Verfahren zur Herstellung von neuen 5-Nitrofuryl-derivaten Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung substituierter heterocyclischer Verbin dungen, die wertvolle pharmakologische Aktivität be sitzen, und insbesondere Nitrofurylderivate von: Oxazo- lidinonen.

geschaffen, worin R Wasserstoff oder einen aliphati schen oder cycloaliphatischen Kohlenwasserstoffrest mit 1 bis 11 Kohlenstoffatomen oder eine Alkoxyalkyl- gruppe mit 2 bis 11 Kohlenstoffatomen bedeutet.
Die Gruppe R kann beispielsweise eine Alkyl gruppe mit 1 bis 11 Kohlenstoffatomen, eine Alkenyl- gruppe mit 2 bis 11 Kohlenstoffatomen oder eine Cycloalkylgruppe mit 3 his 11 Kohlenstoffatomen und vorzugsweise 5 bis 7 Koblenstoffatomen in dem carbo- cyclischen Ring sein.
Wenn die Gruppe R eine Alkylgruppe ist, so kann das Alkyl beispielsweise Methyl, Äthyl, n-Propyl, Iso- propyl, n-Butyl, Isobutyl, sek.-Butyl, tert.-Butyl, n-Pentyl- Isopentyl, n-Hexyl, n-Octyl, Isooctyl oder n-Undecyl sein.
Wenn die Gruppe R eine Alkenylgruppe ist, so kann das Alkenyl beispielsweise Allyl, 2-Methallyl, But-2-enyl (Crotyl), But-3-enyl, Pent-l-enyl, Pent- 2-enyl, Hex-l-enyl, Hexadienyl oder Undecylenyl sein.
Wenn die Gruppe R eine Cycloalkylgruppe ist, kann das Cycloalkyl beispielsweise Cyclopropyl, Cyclo- butyl, Cyclopentyl, Cyclohexyl, Cycloheptyl, Cyclooc- tyl oder Cycloundecyl sein.
Wenn die Gruppe R eine Alkoxyalkylgruppe ist, so kann dies beispielsweise Methoxymethyl, Methoxy- äthyl, Methoxy-n-propyl, Methoxyisopropyl, Methoxy- n-butyl, Methoxy-tert.-butyl, Methoxyhexyl, Meth- oxyoctyl, Methoxydecyl, Äthoxymethyl, Äthoxyäthyl, Isopropoxymethyl, tert.-Butyloxymethyl, Decyloxyme- thyl oder Pentyloxyhexyl sein.

worin R die oben angegebene Bedeutung besitzt, nitriert.
Die Nitrierung kann mit Salpetersäure unter Bedin gungen durchgeführt werden, die bei der Nitrierung von substituierten Furylderivaten üblich sind, beispiels weise durch Umsetzung in Gegenwart eines wasserbin denden Mittels, wobei Glas wasserbindende Mittel bei spielsweise Schwefelsäure sein kann, vorzugsweise jedoch Essigsäureanhydrid ist. Gewünschtenfalls kann ein Anteil an Essigsäure in der Reaktionsmischung vorliegen. Die Nitrierung wird vorzugsweise bei einer 15 C nicht überschreitenden Temperatur unter Ver wendung von konzentrierter oder rauchender Salpeter säure durchgeführt.
Die Umsetzung kann beispielsweise durchgeführt werden, indem eine Mischung von kon zentrierter oder rauchender Salpetersäure, Essigsäure und Essigsäureanhydrid langsam zu einer Suspension oder Lösung des Oxazolidinons der Formel (Il) in einer Mischung von Essigsäure und Essigsäureanhydrid gegeben wird, während die Temperatur bei 5 bis 15 C, vorzugsweisse bei etwa iFI C, gehalten wird. Gewünschtenfalls kann die Verbindung der Formel (II) während der Nitrierung hergestellt werden, indem die Verbindung der Formel III

welches die Gruppierung -CO-R enthält. Das Acylierungsmittel kann beispielsweise eine Carbonsäure, ein Carbonsäu- reanhydrid oder ein gemischtes Anhydrid oder ein Säu ssigsäureanhy- rechlorid sein, ist jedoch vorzugsweise drid oder ein anderes Carbonsäureanhydrid.
Die erfindungsgemässen Verbindungen der Formel (I) zeigen wertvolle antimikrobielle Eigenschaften und insbesondere antibakterielle, anthelminthische, cocci- diostatische, trypanocide und antimalaria-Eigenschaf ten, die in der Human- oder Veterinärmedizin von Wert sind. Die Verbindungen sind besonders wertvoll bei der Behandlung von Infektionen des Intestinal- und Harntrakts. Die Verbindungen können auch dazu ver wendet werden, um hydrophobes oder anderes organi sches Material mit hohem Molekulargewicht, das gegen Zerstörung durch Bakterien oder andere Mikroben empfindlich ist, zu schützen, indem das organische Material mit den Verbindungen kontaktiert, imprä gniert oder in anderer Weise behandelt wird. Die Ver bindungen finden auch Anwendung als wachstumsför dernde Zusätze in Tierfutter.
Erfindungsgemäss wird somit auch ein therapeuti sches Mittel geschaffen, das eine gegen Mikroben wirk i- idenaminooxazo same Menge eines 5-Nitro-2-furfurylidenaminooxazoli- dinons der Formel (I) und einen pharmakologisch ver trägliches flüssiges Verdünnungsmittel enthält.
Die erfindungsgemässen pharmazeutischen Mittel enthalten mindestens eine Verbindung der Formel (I) als aktive Substanz zusammen mit einem herkömm- zwischen lichen pharmazeutischen Träger. Der Typ des tatsäch- kapseln lich verwendeten Trägers hängt in grossem Ausmass von der beabsichtigten Anwendung ab, beispielsweise werden für äusserliche Anwendung bei der Desinfek tion von gesunder Haut, der Desinfektion von Wunden und der Behandlung von Dermatosen und Affektionen der Schleimhäute, die durch Bakterien hervorgerufen sind, insbesondere Salben, Pulverzubereitungen und Tinkturen verwendet.
Die Salbengrundlagen können wasserfrei sein und beispielsweise aus Mischungen von Wollfett und Weichparaffin bestehen oder sie können aus wässrigen Emulsionen bestehen, worin die aktive Substanz suspendiert ist. Geeignete Träger für Pulver zubereitungen sind beispielsweise Reisstärke und an dere Stärken, wobei das Schüttgewicht der Träger ge wünschtenfalls erniedrigt werden kann, beispielsweise durch Zugabe von hochdisperser Kieselsäure oder durch Zugabe von Talk erhöht werden kann.
Die Tinkturen können mindestens einen aktiven Bestand teil der Formel (I) in wässrigem Äthanol, insbesondere 45 bis 75%igem Äthanol, enthalten, dem gewünschten falls 10 bis 2001o Glycerin zugesetzt werden können. Aus Polyäthylenglykol und anderen herkömmlichen Löslichkeitspromotoren und gegebenenfalls auch Emul- giermitteln hergestellte Lösungen können mit besonde rem Vorteil bei der Desinfektion von gesunder Haut verwendet werden. Die Konzentration des aktiven Be standteils in pharmazeutischen Mitteln für äusserliche Anwendung liegt vorzugsweise im Bereich von 0,1 bis 50/0.
Mundwässer oder Konzentrate zu ihrer Herstellung und Tabletten zur langsamen Auflösung im Mund sind für die Desinfektion des Mundes und Rachens geeignet. Erstere werden vorzugsweise aus alkoholischen Lösun gen hergestellt, die 1 bis 50/o an aktiver Substanz enthal ten, denen Glycerin oder Geschmacksstoffe zugesetzt werden können. Pastillen, d. h. feste Dosierungseinhei ten, haben vorzugsweise einen relativ hohen Gehalt an Zucker oder ähnlichen Substanzen und einen relativ niedrigen Gehalt an aktiver Substanz, beispielsweise 0,2 bis 20 Gew.-%, und enthalten die üblichen herkömm lichen Zusätze, wie Bindemittel und Geschmacksstoffe.
Feste Dosierungseinheiten, insbesondere Tabletten, Dragees (mit Zucker überzogene Tabletten) und Kap seln, sind für die Verwendung bei der Intestinaldesin fektion und für die orale Behandlung von Infektionen des Harntrakts geeignet. Diese Einheiten enthalten vor zugsweise 10 bis 900/o der Verbindung der Formel (I), um die Verabreichung von täglichen Dosen von 0,1 bis 2,5 g an Erwachsene oder von in geeigneter Weise ver minderten Dosen an Kinder zu ermöglichen.
Tabletten und Drageekerne werden hergestellt, indem die Verbin dungen der Formel (I) mit festen pulverförmigen Trä gern, wie Lactose, Saccharose, Sorbit, Maisstärke, Kar toffelstärke oder Amylopectin, Zellulosederivaten oder Gelatinen, vorzugweise unter Zugabe von Gleitmitteln, wie Magnesium- oder Calciumstearat oder von Poly- äthylenglykolen mit geeignetem Molekulargewicht, kombiniert werden. Drageekerne können dann überzo gen werden, beispielsweise mit konzentrierten Zuckerlö sungen, die auch Gummiarabikum, Talk und/oder Titandioxyd enthalten können, oder sie können mit einem Lack überzogen werden, der in flüchtigen organi schen Lösungsmitteln oder Mischungen von Lösungs mitteln gelöst ist.
Diesen Überzügen können Farbstoffe zugesetzt werden, beispielsweise zur Differenzierung verschiedenen Dosierungen.
und andere geschlossene Kapseln bestehen bei spielsweise aus einer Mischung von Gelatinen und Gly- cerin und können beispielsweise Mischungen der Ver bindung der Formel (I) mit Polyäthylenglykol enthalten. Hartgelatinekapseln enthalten beispielsweise Granulate einer aktiven Substanz mit festen pulverförmigen Trä gern, beispielsweise Lactose, Saccharose, Sorbit, Man nit, Stärken, wie Kartoffelstärke, Maisstärke oder Amy- lopectin, Zellulosederivate oder Gelatinen und Magne- siumstearat oder Stearinsäure.
In allen Verabreichungsformen können Verbindun gen der Formel (I) als einzige aktive Bestandteile vor liegen oder sie können auch mit anderen bekannten pharmakologisch aktiven und insbesondere antibakte riell und/oder antimykotisch aktiven Substanzen kombi niert sein, beispielsweise um den Anwendungsbereich auszuweiten.
Sie können beispielsweise mit 5,7-Dichlor- 2-methyl-8-chinolinol oder anderen Derivaten des 8-Chinolinols, mit Sulfamerazin oder Sulfafurazol oder anderen Sulfanilamidderivaten, mit Chloramphenicol oder Tetracyclin oder anderen Antibiotika, mit 3,4'-,5-Tribromsalicylanilid oder anderen halogenierten Salicylaniliden, mit halogenierten Carbaniliden, mit halogenierten Benzoxazalen aller Benzoxazolonen, mit Polychlorhydroxydiphenylmethanen, mit Halogendihydroxydiphenylsulfiden, mit 4,4'-Dichlor-2-hydroxydiphenyläther, 2',4,
4'-Trichlor-2-hydroxydiphenyläther oder anderen Polyhalogenhydroxydiphenyläthern, mit bakteriziden, quaternären Verbindungen oder mit be stimmten Dithiocarbaminsäurederivaten, wie Tetrame- thylthiuramidsulfid, kombiniert werden. Es können auch Träger verwendet werden, die selbst günstige pharmakologische Eigenschaften besitzen, beispiels weise Schwefel als: Pulvergrundlage oder Zinkstearat als Komponente von Basengrundlagen.
Erfindungsgemäss wird auch ein Verfahren geschaf fen, um ein organisches Material, das gegen Angriff durch Bakterien oder andere Mikroben empfindlich ist, zu schützen, gemäss dem man das Material mit einem 5-Nitro-2-furfurylidenaminooxazolidinon der Formel (I) behandelt. Das organische Material kann beispielsweise ein natürliches oder synthetisches poly meres Material, eine Protein- oder Kohlehydratsub stanz, eine natürliche oder synthetische Faser oder ein daraus hergestelltes Textilmaterial sein.
Erfindungsgemäss wird auch ein Tierfuttermittel ge schaffen, das ein 5-Nitro-2-furfurylidenaminooxazolidinon der Formel (I) in einer Menge enthält, die ausreicht, um das Wachstum des Tieres zu fördern, das mit dem Mit tel gefüttert wird.
Die nachfolgenden Beispiele sollen die Erfindung weiter veranschaulichen. Prozentangaben sind gewichts bezogen, wenn nichts anderes angegeben ist. <I>Beispiel 1</I> a) Zu einer Lösung von 13,2 g 3-Amino-5-hydroxymethyl-2-oxazolidinon in 100g Wasser werden 9,6 g Furfurol gegeben. Nach dem Stehenlassen wird das Produkt gesammelt und ge trocknet.
Das Produkt ist das 3-Furfurylidenamino- 5-hydroxy-methyl-2-oxazolidinon. b) Eine Mischung von 8,2 g 3-Furfurylidenamino- 5-hydroxymethyl-2-oxazolidinon und 50g Essigsäureanhydrid wird 1 Stunde lang zum Rückfluss erhitzt. Die Reaktionsmischung wird zur Trockene eingedampft und der Rückstand wird aus Äthanol umkristallisiert. Das erhaltene Produkt ist das 5-Acetoxymethyl-3-furfuryliden- amino-2-oxazolidinon.
c) Zu einer Mischung von 10,3g Essigsäureanhy drid und 1,9 g konzentrierter Salpetersäure werden an teilsweise 2,5 g 5-Acetoxymethyl-3-furfuryliden- amino-2-oxazolidinon unter Kühlen gegeben. Die Reaktionsmischung wird stehengelassen und danach wird der erhaltene gelbe Feststoff gesammelt und aus Essigsäureäthylester um kristallisiert.
Als Produkt erhält man das 5 Acetoxymethyl-3-(5-nitrofurfuryliden- amino)-2-oxazolidinon mit einem Smp. = 186 C, das mit dem Produkt von Beispiel 2 identisch äst. d) Zu einer Mischung von 10,3 g Essigsäureanhy drid und 1,9 g konzentrierter Salpetersäure werden rationenweise 2,2 g 3-Furfurylidenamino-5-hydroxy- methyl-2-oxazolidinon unter Kühlen gegeben. Die Reaktionsmischung wird ste hen gelassen, wonach der erhaltene gelbe Feststoff ge- sammelt und aus Äthanol umkristallisiert wird.
Das Produkt ist das 5-Acetoxymethyl-3-(5-nitro- furfurylidenamino)-2-oxazolidinon mit einem Smp. = 186 C.
<I>Beispiel 2</I> Die in Beispiel 1 b) beschriebene Arbeitsweise wird unter Verwendung von Ameisensäure anstelle von Essigsäureanhydrid durchgeführt, wobei die Reaktions bedingungen sonst im wesentlichen die gleichen sind. Das so hergestellte Zwischenprodukt ist das 5-Formyloxymethyl-3-furfuryliden- amino-2-oxazolidinon. Die in Beispiel 1 c) beschriebene Arbeitsweise wird dann unter Verwendung des 5-Formyloxymethyl-3-furfuryliden- amino-2-oxazolidinons durchgeführt, wobei die Reaktionsbedingungen sonst im wesentlichen die gleichen sind.
Das Produkt ist das 5-Formyloxymethyl-3-(5-nitro- furfurylidenamino)-2-oxazolidinon mit einem Smp. = 161' C. <I>Beispiel 3</I> Die in Beispiel 1 b) beschriebene Arbeitsweise wird unter Verwendung von Propionsäureanhydrid anstelle von Essigsäureanhydrid durchgeführt, wobei die Reak tionsbedingungen sonst im wesentlichen die gleichen sind.
Das so hergestellte Zwischenprodukt ist das 3-Furfurylidenamino-5-propionyl- oxymethyl-2-oxazolidinon. Die in Beispiel 1 c) beschriebene Arbeitsweise wird dann unter Verwendung des 3-Furfurylidenamino-5-propionyl- oxymethyl-2-oxazolidinons anstelle des 5-Acetoxymethyl-3-furfurylidenamino- 2-oxazolidinons durchgeführt, wobei die Reaktionsbe dingungen sonst im wesentlichen die gleichen sind.
Das Produkt ist das 3-(5-Nitrofurfurylidenamino)- 5-propionyloxymethyl-2-oxazolidinon mit einem Smp. = 192 C.
<I>Beispiel 4</I> Die in Beispiel 1 b) beschriebene Arbeitsweise wird unter Verwendung von Buttersäureanhydrid anstelle von Essigsäureanhydrid durchgeführt, wobei die Reak tionsbedingungen sonst im wesentlichen die gleichen sind. Das so hergestellte Zwischenprodukt ist das 5-Butyryloxymethyl-3-furfuryliden- amino-2-oxazolidinon. Die in Beispiel 1 c) beschriebene Arbeitsweise wird dann unter Verwendung des 5-Butyryloxymethyl-3-furfuryliden- amino-2-oxazolidinons anstelle des 5-Acetoxymethyl-3 furfuryliden- amino-2-oxazolidinons durchgeführt, wobei die Reaktionsbedingungen sonst im wesentlichen die gleichen sind.
Als Produkt erhält man das 5-Butyryloxymethyl-3-(5-nitrofurfuryliden- amino)-2-oxazolidinon mit einem Smp. = 148 C. <I>Beispiel 5</I> Die in Beispiel 1 b) beschriebene Arbeitsweise wird unter Verwendung von Valeriansäureanhydrid anstelle von Essigsäureanhydrid durchgeführt, wobei die Reak tionsbedingungen sonst im wesentlichen die gleichen sind.
Das so hergestellte Zwischenprodukt ist das 3-Furfurylidenamino-5-valeryl- oxymethyl-2-oxazolidinon. Die in Beispiel 1 c) beschriebene Arbeitsweise wird dann unter Verwendung des 3-Furfurylidenamino-5 valeryl- oxymethyl-2-oxazolidinons anstelle des 5-Acetoxymethyl-3-furfuryliden- amino-2-oxazolidinons durchgeführt, wobei die Reaktionsbedingungen sonst im wesentlichen die gleichen sind.
Das Produkt ist das 3-(5-Nitrofurfurylidenamino)- 5-valeryloxymethyl-2-oxazolidinon mit einem Smp. = 129 C. <I>Beispiel 6</I> Die in Beispiel 1 b) beschriebene Arbeitsweise wird unter Verwendung von Crotonsäureanhydrid anstelle von Essigsäureanhydrid durchgeführt, wobei die Reak tionsbedingungen sonst im wesentlichen die gleichen sind.
Das so hergestellte Zwischenprodukt ist das 5-Crotonyloxymethyl-3-furfuryliden- amino-2-oxazolidinon. Die in Beispiel 1 c) beschriebene Arbeitsweise wird dann unter Verwendung des 5-Crotonyloxymethyl-3-furfuryliden- amino-2-oxazolidinons anstelle des 5-Acetoxymethyl-3-furfuryliden- amino-2-oxazolidinons durchgeführt, wobei die Reaktionsbedingungen sonst im wesentlichen die gleichen sind.
Das Produkt ist das 5-Crotonyloxymethyl-3-(5-nitrofurfuryliden- amino)-2-oxazolidinon mit einem Smp. = 181 C. <I>Beispiel 7</I> Die in Beispiel 1 b) beschriebene Arbeitsweise wird unter Verwendung von Hexansäureanhydrid anstelle von Essigsäureanhydrid durchgeführt, wobei die Reak tionsbedingungen sonst im wesentlichen die gleichen sind.
Das so hergestellte Zwischenprodukt ist das 3-Furfurylidenamino-5-hexanoyl- oxymethyl-2-oxazolidinon. Die in Beispiel 1 c) beschriebene Arbeitsweise wird dann unter Verwendung des 3-Furfurylidenamino-5-hexanoyl- oxymethyl-2-oxazolidinons anstelle des 5-Acetoxymethyl-3-furfuryliden- amino-2-oxazolidinons durchgeführt, wobei die Reaktionsbedingungen sonst im wesentlichen die gleichen sind.
Das Produkt ist das 5-Hexanoyloxymethyl-3-(5-nitro- furfurylidenamino)-2-oxazolidinon mit einem Smp. = 133 C.
