Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer Fluorenverbindungen, welche biologische Aktivität aufweisen und z. B. antidepressiv, anticonvulsisch, anti inflarnmatorisch oder analgetisch wirken.
Die neuen Verbindungen haben die Formel

worin R1 Wasserstoff, Alkyl mit 1 bis 6 C-Atomen ist, Alk Alkylen mit 1 bis 6 C-Atomen bedeutet, R3 und R4 gleich oder verschieden sind und Wasserstoff, Aryl, gegebenenfalls arylsubstituiertes Alkyl mit 1 bis 6 C-Atomen, Acyl, Aroyl, oder zusammen mit dem N-Atom, woran sie gebunden sind, einen gegebenenfalls weitere Heteroatome aufweisenden, 3bis 8gliedrigen Heteroring, der seinerseits mit Alkyl oder Hydroxyalkyl substituiert sein kann, darstellen, sowie Z und Z, gleich oder verschieden sind und Wasserstoff, Halogen, Alkyl mit 1 bis 6 C-Atomen, Alkoxy, Sulfonamido, Trihalogenmethyl, Nitro oder Hydroxy bedeuten, mit der Massgabe, dass wenn Z, Z1 und R1 Wasserstoff sind, die Gruppierung Alk-NR3lR, kein a-Diäthylaminoäthyl, a-Piperidino äthyl, a-Dimethylaminopropyl, Diäthylaminomethyl,
Dimethylaminomethyl, Piperidinomethyl oder Morpholinomethyl bedeutet.
Die in Form der freien Basen erhaltenen neuen Fluorenverbindungen lassen sich gegebenenfalls auch in ihre physiologisch verträglichen Säureadditionssalze überführen.
In den erfindungsgemäss herstellbaren Verbindungen lässt sich die Carbonylgruppe zur Methylengruppe reduzieren. Solche Verbindungen gleichen dem Imipramin in ihren Wirkungen auf das zentrale Nervensystem, sind jedoch praktisch frei von den peripheralen anticholinergischen Wirkungen, welche verantwortlich sind für einen grossen Teil der unerwünschten Nebeneffekte der tricyclischen Antidepressantien beim klinischen Gebrauch, z. B. trockenem Mund, Eingeweide-Störungen und Sehstörungen. So erwies sich z. B.
an Mäusen die Verbindung der Formel II als ebenso aktiv wie Imipramin gegenüber Depression und Hypothermie, verursacht durch die intraventrikulare Injektion von Noradrenalin, sowie gegen plötzliche Anfälle, verursacht durch elektrische Stimulation und gegen Hypothermie, resultierend aus intraventrikularer Injektion von Oxotremorin. Beim letzteren Test, welcher ein Anzeichen für eine zentrale anticholinergische Wirkung darstellt, ergab sich kein Hinweis auf peripherale anticholinergische Wirkungen, verursacht durch die Droge z. B. Mydriasis.

Andere der erfindungsgemäss herstellbaren Verbindungen erweisen sich als wirksam bei analgetischen und antiinflammatorischen Tierversuchen. So zeigte sich z. B. die Verbindung der Formel III als 3- bis Sfach so wirksam wie Codein im Schwanz-Klemm-Test an Mäusen nach (Bianchi, C. und Franceschini, J.: Brit J. Pharmacol. 1954, 9, 280). Die Verbindung war nicht vom narcotisch-analgetischen Typ, da sie einen Straub-Index von < < 0,5 aufwies (vgl. Shemano, I. und Wendel H.: Toxicol. Appl. Pharmacol. 1964, 6, 334) im Vergleich zu Morphin = 30,5, wobei ihre analgetische Wirksamkeit durch Nalorphin nicht antagonisiert wurde. Die antiinflammatorische Wirksamkeit der Verbindung gegenüber carrageenininduziertem modem bei der Ratte (vgl. Winter, C. A., Risley, E. A. und Nuss, G. W.: Proc. Soc. Exp. Biol.
Med., 1962, 111, 544) lag zwischen derjenigen von Phenylbutazon und Indomethacin, war jedoch nicht begleitet von der Salzretention, welche bei diesen letzteren Verbindungen beobachtet wird.
Weiterhin hat sich gezeigt, dass einige der neuen Verbindungen die Aggregation der menschlichen Blutkörperchen vermittels aggregierender Agenzien, wie Collagen und Adrenalin inhibiert. Unter Verwendung der turbidimetrischen Methode von Born (G. V. R. Born und J. Cross. J. Physiol.
1963, 168, 178 bis 195), welche die Aggregation kontinuierlich zu verfolgen gestattet, hat sich ergeben, dass gewisse dieser Verbindungen und insbesondere das N-[4-(dimethylamino)-butyl]-fluoren-9-amin-dihydrochlorid-dihydrat gemäss Beispiel 15 die Aggregation menschlicher Blutkörperchen significant inhibiert, wie sie durch Adenosin-diphosphat verursacht wird. Die Anwendung derartiger Verbindungen empfiehlt sich deswegen in Fällen wie Artherosklerose und coronaren Herzerkrankungen.
Die erfindungsgemäss herstellbaren Verbindungen können z. B. gebraucht werden als antidepressive, anticonvulsische oder analgetischiantiinfiammatorische Arzneien in der Human- oder Veterinärmedizin. Dabei können sie pharmazeutischen Zubereitungen einverleibt werden, welche als aktive Ingredientien die Fluorenderivate gemäss der vorstehenden Formel I, bzw. physiologisch verträgliche Säureadditionsalze derselben enthalten. Derartige Zubereitungen können für den Gebrauch in bekannter Weise beschaffen sein, z. B. unter Zusatz von Trägerstoffen oder Excipientien, sowie Formulierungszusätzen entsprechend ihrem jeweiligen Verwendungszweck mit und ohne zusätzliche Medizinalstoffe.
Es kann sich dabei um feste oder flüssige Zubereitungen für oralen Gebrauch, sowie für Injektionen handeln.
Für orale Verabreichung werden die Substanzen mit Vorteil in Tabletten eingearbeitet, welche sich nach üblichen Verfahren herstellen lassen und gegebenenfalls mit einer Überzugsschicht versehen sein können.
Für die Zwecke der Injektion können die neuen Substanzen in physiologisch verträgliche Trägerstoffe eingearbeitet werden und als Lösungen, Suspensionen oder als trockene Produkte vorliegen, welch letztere unmittelbar vor dem Gebrauch in injizierbare Form übergeführt werden.
Die Dosierungen, mit welchen diese neuen Ingredientien verabreicht werden können, können innerhalb eines weiten Bereiches variieren. Eine geeignete Dosis für mündliche Verabreichung liegt im Bereich von im allgemeinen 10 bis
1000 mg pro Tag, in Abhängigkeit vom Alter, vom Gewicht und vom Zustand des Patienten.
Bevorzugte, nach dem erfindungsgemässen Verfahren herstellbare Verbindungen, sind:
1. N-(Fluoren-9-yl)-2-methylaminoacetamid
2. 2-Amino-N-(fluoren-9-yl)-acetamid
3. N-(Fluoren-9-yl)4-methyl-1-piperazineacetamid
4. 2-Dimethylamino-N-(fluoren-9-yl)-propionamid
5. 2-Acetamido-N-(fiuoren-9-yl)-acetamid
6. N-(Fluoren-9-yl)-N' ,N' dimethyläthylendiamin
7. N-(Fluoren-9-yl)-N,N' ,N' -trimethyläthylendiamin
8. N2-(Fluoren-9-yl)-Nt,Nl-dimethyl-1,2-propandiamin
Das erfindungsgemässe Verfahren zur Herstellung der neuen Fluorenverbindungen ist dadurch gekennzeichnet,

umsetzt. In der letzteren kann gegebenenfalls anschliessend die CO-Gruppe zur CH2-Gruppe reduziert werden.
Verbindungen der Formel VI lassen sich z. B. herstellen durch Umsetzen eines Halogenids einer Halogen-alkancarbonsäure der Formel V mit einem Fluorenderivat der Formel IV.

Erfindungsgemäss herstellbare Verbindungen, worin R3 und R4 Wasserstoffatome darstellen, lassen sich aus Verbindungen der Formel I, worin R3 und R4 Benzylgruppen sind, vermittels Hydrogenolyse mit Edelmetallkatalysatoren herstellen. Oder es kann auch die Halogenverbindung VI kondensiert werden mit einem Alkalimetallsalz von Phthalimid oder Succinimid, worauf die schützende Gruppe durch Hydrazinolyse abgespalten werden kann.
Die nach dem erfindungsgemässen Verfahren erzeugten Amidverbindungen der Formel I werden zur Herstellung entsprechender Amine verwendet, mit der Massgabe, dass wenn Z und Z1 Wasserstoff sind, El Äthyl und Alk Methylen ist R3 und R4 kein Methyl sind; diese Herstellung ist dadurch gekennzeichnet, dass man die Carbonylgruppe im Zwischenglied -N-CO-Alk- entsprechender Verbindungen der Formel I zur Methylengruppe reduziert. Diese Reduktion wird mit Vorteil mit einem komplexen Metallhydrid, z. B.
Lithiumaluminiumhydrid, vorzugsweise in Gegenwart von Aluminiumchlorid, durchgeführt.
Beispiel 1
N-(Fluoren-9 -yl) -N' ,N' -dimethyläthylendiamin a) 2,4 g 2-Dimethylamino-N-(fluoren-9-yl)-acetamid wurden im Soxhlet extrahiert, in eine am Rückfluss siedende Suspension von 1,2 g Aluminium-lithiumhydrid in 170 ml Diäthyläther bis die Reduktion nach 6 Stunden vollständig abgelaufen war. Das Reaktionsgemisch wurde vorsichtig mit Wasser behandelt und die Phasen wurden anschliessend getrennt. Chloroformextrakte der wässrigen Phase wurden mit der ätherischen Schicht vereinigt, gewaschen mit Wasser und getrocknet mit Magnesiumsulfat, worauf das Lösungsmittel im Vakuum abgetrieben wurde. Das zurückbleibende in Äthylacetat gelöste Öl wurde mit 5 % Äthylacetat/HC1 behandelt, wobei das Diamin-dihydrochlorid-monohydrat entstand, welches aus Äthanol/Äther als weisses Pulver isoliert wurde.
Erhalten wurden 1,4 g mit dem Smp. 2402410 C.
b) 440 g N,N-Dimethyläthylendiamin und 1380 g Kaliumcarbonat und Kaliumiodid wurden 2 Stunden lang in Benzol gerührt. Zum Gemisch wurden 1224 g 9-Bromfluoren hinzugegeben und das Gemenge anschliessend 20 Stunden lang unter Rückfluss erhitzt. Schliesslich wurde abgekühlt und filtriert. Das Filtrat wurde im Vakuum eingedampft. Das verbliebene Öl im Gewicht von 947 g, eingetragen in 1000 ml Äthanol, ergab eine Fällung von 701 g mit dem Smp. 218 bis 220" C nach Umsetzung mit Chlorwasserstoff. Umkristallisieren aus Methanol/Isopropylacetat (Aktivkohle) ergab weissliche Kristalle des Dihydrochlorid-monohydratsalzes im Gewicht von 375 g mit einem Smp. von 236 C.
c) Eine Mischung von 45 g 9-Fluorenon, 25 g N,N-Dimethyläthylendiamin und 0,6 g seines Dihydrochloridsalzes wurde unter Rühren in 500 ml Benzol unter azeotropem Rückfluss 28 Stunden lang erwärmt. Anschliessend wurde das Benzol abgetrieben und der harzartige Rückstand in 500 ml Äthanol gelöst. Er wurde unter Wasserstoff mit PdO-C (5 g, 10%) geschüttelt, bis ein Mol Wasserstoff absorbiert worden war. Der Katalysator wurde abfiltriert und das Filtrat auf die Hälfte seines Volumens eingeengt. Behandlung mit Chlorwasserstoff ergab eine rehfarbene Fällung im Gewicht von 24 g mit dem Smp. 230 , welche sich aus Methanol Isopropylacetat als weisse Kristalle des Dihydrochloridsalzes im Gewicht von 20,3 g und mit dem Smp. 240 C abschied.
Andere nach dieser Verfahrensweise hergestellte Stoffe sind in der Tabelle I zusammengestellt.

Beispiel 19 2-Dibenzylamino-N-(fluoren-9-yl) -acetamid
5,15 g 2-Chlor-N-(fluoren-9-yl)-acetamid, 7,9 g Dibenzylamin und 200 ml Dimethylformamid wurden 9 Stunden lang am Rückfluss erwärmt. Das Gemisch wurde im Vakuum konzentriert, verdünnt mit Benzol und filtriert. Das Lösungsmittel wurde ersetzt durch Petroläther (80-100 C), woraus das Dibenzylamino-acetamid sich in Nadeln abschied.
Erhalten wurden 4 g mit dem Smp. 85-86" C. Der Smp. des Hydrochlorids betrug 235-236 (aus Methanol/Äther).
Beispiel 20 2-Acetamido-N-(flouren-9-yl) -acetamid
2,0 g 2-Amino-(N-fiuoren-9-yl) -acetamid-hydrochlorid in 40 ml Pyridin wurden mit 8,0 ml Essigsäureanhydrid bei Zimmertemperatur 20 Stunden lang behandelt und hernach 30 Minuten lang auf 60 C erwärmt. Das Gemisch wurde in 6n Salzsäure eingegossen und das ausgefallene Produkt abfiltriert. Es wurde umkristallisiert aus Äthanol und ergab die gewünschte Verbindung als farblose Mikrokristalle im Gewicht von 1,1 g mit einem Smp. von 249-251" C.
Andere nach den Arbeitsweisen der Beispiele 19 und 20 hergestellten Amide sind in der nachfolgenden Tabelle II beschrieben.

Beispiel 40
2-Amino-N-(fluoren-9-yl)-acetamid
3,9 g 2-Dibenzylamino-N-(Suoren-9-yl)-acetamid-hydro- chlorid in 250 ml Methanol wurden über 8 g 10tonigem PdC hydrogenolysiert.
Der Katalysator wurde abgetrennt und das Produkt auskristallisiert aus Methanol/Äthylacetat. Dabei wurden 2,1 g des primären Amin-hydrochlorids in Form des Monohydrats erhalten. Smp. 2762780 C.
Beispiel 41
N-(Fluoren-9-yl) -N,N' ,N' -trimethyl-äthylendiamin
3,5 g N-(Fluoren-9-yl)-N' ,N' -dimethyläthylendiamin, 30 ml 36%ges wässriges Formaldehyd und 15 ml 96%ige Ameisensäure wurden 6 Stunden lang am Rückfluss erwärmt.
Die freie Base wurde nach Isolierung und Trocknung aus ätherischer Lösung mit Magnesiumsulfat destilliert und ergab das gewünschte Diamin als gelbe Flüssigkeit, im Gewicht von 2,8 g. Sdp. 148-1500 C/0,05 mm, nrl22 = 1,5874.
Beispiel 42 2-[(Fluoren-9-yl)-amino]-acetamid
13,0 g Fluoren-9-yl-amin in 50 ml Acetonitril wurde 18 Stunden lang am Rückfluss mit 2,7 g Chloracetonitril erhitzt.
Die erhaltenen 6,7 g Fluoren-9-yl-amino-hydrochlorid wurden abfiltriert und das Filtrat im Vakuum eingeengt, wobei ein Öl zurückblieb, welches 10,4 g 2-[(Fluoren-9-yl)-amino]- acetonitril-hydrochlorid vom Smp. 207-208 C aus Methanol/ Äther ergab. 2,0 g des Hydrochlorids in 50 ml konzentrierter Salzsäure wurden 18 Stunden lang bei Zimmertemperatur gerührt. Das Amid schied sich in Form farbloser Nadeln als Hydrochlorid im Gewicht von 1,3 g aus Methanol/Äther ab.
Smp. 251" C.
Beispiel 43 N-(Fluoren-9-yl)-äthylendiamin
4,0 g 2-[(Fluoren-9-yl)-amino]-acetonitril in 30 ml Äther wurden tropfenweise unter Rühren zu einer Suspension hinzugefügt, welch letztere hergestellt worden war bei 0 aus 2,4 g Aluminium-lithiumhydrid und 4,85 g Aluminiumchlorid in 250 ml Äther. Das Gemisch wurde am Rückfluss 48 Stunden lang erwärmt und dann zersetzt. Nach Aufarbeitung in üblicher Weise wurden 5,2 g eines gelben, basischen Harzes erhalten, aus welchem das Dihydrochlorid im Gewicht von
2,2 g in Form von Nadeln aus Methanol/Äther sich ergab.
Smp. 2562580 C (Zersetzung).
Beispiel 44 N-j2fluoren-9-yl)-aminoäthylj-acetamid
3,3 g N-(2-Aminoäthyl)-acetamid wurden in 100 ml Benzol mit 5,4 g Fluoren und 2 ml 10toiger Äther-HC1 erwärmt, bis die Entwicklung von Wasser aufgehört hatte, was nacb 24 Stunden der Fall war. Die heisse Lösung wurde filtriert und gekühlt und ergab tiefgelb gefärbte Kristalle im Gewicht von 3,1 g, entsprechend 39 Ausbeute und mit einem Smp. von 1450 C. Umkristallisieren aus Petroläther (Siedebereich 10ab120") ergab gelbe Nadeln vom N-[2-(Fluoren 9-yliden)-aminoäthyl]-acetamid mit einem Smp. von 156 C.
2,5 g des Azomethins wurden geschüttelt mit einem Gramm 10 %igem Pd-C in 125 ml Methanol unter Wasserstoff, bis die theoretische Wasserstoffmenge absorbiert worden war. Das rote Öl, welches sich dabei abschied, ergab nach Filtrieren und Eindampfen eine rote, feste Substanz vermittels Äther HCI. Zweimaliges Umkristallisieren aus Methanol/Äther ergab farblose Prismen des Hydrochlorids im Gewicht von 0,75 g mit dem Smp. 270 C.
Beispiel 45 N2-(Fluoren-9-yl)-N1,N'-dimethyl- 1,2-propandiamin
4,2 g 2-(Fluoren-9-yl)-amino]-N,N-dimethylpropion- amid in 100 ml Äther wurden mit einem Gemisch behandelt, welches hergestellt worden war aus 1,8 g Aluminium-lithiumhydrid und 6,0 g Aluminiumchlorid in 20 ml Äther gemäss Beispiel 43. Das gewünschte Diamin wurde erhalten als farblose Rosetten eines Dihydrochlorids im Gewichts von 1,0 g mit dem Smp. 2192200 C aus Äthanol/Äther.
Beispiel 46
2-(Fluoren-9-yl-amino) -N,N-dimethylacetamid
2,43 g 2-Chlor-N,N-dimethylacetamid wurden erwärmt mit 7,24 g Fluoren-9-yl-amin in 200 ml Benzol am Rückfluss während 4 Stunden. Das Benzol wurde im Vakuum abgetrieben und der Rückstand in Äther extrahiert. Bei Zugabe von Chlorwasserstoff schied sich das Hydrochlorid-hemihydrat ab und wurde umkristallisiert aus Äthanol/Äther. The present invention relates to a process for the preparation of new fluorene compounds which have biological activity and are e.g. B. have an antidepressant, anticonvulsant, anti-inflammatory or analgesic effect.

The new fluorene compounds obtained in the form of the free bases can, if appropriate, also be converted into their physiologically tolerable acid addition salts.
In the compounds which can be prepared according to the invention, the carbonyl group can be reduced to the methylene group. Such compounds are similar to imipramine in their effects on the central nervous system, but are practically free of the peripheral anticholinergic effects which are responsible for a large part of the undesirable side effects of tricyclic antidepressants in clinical use, e.g. B. dry mouth, intestinal disorders and visual disorders. So it turned out z. B.
on mice the compound of formula II as active as imipramine against depression and hypothermia caused by the intraventricular injection of noradrenaline, as well as against sudden attacks caused by electrical stimulation and against hypothermia resulting from intraventricular injection of oxotremorine. In the latter test, which is an indication of a central anticholinergic effect, there was no indication of peripheral anticholinergic effects caused by the drug e.g. B. Mydriasis.

Other compounds that can be prepared according to the invention are found to be effective in analgesic and anti-inflammatory animal experiments. So it was shown z. For example, the compound of the formula III is 3 to 5 times as effective as codeine in the tail-clamping test on mice (Bianchi, C. and Franceschini, J .: Brit J. Pharmacol. 1954, 9, 280). The compound was not of the narcotic-analgesic type, since it had a Straub index of <<0.5 (cf. Shemano, I. and Wendel H .: Toxicol. Appl. Pharmacol. 1964, 6, 334) in comparison to Morphine = 30.5, its analgesic effectiveness not being antagonized by nalorphine. The anti-inflammatory activity of the compound against carrageenin-induced modems in the rat (cf. Winter, C. A., Risley, E. A. and Nuss, G. W .: Proc. Soc. Exp. Biol.
Med., 1962, 111, 544) was between that of phenylbutazone and indomethacin, but was not accompanied by the salt retention observed with these latter compounds.
Furthermore, it has been shown that some of the new compounds inhibit the aggregation of human blood cells by means of aggregating agents such as collagen and adrenaline. Using Born's turbidimetric method (G. V. R. Born and J. Cross. J. Physiol.
1963, 168, 178 to 195), which allows the aggregation to be followed continuously, it has been found that certain of these compounds and in particular N- [4- (dimethylamino) butyl] fluorene-9-amine dihydrochloride dihydrate according to Example 15 significantly inhibited human blood cell aggregation such as that caused by adenosine diphosphate. The use of such compounds is therefore recommended in cases such as atherosclerosis and coronary heart disease.
The compounds which can be prepared according to the invention can, for. B. are used as antidepressant, anticonvulsant or analgesic antiinfiammatory drugs in human or veterinary medicine. They can be incorporated into pharmaceutical preparations which contain, as active ingredients, the fluorene derivatives according to the above formula I, or physiologically acceptable acid addition salts thereof. Such preparations can be designed for use in a known manner, e.g. B. with the addition of carriers or excipients, as well as formulation additives according to their respective intended use with and without additional medicinal substances.
These can be solid or liquid preparations for oral use as well as for injections.
For oral administration, the substances are advantageously incorporated into tablets, which can be produced by customary methods and optionally provided with a coating layer.
For the purpose of injection, the new substances can be incorporated into physiologically compatible carrier substances and present as solutions, suspensions or as dry products, the latter being converted into injectable form immediately before use.
The dosages with which these new ingredients can be administered can vary within a wide range. A suitable dose for oral administration is in the range of generally 10 to
1000 mg per day, depending on the age, weight and condition of the patient.
Preferred compounds which can be prepared by the process according to the invention are:

c) A mixture of 45 g of 9-fluorenone, 25 g of N, N-dimethylethylenediamine and 0.6 g of its dihydrochloride salt was heated with stirring in 500 ml of benzene under azeotropic reflux for 28 hours. The benzene was then driven off and the resinous residue was dissolved in 500 ml of ethanol. It was shaken with PdO-C (5 g, 10%) under hydrogen until one mole of hydrogen had been absorbed. The catalyst was filtered off and the filtrate was concentrated to half its volume. Treatment with hydrogen chloride resulted in a fawn-colored precipitate weighing 24 g and melting point 230, which separated out from methanol isopropyl acetate as white crystals of the dihydrochloride salt weighing 20.3 g and having a melting point of 240 ° C.

Example 19 2-Dibenzylamino-N- (fluoren-9-yl) -acetamide
5.15 g of 2-chloro-N- (fluoren-9-yl) acetamide, 7.9 g of dibenzylamine and 200 ml of dimethylformamide were refluxed for 9 hours. The mixture was concentrated in vacuo, diluted with benzene and filtered. The solvent was replaced by petroleum ether (80-100 C), from which the dibenzylamino-acetamide separated out in needles.
This gave 4 g with a melting point of 85-86 "C. The melting point of the hydrochloride was 235-236 (from methanol / ether).
Example 20 2-Acetamido-N- (fluor-9-yl) -acetamide
2.0 g of 2-amino- (N-fluorene-9-yl) acetamide hydrochloride in 40 ml of pyridine were treated with 8.0 ml of acetic anhydride at room temperature for 20 hours and then heated to 60 ° C. for 30 minutes. The mixture was poured into 6N hydrochloric acid and the precipitated product was filtered off. It was recrystallized from ethanol and gave the desired compound as colorless microcrystals weighing 1.1 g with a m.p. of 249-251 "C.

2-amino-N- (fluoren-9-yl) acetamide
3.9 g of 2-dibenzylamino-N- (suoren-9-yl) acetamide hydrochloride in 250 ml of methanol were hydrogenolyzed over 8 g of 10-tone PdC.
The catalyst was separated off and the product crystallized from methanol / ethyl acetate. 2.1 g of the primary amine hydrochloride were obtained in the form of the monohydrate. M.p. 2762780 C.

N- (Fluoren-9-yl) -N, N ', N' -trimethyl-ethylenediamine
3.5 g of N- (fluoren-9-yl) -N ', N'-dimethylethylenediamine, 30 ml of 36% total aqueous formaldehyde and 15 ml of 96% formic acid were refluxed for 6 hours.
After isolation and drying, the free base was distilled with magnesium sulfate from an ethereal solution and gave the desired diamine as a yellow liquid, weighing 2.8 g. Bp 148-1500 C / 0.05 mm, nrl22 = 1.5874.
Example 42 2 - [(Fluoren-9-yl) -amino] -acetamide
13.0 g of fluoren-9-ylamine in 50 ml of acetonitrile were refluxed with 2.7 g of chloroacetonitrile for 18 hours.
The 6.7 g of fluoren-9-ylamino hydrochloride obtained were filtered off and the filtrate was concentrated in vacuo, leaving an oil which contained 10.4 g of 2 - [(fluoren-9-yl) amino] acetonitrile hydrochloride of m.p. 207-208 C from methanol / ether resulted. 2.0 g of the hydrochloride in 50 ml of concentrated hydrochloric acid were stirred for 18 hours at room temperature. The amide separated out in the form of colorless needles as a hydrochloride weighing 1.3 g from methanol / ether.
M.p. 251 "C.
Example 43 N- (fluoren-9-yl) ethylenediamine
4.0 g of 2 - [(fluoren-9-yl) -amino] -acetonitrile in 30 ml of ether were added dropwise with stirring to a suspension, which the latter had been prepared at 0 from 2.4 g of aluminum-lithium hydride and 4, 85 g aluminum chloride in 250 ml ether. The mixture was heated to reflux for 48 hours and then decomposed. After working up in the usual way, 5.2 g of a yellow, basic resin were obtained, from which the dihydrochloride weighing
2.2 g in the form of needles from methanol / ether resulted.

Example 44 N-j2fluoren-9-yl) -aminoethylj-acetamide
3.3 g of N- (2-aminoethyl) acetamide were heated in 100 ml of benzene with 5.4 g of fluorene and 2 ml of 10-carbon ether HC1 until the development of water had ceased, which was the case after 24 hours. The hot solution was filtered and cooled and gave deep yellow colored crystals weighing 3.1 g, corresponding to 39 yield and with a melting point of 1450 C. Recrystallization from petroleum ether (boiling range 10ab120 ") gave yellow needles of N- [2- ( Fluorene-9-ylidene) aminoethyl] acetamide with a melting point of 156 C.
2.5 g of the azomethine was shaken with one gram of 10% Pd-C in 125 ml of methanol under hydrogen until the theoretical amount of hydrogen had been absorbed. The red oil, which separated out, gave a red, solid substance after filtration and evaporation by means of ether HCl. Recrystallization twice from methanol / ether gave colorless prisms of the hydrochloride weighing 0.75 g with a melting point of 270 C.
Example 45 N2- (Fluoren-9-yl) -N1, N'-dimethyl-1,2-propanediamine
4.2 g of 2- (fluoren-9-yl) -amino] -N, N-dimethylpropionamide in 100 ml of ether were treated with a mixture which had been prepared from 1.8 g of aluminum-lithium hydride and 6.0 g of aluminum chloride in 20 ml of ether according to Example 43. The desired diamine was obtained as colorless rosettes of a dihydrochloride weighing 1.0 g with a melting point of 2192200 C from ethanol / ether.
Example 46
2- (Fluoren-9-yl-amino) -N, N-dimethylacetamide
2.43 g of 2-chloro-N, N-dimethylacetamide were heated with 7.24 g of fluoren-9-yl-amine in 200 ml of benzene at reflux for 4 hours. The benzene was driven off in vacuo and the residue extracted into ether. Upon addition of hydrogen chloride, the hydrochloride hemihydrate separated out and was recrystallized from ethanol / ether. 4.7 g of microcrystals with the melting point 201-203 C.
