Die Herstellung der Ausgangsverbindungen der Formel (II) kann beispielsweise nach dem Verfahren des Schweizer Patentes Nr. 539 645 erfolgen.
Die Wasserabspaltung wird zweckmässig bei höheren Temperaturen durchgeführt, beispielsweise in einem Tempe raturbereich von 20 bis 150 C. Vorzugsweise werden Lö sungsmittel, wie z. B. Eisessig, Benzol, Dioxan usw. verwen det.
Als wasserabspaltende Mittel kommen beispielsweise in Betracht: Mineralsäuren, wie Schwefelsäure oder Halogen wasserstoffsäuren; organische Säuren, wie Oxalsäure, Amei sensäure; Thionylchlorid; Aluminiumchlorid; Zinkchlorid; Zinnchlorid; Bortrifluorid; Kaliumhydrogensulfat; Alumi niumoxid; Phosphorpentoxid; Säurechlorid; roter Phosphor Jod in Gegenwart von Wasser.
Diejenigen Verbindungen, die asymmetrische Kohlenstoff atome enthalten und in der Regel als Racemate anfallen, kön nen in an sich bekannter Weise, beispielsweise mittels einer optisch aktiven Säure in die optisch aktiven Isomeren gespal ten werden. Es ist aber auch möglich, von vornherein optisch aktive bzw. auch diastereomere Ausgangsstoffe einzusetzen, wobei dann als Endprodukt eine entsprechende reine optisch aktive Form bzw. diastereomere Konfiguration erhalten wird.
Die Überführung in die Salze erfolgt nach bekannten Methoden; als Anionen für die Salze kommen die hierfür delt es sich um solche mit 1-6 Kohlenstoffatomen. Ein Bei spiel für die Halogenalkylgruppe ist die Trifluormethylgruppe.
Die erfindungsgemäss zugänglichen Verbindungen sind pharmakologisch wirksam, insbesondere bei Herz- und Kreis laufkrankheiten. Sie bewirken eine Erweiterung der Coronar- gefässe und steigern die periphere und cerebrale Durchblu tung. Diese Wirkung wird in einigen Fällen von einem posi tiv inotropen Effekt begleitet.
Gemäss der Erfindung können die neuen Verbindungen dadurch hergestellt werden, dass man in. an sich bekannter Weise einen Alkohol der allgemeinen Formel bekannten und therapeutisch verwendbaren Säurereste in Frage.
Die erfindungsgemäss erhältlichen Verbindungen sind zur Herstellung pharmazeutischer Zusammensetzungen und Zu bereitungen geeignet. Die pharmazeutischen Zusammenset zungen bzw. Arzneimittel enthalten als Wirkstoff einen oder mehrere der erfindungsgemäss zugänglichen Verbindungen, gegebenenfalls in Mischung mit anderen pharmakologisch wirksamen Stoffen. Die Herstellung der Arzneimittel kann unter Verwendung der bekannten und üblichen pharmazeuti schen Trägermittel und Zuschläge erfolgen. Die Arzneimittel können enteral, parenteral, oral, perlingual oder in Form von Sprays angewendet werden.
Die Verabreichung kann in Form von Tabletten, Kap seln, Pillen, Dragees, Zäpfchen, Liquida oder Aerosolen er folgen. Als Liquida kommen z. B. in Frage: ölige oder wässri- ge Lösungen oder Suspensionen, Emulsionen, injizierbare wässrige oder ölige Lösungen oder Suspensionen.

In eine Aufschlämmung von 13 g (0,0275 Mol) d, i (1,1-Dithienyl-(2)-1-hydroxy-propyl-(3)]-[1-phenyl-propyl-(2)]- amin-maleat in 100 ml Eisessig wird bis zur vollständigen Lösung Chlorwasserstoff eingeleitet.

Aus 7,2 g (0,3 Mol) Magnesium und 49 g (0,3 Mol) 2- Bromthiophen wird in 100 ml absolutem Äther die Grignard- verbindung bereitet.
Zu dieser Lösung werden 17 g (0,05 Mol) 2-ss-[1-Phenyl-l-hydroxypropyl-(2)-methylamino]-propiothie- non-(3)-HCl (F. 145 C unter Sintern; hergestellt durch Ko chen von 3-Acetyl-thiophen, Paraformaldehyd und ±-Ephe- drin-HC1 in Isopropano1), in 100 ml absolutem Benzol aufge schlämmt, gegeben und 3 Stunden unter Rückfluss gekocht. Nach dem Zersetzen mit Eis und 50 g Ammoniumchlorid wird die organische Phase abgetrennt, mit Kaliumcarbonat getrocknet und das Lösungsmittel abdestilliert. Die zurück bleibende Base wird in Äther gelöst und mit isopropanolischer HCl angesäuert.
Das sirupös anfallende HCl-Salz wird in 50 ml Eisessig gelöst, 15 Minuten mit HCl-Gas behandelt und das Lösungsmittel abdestilliert. Der Rückstand wird mit 25%igem Ammoniak behandelt, die Base in Äther gelöst und mit isopropanolischer HCl neutralisiert. Das HCl-Salz wird aus Isopropanol umkristallisiert. Ausbeute: 3g. Die Base wird in Äther gelöst und mit isopropanolischer HCl in das HCl-Salz übergeführt. Das sirupös anfallende Salz wird mit Aceton zur Kristallisation gebracht und aus Isopropanol umkristallisiert.
Ausbeute: 8 g. F. 200-201 C. Beispiel 6 d,±-[1-Thienyl-(2)-1-thienyl-(3)-propen-(1)-yl-(3)]-[1-phe- iyl-l-hydroxy-propyl-(2)]-amin

Das HCl-Salz wird aus Isopropanol umkristallisiert. Ausbeute 12 g. F. 206 C.

Der Rückstand wird in 100 ml Chloroform gelöst, 30 Minuten mit HCl-Gas be handelt, nach dem Entfernen des Lösungsmittels mit 10%iger Natronlauge alkalisiert, die Base in Äther gelöst und mit iso- propanolischer HCl neutralisiert. Das HCl-Salz wird aus Äthanol umkristallisiert. Ausbeute 4 g. F. 184 C. organische Phase abgetrennt, mit Kaliumcarbonat getrocknet und das Lösungsmittel abdestilliert. Der Rückstand wird in 100 ml Chloroform gelöst, 30 Minuten mit HCl-Gas behan delt, nach dem Entfernen des Lösungsmittels mit l0%iger Natronlauge alkalisiert, die Base in Äther gelöst und mit iso- propanolischer HCl neutralisiert.
Das HCl-Salz wird aus Iso- propanol umkristallisiert. Ausbeute 8 g. F. 197 C. Phase abgetrennt, mit Kaliumcarbonat getrocknet und das Lösungsmittel abdestilliert. Der Rückstand wird in 100 ml Chloroform gelöst, 30 Minuten mit HCl-Gas behandelt, nach dem Entferhen des Lösungsmittels mit 10%iger Na tronlauge alkalisiert, die Base in Äther gelöst und mit iso- propanolischer HCl neutralisiert. Das HCl-Salz wird aus Iso- propanol umkristallisiert. Ausbeute 6 g. F. 180-182 C.
Beispiel 14 d,$-[l,l-Dithienyl-(3)-propen-(1)-yl-(3)]-[1-(4-methyl-phenyl)-1-hydroxy-propyl-(2)]-amin

Das zurückbleibende rohe d, ±-[1,1-Di thienyl-(3)-1-hydroxy-propyl-(3)]-[1-(4-methyl-phenyl)-1-hy- droxy-propyl-(2)]-amin wird in 100 ml Chloroform gelöst und 30 Minuten HCl-Gas eingeleitet.
Nach dem Entfenen des Lösungsmittels wird der Rückstand mit 10%iger Natronlauge versetzt, die Base in Äther gelöst und mit isopropanolischer HCl neutralisiert. Das Hydrochlorid der Titelsubstanz wird aus Isopropanol umkristallisiert. F. 183-184 C. Ausbeute 6 g. destilliert. Das zurückbleibende rohe d,±-[l,l-Dithienyl-(3) 1-hydroxy-propyl-(3)]-[2-(3-trifluormethyl-phenyl)-2-hydroxy- äthyl]-amin wird in 75 ml Chloroform gelöst und 30 Minuten HCl-Gas eingeleitet. Nach dem Entfernen des Lösungsmittels wird der Rückstand mit l0%iger Natronlauge versetzt, die Base in Äther gelöst und mit isopropanolischer HCl neutra lisiert. Das Hydrochlorid der Titelsubstanz wird aus Iso- propanol umkristallisiert. F. 1811 C.
Ausbeute 2,5 g. niumchloridlösung wird die organische Phase abgetrennt, mit Kaliumcarbonat getrocknet und das Lösungsmittel abdestil liert. Die zurückbleibende Base, die neben der Titelverbin dung Q-[l,l-Dithienyl-(2)-1-hydroxy-propyl-(3)]-[1-phenyl-l- hydroxy-propyl-(2)]-amin enthält, wird zur vollständigen De- hydratisierung in 100 ml Äthanol und 10 ml 7-n isopropano- lischer HCI 15 Minuten gekocht. Das HCl-Salz wird nach Ab- destillieren des Lösungsmittels mit Essigester zur Kristallisa tion gebracht und aus Isopropanol umkristallisiert.
Ausbeute 4 g. F. 189-190<B>1</B> C.
Das ±-ss-[1-Phenyl-l-hydrocy-propyl-(2)-amino]-propion- säurechlorid-HCl wird durch Umsetzung von #-ss-[1-Phenyl- 1-hydroxy-propyl-(2)-amino]-propionsäure-HCl bei Raumtem peratur in Chloroform mit der berechneten Menge Phosphor- pentachlorid in Gegenwart von wenig Dimethylformamid ge wonnen und als Rohprodukt (F. 152 C) bei der obigen Re aktion eingesetzt.
