Verfahren zur Herstellung von 5α-Brom-6ss-hydroxy-steroiden 5α-Brom-6ss-hydroxy-steroide sind wertvolle Zwi schenprodukte für die Herstellung wichtiger pharmoko logischer Verbindungen, wie die 19-nor-Steroide. Die erfindungsgemäss hergestellten 5α-Brom-6ss-hydroxy-ste- roide können nach bekannten Verfahren, wie sie z. B. in den USA-Patentschriften Nrn. 3 033 862, 3 206 460 und 3 235 574 beschrieben sind, in wertvolle 19-nor- Steroide umgewandelt werden.
Bisher wurden 5α-Brom-6ss-hydroxy-steroide herge stellt, indem man ein ^5-Steroid mit unterbromiger Säure oder einem unterbromige Säure liefernden Mittel in Anwesenheit einer starken anorganischen Säure als Ka talysator, wie Perchlorsäure, Schwefelsäure usw., in einem wässrigen organischen Lösungsmittelmedium, wie z. B. wässriges Tetrahydrofuran, Aceton, Dioxan, alipha tische Äther, Äthylacetat, tertiäre Alkohole usw., um setzte.
Die bekannten Verfahren waren schwer zu hand haben oder durchzuführen und lieferten ein Produkt, das erhebliche Mengen von schwierig zu entfernenden Nebenprodukten enthielt. Weiterhin. lieferten die be kannten Verfahren eine niedrige Ausbeute an reinem 5α-Brom-6ss-hydroxy-steroid, das heisst etwa 60-65 %.
Hauptziel der vorliegenden Erfindung ist die Schaf fung eines neuen Verfahrens zur Herstellung von 5a Brom-6ss-hydroxy-steroiden und Zwischenprodukten da für, das die oben erwähnten Nachteile nicht aufweist.
Das erfindungsgemässe Verfahren zur Herstellung von 5α-Brom-6ss-hydroxy-steroiden kann weiterhin bei Zimmertemperatur durchgeführt werden, ist einfach in der Handhabung, liefert eine hohe Ausbeute des ge wünschten 5α-Brom-6ss-hydroxy-steroids und ergibt ein Minimum an Nebenprodukten.
Ausserdem können aus den entstandenen und in den Mutterlaugen enthaltenen Nebenprodukten wieder leicht die 5ss,6ss-Oxido-Zwischenprodukte zwecks Rückfüh rung in das Verfahrens zurückgebildet werden.
Die Herstellung der Ausgangsstoffe sowie die Bil dung der 5α-Brom-6ss-hydroxy-steroide kann ausgehend von Verbindungen der Formel I bzw. II wie folgt dar- gestellt werden, wobei nur die Ringe A und B des Steroidkernes dargestellt sind:

In den obigen Teilformeln steht X für Chlor oder Brom; R steht z. B. für Wasserstoff, Halogen, z. B. Chlor, eine Hydroxylgruppe oder eine Acyloxygruppe mit weniger als 12 Kohlenstoffatomen, vorzugsweise eine Acylaxygruppe, und R' steht für eine Acylgruppe mit weniger als 12 Kohlenstoffatomen. Typische Ester gruppen umfassen Acetat, Propionat, Formfiat, Butyrat, Benzoat, Cyclopentylpropionat usw.
Ist in der 3-Stellung eine Estergruppe anwesend, so kann diese gleich oder von der Estergruppe in der 6-Stellung verschieden sein (II; R' = Acyl).
Bei der Durchführung des erfindungsgemässen Ver fahrens kann man in der Weise vorgehen, dass man erst ein ^5-Steroid, mit 1-10molaren Äquivalenten, vorzugs weise etwa 1-3molaren Äquivalenten eines, positives Brom oder Chlor freisetzenden Mittels in einem Kohlen- wasserstoffcarbonsäurereaktionsmediums behandelt.
Die Reaktion kann bei einer Temperatur zwischen etwa 10 C (oder etwas niedriger) bis zu etwa 40 C oder mehr, vorzugsweise bei Zimmertemperatur (etwa 20 bis <B>25'</B> C) für etwa 15 Minuten bis etwa 2 Stunden oder mehr durchgeführt werden. Es können höhere Tempera turen angewendet werden, was jedoch im allgemeinen keinen Vorteil mit sich bringt.
Gewöhnlich sollten höhere Temperaturen vermieden werden, um die mögliche Bil- dung unerwünschter Nebenprodukte zu verringern. Die zweckmässigste Temperatur und Reaktionszeit sind durch übliche Versuche leicht zu bestimmen, wobei das ver wendete ^5-Steroid und die Konzentration der Reak- tionsteilnehmer in Betracht gezogen werden sollen.
Diese Reaktion liefert eine Ausbeute von etwa 80-909ö des gewünschten Ausgangsstoffs, nämlich des 5α-Halogen- 6ss-acyloxy-steroids (II), das leicht aus der Reaktions mischung gewonnen oder abgetrennt werden kann.
Die während der obigen Reaktion gebildeten Neben produkte, die hauptsächlich aus dem entsprechenden 5α,6ss-Dibrom- oder 5α,6ss-Dichlor-steroid und dem 5a- Acyloxy-6ss-brom- oder -chlor-steroid bestehen, können durch Behandlung mit Zink in Anwesenheit einer nied rigen Kohlenwasserstoffcarbonsäure, wie Essigsäure, Propionsäure, Ameisensäure, Buttersäure usw., vorzugs weise Essigsäure, leicht in die ursprüngliche Ausgangs verbindung, das heisst das ^5-Steroid, umgewandelt wer den. Diese Rückbildung den ^5-Steroids erfolgt zweck mässig, indem man fein zerteiltes Zinkpulver und eine Carbonsäure, z. B.
Essigsäure, zu den Mutterlaugen zu fügt, die in der obigen Reaktion erhalten wurden. Auf diese Weise werden bis zu 13 3ö des ursprünglichen d5 Steroids zur Rückführung zurückgewonnen, was die durch diese Reaktion erzielbare Gesamtausbeute wirksam erhöht. Diese Rückgewinnung kann gegebenenfalls zur weiteren Erhöhung der Gewinnung an Ausgangsmaterial aus den in den Mutterlaugen enthaltenen Nebenproduk ten wiederholt werden. Das Verhältnis von Zink zu Mutterlauge sollte zwischen etwa 0,5:45 bis 4:45 Gewichtsteilen, vorzugsweise zwischen etwa 0,9 : 45 bis etwa 1,2 : 45, liegen.
Das Verhältnis von Carbonsäure zu Mutterlaugen sollte zwischen etwa 0,5 : 9 bis etwa 4:9, vorzugsweise ewa 0,8:9 bis etwa 1,5 :9, Ge wichtsteilen liegen.
Als positives Halogen freisetzendes Mittel kann unterbromige Säure, N,N-Dichlordimethylhydantoin, N,N-Dibromdimethylhydantoin und die N-Brom- und N-Chlorderivate von niedrigen aliphatischen Carbon- säureamiden oder -imiden verwendet werden, wie z. B. N-Bromacetamid, N-Chloracetamid, N-Bromsuccinimid, N-Chlorsuccinimid usw.
Das ^5-Steroid kann, wie gesagt, mit einem, positives Chlor oder Brom freisetzenden Mittel in einem Kohlen- wasserstoffcarbonsäurereaktionsmedium umgesetzt wer den. Als Carbonsäuremedium wird zweckmässig eine niedrige Monocarbonsäure, wie Ameisensäure, Essig säure oder Mischungen derselben, verwendet; es können jedoch auch andere Carbonsäuren mit weniger als 12 Kohlenstoffatomen verwendet werden, wie Propion- säure, Buttersäure, Pentansäure, Benzoesäure, Cyclo- pentylpropionsäure usw. Weiterhin kann auch eine Mi schung von Carbonsäuren, z. B. Ameisensäure und Essigsäure, verwendet werden.
Die Carbonsäure oder Mischung von Carbonsäuren ist vorzugsweise wasserfrei; es können jedoch auch praktisch wasserfreie, das heisst technisch reine Carbonsäuren, verwendet werden. Falls die verfügbare, als Reaktionsmedium gewünschte Car- bonsäure Wasser enthält, kann das Wasser vor Beginn der Reaktion entfernt werden, indem man eine entspre chende Menge eines einfachen oder gemischten Carbon- säureanhydrids, wie z. B.
Essigsäureanhydrid, Propion- säureanhydrid, Buttersäureanhydrid usw., zufügt, das unter Bildung der entsprechenden Säure oder Säuren mit Wasser reagiert und dadurch das Reaktionsmedium wasserfrei macht. Daher kann diese Reaktion eine Mi schung von 6ss-Estern der Ausgangssteroide oder einen einzigen solchen 6ss-Ester ergeben.
Die in der vorliegen den Beschreibung verwendete Bezeichnung Kohlen- wasserstoffcarbonsäurereaktionsmedium umfasst Car- bonsäuren und Mischungen derselben, die durch die Zugabe eines Carbonsäureanhydrids wasserfrei gemacht worden sind.
Das Reaktionsmedium kann gegebenenfalls mit einem organischen Lösungsmittel, vorzugsweise einem wasserfreien organischen Lösungsmittel, verdünnt wer den, das ein Lösungsmittel für das Steroid und/oder die Säure ist und gegenüber der Reaktion inert ist. Geeig nete organische Lösungsmittel umfassen die aroma tischen Kohlenwasserstoffe, wie Benzol, Toluol, Xylol usw., chlorierte Kohlenwasserstoffe, wie Methylenchlo- rid, Chloroform, Chlorbenzol oder Tetrachlorkohlen stoff, Mischungen chlorierter Kohlenwasserstoffe mit Alkanen, wie Pentan, Hexan, Heptan, z.
B. eine Mi schung aus Chloroform und Hexan, Äther, wie Diäthyl- äther, Dioxan oder Tetrahydrofuran, und andere, be kannte, organische Lösungsmittel, wie Aceton, Äthyl- acetat, Dimethylsulfoxyd usw. Die Menge des inerten organischen Lösungsmittels, vorzugsweise des wasser freien organischen Lösungsmittels, kann beträchtlich va riieren, sollte jedoch nicht mehr als etwa 60 Vol.% des gesamten Reaktionsmediums, vorzugsweise nicht mehr als<B>30%,</B> betragen.
Gegebenenfalls kann zur Katalyse der Reaktion eine geringe Menge einer starken Säure, wie Perchlorsäure, Schwefelsäure usw., in der Reaktionsmischung mitver wendet werden. Gewöhnlich sollte der Säurekatalysator nicht mehr als etwa 1 Vol.% des gesamten Reaktions gemisch übersteigen. Die Anwesenheit einer katalytischen Menge einer starken Säure ist im Hinblick auf das be reits saure Reaktionsmedium nicht entscheidend oder notwendig.
Die Konzentration der Reaktionsmischung kann be trächtlichen variiert werden. Aus praktischen oder wirt schaftlichen Gründen wird vorzugsweise die geringst mögliche Carbonsäuremenge verwendet. Im allgemeinen sollte die Steroidkonzentration zwischen etwa 2 bis 50 Gew.% der gesamten Reaktionsmischung, vorzugs weise 5-35 %, betragen. Das Steroidausgangsmaterial braucht nicht im Reaktionsmedium gelöst zu sein, da sich auch eine Suspension des Steroids in befriedigender Weise in der Reaktion verhält.
Die erste Stufe des erfindungsgemässen Verfahrens ist die Umwandlung des 5α-Halogen-, insbesondere des 5α-Brom- oder 5α-Chlor-6ss-acyloxy-steroids (II) in das Epoxyd, das heisst das 5ss,6ss-Oxido-steroid (11I). Diese Reaktion erfolgt durch Behandlung des 5a-Halogen-6ss- acyloxy-steroids (II) mit einer Base in Anwesenheit eines mit Wasser mischbaren organischen Lösungsmittels.
Ge- eignete, mit Wasser mischbare organische Lösungsmittel umfassen die niedrigen Alkohole, vorzugsweise einwer tige Alkohole, wie z. B. Methanol oder Athanol, sodann Äther, wie Tetrahydrofuran, Dioxan usw., Dimethyl- sulfoxyd, Dimethylformamid, Dimethylacetamid usw. Die Base kann zweckmässig ein Alkali- oder Erdalkali- rnetallhydroxyd, -carbonat, -bicarbonat oder -acetat, vorzugsweise eine Alkalimetallbase, wie Kalium- oder Natriumhydroxyd, oder die Bicarbonate, Carbonate oder Acetate derselben sein.
Die in dieser Reaktion ver wendete Menge an Bass, sollte zwischen etwa 2 bis 25molaren Äquivalenten pro Mol Steroid vorzugs weise zwischen etwa 2 bis 20molaren Äquivalenten liegen. Gegebenenfalls können auch grössere Mengen an Base verwendet werden, was gewöhnlich jedoch keine Vorteile mit sich bringt. Die in der Reak tionsmischung anwesende Menge an Steroidmaterial sollte zwischen etwa 2 bis. 50 Gew.%, vorzugsweise 20 Gew.%, bezogen auf die gesamte Reaktionsmischung, betragen. Die Reaktionstemperatur kann von etwa Zim mertemperatur bis zur Rückflusstemperatur des verwen deten Lösungsmittels liegen.
Diese Stufe ergibt eine prak tisch quantitative Ausbeute der gewünschten 5ss,6ss- Oxidoverbindung (III).
Ist bei der Durchführung der ersten Stufe in der 3-Stellung eine Estergruppe anwesend (Il; R=Acyloxy), so wird die Estergruppe aufgrund der basischen Reak tionsbedingungen in eine Hydroxylgruppe umgewandelt. Das erhaltene 3ss-Hydroxy-5ss,6ss-oxido-steroid kann in der nächsten Stufe des Verfahrens verwendet werden; vor der Durchführung der nächsten Stufe wird die Hy droxylgruppe jedoch zweckmässig in eine Estergruppe umgewandelt. Diese Umwandlung oder Veresterung er folgt in üblicher Weise durch Behandlung des 3ss-Hy- droxy-steroids mit einem Acylhalogenid, vorzugsweise dem Chlorid, z. B. Benzoylchlorid, oder einem Anhydrid einer Kohlenwasserstoffcarbonsäure mit weniger als 12 Kohlenstoffatomen, z. -B.
Essig-, Propion-, Butter- oder Capronsäureanhydrid usw., in Anwesenheit von Pyridin, Collidin, Lutidin usw. Es können Mischungen der obigen Lösungsmittel allein oder zusammen mit einem oder mehreren anderen inerten, organischen Lö sungsmitteln, wie Benzol, Toluol, Xylol usw., verwen det werden. Das Acylhalogenid oder Carbonsäure- anhydrid sollte in einer Menge von mindestens einem molaren Äquivalent pro Mol des Steroids, vorzugsweise in einem leichten Überschuss anwesend sein. Diese Reaktion erfolgt vorzugsweise in Pyridin unter Verwen dung von Essigsäureanhydrid.
Die Reaktion kann bei einer Temperatur zwischen Zimmertemperatur oder dar unter bis zur Rückflusstemperatur durchgeführt werden; die Reaktionszeit kann etwa 30 Minuten bis etwa 15 Stunden oder mehr betragen. Die Konzentration des in der Reaktionsmischung anwesenden Steroids kann beträchtlich variiert werden. Gewöhnlich wird eine Kon zentration von 2 bis 50 Gew.%, vorzugsweise etwa 20 Gew.%, der gesamten Reaktionsmischung verwendet.
In der nächsten Stufe des erfindungsgemässen Ver fahrens wird das 5ss,6ss-Oxido-steroid (III) einer Hydro- bromierung unterworfen. Diese Stufe erfolgt durch Be handlung des Steroids mit Bromwasserstoff in einem inerten organischen Lösungsmittelmedium zur Umwand lung des Epoxyds in das gewünschte 5α-Brom-6ss- hydroxyd-steroid (IV). Diese Reaktion wird in der Regel in der Weise durchgeführt, indem man das Steroid in einem organischen Lösungsmittel, das gegenüber trockenem Bromwasserstoff praktisch inert ist, vorzugs weise einem praktisch nichtpolaren organischen Lösungs mittel oder einer Lösungsmittelmischung, z.
B. einem aromatischen Lösungsmittel, wie Benzol, Toluol oder Xylol, einem chlorierten Kohlenwasserstoff, wie Methy- lenchlorid, Chloroform, Tetrachlorkohlenstoff oder Chlorbenzol, Mischungen chlorierter Kohlenwasserstoffe mit Alkanen, wie Pentan, Hexan oder Heptan, z. B. eine Mischung aus Chloroform und Hexan, einem Äther, wie Diäthyläther, Dioxan oder Tetrahydrofuran usw., sowie Mischungen der obigen Lösungsmittel, wie Toluol und Hexan, löst und die Lösung mit einem molaren Äqui valent oder mehr wasserfreiem Bromwasserstoff behan delt.
Der Bromwasserstoff kann als Lösung in einem oder mehreren der oben genannten Lösungsmittel zuge fügt werden; er wird vorzugsweise jedoch in gasförmi- ger Form in das Reaktionssystem oder -medium einge führt. Gewöhnlich wird eine Reaktionstemperatur zwi schen etwa -10 C oder weniger bis etwa 30 C, vor- zugsweise zwischen etwa<B>15-25'</B> C, angewendet; gege- benenfalls können jedoch auch höhere und niedrigere Temperaturen verwendet werden, wobei die Reaktions geschwindigkeit entsprechend beeinflusst wird.
Die Re aktion sollte fortgesetzt werden, bis keine weitere Brom wasserstoffaufnahme mehr erfolgt. Die Durchführung der Reaktion bei einer Temperatur von etwa 15-25 C und die Einführung von trockenem Bromwasserstoff bei einer Geschwindigkeit von etwa 0,1 bis 1;0 Liter/Min. oder mehr erfordert gewöhnlich eine Reaktionszeit zwi schen 10-60 Minuten für einen Ansatz von 100 g Steroid. Die Steroidkonzentration in der Reaktions mischung liegt zweckmässig zwischen etwa 2-50 Gew.%, vorzugsweise zwischen etwa 5-20 Gew.%, der gesamten Reaktionsmischung.
Die obige Hydrobromierung verläuft mit ausgezeich neter Ausbeute, das heisst etwa 90-95 A des gewünschten 5α-Brom-6ss-hydroxy-steroids (IV), das, z. B. durch Fil trieren, leicht gewonnen werden kann. Die Nebenpro dukte dieser Reaktion, die hauptsächlich das entspre chende 5ss-Hydroxy-6α-brom-steroid umfassen, verblei ben in Lösung; durch Behandlung der Lösung (Mutter lauge aus der Hydrobromierung) mit einer Base, vor zugsweise einer starken Base, wie Natrium- oder Ka liumhydroxyd, in Anwesenheit eines mit Wasser misch baren, organischen Lösungsmittels, z. B. Methanol, Äthanol usw., werden die Nebenprodukte zwecks Rück führung in das Verfahren in das 5ss,6ss-Oxido-steroid (III) zurückverwandelt.
Die Umwandlung der Neben produkte erfolgt zweckmässig, indem man die aus der Hydrobromierungsstufe erhaltenen Mutterlaugen konzen triert, dem Konzentrat eine stark basische Lösung, z. B. eine 5-0%ige Natriumhydroxydlösung und ein mit Was ser mischbares, organisches Lösungsmittel, wie Metha nol, zufügt und für die Dauer von etwa 30 Minuten bis 15 Stunden oder mehr auf eine Temperatur oberhalb Zimmertemperatur erhitzt. Es kann eine Temperatur zwischen etwa Zimmertemperatur bis zur Rückflusstem peratur des verwendeten Lösungsmittels angewendet werden.
Die Menge der verwendeten, starken Base in bezug auf die Mutterlauge aus der Hydrobromierung sollte zwischen etwa 0,5 : 150 bis etwa 4:150 Gewichts teilen, vorzugsweise etwa 1 : 150, liegen. Im allgemei nen sollte die Menge des mit Wasser mischbaren, orga nischen Lösungsmittels in bezug auf die Mutterlauge aus der Hydrobromierung vor der Konzentration dieser Mutterlauge zwischen etwa 0,5 : 20 bis etwa 4 : 20 Ge wichtsteilen betragen.
Zur Herstellung der Ausgangsmaterialien kommen monoungesättigte, nicht konjugierte d5-Steroide mit der äthylenisch ungesättigten Bindung zwischen dem 5-Koh- lenstoff und 6-Kohlenstoff in Betracht. Diese 45-Steroide können z. B. in 3-Stellung Acyloxygruppen aufweisen; z. B. sind die 3-Acylate vorzugsweise die 3-Acetate von ^5-Steroiden der Androstan-, Pregnan-, Sapogenin-, Alkaloid- und Cholestan-Reihe. Typische 3ss-Acyloxy- ^5-steriode sind z. B.
3ss-Acetoxy-androst-5-en-17-on, 3ss,17ss-Dihydroxy-androst-5-en-3-acetat-17-benzoat, 3ss,17ss-Diacetoxy-androst-5-en, 3ss-Acetoxy-pregn-5-en-20-on, 3ss-Acetoxy-cholest-5-en, die Diacetate und Dibenzoate von Cholest-5-en-3ss,17ss- diol und Cholest-5-en-3ss,7ss-diol, 3ss-Acetoxy-^5-bis- nor-cholensäure, das Acylat, vorzugsweise das Acetat von Campesterol, ss-Sitosterol, Clionasteriol, α-Sitosterol, und die Acylate, vorzugsweise das Acetat, von ^5-Sapo- geninen, z. B.
Diosgenin, Yamogenin, Ruscogenin, Boto- genin usw., und von ^5-Alkaloiden, wie Solanidin, Sola- sodin, Rubijervin, Isorubijervin usw. Andere, als erfin dungsgemässe Ausgangsmaterialien geeignete ^5-Steroide umfassen die A5-, 3-Halogen-^5- und 3-Hydroxy-^5- steroide, wie Androst-5-en-17-on, 3ss-Chlor-androst-5- en-17-on, 3ss-Hydroxy-androst-5-en-17-on, Pregn-5-en- 20-on usw.
Die folgenden Abschnitte betreffen die Ausgangs- stoffherstellung. <I>Abschnitt 1</I> Zu 187 g Ameisensäure (90%ig) wurden 113 g Essig säureanhydrid zugegeben, wobei die Temperatur etwa 10 Minuten auf etwa 55-65 C gehalten wurde. Dann wurde die Mischung auf etwa 20 C abgekühlt, und unter Rühren wurden 100 g 3ss-Acetoxy-androst-5-en-17-on zugefügt.
Anschliessend wurden 59g N-Bromsuccinimid zugegeben, wobei die Temperatur mittels eines Eisbades auf etwa<B>20-25'</B> C gehalten wurde. Dann wurde die erhaltene Mischung eine Stunde gerührt, wobei die Tem peratur auf<B>20-25'</B> C gehalten wurde. Anschliessend wurden etwa 10 g Natriumbisulfit in 50 cm3 Wasser zugefügt und die Reaktionsmischung wurde in etwa 850 cm3 Wasser gegossen, gerührt und filtriert; so wurde eine Mischung des 6ss-Acetates und 6ss-Formiates von 5α-Brom-3ss,6ss-dihydroxy-androstan-17-on-3-acetat er halten, die wie folgt gereinigt wurde.
Die rohen ge mischten Ester wurden mit Wasser gewaschen und dann in 700 cm3 Methylenchlorid gelöst; die wässrige Schicht wurde abgetrennt und mit Methylenchlorid extrahiert. Die organischen Phasen wurden dann vereinigt und bei atmosphärischem Druck konzentriert. Der Rückstand wurde in 600 cm3 Methanol aufgenommen und die Lö sung unter vermindertem Druck destilliert, bis das Vo lumen auf 400 cm3 verringert war. Die erhaltene Lösung wurde auf etwa 15-20 C abgekühlt und dann filtriert.
Der Niederschlag wurde mit 150 cm3 Methanol ge waschen und lieferte eine reine Mischung des 6ss-Ace- tates und 6ss-Formiates des 5α-Brom-3ss,6ss-dihydroxy- androstan-17-on-3-acetates.
Durch Wiederholung der obigen Verfahrensweise unter Verwendung von N-Chlorsuccinimid anstelle von N-Bromsuccinimid wurden die entsprechenden 5a-Chlor- verbindungen erhalten.
Die aus der Reinigung der gemischten Ester aus der obigen Reaktion erhaltenen Mutterlaugen (vereinigtes Filtrat und Waschwasser), 5 g Zinkpulver und 50 g Essigsäure wurden auf etwa 85-90 C erhitzt. Dann wurden weitere 5 g Zinkpulver zugegeben und die Reak tionsmischung etwa eine Stunde auf einem Wasser- dampfbad (85-95 C) erhitzt. Der grösste Teil des rest lichen Lösunggmittels wurde dann abgedampft und 100 cm3 Benzol zugefügt. Die erhaltene Mischung wurde abgekühlt (auf etwa 20 C), und es wurden 100g Wasser zugefügt. Dann wurde die Mischung filtriert und das zurückgewonnene Zink wurde mit etwa 50 cm3 Benzol gewaschen. Die wässrige Phase wurde vom Filtrat abge trennt und verworfen.
Die verbleibende organische Phase wurde mit Wasser gewaschen, abgetrennt und auf ein kleines Volumen eingedampft. Das kleine Volumen des Konzentrates wurde dann in 100 cm3 Methanol auf genommen und die erhaltene Mischung auf ein kleines Volumen eingedampft. Das erhaltene Konzentrat wurde anschliessend in etwa 80 cm3 Methanol gelöst, die erhal tene Lösung bis zur Sättigung konzentriert, auf etwa 20 C gekühlt und filtriert. Das erhaltene feste Produkt wurde mit Methanol gewaschen und lieferte 3ss-Acetoxy- androst-5-en-17-on mit einem F. von 168,5-170 C.
Eine zweite Ausbeute des obigen ^5-Steroids wurde er halten, indem die Mutterlauge eingedampft, abgekühlt, filtriert und gewaschen wurde; dieses Ausbeute hatte einen F. von 164,5-167 C.
<I>Abschnitt 2</I> Eine Mischung aus 100 g 3ss,17ss-Diacetoxy-androst- 5-en, 60 g N-Bromsuccinimid und 300 cm3 wasserfreier Essigsäure wurde unter Rühren etwa 70 Minuten bei Zimmertemperatur<B>(20-25'</B> C) gehalten. Dann wurde die Reaktionsmischung in 1 Liter Wasser, das etwa 12g Natriumbisulfit enthielt, gegossen und die erhaltene Mischung filtriert. Das feste Produkt wurde mit Wasser gewaschen, in Methylenchlorid gelöst und die organische Phase wurde abgetrennt und zur Trockne konzentriert. So wurde 5α-Brom-3ss,6ss,17ss-triacetoxy-androstan er halten, das gegebenenfalls wie in Abschnitt 1 weiter ge reinigt werden kann.
<I>Abschnitt 3</I> Durch Wiederholung von Abschnitt 1 unter Verwen dung von 3ss-Acetoxy-17ss-benzoxy-androst-5-en, 3ss-Acetoxy-pregn-5-en, 3ss-Acetoxy-pregn-5-en-20-on und 3ss-Acetoxy-chloest-5-en als Ausgangsmaterial wurde eine Mischung des entspre chenden 5α-Brom-6ss-formiates und 6ss-Acetates jedes der oben genannten ^5-Steroide erhalten.
<I>Abschnitt 4</I> Zu einer Mischung aus 10 g 3ss-Aectoxy-androst-5- en-17-on und 10 cm3 97%iger Ameisensäure wurden 6,24 g N-Bromsuccinimid, in 20 cm3 Methylenchlorid suspendiert, zugefügt. Dann wurde die erhaltene Mi schung etwa 1 Stunde und 50 Minuten bei 15-20 C gerührt. Zur Reaktionsmischung wurde etwa 1 g Na- triumbisulfit in 50 cm3 Wasser zugefügt, die dann zwei mal mit Methylenchlorid extrahiert wurde.
Die orga nischen Extrakte wurden dann mit Wasser gewaschen, getrocknet und zur Trockne eingedampft; so wurde 3ss- Acetoxy-5a-brom-6ss-formoxy-androstan-17-on mit ei nem F. von 210-212 C erhalten.
<I>Abschnitt 5</I> Zu einer Mischung aus 1 g 3ss-Acetoxy-androst-5-en- 17-on und 15 cms wasserfreier Essigsäure wurden 1,2molare Äquivalente N-Cholacetamid zugefügt. Die erhaltene Mischung wurde etwa 2 Stunden bei 20 ge rührt, dann in 50 cm3 Wasser gegossen und die erhal tene Mischung mit Methylenchlorid extrahiert. Die Methylenchloridextrakte wurden mit Wasser gewaschen, getrocknet und zur Trockne eingedampft, worauf 3ss,6ss- Diacetoxy-5α-chlor-androstan-17-on erhalten wurde.
<I>Beispiel 1</I> Zu einer Lösung aus 500, cm3 Methanol und den gemäss Abschnitt 1 erhaltenen, gemischten Estern (6ss- Formiat und 6ss-Acetat von 5α-Brom-3ss,6ss-dihydroxy- androstan-17-on-3-acetat) wurden 100 g wässrige Na triumhydroxydlösung (50%ig) zugegeben, wobei die Temperatur auf etwa 35--45 C gehalten wurde. Dann wurde die erhaltene Mischung etwa 30 Minuten auf 30 bis 45 C gehalten.
Die Reaktionsmischung wurde durch Zugabe von verdünnter wässriger Essigsäure neutralisiert und bis zu einer inneren Temperatur von 90 C destil liert, worauf 300 cm3 Toluol zugefügt wurden. Dann wurde die Mischung unter Rühren zum Rückfluss erhitzt, bis keine ungelösten Feststoffe zurückblieben und sich die organischen und wässrigen Schichten trennten. Die organische, 3ss-Hydroxy-5ss,6ss-oxido-androstan-17-on enhaltende Schicht wurde dann auf etwa 250-300 cm- konzentriert und es wurden 50 g Pyridin und 75 g Ess.ig- säureanhydrid zugefügt.
Die Temperatur wurde eine Stunde auf 85-95 C gehalten, dann wurden 600 cm3 Toluol zugegeben und die Mischung auf etwa 20 C ab gekühlt. Die Mischung vurde dann mit Wasser, ver dünnter wässriger Essigsäure (50 cm3 Essigsäure: 350 cm3 Wasser) und Wasser zur Entfernung des Pyridins ge waschen und die organische Phase wurde auf ein Ge samtvolumen von etwa 700 cm3 konzentriert. Zu dieser, 3ss-Acetoxy-5ss,6ss-oxido-androstan-17-on (F. 187-188 ) enthaltenden Lösung wurden 100 cm3 Toluol und 500 cm3 Hexan zugegeben und die erhaltene Mischung auf 15-18 C abgekühlt.
Während die Temperatur auf 15-18 C gehalten wurde, wurden 21,5 g trockener Bromwasserstoff langsam über der Lösung unter Rüh ren eingeführt, bis ein leichter Überschuss vorlag.
Die Bromwasserstoffzugabe erforderte etwa 45 bis 60 Minuten. Die Reaktionsmischung wurde dann filtriert und das feste Produkt mit einer Mischung aus 100 cm! Toluol und 100 cm3 Hexan und dann zweimal mit 200 cm3-Anteilen Hexan gewaschen; das Produkt wurde bei etwa 25 C getrocknet. So wurde das 3ss-Acetoxy- 5α-brom-6ss-hydroxy-androstan-17-on mit einem F. von 180,5 bis <B>1811</B> C erhalten.
Die Mutterlauge (Filtrat und Waschwasser aus dem Endprodukt) wurden dann mit 100 g Wasser, das 5 g Natriumbicarbonat enthielt, gewaschen und die wässrige Phase abgetrennt und verworfen. Die organische Phase wurde auf ein kleines Volumen eingedampft. Etwa 60 cm3 Methanol wurden zum Konzentrat zugefügt, dann. wurden 12 g Natriumhydroxydlösung (50% Gew.%/ Gew.) unter Rühren zugegeben, wobei die Temperatur auf 35-45 C gehalten wurde. Diese Temperatur wurde etwa 30 Minuten aufrechterhalten.
Dann wurde die Re aktionsmischung abgekühlt, durch Zugabe von wässriger Essigsäure neutralisiert; dann wurde der neutralisierten Mischung Toluol zugefügt und die wässrige Phase wurde abgetrennt und verworfen. Die orgasniche Phase wurde gewaschen, getrocknet und eingedampft und ergab 3ss- Hydroxy-5ss,6ss-oxido-androstan-17-on, das durch Be handlung mit Essigsäureanhydrid in Pyridin in oben be-, schriebener Weise in das entsprechende 3-Acetat umge wandelt werden kann.
<I>Beispiel 2</I> Eine Mischung aus 1 g Androst-5-en-17-on, 2mo- laren Äquivalenten N-Bromsuccinimid und 50 cm3 was serfreier Essigsäure wurde 1,3 Stunden gerührt, wobei die Temperatur auf etwa 20 C gehalten wurde. Dann wurden 15 cm-' Wasser, die 1 g Natriumbisulfit enthiel ten, zugegeben und die Reaktionsmischung in 150 cm' Wasser gegossen und gerührt. Der Niederschlag wurde abfiltriert, mit Wasser gewaschen und getrocknet; er lieferte das 5α-Brom-6ss-acetoxy-androstan-17-on.
Eine Mischung des so hergestellten Steroids, 50 cm3 Methanol und 10 g wässriger Kaliumhydroxydlösung (50%ig) wurde 35 Minuten auf etwa 35 C gehalten. Dann wurde die Reaktionsmischung durch Zugabe von verdünnter wässriger Essigsäure neutralisiert, die erhal tene Mischung wurde in 100 cm3 Wasser gegossen und der organische Niederschlag abfiltriert, gewaschen und getrocknet; so wurde das 5ss,6ss-Oxido-androstan-17-on erhalten.
Das so erhaltene Epoxyd wurde in 15 cm3 Toluol gelöst und die Lösung auf etwa 15-18 C abgekühlt; unter Aufrechterhaltung dieser Temperatur wurden 1,5molare Äquivalente trockener Bromwasserstoff lang sam über der Lösung unter Rühren eingeführt, bis ein leichter Überschuss anwesend war. Dann wurde die Reaktionsmischung filtriert und das gesammelte feste Produkt mit einer Mischung (1 : 1) aus Toluol und Hexan und dann mit Hexan gewaschen und das Pro dukt unter vermindertem Druck getrocknet. So wurde das 5α-Brom-6ss-hydroxy-androstan-17-on erhalten.

The process of the present invention for the preparation of 5α-bromo-6ss-hydroxy-steroids can furthermore be carried out at room temperature, is easy to use, provides a high yield of the desired 5α-bromo-6ss-hydroxy-steroid and gives a minimum of by-products.

The reaction can be carried out at a temperature between about 10 C (or slightly lower) up to about 40 C or more, preferably at room temperature (about 20 to 25 ° C) for about 15 minutes to about 2 hours or more more to be done. The by-products formed during the above reaction, which mainly consist of the corresponding 5α, 6ss-dibromo or 5α, 6ss-dichloro steroid and the 5a-acyloxy-6ss-bromo or chloro-steroid, can be treated by treatment with zinc in the presence of a lower hydrocarbyl carboxylic acid such as acetic acid, propionic acid, formic acid, butyric acid, etc., preferably acetic acid, easily converted into the original starting compound, i.e. the ^ 5 steroid. Diosgenin, Yamogenin, Ruscogenin, Botogenin, etc., and of ^ 5-alkaloids such as solanidine, solasodin, rubijervin, isorubijervin etc. Other ^ 5-steroids suitable as starting materials according to the invention include the A5-, 3-halogen - ^ 5- and 3-hydroxy- ^ 5- steroids, such as androst-5-en-17-one, 3ss-chloro-androst-5- en-17-one, 3ss-hydroxy-androst-5-en-17 -on, Pregn-5-en- 20-on etc.
The following sections concern the production of raw materials. <I> Section 1 </I> To 187 g of formic acid (90%) were added 113 g of acetic anhydride, the temperature being kept at about 55-65 ° C. for about 10 minutes. The mixture was then cooled to about 20 ° C. and 100 g of 3ss-acetoxy-androst-5-en-17-one were added with stirring.
59 g of N-bromosuccinimide were then added, the temperature being kept at about 20-25 ° C. by means of an ice bath. The mixture obtained was then stirred for one hour, the temperature being kept at 20-25 ° C. Then about 10 g of sodium bisulfite in 50 cm3 of water were added and the reaction mixture was poured into about 850 cm3 of water, stirred and filtered; thus a mixture of the 6ss acetate and 6ss formate of 5α-bromo-3ss, 6ss-dihydroxy-androstane-17-one-3-acetate was obtained, which was purified as follows.
The crude mixed esters were washed with water and then dissolved in 700 cc of methylene chloride; the aqueous layer was separated and extracted with methylene chloride. The organic phases were then combined and concentrated at atmospheric pressure. The residue was taken up in 600 cm3 of methanol and the solution was distilled under reduced pressure until the volume was reduced to 400 cm3. The resulting solution was cooled to about 15-20 ° C. and then filtered.
The precipitate was washed with 150 cm3 of methanol and yielded a pure mixture of the 6ss acetate and 6ss formate of 5α-bromo-3ss, 6ss-dihydroxy-androstane-17-one-3-acetate.
By repeating the above procedure using N-chlorosuccinimide instead of N-bromosuccinimide, the corresponding 5α-chloro compounds were obtained.
The mother liquors obtained from purifying the mixed esters from the above reaction (combined filtrate and wash water), 5 g zinc powder and 50 g acetic acid were heated to about 85-90 ° C. A further 5 g of zinc powder were then added and the reaction mixture was heated on a steam bath (85-95 ° C.) for about an hour. Most of the remaining solvent was then evaporated and 100 cm3 of benzene was added. The resulting mixture was cooled (to about 20 ° C.) and 100 g of water were added. The mixture was then filtered and the recovered zinc was washed with about 50 cc of benzene. The aqueous phase was separated from the filtrate and discarded.
The remaining organic phase was washed with water, separated and evaporated to a small volume. The small volume of the concentrate was then taken up in 100 cm3 of methanol and the resulting mixture was evaporated to a small volume. The concentrate obtained was then dissolved in about 80 cm3 of methanol, the solution obtained was concentrated to saturation, cooled to about 20 ° C. and filtered. The solid product obtained was washed with methanol and gave 3ss-acetoxy-androst-5-en-17-one with a F. of 168.5-170 C.
A second crop of the above ^ 5 steroid was obtained by evaporating the mother liquor, cooling, filtering and washing; this yield had an F. of 164.5-167 C.
<I> Section 2 </I> A mixture of 100 g 3ss, 17ss-diacetoxy-androst-5-en, 60 g N-bromosuccinimide and 300 cm3 anhydrous acetic acid was stirred for about 70 minutes at room temperature <B> (20- 25 'C). The reaction mixture was then poured into 1 liter of water containing about 12 g of sodium bisulfite, and the resulting mixture was filtered. The solid product was washed with water, dissolved in methylene chloride and the organic phase was separated and concentrated to dryness. Thus 5α-bromo-3ss, 6ss, 17ss-triacetoxy-androstane was obtained, which can be further purified as in section 1 if necessary.
<I> Section 3 </I> By repeating Section 1 using 3ss-Acetoxy-17ss-benzoxy-androst-5-ene, 3ss-Acetoxy-pregn-5-ene, 3ss-Acetoxy-pregn-5- en-20-one and 3ss-acetoxy-chloest-5-ene as the starting material, a mixture of the corresponding 5α-bromo-6ss-formate and 6ss-acetate of each of the above-mentioned ^ 5-steroids was obtained.
<I> Section 4 </I> 6.24 g of N-bromosuccinimide were suspended in 20 cm3 of methylene chloride to a mixture of 10 g of 3ss-aectoxy-androst-5-en-17-one and 10 cm3 of 97% formic acid, added. The mixture obtained was then stirred at 15-20 ° C. for about 1 hour and 50 minutes. About 1 g of sodium bisulfite in 50 cm3 of water was added to the reaction mixture, which was then extracted twice with methylene chloride.
The organic extracts were then washed with water, dried and evaporated to dryness; so 3ss-acetoxy-5a-bromo-6ss-formoxy-androstan-17-one with a F. of 210-212 ° C. was obtained.
<I> Section 5 </I> To a mixture of 1 g of 3ss-acetoxy-androst-5-en-17-one and 15 cms of anhydrous acetic acid, 1.2 molar equivalents of N-cholacetamide were added. The mixture obtained was stirred at 20 for about 2 hours, then poured into 50 cm3 of water and the mixture obtained was extracted with methylene chloride. The methylene chloride extracts were washed with water, dried and evaporated to dryness to give 3ss, 6ss-diacetoxy-5α-chloro-androstan-17-one.
<I> Example 1 </I> To a solution of 500 cm3 of methanol and the mixed esters obtained according to Section 1 (6ss-formate and 6ss-acetate of 5α-bromo-3ss, 6ss-dihydroxy-androstane-17- on-3-acetate) 100 g of aqueous sodium hydroxide solution (50%) were added, the temperature being kept at about 35-45 C. Then the resulting mixture was kept at 30 to 45 ° C for about 30 minutes.
The reaction mixture was neutralized by adding dilute aqueous acetic acid and distilled to an internal temperature of 90 ° C., whereupon 300 cm 3 of toluene were added. The mixture was then heated to reflux with stirring until no undissolved solids remained and the organic and aqueous layers separated. The organic layer containing 3ss-hydroxy-5ss, 6ss-oxido-androstan-17-one was then concentrated to about 250-300 cm- and 50 g of pyridine and 75 g of acetic anhydride were added.
The temperature was held at 85-95 ° C. for one hour, then 600 cm3 of toluene were added and the mixture was cooled to about 20 ° C. The mixture was then washed with water, dilute aqueous acetic acid (50 cm3 acetic acid: 350 cm3 water) and water to remove the pyridine, and the organic phase was concentrated to a total volume of about 700 cm3. 100 cm3 of toluene and 500 cm3 of hexane were added to this solution containing 3ss-acetoxy-5ss, 6ss-oxido-androstan-17-one (F. 187-188) and the mixture obtained was cooled to 15-18 ° C.
While maintaining the temperature at 15-18 C, 21.5 g of dry hydrogen bromide was slowly introduced over the solution with stirring until a slight excess was present.
The hydrogen bromide addition took about 45 to 60 minutes. The reaction mixture was then filtered and the solid product with a mixture of 100 cm! Toluene and 100 cm3 hexane and then washed twice with 200 cm3 portions of hexane; the product was dried at about 25 ° C. The 3ss-acetoxy-5α-bromo-6ss-hydroxy-androstan-17-one with an F. of 180.5 to 1811 C was obtained.
The mother liquor (filtrate and washing water from the end product) were then washed with 100 g of water containing 5 g of sodium bicarbonate, and the aqueous phase was separated off and discarded. The organic phase was evaporated to a small volume. About 60 cc of methanol was added to the concentrate, then. 12 g of sodium hydroxide solution (50% wt.% / wt.) were added with stirring, the temperature being kept at 35-45 ° C. This temperature was maintained for about 30 minutes.
Then the reaction mixture was cooled, neutralized by adding aqueous acetic acid; then toluene was added to the neutralized mixture and the aqueous phase was separated and discarded. The organic phase was washed, dried and evaporated and gave 3ss-hydroxy-5ss, 6ss-oxido-androstan-17-one, which was converted into the corresponding 3-acetate by treatment with acetic anhydride in pyridine in the manner described above can be.
<I> Example 2 </I> A mixture of 1 g of androst-5-en-17-one, 2 molar equivalents of N-bromosuccinimide and 50 cm3 of anhydrous acetic acid was stirred for 1.3 hours, the temperature increasing to about 20 C was held. Then 15 cm- 'of water containing 1 g of sodium bisulfite were added and the reaction mixture was poured into 150 cm' of water and stirred. The precipitate was filtered off, washed with water and dried; it provided the 5α-bromo-6ss-acetoxy-androstan-17-one.
A mixture of the steroid so produced, 50 cm3 of methanol and 10 g of aqueous potassium hydroxide solution (50%) was kept at about 35 ° C. for 35 minutes. The reaction mixture was then neutralized by adding dilute aqueous acetic acid, the mixture obtained was poured into 100 cm3 of water and the organic precipitate was filtered off, washed and dried; so the 5ss, 6ss-oxido-androstan-17-one was obtained.
The epoxy obtained in this way was dissolved in 15 cm3 of toluene and the solution was cooled to about 15-18 ° C; while maintaining this temperature, 1.5 molar equivalents of dry hydrogen bromide were slowly introduced over the solution with stirring until a slight excess was present. Then the reaction mixture was filtered and the collected solid product was washed with a mixture (1: 1) of toluene and hexane and then with hexane, and the product was dried under reduced pressure. Thus the 5α-bromo-6ss-hydroxy-androstan-17-one was obtained.
