulfate and evaporated to give an oily residue. The oil obtained is treated with anhydrous hydrogen chloride in ether, crystals of γ-piperidino-2-chlorobutyrophenone hydrochloride being obtained which are recrystallized from ethanol. Example 2
A solution of 1.52 g of sodium nitrite in 10 cm3 of water is added to a cooled solution of 7.1 g of y- (4-hydroxy-4-phenylpiperidino) -2-amino-5-fluorobutyrophenone in 200 cm3 of 2N hydrochloric acid, with stirring at a temperature of less than 0 C. The diazonium salt solution obtained is added to a cooled suspension of 2.0 g of cuprous chloride in 20 cm3 of concentrated hydrochloric acid with vigorous stirring. Stirring is continued for a further 2 hours at room temperature and then for a further 2 hours at 55 to 60 ° C. After cooling the reaction mixture, it is made alkaline by adding an aqueous ammonium hydroxide solution and then extracted with chloroform.
The extract is washed with water, dried over anhydrous sodium sulfate and evaporated to give r- (4-hydroxy-4-phenyl) -2-chloro-5-fluorobutyrophenone. The melting point of the chlorohydrate is 197.5 to 2000C (decomp.).

Example 3 Stage I

hydrochloride is obtained, which is recrystallized from isopropyl alcohol. Melting point: 185.5 to 188.0 C (with decomposition).
Example 4 Stage 1
