Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung von 6-(o-Halogenphenyl)- 1 -methyl- 4H-s-triazolo[4,3-al[ 1,4]benzodiazepinen der folgenden Formel

worin Hal Halogen bedeutet mit einer Atomnummer bis zu 35, nämlich Fluor, Chlor oder Brom. Die erfindungsgemäss erhältlichen Verbindungen können in ihre pharmakologisch annehmbaren Säureadditionssalze überführt werden. Die neuen Verbindungen sind insbesondere nützlich als Entspannungsmittel für die Muskulatur sowie auch als anxiolytisches Mittel.
Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass man eine Verbindung der Formel

Die Ausgangsmaterialien für das erfindungsgemässe Verfahren können nach einer Methode von G.A. Archer und L.H. Sternbach im J. Org. Chem. 29, 231 (1964) und US Patent 3 422 091 hergestellt werden. Diese Ausgangsmaterialien erhält man üblicherweise durch die Umsetzung bekannter entsprechender 1 ,3-Dihydro-5-(o-halogenphenyl)- 2H- 1 ,4-benzodiazepin-2-one mit Phosphorpentasulfid in Pyridin durch Erhitzen während 45 Minuten (Archer et al., ibid.).
Präparat
1,3-Dihydro-5-(o-chlorphenyl) 2H-1 ,4-benzodiazepin-2-thion
Eine Mischung, die aus 23 g (0,085 Mol) 1,3-Dihydro 5-(o-chlorphenyl)-9H-1,4-benzodiazepin-2-on [Sternbach et al., J. Med. Chem. 6, 261 (1963)1 und 21 g Phosphorpentasulfid in 1500 ml Pyridin bestand, wurde schnell auf die Rückflusstemperatur erhitzt und dann 45 Minuten lang am Rückfluss gekocht. Die Reaktionsmischung wurde anschliessend zur Trockne im Vakuum konzentriert und man behandelte den Rückstand mit 150 ml gesättigter Natriumchloridlösung und extrahierte dreimal mit Methylenchlorid. Die Methylenchloridlösung wurde mit Wasser gewaschen und über trockenem Magnesiumsulfat getrocknet. Man filtrierte ab und dampfte zur Trockne ein.
Der Rückstand wurde in 1 Liter absolutem Äthanol gelöst und mit Darco behandelt und anschliessend heiss filtriert. Dann konzentrierte man die Lösung auf ein Volumen von 200 ml ein und liess abkühlen. Man erhielt 19 g (78%) einer gelben, kristallinen Verbindung, die einen Schmelzpunkt von 228 bis 229C hatte. Durch IR-Spektren sowie auch durch NMR-Spektren konnte man die Struktur beweisen.

Beispiel 6-(o-Chlorphenyl)-1-methyl-4H-s-triazolo- [4,3-a] [1 ,4]-benzodiazepin
Eine Mischung, die aus 14 g (0,05 Mol) 1,3-Dihydro-5 (o-chlorphenyl)-2H-1,4-benzodiazepin-2-thion und 10 g (0,13 Mol) Acethydrazid in 1400 ml normalem Butanol bestand, wurde 24 Stunden lang am Rückfluss erhitzt. Während dieser Zeit leitete man Stickstoff durch die Reaktionsmischung. Diese wurde dann zur Trockne im Vakuum eingedampft und man behandelte den erhaltenen Rückstand mit Wasser, wobei eine gelbe feste Substanz auftrat. Diese wurde gesammelt und an Luft getrocknet. Man erhielt 12 g. Die Verbindung wurde kristallisiert, indem man sie in 700 ml Äthylacetat löste und konzentrierte anschliessend auf ein Volumen von 150 ml.
Durch Abkühlen erhielt man 11 g (73 O/o) einer weissen kristallinen Verbindung, die einen Schmelzpunkt von 211,50 bis 2130C hatte. Diese Verbindung zeigte nur einen einzigen Fleck während Dünnschichtchromatographie (10% MeOH/CHCl3).
Die IR- und NMR-Spektren bewiesen die Struktur.

The starting materials for the process according to the invention can be prepared by a method of G.A. Archer and L.H. Sternbach in J. Org. Chem. 29, 231 (1964) and US Patent 3,422,091. These starting materials are usually obtained by reacting corresponding known 1,3-dihydro-5- (o-halophenyl) -2H-1,4-benzodiazepin-2-ones with phosphorus pentasulfide in pyridine by heating for 45 minutes (Archer et al., ibid.).

1,3-dihydro-5- (o-chlorophenyl) 2H-1,4-benzodiazepine-2-thione
A mixture consisting of 23 g (0.085 mol) 1,3-dihydro 5- (o-chlorophenyl) -9H-1,4-benzodiazepin-2-one [Sternbach et al., J. Med. Chem. 6, 261 (1963) 1 and 21 g of phosphorus pentasulfide in 1500 ml of pyridine, was quickly heated to reflux temperature and then refluxed for 45 minutes. The reaction mixture was then concentrated to dryness in vacuo and the residue was treated with 150 ml of saturated sodium chloride solution and extracted three times with methylene chloride. The methylene chloride solution was washed with water and dried over dry magnesium sulfate. It was filtered off and evaporated to dryness.
The residue was dissolved in 1 liter of absolute ethanol and treated with Darco and then filtered while hot. The solution was then concentrated to a volume of 200 ml and allowed to cool. 19 g (78%) of a yellow, crystalline compound were obtained which had a melting point of 228 to 229C. The structure could be proven by IR spectra as well as by NMR spectra.

Example 6- (o-Chlorophenyl) -1-methyl-4H-s-triazolo- [4,3-a] [1,4] -benzodiazepine
A mixture consisting of 14 g (0.05 mol) 1,3-dihydro-5 (o-chlorophenyl) -2H-1,4-benzodiazepine-2-thione and 10 g (0.13 mol) acethydrazide in 1400 ml normal butanol was refluxed for 24 hours. During this time nitrogen was bubbled through the reaction mixture. This was then evaporated to dryness in vacuo and the residue obtained was treated with water, a yellow solid substance appearing. This was collected and air dried. 12 g were obtained. The compound was crystallized by dissolving it in 700 ml of ethyl acetate and then concentrated to a volume of 150 ml.
Cooling gave 11 g (73%) of a white crystalline compound which had a melting point of 211.50 to 2130C. This compound showed only a single spot during thin layer chromatography (10% MeOH / CHCl3).
The IR and NMR spectra proved the structure.
