Die neuen Verbindungen zeichnen sich gegenüber dem strukturell ähnlichen und bekannten 2-Benzolsulfonamido-5-methoxy-äthoxypyrimidin durch eine um ein Vielfaches gesteigerte blutzuckersenkende Wirkung bei oraler Applikation aus bei etwa gleicher Toxizi- tät. Die Tabelle 1 zeigt die starke Wirksamkeit der neuen Verbindungen an Hand einiger ausgewählter Beispiele. Die Vergleichssubstanz (2-Benzolsulfonamido-5-methoxy-äthoxypyrimidin) ist bei einer angewandten Dosis von 2 mg/kg unwirksam. Die neuen Verbindungen zeigen zum Teil sogar noch gute Wirk samkeit bei einer Dosis von nur 0,5 mg/kg.
Die neuen Verbindungen zeigen auch beim Menschen eine ausgezeichnete blutzuckersenkende Wirkung nach oraler Applikation, wie die Tabelle 2 an Hand zweier Beispiele zeigt. Besonders wirksam sind diejenigen Verbindungen der allge meinen Formel I, worin W eine direkte C-C-Bindung oder ein Sauerstoffatom, X eine direkte C-C-Bindung, Y eine CHz- kende Wirkung aufweisen.
Die erfindungsgemäss herstellbaren Sulfonamide besitzen die folgende Formel Z Wasserstoff oder eine niedere Alkylgruppe, R einen Kohlenwasserstoffrest mit 1 bis 8 C-Atomen, der auch ringgeschlossen und/oder durch ein oder mehrere Sauer stoffatome unterbrochen sein kann, oder einen durch Halogen, Alkoxy oder Alkylmercapto substituierten Phenylkern und A einen mono- oder bicyclischen Arylrest, einen heterocy- clischen Kern oder einen araliphatischen Rest bedeuten,
sowie deren Salze mit physiologisch verträglichen Basen.
Das erfindungsgemässe Verfahren ist dadurch gekennzeich net, dass man eine Verbindung der Formel Gruppe, Z ein Wasserstoffatom oder eine Methylgruppe, A einen Benzolkern, R, R', R Wasserstoffatome, Chloratome, Alkyl- oder Alkoxygruppen mit 1-6 C-Atomen und Ri eine Alkylgruppe mit 1-6 C-Atomen oder eine Phenylgruppe bedeuten.
Die neuen Substanzen sollen vorzugsweise in Dosen von etwa 0,01-1 mg/kg verabfolgt werden.
Zum therapeutischen Gebrauch können die erfindungsge- mäss erhältlichen Substanzen verabreicht werden als freie Sulfonamide, als Salze mit physiologisch vertretbaren anorga nischen und/oder organischen Basen wie z. B. Natrium-, Lithium-, Calcium-, Ammoniumhydroxyd, Aminen wie Methylglukamin, Morpholin, Äthanolamin u. a., oder auch in Form von Mischungen der freien Sulfonamide mit einem geeigneten Alkalihydrogenkarbonat bzw. Karbonat.
Besonders geeignet sind Basen, die selbst blutzuckersenkend wirken, wie z. B. Butylbiguanid. Die Konfektionierung der Substanzen kann ohne oder mit den in der galenischen Pharmazie üblichen Zusätzen, Trägersubstanzen, Geschmackskorrigenzien u. ä. erfolgen, und zwar beispielsweise in Pulverform, als Tabletten, Dragees, Kapseln, Pillen, in Form von Suspensionen oder Lösungen.
