Automatisch arbeitendes Gerät zur Analyse von flüssigen Proben
Die vorliegende Erfindung betrifft ein automatisch arbeitendes Gerät zur Analyse von flüssigen Proben, welche durch Beimischen von Reagenzlösungen zubereitet werden, bestehend aus einer periodisch arbeitenden Druckluftquelle, einer Flüssigkeitsabsperrvorrichtung, einer Mischabmessvorrichtung für Probe und Reagenzlösungen, Überlaufpipetten für das Abmessen der Reagenzlösungen und ferner einer Photometerküvette mit einer Entleervorrichtung.
Die technische Entwicklung von Analysatoren, bei denen ein Abmessen, Umfüllen, Mischen und Ablassen der Flüssigkeiten nötig ist, bestand hauptsächlich im Suchen geeigneter Elemente für die Ausführung dieser Aufgaben. Zu diesem Zwecke wurden Verschlussorgane, wie elektrisch betätigte Quetschhähne, elektromagnetische Solenoidventile und pneumatisch gesteuerte Membranventile entwickelt, wobei eine zuverlässige Funktion unter Miniaturbedingungen selbst bei Anwendung aggressiver und korrodierender Lösungen sichergestellt werden sollte. Mit Hilfe dieser Verschlussorgane war es möglich, die gewünschten Leistungen mit Hilfe der Schwerkraft zu erzielen, die hierzu nötigen Apparaturen waren jedoch sehr kompliziert, teuer und störanfällig.
Unabhängig davon wurden daher kontinuierlich arbeitende Apparate entwickelt, die mittels peristaltischer Mikropumpen die Durchführung einer Reihe analytischer Operationen, wie dies die Automatisierung analytischer Prozesse erfordert, ermöglichen.
Schwierigkeiten verursachen hier die in diesen Mikropumpen verwendeten Schläuche, da sie bei jedem gewählten Material nach einiger Zeit ihre Elastizität einbüssen und die Zuverlässigkeit bei Anwendung dieser Apparaturen in Frage stellen.
Um diese Nachteile zu vermeiden, wird nunmehr ein automatisch arbeiten des Gerät zur Analyse von flüssigen Proben, welche durch Beimischen von Reagenzlösungen zubereitet werden, bestehend aus einer periodisch arbeitenden Druckluftquelle, einer Flüssigkeitsabsperrvorrichtung, einer Mischabmessvorrichtung für Probe und Reagenzlösungen, Überlaufpipetten für das Abmessen der Reagenzlösungen und ferner einer Photometerküvette mit einer Entleervorrichtung vorgeschlagen, das sich erfindungsgemäss dadurch auszeichnet, dass das gesamte Abmessen der Probe und Reagenzlösungen und ihr gemeinsames Durchmischen periodisch durch Ein- und Ausschalten der Druckluftquelle erfolgt;
dass der erzeugte Luftstrom in der Flüssigkeitsabsperrvorrichtung das Überströmen der aus einer Zuleitung kontinuierlich zufliessenden Probe in einen Füll trichter verursacht, von wo die mit einem U-förmigen Entleerungsrohr versehene Mischabmessvorrichtung angefüllt wird, deren Rohr ein von der genannten Druckluftquelle abgezweigter Luftstrom periodisch zugeführt wird, der einerseits das Vermischen der in der Mischabmessvorrichtung angesammelten Flüssigkeit bewirkt, andererseits den Abfluss der Flüssigkeit aus der Misch abmessvorrichtung verhindert; und dass der in der Austrittsleitung der Druckluftquelle entstehende Überdruck gleichzeitig zur Betätigung der pneumatischen tXberlauf- pipetten ausgenützt wird.
Der Vorteil eines solchen Analysators besteht vor allem darin, dass sämtliche nötigen Handhabungen mit der Probe und den Reagenzlösungen mittels einer zeitweilig eingeschalteten und ausgeschalteten kleinen Luftpumpe erfolgen können. Ausserdem ist das hydraulischpneumatische System vollkommen störungsfrei, denn es hat keine beweglichen Teile, die einer Abnützung unterliegen.
Eine beispielsweise Ausführungsform des Erfindungsgegenstandes wird nachfolgend anhand der Zeichnung näher erläutert. Es zeigen:
Fig. 1 eine Flüssigkeitsabsperrvorrichtung und
Fig. 2 die Kombination einer Flüssigkeitsabsperrvorrichtung mit einer Mischabmesseinrichtung mit pneu matischen Überlaufpip etten, sowie einer Photometerküvette mit einer Entleereinrichtung und mit einer periodisch ein und ausschaltbaren Luftpumpe.
Einen wesentlichen Bestandteil des selbsttätigen Analysators bildet die Flüssigkeitsabsperrvorrichtung, hier ein System von Kanälen, in dem durch Einführung strömender Luft die Ausflussstelle der Flüssigkeit geändert wird, die der Flüssigkeitsabsperrvorrichtung kontinuierlich zufliesst. Die Flüssigkeitsabsperrvorrichtung ist in Fig. 1 dargestellt. In den Behälter 1 der Flüssigkeitsabsperrvorrichtung 22 fliesst die Probe durch die Zuleitung 3 kontinuierlich zu. Der Behälter 1 ist durch einen Überlauf 2 der gehobenen Probe mit einer Sammelleitung 4 verbunden. Aus dem Boden des Behälters 1 ist ein Verbindungskanal 5 in Form einer U-Schleife herausgeführt und geht in ein Knie 6 über, in das ein Luftzufuhrrohr 7 mündet. Durch ein U-förmiges Abfuhrrohr 9 ist das Knie 6 über einen zweiten Uberlauf
11 mit einem Ablauf 10 verbunden.
Das Luftzufuhrrohr 7 ist an die Luftpumpe über eine Verengung 8 angeschlossen. Zwischen der Verengung 8 und dem Knie 6 ist an das Luftzufuhrrohr 7 ein Rohr 12 angeschlossen, das in einen Behälter 13 des Überdruckbegrenzers 14 mündet. In der Abbildung ist eine Alternative dargestellt, wo der Behälter 13 des Überdruckbegrenzers 14 zwischen das U-förmige Abfuhrrohr 9 und den zweiten Überlauf 11 gelegt ist. Vor der Verengung 8 kann das Luftzufuhrrohr 7 durch die Kapillare 18 mit der Aussenluft verbunden sein.
Die Flüssigkeitsabsperrvorrichtung arbeitet wie folgt: Falls die Luftpumpe nicht eingeschaltet ist, fliesst die durch die Zuleitung 3 zufliessende Probe durch den schleifenförmigen Verbindungskanal 5 und das Uförmige Abfuhrrohr 9 in den Abfall 10. Dabei füllt die Probe den Behälter 13 des Überdruckbegrenzers 14 an, das U-förmige Abfuhrrohr 9 und den schleifenförmigen Verbindungskanal 5 an, falls diese nicht schon früher angefüllt wurden. Es wird vorausgesetzt, dass die Kanäle eine genügende lichte Weite haben, die der abfliessenden Flüssigkeit praktisch keinen Widerstand entgegensetzen, so dass beim Abfliessen der Probe über den zweiten Überlauf 11 in den Ablauf 10 der Behälter
1 leer ist.
Sobald die Luftpumpe in Betrieb versetzt wird, entsteht im Luftzufuhrrohr 7 ein Üb erd ruck, der konstant bleibt, da die überflüssige Luft durch das Rohr 12 im Behälter 13 des Überdruckbegrenzers 14 entweicht.
Die Verengung 8 reduziert die Luftzufuhr in dem Masse, dass im Rohr 12 kein Druckabfall entsteht und im Luftzufuhrrohr 7 herrscht ein Überdruck, der der Wassersäule von der Tiefe 15 entspricht. Im U-förmigen Abfuhrrohr 9 entsteht ein Luftpolster, der den Durchfluss der Probe verhindert und die deshalb den Behälter 1 anfüllt, so dass sie schliesslich über den Überlauf 2 in die Sammelleitung 4 abfliesst. Das Niveau der überströmenden Probe ist dabei ruhig, da der Luftüberdruck in dem Luftzufuhrrohr 7 nicht genügt, um die Luft durch den schleifenförmigen Verbindungskanal 5 durchzudrücken. Voraussetzung ist, dass die Höhe 16 des Überlaufs 2 über der oberen Wand des Verbindungskanals 5 grösser ist als die Tiefe 15.
Um das Anstauen der Flüssigkeit zu ermöglichen, muss das Flüssigkeitsniveau im Verbindungskanal 5 tiefer liegen als die Trennwand im Knie 6, die von der unteren Wand des Verbindungskanals 5 gebildet wird. Diese Trennwand und auch der zweite Überlauf 11 liegen im Niveau des Bodens des Behälters 1. Deshalb muss die Höhe 17 des Überlaufs 2 der gehobenen Flüssigkeit über der unteren Wand des Knies 6 geringer sein als die Tiefe 15.
Wenn die Luftpumpe zu arbeiten aufhört, entweicht der Überdruck aus dem Luftzufuhrrohr 7 durch die Kapillare 18, wodurch der Abflussweg für die im Behälter 1 angesammelte Flüssigkeit frei wird. Falls die lichten Weiten des schleifenförmigen Verbindungskanals 5 und des U-förmigen Abfuhrrohres 9 genügend gross sind, kann bei genügender Weite der Verengung 8 und des Kapillarrohres 18 das Niveau im Behälter 1 nach Abstellung der Luftpumpe beinahe sofort absinken. Die Füllgeschwindigkeit des Behälters 1 hängt von der Menge der zufliessenden Probe und dem Fassungsraum des Behälters 1 ab.
Der nach Fig. 2 ausgeführte Analysator ist im wesentlichen mit einer Dosiereinrichtung versehen, die aus einer Flüssigkeitsabsperrvorrichtung 22, einer pneumatischen Überlaufpipette 38 für das Abmessen der Reagenzlösung oder mehreren parallel geschalteten Überdruckpipetten für verschiedene Reagenzlösungen, einer Mischabmessvorrichtung 23, einer Photometerküvette 24, einer Entleervorrichtung 25 und einer periodisch ein- und ausgeschalteten kleinen Luftpumpe 26 besteht. Der Behälter 1 ist durch den Überlauf 2 mit dem Fülltrichter 27 verbunden, der in das schräge Füllrohr 28 übergeht. Dieses hat oben einen Überlaufansatz 29 mit einer Überlaufkante 30 und geht in das Überlaufrohr 31 über. Mit dem anderen Ende mündet das Füllrohr 28 in die Mischabmessvorrichtung 23.
Sie dient zum Mischen und Abmessen der Probe und Reagenzlösung und hat im Oberteil über dem Niveau des Füllrohres 28 einen verengten Querschnitt. Durch diese Verengung wird eine vollkommene Reproduzierbarkeit der abgemessenen Probe und Reagenzlösung erzielt, falls die Niveauhöhe der Flüssigkeit beim Anfüllen nicht genau eingehalten wird, äussert sich dies in geringeren Unterschieden der Abmessung. Der abmessende Teil der Mischabmessvorrichtung 23 besteht aus dem U-förmigem Entleerrohr 33, an das oben das Rohr 34 für die Zufuhr der Druckluft angeschlossen ist. Mit seinem zweiten Ende mündet das Entleerrohr 33 in die Rohrleitung 52 für die verarbeitete Probe. Durch den engen Kanal 36 ist das Entleerrohr 33 mit dem Boden der Mischabmessvorrichtung 23 verbunden. In diese mündet der Flüssigkeitsverschluss 37 der Überlaufpipette 38.
Den Flüssigkeitsverschluss 37 bildet wieder ein U-förmiges Rohr, das mit einem Ende in die Seitenwand der Mischabmessvorrichtung 23 und mit seinem zweiten Ende in den Körper 39 der Überlaufpipette 38 übergeht.
Der Körper 39 der Überlaufpipette 38 ist oben mit der Rohrleitung 40 an das Rohr 41 angeschlossen und in den Körper 39 mündet seitlich das Füllrohr 42, das Uförmig und mit seinem zweiten Ende mit dem Boden des Vorratsbehälters 43 verbunden ist, in dem der Flüssigkeitsspiegel auf konstanter Niveauhöhe 44 gehalten wird. Im Vergleich zur Niveauhöhe 44 liegt die Mündung des Füllrohres 42 in den Körper 39 der Überlaufpipette 38 tiefer, die Mündung des Flüssigkeitsverschlusses 37 in den Abmessbehälter 32 jedoch höher. Ebenso liegt die Überlaufkante 30 des Überlaufansatzes 29 tiefer als die erwähnte Mündung des Flüssigkeitsverschlusses 37. Die der Luftzufuhr zur Pipette 38 dienende Rohrleitung 40 und das Rohr 34 für die Betätigungsluft der Mischabsperrvorrichtung 23 sind gemeinsam über die Verengung 45 an den Druckstutzen 46 der Luftpumpe 26 angeschlossen.
An diesen Druck stutzen 46 ist gleichzeitig über die Verengung 8 auch das Luftzufuhrrohr 7 für die Flüssigkeitsabsperrvorrichtung 22 angeschlossen. Die pneumatischen Impulse in der Austrittsleitung 48 werden durch die Luftpumpe 26 geschaffen, die z. B. durch den Schalter 49 periodisch einund ausgeschaltet werden kann. An die Austrittsleitung 48 kann über die Verengung 50 die Rohrleitung 51 für die Luftzufuhr zur Entleervorrichtung 25 der Photometerküvette 24 angeschlossen sein. So kann mit demselben pneumatischen Impuls die Photometerküvette 24 entleert werden, bevor die neu gemischte Reagenzlösung bereitgestellt ist, die dann in die Photemeterküvette 24 aus der Rohrleitung 52 für die verarbeitete Lösung zufliesst.
An die Austrittsleitung 48 kann auch das enge in die Aussenluft mündende Kapillarrohr 53 angeschlossen sein, das in der Ruhezeit der Luftpumpe 26 den Überdruck in der Austrittsleitung 48 gegenüber der Aussenluft ausgleicht. Ausser der Überdruckpipette 38 können an die Mischabsperrvorrichtung 23 auch die Flüssigkeitsverschlüsse mehrerer Pipetten angeschlossen sein, die gleich wie die Überlaufpipette 38 angeordnet und an die gleiche pneumatische Rohrleitung angeschlossen sind, die durch die Rohrleitung 40 und das Rohr 34 charakterisiert ist.
Die in Fig. 2 dargestellte Apparatur arbeitet folgendermassen: Zu Beginn, wenn die Luftpumpe 26 in Ruhe ist, fliesst die Probe durch den Behälter 1, den Verbindungskanal 5, das Abfuhrrohr 9 und den Behälter 13 in den Ablauf 10. Die Überlaufpipette 38, ihr Füllrohr 42 und der Flüssigkeitsverschluss 37 sind mit zur Abmessung bestimmter Flüssigkeit bis zum konstanten Niveau 44 angefüllt. Schon früher wurde das Entleerrohr 33 bis zur Überlaufhöhe mit alter Flüssigkeit und auch die Photometerküvette 24 angefüllt.
Wenn der Schalter 49 die Luftpumpe 26 in Betrieb setzt, führt die Austrittsleitung 48 die Druckluft in alle Organe. Die entsprechenden Verengungen 8, 45 und 50 verteilen die Luft in einem Verhältnis wie dies die einzelnen Organe verlangen.
Im System mit der Verengung 45 entsteht anfangs praktisch kein Überdruck, solange die Luft durch den engen Kanal 36 entweichen kann. Die in die Flüssigkeitsabsperrvorrichtung 22 strömende Luft bildet im Verbindungskanal 5 und im Abfuhrrohr 9 einen Luftpolster, der das Herausfliessen der Probe in den Ablauf 10 verhindert. Der Flüssigkeitsspiegel im Behälter 1 steigt an und die Probe fliesst schliesslich über den Überlauf 2 in den Fülltrichter 27 ab und gelangt durch das Füllrohr 28 in die Mischabmessvorrichtung 23.
Von hier kann die Probe nicht abfliessen, da in dem Entleerungsrohr 33 auch ein Luftpolster entsteht und die gesamte hergeleitete Luft durch den engen Kanal 36 entweicht. Dieser ist mit Luft ganz angefüllt und falls die Luft durch den Kanal 36 in genügender Menge zufliesst, kann die Flüssigkeit nicht durchdringen. In dem Masse wie der Flüssigkeitsspiegel, der von Luftblasen durchdrungenen Probe in der Mischabmessvorrichtung 23 ansteigt, steigt auch der Überdruck in der Rohrleitung 40 und dem Rohr 34 an und aus der Überlaufpipette 38 wird die Lösung herausgedrückt.
Im Augenblick, wenn das Niveau der Reagenzlösung im Körper
39 der Überlaufpipette 38 unter die Mündung des Füllrohres 42 abgesunken ist, wird der weitere Abfluss der Reagenzlösung in den Vorratsbehälter 43 verhindert und die im Körper 39 eingeschlossene Reagenzlösung wird nun durch den Flüssigkeitsverschluss 37 in die Mischabmessvorrichtung 23 gedrückt, wo sie durch die aufsteigende Luft mit der hier bereits vorhandenen Probe vermengt wird. Wenn die Mischabmessvorrichtung 23 bis zum Niveau der Überlaufkante 30 angefüllt ist, beginnt die durch das Füllrohr 28 zufliessende Probe in das Ablaufrohr 31 zu fliessen und das Niveau in der Mischabmessvorrichtung 23 bleibt auf dieser Höhe stehen.
In diesem Zustand ist in der Rohrleitung 40 und im Rohr 34 der Überdruck am grössten, der das Mass des Herausdrückens der Reagenzlösung aus der Überlaufpipette 38 reproduzierbar bestimmt und daher für die Menge der von der Überlaufpipette 38 abgemessenen Reagenzlösung entscheidend ist. Der Flüssigkeitsverschluss 37 muss natürlich so tief sein, dass die Druckluft nicht die gesamte Lösung herausdrückt. Wenn dann der Schalter 49 die Luftpumpe 26 von dem Netz 54 abschaltet, wird der Zufluss der Probe aus dem Behälter 1 unterbrochen und die mit dem Reagenzmittel vermischte Probe fliesst durch das Entleerrohr 33 in die Rohrleitung 52 für die verarbeitete Probe. Der Überdruck in der Austrittsleitung 48 wird rasch beseitigt, da die überflüssige Luft durch das Kapillarrohr 53 in die Aussenluft entweicht.
Selbstverständlich wird die Überlaufpipette 38 mit einer Reagenzlösung aus dem Vorratsbehälter 43 angefüllt. Die Photometerküvette 24 kann als Durchflussküvette ausgeführt sein. In diesem Falle wird die photometriente Lösung aus ihr in den Ablauf geleitet. Es kann jedoch auch eine selbsttätige Entleervorrichtung 25 installiert sein, wie dies in Fig. 2 angedeutet ist. In diesem Falle ist aus dem Photometerbehälter 24 unten ein doppelt gebogenes Abflussrohr 55 mit einem absteigenden Teil 56, unteren Knie 57 und einem aufsteigenden Teil 58 herausgeführt, das mit seinem oberen Knie 59 einen Überlauf bildet und in das in den Ablauf mündende Ablaufrohr 60 übergeht. In den unteren Teil des aufsteigenden Teiles 58 des doppelt gebogenen Abflussrohres 55 mündet das enge Rohr 61 für die Luftzufuhr mit dem nach oben gerichteten Ende 62.
Das enge Rohr 61 ist über die Verengung 50 an die Austrittsleitung 48 der Luftpumpe 62 angeschlossen.
Beim Einschalten der Luftpumpe 26 strömt die Druckluft durch das enge Rohr 61 und das nach oben gerichtete Ende 62 in den aufsteigenden Teil 58 und befördert das Gemisch von Flüssigkeit und Luft durch das obere Knie 59 und den absteigenden Teil 56 in das Ablaufrohr 60 und weiter in den Ablauf.
Die beschriebenen Einrichtungen können z.B. bei selbsttätigen Analysatoren für die Kontrolle der Wasserzusammensetzung genützt werden.
Automatic device for the analysis of liquid samples
The present invention relates to an automatically operating device for analyzing liquid samples, which are prepared by adding reagent solutions, consisting of a periodically operating compressed air source, a liquid shut-off device, a mixing measuring device for sample and reagent solutions, overflow pipettes for measuring the reagent solutions and also a photometer cuvette an emptying device.
The technical development of analyzers that require measuring, transferring, mixing and draining of the liquids has mainly consisted of finding suitable elements to carry out these tasks. For this purpose, closing devices such as electrically operated pinch cocks, electromagnetic solenoid valves and pneumatically controlled diaphragm valves were developed, whereby a reliable function should be ensured under miniature conditions even when using aggressive and corrosive solutions. With the help of these locking devices it was possible to achieve the desired performance with the help of gravity, but the equipment required for this was very complicated, expensive and prone to failure.
Irrespective of this, continuously operating apparatuses were therefore developed which, by means of peristaltic micropumps, enable a series of analytical operations to be carried out, as required by the automation of analytical processes.
The hoses used in these micropumps cause difficulties here, since they lose their elasticity after some time with any chosen material and the reliability when using this apparatus is questionable.
In order to avoid these disadvantages, the device now works automatically for the analysis of liquid samples, which are prepared by adding reagent solutions, consisting of a periodically operating compressed air source, a liquid shut-off device, a mixing device for measuring the sample and reagent solutions, and overflow pipettes for measuring the reagent solutions and furthermore a photometer cuvette with an emptying device is proposed, which is characterized according to the invention in that the entire measurement of the sample and reagent solutions and their mutual mixing takes place periodically by switching the compressed air source on and off;
that the air flow generated in the liquid shut-off device causes the sample flowing continuously from a supply line to overflow into a filling funnel, from where the mixing measuring device, which is provided with a U-shaped discharge tube, is filled, the tube of which is periodically fed an air flow branched off from the compressed air source mentioned on the one hand causes the mixing of the liquid collected in the mixing measuring device, on the other hand preventing the outflow of the liquid from the mixing measuring device; and that the overpressure created in the outlet line of the compressed air source is used to actuate the pneumatic overflow pipettes.
The main advantage of such an analyzer is that all the necessary manipulations with the sample and the reagent solutions can be carried out by means of a small air pump that is temporarily switched on and switched off. In addition, the hydraulic-pneumatic system is completely trouble-free because it has no moving parts that are subject to wear and tear.
An example embodiment of the subject matter of the invention is explained in more detail below with reference to the drawing. Show it:
Fig. 1 shows a liquid shut-off device and
Fig. 2 the combination of a liquid shut-off device with a mixing meter with pneumatic overflow pipettes, and a photometer cuvette with an emptying device and with an air pump that can be switched on and off periodically.
The liquid shut-off device forms an essential part of the automatic analyzer, here a system of channels in which the outflow point of the liquid is changed by introducing flowing air, which flows continuously to the liquid shut-off device. The liquid shut-off device is shown in FIG. The sample flows continuously through the supply line 3 into the container 1 of the liquid shut-off device 22. The container 1 is connected to a collecting line 4 through an overflow 2 of the raised sample. A connecting channel 5 in the form of a U-loop is led out of the bottom of the container 1 and merges into a knee 6 into which an air supply pipe 7 opens. The knee 6 is through a U-shaped discharge pipe 9 via a second overflow
11 connected to a drain 10.
The air supply pipe 7 is connected to the air pump via a constriction 8. Between the constriction 8 and the knee 6, a pipe 12 is connected to the air supply pipe 7, which pipe opens into a container 13 of the overpressure limiter 14. The figure shows an alternative where the container 13 of the overpressure limiter 14 is placed between the U-shaped discharge pipe 9 and the second overflow 11. In front of the constriction 8, the air supply pipe 7 can be connected to the outside air through the capillary 18.
The liquid shut-off device works as follows: If the air pump is not switched on, the sample flowing in through the supply line 3 flows through the loop-shaped connecting channel 5 and the U-shaped discharge pipe 9 into the waste 10. The sample fills the container 13 of the overpressure limiter 14, the U -shaped discharge pipe 9 and the loop-shaped connecting channel 5, if these were not filled earlier. It is assumed that the channels have a sufficient internal width that offer practically no resistance to the flowing liquid, so that when the sample flows out via the second overflow 11 into the outlet 10, the container
1 is empty.
As soon as the air pump is put into operation, an overpressure occurs in the air supply pipe 7, which remains constant since the excess air escapes through the pipe 12 in the container 13 of the overpressure limiter 14.
The constriction 8 reduces the air supply to the extent that there is no pressure drop in the pipe 12 and there is an overpressure in the air supply pipe 7 which corresponds to the water column from the depth 15. An air cushion is created in the U-shaped discharge pipe 9, which prevents the sample from flowing through and which therefore fills the container 1 so that it finally flows off via the overflow 2 into the collecting line 4. The level of the sample flowing over is steady, since the excess air pressure in the air supply pipe 7 is not sufficient to force the air through the loop-shaped connecting channel 5. In order to enable the liquid to accumulate, the liquid level in the connecting channel 5 must be lower than the partition wall in the knee 6, which is formed by the lower wall of the connecting channel 5. This partition wall and also the second overflow 11 are at the level of the bottom of the container 1. Therefore, the height 17 of the overflow 2 of the lifted liquid above the lower wall of the knee 6 must be less than the depth 15.
When the air pump stops working, the overpressure escapes from the air supply pipe 7 through the capillary 18, whereby the drainage path for the liquid accumulated in the container 1 becomes free. If the clear widths of the loop-shaped connecting channel 5 and the U-shaped discharge pipe 9 are sufficiently large, the level in the container 1 can drop almost immediately after the air pump is switched off if the narrowing 8 and the capillary tube 18 are sufficiently wide. The filling speed of the container 1 depends on the amount of the sample flowing in and the capacity of the container 1.
The analyzer executed according to FIG. 2 is essentially provided with a metering device, which consists of a liquid shut-off device 22, a pneumatic overflow pipette 38 for measuring the reagent solution or several overpressure pipettes connected in parallel for different reagent solutions, a mixing measuring device 23, a photometer cuvette 24, an emptying device 25 and a small air pump 26 which is switched on and off periodically. The container 1 is connected by the overflow 2 to the filling funnel 27, which merges into the inclined filling pipe 28. This has an overflow extension 29 at the top with an overflow edge 30 and merges into the overflow pipe 31. At the other end, the filling pipe 28 opens into the mixing measuring device 23.
It is used to mix and measure the sample and reagent solution and has a narrowed cross-section in the upper part above the level of the filling tube 28. This narrowing achieves complete reproducibility of the measured sample and reagent solution, if the level of the liquid is not precisely adhered to when filling, this manifests itself in smaller differences in the dimensions. The measuring part of the mixing measuring device 23 consists of the U-shaped discharge pipe 33 to which the pipe 34 for the supply of compressed air is connected at the top. At its second end, the emptying tube 33 opens into the pipeline 52 for the processed sample. The emptying pipe 33 is connected to the bottom of the mixing measuring device 23 through the narrow channel 36. The liquid closure 37 of the overflow pipette 38 opens into this.
The liquid seal 37 is again formed by a U-shaped tube which merges with one end into the side wall of the mixing measuring device 23 and with its second end into the body 39 of the overflow pipette 38.
The body 39 of the overflow pipette 38 is connected at the top with the pipeline 40 to the pipe 41 and the filling pipe 42 opens laterally into the body 39, which is U-shaped and connected at its second end to the bottom of the storage container 43, in which the liquid level is constant Level 44 is maintained. Compared to the level 44, the opening of the filling pipe 42 in the body 39 of the overflow pipette 38 is lower, but the opening of the liquid closure 37 in the measuring container 32 is higher. Likewise, the overflow edge 30 of the overflow extension 29 is lower than the mentioned mouth of the liquid seal 37. The pipe 40 serving to supply air to the pipette 38 and the pipe 34 for the actuating air of the mixing shut-off device 23 are jointly connected via the constriction 45 to the pressure port 46 of the air pump 26 .
At the same time, the air supply pipe 7 for the liquid shut-off device 22 is connected to this pressure port 46 via the constriction 8. The pneumatic pulses in the outlet line 48 are created by the air pump 26 which, for. B. can be switched on and off periodically by the switch 49. The pipe 51 for the air supply to the emptying device 25 of the photometer cuvette 24 can be connected to the outlet line 48 via the constriction 50. The photometer cuvette 24 can thus be emptied with the same pneumatic impulse before the newly mixed reagent solution is provided, which then flows into the photometer cuvette 24 from the pipe 52 for the processed solution.
The narrow capillary tube 53, which opens into the outside air and which, when the air pump 26 is idle, can also be connected to the outlet line 48, compensates for the excess pressure in the outlet line 48 with respect to the outside air. In addition to the overpressure pipette 38, the liquid closures of several pipettes can also be connected to the mixing shut-off device 23, which are arranged in the same way as the overflow pipette 38 and are connected to the same pneumatic pipeline, which is characterized by the pipeline 40 and the pipe 34.
The apparatus shown in Fig. 2 works as follows: At the beginning, when the air pump 26 is at rest, the sample flows through the container 1, the connecting channel 5, the discharge pipe 9 and the container 13 into the outlet 10. The overflow pipette 38, you The filling tube 42 and the liquid closure 37 are filled with liquid intended for the measurement up to the constant level 44. The emptying pipe 33 was already filled with old liquid up to the overflow level and the photometer cuvette 24 was also filled earlier.
When the switch 49 puts the air pump 26 into operation, the outlet line 48 leads the compressed air into all organs. The corresponding constrictions 8, 45 and 50 distribute the air in a ratio as required by the individual organs.
In the system with the constriction 45 there is practically no overpressure at the beginning as long as the air can escape through the narrow channel 36. The air flowing into the liquid shut-off device 22 forms an air cushion in the connecting channel 5 and in the discharge pipe 9, which prevents the sample from flowing out into the outlet 10. The liquid level in the container 1 rises and the sample finally flows over the overflow 2 into the filling funnel 27 and passes through the filling tube 28 into the mixing measuring device 23.
The sample cannot flow away from here, since an air cushion is also created in the emptying pipe 33 and all of the extracted air escapes through the narrow channel 36. This is completely filled with air and if the air flows in in sufficient quantity through the channel 36, the liquid cannot penetrate. As the liquid level rises in the sample through which air bubbles have penetrated in the mixing measuring device 23, the excess pressure in the pipeline 40 and the pipe 34 also rises and the solution is pressed out of the overflow pipette 38.
At the moment when the level of the reagent solution in the body
39 of the overflow pipette 38 has sunk below the mouth of the filling tube 42, the further outflow of the reagent solution into the storage container 43 is prevented and the reagent solution enclosed in the body 39 is now pressed through the liquid closure 37 into the mixing measuring device 23, where it is carried along by the rising air the sample already present here is mixed. When the mixing measuring device 23 is filled to the level of the overflow edge 30, the sample flowing through the filling pipe 28 begins to flow into the drainage pipe 31 and the level in the mixing measuring device 23 remains at this height.
In this state, the overpressure is greatest in the pipeline 40 and in the pipe 34, which reproducibly determines the degree of the reagent solution being pressed out of the overflow pipette 38 and is therefore decisive for the amount of the reagent solution measured by the overflow pipette 38. The liquid seal 37 must of course be deep enough that the compressed air does not force out all of the solution. If the switch 49 then switches off the air pump 26 from the network 54, the flow of the sample from the container 1 is interrupted and the sample mixed with the reagent flows through the emptying pipe 33 into the pipe 52 for the processed sample. The overpressure in the outlet line 48 is quickly eliminated, since the excess air escapes through the capillary tube 53 into the outside air.
Of course, the overflow pipette 38 is filled with a reagent solution from the storage container 43. The photometer cuvette 24 can be designed as a flow cell. In this case, the photometric solution is passed from it into the drain. However, an automatic emptying device 25 can also be installed, as indicated in FIG. 2. In this case, a double-curved drainage pipe 55 with a descending part 56, lower knee 57 and an ascending part 58 is led out of the photometer container 24, which forms an overflow with its upper knee 59 and merges into the drainage pipe 60 opening into the drain. In the lower part of the ascending part 58 of the double-curved drainage pipe 55, the narrow pipe 61 for the air supply opens with the upwardly directed end 62.
The narrow tube 61 is connected to the outlet line 48 of the air pump 62 via the constriction 50.
When the air pump 26 is switched on, the compressed air flows through the narrow tube 61 and the upwardly directed end 62 into the ascending part 58 and carries the mixture of liquid and air through the upper knee 59 and descending part 56 into the drain pipe 60 and further in process.
