Verfahren zur Herstellung von Enzymtrockenpräparaten
Es ist bekannt, dass Eiweissstoffe aus wässrigen Medien mit Tannin in Form von wasserunlöslichen Verbindungen ausgefällt werden können. Nach einem bekannten Verfahren werden aus den wasserlöslichen Tannaten anorganische Salze mit Wasser entfernt und anschliessend die Enzyme durch Behandeln der Tannate mit Mischungen aus Wasser und organischen Lösungsmitteln extrahiert. Oemäss einem anderen Verfahren wird über die Entfernung der Amyloglucosidase mit Hilfe von Lignin und/oder Gerbsäure von solchen Enzymen berichtet, die die Stärkekonvertierung behindern.
Es wurde nun gefunden, dass Enzyme aus wässri- gen Medien, beispielsweise aus mikrobiellen Kultur- lösungen, mit synthetischen Gerbstoffen in vortelhafter Weise gefällt werden können.
Bei den für das erfindungsgemässe Verfahren zur Fällung der Enzyme verwendeten synthetischen Gerbstoffen handelt es sich vorzugsweise um Kondensationsprodukte von ein- und mehrwertigen Phenolen, Phenolcarbonsäuren, Phenolsulfonsäuren sowie Phenolsulfen- amiden, Ligninsulfonsäuren und aromatischen Aminen, die im Molekül Kohlenstoff-, Sulfon-, Sulfoxy-, Sulfonimidbrücken enthalten.

Gegenstand der Erfindung ist die Verwendung von anionischen und kationischen synthetischen Gerbstoffen zur Fällung von Enzymen aus mikrobiellen Kulturlösungen oder anderweitigen pflanzlichen oder tierischen Extraktlösungen als Enzym-Gerbstoff-Addukte.
Während die Fällung von Eiweissstoffen bisher mit Tannin, Lignin oder natürlichen Gerbsäuren erfolgte, werden hier synthetische Gerbstoffe verwendet, die gleiche Fällungsergebnisse bei wesentlich geringeren
Kosten gestatten.
Das erfindungsgemässe Verfahren ist dadurch ge kennzeichnet, dass Enzyme mittels anionischer, kationi scher oder amphoterer synthetischer Gerbstoffe aus mikrobiellen Kulturlösungen oder -filtraten sowie pflanzlichen oder tierischen Extrakten als Enzym-Gerb stoffaddukte ausgefällt und aus dem erhaltenen Niederschlag anorganische Salze und Gerbstoffe mit Wasser bzw. mit Gemischen aus Wasser und organischen Lösungsmitteln entfernt werden und der gereinigte Niederschlag getrocknet wird.
Vorzugsweise werden die Kulturlösungen mit 0,5 bis
5 O/o und mehr synthetischen Gerbstoffen versetzt. Die Fällung kann in der Wärme (bis 50 C), bei Zimmertemperatur oder unter Kühlung vorgenommen werden.
Das unter Rühren ausgefällte Eiweiss-Gerbstoffaddukt kann durch Zentrifugieren, Filtrieren oder Dekantieren vom Überstand abgetrennt und anschliessend entweder mit Wasser oder mit organischen Lösungsmitteln versetzt werden. Vermittels des Wassers werden überschüssige Gerbstoffgaben sowie anorganische Salze aufgenommen, und durch die Verwendung organischer Lösungsmittel werden weitere Salze und die gebundenen Gerbstoffe aus dem Fällungsprodukt entfernt. Als organische Lösungsmittel bewährten sich ein- und mehrwertige Alkohole sowie mit Wasser mischbare Ketone, Glycoläther, Dioxan und Tetrahydrofuran. Um einen Aktivitätsabfall der enzytuhaltigen Fällungsprodukte zu vermeiden, hat die Extraktion der synthetischen Gerbstoffe in der Kälte bevorzugt zwischen ODC und + 10 C zu erfolgen.
Das erhaltene Fällungsprodukt kann je nach den Reinheitsforderungen entweder erneut mit organischen Lösungsmitteln gewaschen oder sofort in üblicher Weise getrocknet werden.
Beispiel 1
Für die Fällung von Amylase wird das Kulturfiltrat vom Bacillus subtilis benutzt. 1,5 1 eines amylasehaltigen Kulturfiltrates werden mit Natriumcarbonatlösung auf pH 7,0 eingestellt. Danach erfolgt unter Rühren die Zugabe einer Lösung eines synthetischen Gerbstoffes vom Typ der Polykondensationsprodukte von einwertigen Phenolen mit Kohlenstoffbrücken (z. B. Skytan).
Die Gerbstoffkonzentration in der Mischung beträgt 2 /1,. Der Niederschlag wird abgetrennt und zweimal mit Aceton bei 0 C bis +5 OC gewaschen. Beim Trocknen entstehen 3,70g Trockenprodukt. Die Gesamtaktivität im Kulturfiltrat beträgt 2 140 500 Amylaseeinheiten (IE nach IUB), die Gesamtaktivität des Trockenproduktes 1 765 000 Amylaseeinheiten. Daraus errechnet sich eine Aktivitätsausbeute von 82,5 010.
Beispiel 2
Für die Fällung von Protease wird das Kulturfiltrat vom Bacillus subtilis benutzt. Zu 1000 ml Filtrat mit einer Gesamtaktivität von 4780 Proteaseeinheiten (PU(. ,s) wird bei pH 7,0 eine Lösung eines synthetischen Gerbstoffes gegeben, dessen pH-Wert ebenfalls auf pH 7,0 eingestellt ist. Der Gerbstoff entspricht dem Typ der Polykondensationsprodukte von mehrwertigen Phenolen und aromatischen Sulfonsäuren (z. B. Skytan). Nach zweistündigem Rühren bei 30 C wird vom Niederschlag abgetrennt und der Niederschlag zweimal mit Aceton extrahiert und anschliessend getrocknet. Es wird 2,20 g Trockenprodukt mit einer Gesamtaktivität von 3771 Proteaseeinheiten erhalten, die Aktivitätsausbeute beträgt damit 78,9 /o.
