Procédé de préparation de benzo [b] thiophéne substitué La présente invention a trait à un procédé de pré paration de dérivés pharmacologiquement actifs du benzo[b]thiophène, appelés ci-après dérivés de benzo- thiophène.
L'invention a pour objet un procédé de préparation de benzothiophènes qui sont (a) substitués par au moins un groupe de la for mule (B)n-Y I où n est 0 ou 1, B est une chaîne alkylène de 1 à 4 car bones et Y est choisi parmi les groupes de formule (i) -C (: NH) - OR, II (iii) -C(:
NORA) - NH2 où Ri est alkyle ou aryle, R2 est de l'hydrogène, alkyle ou aryle, et les dérivés N-alkyle, N-aryle, N-alcoxycar- bonyle et N-aryloxycarbonyle du groupe (iii) ; et (b) non substitués ou substitués par au moins un halo gène, alkoxy, alkyle, halogénoalkyle, carboxyle, alcoxy- carbonyle, aryloxycarbonyle, nitro, amino, cyano, alkyl- amino, halogénoalkylamino, acylamino, arylacylamino, ou aryloxyacylamino ;
et leurs sels d'addition acides, caractérisé par le fait qu'un benzo[b]thiophène substitué ou non par un groupe tel que défini en (b) ci-dessus et ayant au moins un substituant de formule -(B)n-CN où B et n sont tels que définis ci-dessus est amené à ré agir avec un alcool R10H ou un dérivé de l'hydroxy- amine H2NOR2 où Ri et R2 sont définis comme ci-des sus, ou un dérivé N-alkylé, N-arylé, N-alcoxycarbonylé ou N-aryloxycarbonylé de l'hydroxylamine.
Le composé dans lequel Y est représenté par la for mule II (i), ci-dessus, est utilisé pour la préparation, par réaction avec l'ammoniac ou une amine, du com posé dans lequel Y est un reste de formule (ii) -C(: NH) -NH2 et ses dérivés N-alkylés, N-arylés, N-alcoxycarbonylés et N-aryloxycarbonylés.
Des exemples des substituants alkoxy susmentionnés sont le méthoxy, l'éthoxy, le propoxy, le butoxy, l'amyl- oxy et l'hexyloxy. Les radicaux alkyles comportent le méthyle, l'éthyle, le propyle, le butyle, l'amyle, l'hexyle, l'heptyle, l'octyle, le nonyle, le décyle, l'undécyle et le dodécyle.
Les substituants halogènes sont le chloro, le bromo, l'iodo et le fluoro alors que les substituants halogénoalkyles comportent le 2-chloro-éthyle, le 2-bro- moéthyle, le 2-iodoéthyle, le 2-fluoroéthyle, le 2-chloro- propyle, le 3-fluoropropyle, le 4-trifluorométhyibutyle, le 5-bromoamyle, le 2-bromo-méthyl-3-bromopropyle, le 3- méthyl-5-iodopentyle et autres.
Les substituants aryles et aralkyles sont, de préfé rence, ceux qui comportent 6 à 12 carbones, à savoir le phényle, l'a-naphtyle, le -naphtyle, le benzyle, le tolyle, le xylyle, le cuményle, l'alcoxyphényle, l'halogénophé- nyle, l'halogénobenzyle, l'α-naphtyl-méthyl, le phénopro- pyle, le phénobutyle, l'alcoxyphénoéthyle, le tolylpropyle et autres.
Les substituants de l'alcoxycarbonyle et de l'aryl- oxycarbonyle sont, par exemple, le méthoxycarbonyle, l'éthoxycarbonyle, le propoxycarbonyle, l'amyloxycarbo- nyle, le phénoxycarbonyle, l'halogénophénoxycarbonyle, l'alcoxyphénoxycarbonyle, le benzyloxycarbonyle, le to- luyloxycarbonyle et autres.
Des exemples d'alkylamino, d'halogénoalkylamino, d'acylamino, d'arylacylamino et d'aryloxyacylamino sont le méthylamino, le diméthyl- amino, le diéthylamino, le butylamino, le 2-chloroéthyl- amino, le 3-bromopropylamino, le N,N-di-(2-fluoro- éthyle)amino, l'acétylamino, le valérylamino, le phényl- propionylamino, le phénylacétylamino, l'alkoxyphényl- acétylamino, le phénoxyacétylamino et le phénoxybuty- rylamino.
Alors que, tout au long de la présente description, on se réfère à un substituant sans qu'il soit fait mention de son état isomérique, il est à noter que ce substituant comporte tous ses isomères ; ainsi, par exemple, lors qu'on se réfère au butyle, cela inclut le n-butyle, l'isobu- tyle, le s-butyle, et le t-butyle.
Le terme d' inférieur qualifiant les différents grou pes est utilisé ici pour indiquer que ces groupes con tiennent jusqu'à 6 atomes de carbone.

des sulfonates de p-toluyl-mé- thyle ou éthyle des halogénures de phényl-alkyles infé rieurs, par exemple le chlorure de benzyle ou de phéné- thyle, les bromures ou les iodures correspondants. Les hydroxydes -quaternaires d'ammonium sont également inclus de même que les composés quaternaires d'am monium- ayant comme anions ceux des autres sels in organiques ou organiques, par exemple ceux des acides utilisés pour la préparation des sels acides d'addition mentionnés précédemment.
Les benzothiophènes préparés suivant la présente invention présentent une action pharmacologique utile, en particulier une action antimicrobienne et une-acti vité anti-hypertensive. Par le terme antimicrobien , il est convenu d'entendre les actions antibactériennes anti virales, antifongiques et anthelmintiques.
Pour autant que cela soit connu, les présents benzo- thiophènes et leurs dérivés, à l'exception du 3-benzo[b] thiénylacétimidate d'éthyle sont des composés nouveaux.
Les produits de base pour la mise en ouvre du présent procédé sont les benzothiophènes substitués par au moins un groupe de la formule (B)n-CN, où B et n sont tels que définis dans la formule 1.
De tels produits de base sont soit connus en soi, soit peuvent être préparées par des méthodes connues.
Les groupes de la formule II (i) sont obtenus par réaction du composé cyanuré avec un alcool de la for mule R10H, où R1 est tel que défini dans la formule II, la réaction étant de préférence effectuée dans un sol vant ou un mélange de solvants tel que du benzène ou du dioxane. Le groupe résultant II (i) est ensuite aisé ment converti en groupe Il (ü) par réaction avec des amines ou l'ammoniac de préférence en présence d'un solvant convenable tel qu'un alcool, par exemple de l'éthanol.
Des groupes de formule II (iii) sont obtenus par réaction du composé cyanuré correspondant avec une hydroxylamine substituée ou non en présence d'un solvant tel que l'eau ou l'éthanol aqueux, la réaction étant normalement effectuée à température élevée.
Lorsque des dérivés N-alkyle ou N-aryle des groupes (i) à (iii) sont désirés, ils sont normalement obtenus en effectuant les réactions susmentionnées avec les inter médiaires substitués N-alkyle ou N-aryle appropriés. Lorsque des dérivés N-alcoxycarbonyle ou aryloxycar- bonyle sont désirés, ils peuvent être normalement pré parés par une réaction subséquente du composé corres- pondant N- non substitué avec un chloroformate d'al kyle ou d'aryle.

Du 5-méthyl-3-cyanométhylbenzo[b]thiophène (0,014 mole) a été chauffé sur un bain de vapeur pendant 90 minutes avec une solution de carbonate de soude an hydre (2,5 g) et de chlorhydrate d'hydroxylamine (3,5 g) dans de l'eau (38 ml), de l'éthanol étant ajouté en quan tité suffisante pour maintenir la solution claire. Du 5-chloro-3-cyanométhylbenzo[b]thiophène (0,02 mole) et de l'éthanol sec (1,0g, 0,021 mole) ont été dissous dans un mélange de benzène sec (20-40 ml) et en présence de dioxanne pur en quantité juste suffisante pour empêcher une précipitation à 00 C. Examples of the aforementioned alkoxy substituents are methoxy, ethoxy, propoxy, butoxy, amyloxy and hexyloxy. Alkyl radicals include methyl, ethyl, propyl, butyl, amyl, hexyl, heptyl, octyl, nonyl, decyl, undecyl and dodecyl.
The halogen substituents are chloro, bromo, iodo and fluoro while the haloalkyl substituents include 2-chloro-ethyl, 2-bromoethyl, 2-iodoethyl, 2-fluoroethyl, 2-chloro - propyl, 3-fluoropropyl, 4-trifluoromethyibutyl, 5-bromoamyl, 2-bromo-methyl-3-bromopropyl, 3-methyl-5-iodopentyl and others.
The aryl and aralkyl substituents are preferably those which contain 6 to 12 carbons, namely phenyl, α-naphthyl, -naphthyl, benzyl, tolyl, xylyl, cumenyl, alkoxyphenyl, halophenyl, halobenzyl, α-naphthyl-methyl, phenopropyl, phenobutyl, alkoxyphenoethyl, tolylpropyl and the like.
The substituents of alkoxycarbonyl and aryloxycarbonyl are, for example, methoxycarbonyl, ethoxycarbonyl, propoxycarbonyl, amyloxycarbonyl, phenoxycarbonyl, halogenophenoxycarbonyl, alkoxyphenoxycarbonyl, benzyloxycarbonyl, to - luyloxycarbonyle and others.

The benzothiophenes prepared according to the present invention exhibit useful pharmacological action, in particular antimicrobial action and anti-hypertensive activity. By the term antimicrobial, it is agreed to understand the antibacterial, anti viral, antifungal and anthelmintic actions.
As far as is known, the present benzothiophenes and their derivatives, with the exception of ethyl 3-benzo [b] thienylacetimidate, are novel compounds.
The basic products for the implementation of the present process are the benzothiophenes substituted by at least one group of the formula (B) n-CN, where B and n are as defined in the formula 1.
Such basic products are either known per se or can be prepared by known methods.
The groups of formula II (i) are obtained by reacting the cyanide compound with an alcohol of the formula R10H, where R1 is as defined in formula II, the reaction being preferably carried out in a solvent or a mixture of solvents such as benzene or dioxane. The resulting group II (i) is then easily converted to group II (ü) by reaction with amines or ammonia, preferably in the presence of a suitable solvent such as an alcohol, for example ethanol.
Groups of formula II (iii) are obtained by reacting the corresponding cyanide compound with a substituted or unsubstituted hydroxylamine in the presence of a solvent such as water or aqueous ethanol, the reaction normally being carried out at elevated temperature.
When N-alkyl or N-aryl derivatives of groups (i) to (iii) are desired, they are normally obtained by carrying out the aforementioned reactions with the appropriate N-alkyl or N-aryl substituted intermediates. When N-alkoxycarbonyl or aryloxycarbonyl derivatives are desired, they can normally be prepared by a subsequent reaction of the corresponding N- unsubstituted compound with an alkyl or aryl chloroformate.

5-Chloro-3-cyanomethylbenzo [b] thiophene (0.02 mole) and dry ethanol (1.0g, 0.021 mole) were dissolved in a mixture of dry benzene (20-40 ml) and in the presence of pure dioxane in an amount just sufficient to prevent precipitation at 00 C. The solution was saturated with dry hydrochloric gas, and at 00 C, and kept overnight in the refrigerator.
