Procédé de préparation de nouveaux 1,3-diazafluoranthène 1-oxydes 2-substitués
La présente invention est relative à la préparation de nouveaux 1,3-diazafluoranthène 1-oxydes 2-substitués répondant à la formule générale suivante:

dans laquelle R3 et Ra représentent de l'hydrogène ou un groupe alkyle inférieur, R4 représente de l'hydrogène ou un halogène, tel que le chlore en position 4 ou 6, et

représente un groupe hétérocyclique azoté qui peut être substitué, tel qu'un groupe morpholino, pipéridino, hexaméthylèneimino, pipérazino ou pipérazino substitué.
Parmi les composés de formule I, les composés suivants, de même que leurs chlorhydrates, peuvent être cités comme exemple:
2-pipéridinométhyl- 1,3-diazafluoranthène-1 -oxyde,
2-hexaméthylèneiminométhyl- 1,3 -diazafluoroanthène-
1-oxyde,
2-morpholinométhyl- 1,3 -diazafluoranthène- 1-oxyde,

dans laquelle Rs, RA et Ra ont les significations indiquées plus haut, tandis que X désigne un halogène, tel que le chlore, par réaction avec un composé hétérocyclique azoté de préférence en présence de tétrahydrofurane.
Les composés de formule (III) sont également de nouveaux composés. Parmi ces composés de formule
III, on peut citer les suivants, à titre d'exemples:

Les composés de formule VIII peuvent être obtenus par réaction de chlorhydrate d'hydroxylamine (NH2OH. HCî) avec une aminofluorénone de formule (IX) comme indiqué par le schéma suivant:

Dans ces formules, R4 représente de l'hydrogène ou un halogène tel que le chlore ou le brome.
Les composés de formule (IX) peuvent également être utilisés pour obtenir les composés de formule (VI) par réaction avec des halogènures de formule (VII).

La 1-aminofluorénone est un composé connu, mais la 2-chloro-1-aminofluorénone et la 4-chloro-1-aminofluorénone sont de nouveaux composés que l'on peut préparer par chloration de 1-aminofluorénone par divers procédés. Ainsi, la chloration peut se faire dans du chloroforme en présence de pyridine à une température comprise entre 00 C et 200 C. Un mélange de 2-chloro-1aminofluorénone et de 4-chloro- 1 -aminofluorénone est obtenu. Ces deux composés peuvent être séparés par chromatographie ou par cristallisation fractionnée.
La préparation de divers composés de formule I sera décrite à présent dans les exemples suivants:
Exemple 1:
Préparation
de 2-pipéridinométhyl-1,3-diazafluoranthène-1-oxyd -oxyde
et son chlorhydrate
Un mélange de 0,54g de 2-chlorométhyl-1,3-diazafluoranthène-1-oxyde, de 25 mol de tétrahydrofurane et de 0,4 ml de pipéridine est chauffé au reflux pendant 30 minutes. La suspension est alors versée sur de la glace et le précipité est filtré, lavé, séché et recristallisé dans du cyclohexane: P.F. 155-1570 C.
On prépare le chlorhydrate en ajoutant une solution 2 N de HC1 dans de l'éthanol à une solution de la base dans du chloroforme, P.F. 192-1940 C (décomposition) après recristallisation dans de l'éthanol.
Exemple 2:
Préparation
de 2-hexaméthylèneiminométhyl-1,3-diazafluoranthène- oxyde
Un mélange de 0,54 g de 2-chlorométhyl-1,3-diazafluoranthène-1-oxyde, de 30ml de tétrahydrofurane et de 0,45 ml d'hexaméthylèneimine est chauffé au reflux pendant 30 minutes. La suspension est versée sur de la glace et le précipité obtenu est filtré, lavé et séché. On obtient des micro-cristaux fondant à 146 -148 C après plusieurS recristallisations dans du cyclohexane.
Exemple 3:
Préparation
de 2-morpholinométhyl-1,3-diazafluoranthène-1-oxyde
et de son chlorhydrate
Un mélange de 0,5 g de 2-chlorométhyl- 1,3-diaza- fluoranthène-1-oxyde, de 2 mol de morpholine est agité pendant 64 heures à température ambiante dans 20 ml de méthanol, on chauffe au reflux dans 35 ml de tétrahydrofurane pendant 1 heure. La solution est alors concentrée jusqu'à siccité et le résidu est traité à l'eau et filtré. Le produit obtenu est recristallisé dans du méthanol et fond à 168-170"C (cristaux orange).
Le chlorhydrate se prépare en dissolvant la base dans du méthanol et en ajoutant une solution éthanolique d'acide chlorhydrique jusqu'à ce que le pH atteigne 3,5-4. Le mélange est alors concentré jusqu'à siccité et recristallisé dans un mélange de méthanol et de méthyléthyl cétone. Le produit fond à 199-2010 C en se décomposant. Les micro-cristaux jaune orange sont solubles dans l'eau.
Exemple 4:
Préparation
de 4-chloro-2-morpholinométhyl-1 ,3-diazafluoranthène-
I-oxyde et de son chlorhydrate
On fait réagir 0,6 g de 4-chloro-2-chlorométhyl-1,3- diazafluoranthène-1-oxyde avec 0,35ml de morpholine dans 35 ml de tétrahydrofurane. Le mélange est chauffé au reflux et agité pendant 2 heures. On obtient des cristaux orange jaune fondant à 172-174 C.
Le chlorhydrate de ce composé, préparé de la manière habituelle, fond à 223-2250 C (déc.).
Les composés préparés conformément au procédé selon l'invention exercent une action sédative sur le système nerveux central. Ils peuvent donc être utilisés, en thérapeutique humaine et animale, notamment comme sédatifs, hypnotiques, tranquillisants, anticonvulsivants, antiparkinsoniens, etc.
Process for the preparation of novel 2-substituted 1,3-diazafluoranthene 1-oxides
The present invention relates to the preparation of novel 2-substituted 1,3-diazafluoranthene 1-oxides corresponding to the following general formula:

in which R3 and Ra represent hydrogen or a lower alkyl group, R4 represents hydrogen or a halogen, such as chlorine in position 4 or 6, and

represents a nitrogenous heterocyclic group which may be substituted, such as a morpholino, piperidino, hexamethyleneimino, piperazino or substituted piperazino group.
Among the compounds of formula I, the following compounds, as well as their hydrochlorides, can be cited as an example:
2-piperidinomethyl- 1,3-diazafluoranthene-1 -oxide,
2-hexamethyleneiminomethyl- 1,3 -diazafluoroanthene-
1-oxide,
2-morpholinomethyl- 1,3 -diazafluoranthene- 1-oxide,
4-chloro-2-morpholinomethyl-1,3-diazafluoranthene oxide.
The new compounds of formula I are prepared from compounds of the following general formula:

in which Rs, RA and Ra have the meanings indicated above, while X denotes a halogen, such as chlorine, by reaction with a nitrogenous heterocyclic compound, preferably in the presence of tetrahydrofuran.
The compounds of formula (III) are also new compounds. Among these compounds of formula

2- (a-chloroethyl) - 1,3 -diazafluoranthene- 1-oxide,

The compounds of formula (V) can be obtained by reaction of hydroxylamine hydrochloride (NH2 OH. HCl) with new compounds which are the acylaminofluorenones of formula (VI). This reaction can be schematized as follows:

The compounds of formula VIII can be obtained by reaction of hydroxylamine hydrochloride (NH2OH. HCl) with an aminofluorenone of formula (IX) as indicated by the following scheme:

In these formulas, R4 represents hydrogen or a halogen such as chlorine or bromine.
The compounds of formula (IX) can also be used to obtain the compounds of formula (VI) by reaction with halides of formula (VII).

1-Aminofluorenone is a known compound, but 2-chloro-1-aminofluorenone and 4-chloro-1-aminofluorenone are new compounds which can be prepared by chlorinating 1-aminofluorenone by various methods. Thus, the chlorination can be carried out in chloroform in the presence of pyridine at a temperature between 00 C and 200 C. A mixture of 2-chloro-1aminofluorenone and 4-chloro-1 -aminofluorenone is obtained. These two compounds can be separated by chromatography or by fractional crystallization.
The preparation of various compounds of formula I will now be described in the following examples:
Example 1:
Preparation
2-piperidinomethyl-1,3-diazafluoranthene-1-oxid -oxide
and its hydrochloride
A mixture of 0.54 g of 2-chloromethyl-1,3-diazafluoranthene-1-oxide, 25 mol of tetrahydrofuran and 0.4 ml of piperidine is heated under reflux for 30 minutes. The suspension is then poured onto ice and the precipitate is filtered, washed, dried and recrystallized from cyclohexane: m.p. 155-1570 C.
The hydrochloride is prepared by adding a 2N solution of HCl in ethanol to a solution of the base in chloroform, m.p. 192-1940 C (decomposition) after recrystallization from ethanol.
Example 2:
Preparation
2-hexamethyleneiminomethyl-1,3-diazafluoranthene-oxide
A mixture of 0.54 g of 2-chloromethyl-1,3-diazafluoranthene-1-oxide, 30 ml of tetrahydrofuran and 0.45 ml of hexamethyleneimine is heated under reflux for 30 minutes. The suspension is poured onto ice and the precipitate obtained is filtered, washed and dried. Microcrystals are obtained, melting at 146 -148 C after several recrystallizations from cyclohexane.
Example 3:
Preparation
2-morpholinomethyl-1,3-diazafluoranthene-1-oxide
and its hydrochloride
A mixture of 0.5 g of 2-chloromethyl-1,3-diaza-fluoranthene-1-oxide, 2 mol of morpholine is stirred for 64 hours at room temperature in 20 ml of methanol, heated to reflux in 35 ml. of tetrahydrofuran for 1 hour. The solution is then concentrated to dryness and the residue is treated with water and filtered. The product obtained is recrystallized from methanol and melts at 168-170 ° C (orange crystals).
The hydrochloride is prepared by dissolving the base in methanol and adding an ethanolic solution of hydrochloric acid until the pH reaches 3.5-4. The mixture is then concentrated to dryness and recrystallized from a mixture of methanol and methyl ethyl ketone. The product melts at 199-2010 C as it decomposes. The yellow orange micro-crystals are soluble in water.
Example 4:
Preparation
4-chloro-2-morpholinomethyl-1, 3-diazafluoranthene-
I-oxide and its hydrochloride
0.6 g of 4-chloro-2-chloromethyl-1,3-diazafluoranthene-1-oxide is reacted with 0.35 ml of morpholine in 35 ml of tetrahydrofuran. The mixture is heated to reflux and stirred for 2 hours. Yellow orange crystals are obtained, melting at 172-174 C.
The hydrochloride of this compound, prepared in the usual manner, melts at 223-2250 C (dec.).
The compounds prepared in accordance with the process according to the invention exert a sedative action on the central nervous system. They can therefore be used in human and animal therapy, in particular as sedatives, hypnotics, tranquilizers, anticonvulsants, antiparkinson drugs, etc.
