Verfahren zur Herstellung von therapeutisch wirksamen Hydroxycyclohexancarbonsäureestern Die Erfindung betrifft ein Verfahren zur Herstellung einer Reihe therapeutisch wirksamer Hydroxycyclohexancarbon- säureestern sowie deren Salzen.

Die neuen Verbindungen werden durch die folgende allge meine Formel dargestellt:

darstellt. Auch die Salze dieser basischen Ester mit einigen anor ganischen und organischen Säuren (HCl, H2SO4, Zitronen säure, Malonsäure, Weinsäure) können erhalten werden. Ferner dienen diese Ester zur Herstellung entsprechender quaternärer Verbindungen mit Alkylhalogeniden, wie CH3Br, C2H5Br, C4H9Br, C8H17Br und Methylparatoluolsulfonat.
Die erfindungsgemäss hergestellten Verbindungen, welche antispastische Wirkung aufweisen, können mit anderen Stof fen, wie z. B. inerten Füllstoffen, beispielsweise Milchzucker, Zucker, mikrokristaller Gelatine, Talkum, Magnesiumstearat, Sorbitol, Stärke oder mit anderen Hilfsmitteln und/oder zu sätzlichen Wirkstoffen, zu pharmazeutischen Präparaten ver arbeitet werden.
Das Verfahren gemäss der Erfindung ist dadurch gekenn zeichnet, dass man ein Alkalisalz einer 2-R-2-Hydrocyclo- hexancarbonsäure, worin R=Phenyl oder Cyclohexyl, mit einer Verbindung der Formel Hal-R' verestert, in welcher Hal=Halogen und R' einer der oben angegebenen Reste be deutet. Diese Ester werden zur Herstellung entsprechender quaternisierter Verbindungen verwendet.
Die Ausgangsverbindung lässt sich herstellen, indem man 2 Hydroxymethylcyclohexanon mit einer Grignardverbindung der Formel R-Mg-Hal umsetzt, in welcher R dieselbe Be deutung hat wie in Formel I und Hal=Halogen bedeutet,

das erhaltene 2-R 2-Hydroxycyclohexylmethanol oxydiert und die erhaltene Säure in ihr Alkalisalz überführt.
Die antispastische Wirkung der Ester und Salze wurde auf Segmenten verschiedener gesonderter Organe bestimmt, und es konnte beobachtet werden, dass der grösste Teil der getesteten Verbindungen die Kontraktion verschiedener Krampferzeuger, sowohl hormonaler wie gemütsmässiger oder sonstiger Herkunft herabsetzt oder verhindert, wobei diese Wirkung bereits bei Konzentrationen von 1 x 10-5 und 1 x 10' g/ml auftritt.
Die antispastische Wirkung der erfindungsgemäss erhal tenen Verbindungen ist deutlicher in der folgenden Tabelle dargestellt, in welcher als Konzentrationsangaben die ED50 angeführt sind, welche die Zuckungen isolierter Organe (Endkrummdarm eines Meerschweinchens, aufsteigendes Kolon der Ratte, Leerdarm der Ratte, Uterus der Ratte, hervorgerufen durch einige zuckungserzeugende Wirkstoffe, d.h.
Histamin (Rist.), 5-Hydroxy-triptamin (5 Ht), Acetyl- cholin und BaC12; in der Tabelle sind ausser den erfindungs- gemäss hergestellten Verbindungen auch einige der im Han del befindlichen antispatischen Mittel angegeben, und die entsprechenden Werte des ED5O zeigen, dass die neuen Ver bindungen vorteilhaft diesen letzteren gleichgesetzt werden können.

Die neuen Verbindungen weisen keine hypotensive Wir kungen auf und sind besonders zur Behandlung von Spasmen in den Gallenwegen und im Darm angebracht, weil sie auf dem Gallenwege ausgeschieden werden, und wegen ihrer ausgesprochenen antiacetylcholinischen Wirkung, welche in der gleichen Grössenordnung wie diejenige des Atropins ein tritt.
Aus diesen Gründen bilden die neuen Verbindungen nützliche Heilmittel zur Behandlung zahlreicher krankhafter Zustände der Gallenwege und der Leber sowie zur Behand lung spastischer Zustände der Verdauungsorgane, der Gallen wege, der Urinwege und der Geschlechtsorgane.

bei welchen Leiden ein Verschwinden der Beschwerden schon nach den ersten Tagen der Verab reichung festzustellen war, während eine merkliche Besserung der Verdauungsfunktion und der Leber-Gallen-Tätigkeit zu verzeichnen ist. Nicht konstante Ergebnisse wurden bei uro logischen und gynäkologischen Beschwerden erhalten. Ein wandfrei ist die lokale und allgemeine Toleranz in Abwesen heit von Nebenwirkungen und atropinartigen Erscheinungen. Dieses Erzeugnis kristallisiert durch Lösung in Äther und Fällung mit Petroläther, unter Erhalt von 7 g 1-Phenyl-2- hydroxymethylcyclohexanol. Schmelzpunkt (Koffer) 81 bis 83 C.
Der so erhaltene Alkohol wird nach Trocknung und feiner Zerpulverung in 1,4 1 einer wässrigen Lösung aus 14 g KMn04 und 7 g Ka2CO3 suspendiert und die Suspension 1 Tag lang heftig gerührt.
Alsdann wird das entstandene Mn02 gefiltert, und dem Filtrat wird etwas Na2S03 bis zum Verschwinden der violet ten Färbung zugesetzt; alsdann wird erneut das Mn02 abfil- triert und die alkalische Lösung mit konz. HCl angesäuert.

B. Herstellung von weinsaurem 2-Phenyl-2-hydroxycyclo- hexancarboxylat des N-Äthyl-3-piperidinols Aus der sich ergebenden Lösung scheidet sich ein Öl ab, welches nach wiederholtem Waschen mit Äther farblose hygroskopische Kristalle ergibt (Schmelzpunkt 65-70'C). Waschen mit Äther und Kristallisieren aus Isopropanol die Form hellgelber Kristalle mit Schmelzpunkt 143-5'C auf weist.
