Procédé de préparation de dérivés du xanthène
La présente invention a pour objet un procédé de préparation de nouveaux dérivés du xanthène qui sont utiles pour le traitement des ulcères gastriques.
Les ulcères gastriques sont assez fréquents et le traitement de cet état est par la chirurgie, par la neutralisation de l'acide gastrique en utilisant des milieux alcalins, par exemple l'alumine et la magnésie, ou par l'administration d'agents adsorbants tels que le trisilicate de magnésium, ou par l'administration de produits pharmaceutiques qui diminuent la sécrétion d'acide dans l'estomac. Il est évident qu'il est avantageux d'éviter la chirurgie si cela est possible, mais jusqu'à maintenant les traitements anti-acide et ceux inhibant la sécrétion n'ont pas été entièrement satisfaisants. L'action des antiacides est presque immédiate et de courte durée; cela rend difficile le contrôle de la sécrétion gastrique nocturne par l'utilisation d'anti-acides.
Un grand nombre des agents inhibant la sécrétion sont des substances anticholinergiques qui donnent naissance à des effets secondaires indésirables tels que la sécheresse de la bouche, la mydriase et d'autres effets analogues à ceux dus à l'atropine.
Selon le procédé de la présente invention, on prépare les composés de formule générale I:

Les noyaux A et B peuvent éventuellement contenir des substituants choisis parmi les suivants: halogène, alcoyle, alcoxy et hydroxy.
Le procédé selon l'invention est caractérisé en ce que l'on fait réagir un composé de formule générale II:

Si désiré, on peut transformer un produit contenant un groupe carboxyle ou un atome d'azote basique en un sel.
On a trouvé que les composés de formule générale I sont des agents inhibant la sécrétion, ayant une activité spécifique envers la sécrétion gastrique et sans aucune activité anticholinergique.
On a en outre constaté que les composés dans lesquels R, est de l'hydrogène ne manifestent un degré utile d'activité antisécrétoire qu'à condition que R2 soit un groupe hydroxy ou acyloxy.

Les expressions alcoyle , alcényle et aralcoyle utilisées ici signifient des groupes de ce genre contenant jusqu'à 7 atomes de carbone dans le reste
aliphatique.
L'expression acyle utilisée ici indique le reste acyle d'un acide carboxylique, qui peut être un acide aliphatique, un acide aromatique, un acide carbamique N-substitué, un acide carbonique ou un acide carboxylique hétérocyclique. Des exemples de tels groupes acyle sont les suivants:

Les composés de a à h dans lesquels le reste xanthényle est remplacé par le groupe thiaxanthényle correspondant:

Le procédé selon l'invention est de préférence mis en oeuvre en présence d'un acide dans un solvant inerte.
L'acide acétique est un acide commode et préféré. Lorsqu'il est mis en oeuvre en présence d'un acide, il est possible que le procédé s'effectue par formation intermédiaire d'un ester de xanthydrol ou de thiaxanthydrol, par exemple de l'acétate, et que c'est cet ester, formé in situ, qui se condense en réalité avec l'urée ou la thiourée de formule III. Les conditions exactes de la réaction apparaîtront facilement à l'homme du métier, à la lumière de la littérature antérieure et des exemples ciaprès.
L'activité inhibant la sécrétion des composés préparés par le procédé selon l'invention a été démontrée chez le rat stimulé, dont le pylore a été ligaturé, et elle varie selon la valeur de X1, X2 et Rl à R3 et selon la nature et les positions des substituants des noyaux A et B. Nous avons trouvé en général que les composés dans lesquels Xl est un atome d'oxygène sont plus actifs que ceux dans lesquels Xl est un atome de soufre, et que les composés dans lesquels X2 est un atome d'oxygène sont plus actifs que ceux dans lesquels X est un atome de soufre.
En général la substitution dans les noyaux A et B diminue l'activité. Un groupe préféré de composés de formule générale I est celui de formule générale IV:

Les groupes que l'on préfère particulièrement sont les composés de formule générale IV dans laquelle a) Rl est un groupe méthyle, R2 est un groupe alcoyle, et R8 est un atome d'hydrogène, et b) Ri est un groupe hydroxy ou alcoxy, R2 est un groupe alcoyle ou un atome d'hydrogène, et R8 est un atome d'hydrogène.

Les composés préparés par le procédé selon l'invention peuvent être administrés par voie orale, rectale ou parentérale, de préférence par voie orale; le dosage optimum varie suivant l'activité des composés. Un dosage pour l'administration orale que l'on préfère est de l'ordre de 0,25 à 4 g par jour, éventuellement en plusieurs fois.
Pour l'utilisation, les composés de formule I sont administrés dans des compositions usuelles en association avec des excipients pharmaceutiques connus pour la préparation de compositions appropriées à l'administration par voie orale, rectale ou parentérale.
Les matières de départ de formules générales II et
III utilisées pour la préparation de composés de formule générale I sont dans de nombreux cas des composés connus; lorsqu'ils sont nouveaux, on les prépare selon des procédés analogues à ceux utilisés pour les composés connus et ainsi seront clairs pour ceux du métier.
Exemple 1:
On laisse reposer pendant une nuit à la température ordinaire une solution de 20g de xanthydrol et 7,6 g d'hydroxyurée dans 200 ml d'acide acétique éthanolique (1:1). On lave à l'alcool le solide qui se sépare puis le sèche pour obtenir la N-hydroxy-N-9-xanthénylurée qui fond à 1850 C.

Exemple 2:
On chauffe à reflux pendant 20 minutes un mélange de 2 g de xanthydrol, 1 g de N,N'-diméthylurée, 15 ml de toluène et 0,6 ml d'acide acétique, on évapore à sec, lave le résidu à l'eau puis le sèche. On le recristallise dans du benzène et on sèche les cristaux à 900 C/2 mm pour obtenir la N,N'-diméthyl-N-9-xanthénylurée qui fond de 169 à 1700C.
Par des méthodes similaires à celles décrites cidessus, on prépare les composés suivants:

Process for the preparation of xanthene derivatives
The present invention relates to a process for the preparation of novel xanthene derivatives which are useful for the treatment of gastric ulcers.
Gastric ulcers are quite common and the treatment for this condition is by surgery, by neutralization of stomach acid using alkaline media, eg alumina and magnesia, or by administration of adsorbents such as as magnesium trisilicate, or by the administration of pharmaceuticals which decrease the secretion of acid in the stomach. Obviously, it is advantageous to avoid surgery if possible, but so far anti-acid and secretion-inhibiting treatments have not been entirely satisfactory. The action of antacids is almost immediate and of short duration; this makes it difficult to control nocturnal gastric secretion by the use of antacids.
Many of the secretion inhibitors are anticholinergic substances which give rise to unwanted side effects such as dry mouth, mydriasis, and other atropine-like effects.
According to the process of the present invention, the compounds of general formula I are prepared:

The rings A and B may optionally contain substituents chosen from the following: halogen, alkyl, alkoxy and hydroxy.
The process according to the invention is characterized in that a compound of general formula II is reacted:

If desired, a product containing a carboxyl group or a basic nitrogen atom can be converted into a salt.
The compounds of general formula I have been found to be secretion inhibiting agents, having specific activity towards gastric secretion and without any anticholinergic activity.
It has further been found that compounds in which R 3 is hydrogen only exhibit a useful degree of antisecretory activity provided that R 2 is hydroxy or acyloxy.
The most accessible compounds of general formula I, and therefore a preferred group, are those containing the following groups:

The expressions alkyl, alkenyl and aralkyl used herein mean such groups containing up to 7 carbon atoms in the remainder

The process according to the invention is preferably carried out in the presence of an acid in an inert solvent.
Acetic acid is a convenient and preferred acid. When it is carried out in the presence of an acid, it is possible that the process is carried out by intermediate formation of an ester of xanthydrol or of thiaxanthydrol, for example acetate, and that it is this ester , formed in situ, which actually condenses with urea or thiourea of formula III. The exact reaction conditions will be readily apparent to those skilled in the art in light of the prior literature and the examples below.
The activity inhibiting the secretion of the compounds prepared by the method according to the invention has been demonstrated in the stimulated rat, whose pylorus has been ligated, and it varies according to the value of X1, X2 and Rl to R3 and according to the nature and the positions of the substituents of the rings A and B. We have found in general that the compounds in which X1 is an oxygen atom are more active than those in which X1 is a sulfur atom, and that the compounds in which X2 is an oxygen atom. oxygen atom are more active than those in which X is a sulfur atom.
In general, substitution in the A and B rings decreases the activity. A preferred group of compounds of general formula I is that of general formula IV:

in which Rl, R2 and R8 have the meaning indicated above. Particularly preferred groups are those compounds of general formula IV wherein a) R1 is methyl, R2 is alkyl, and R8 is hydrogen, and b) R1 is hydroxy or alkoxy. , R2 is an alkyl group or a hydrogen atom, and R8 is a hydrogen atom.

The compounds prepared by the process according to the invention can be administered orally, rectally or parenterally, preferably orally; the optimum dosage varies according to the activity of the compounds. A preferred dosage for oral administration is in the range of 0.25 to 4 g per day, optionally in several portions.
For use, the compounds of formula I are administered in customary compositions in combination with known pharmaceutical excipients for the preparation of compositions suitable for oral, rectal or parenteral administration.
The starting materials of general formulas II and
III used for the preparation of compounds of general formula I are in many cases known compounds; when they are new, they are prepared according to methods analogous to those used for the known compounds and thus will be clear to those skilled in the art.
Example 1:
A solution of 20 g of xanthydrol and 7.6 g of hydroxyurea in 200 ml of ethanolic acetic acid (1: 1) is left to stand overnight at room temperature. The solid which separates is washed with alcohol and then dried to obtain N-hydroxy-N-9-xanthenylurea which melts at 1850 C.

Example 2:
A mixture of 2 g of xanthydrol, 1 g of N, N'-dimethylurea, 15 ml of toluene and 0.6 ml of acetic acid is refluxed for 20 minutes, the mixture is evaporated to dryness, the residue washed with water. water and then dry it. It is recrystallized from benzene and the crystals are dried at 900 C / 2 mm to obtain N, N'-dimethyl-N-9-xanthenylurea which melts at 169 to 1700C.
