Verfahren zur Herstellung von neuen Dibenzo-oxepinen, -thiepinen und -azepinen
Erfindungsgemäss werden neue Dibenz-oxepine, -thie- pine und -azepine hergestellt, welche psychotrope und neurotrope Wirkungen ausüben und der folgenden allgemeinen Formel entsprechen:

In der obigen Definition bedeutet der Ausdruck nie- der , wie er im Zusammenhang mit dem Alkanteil Ver wendung findet, eine gerade oder eine verzweigte Kette mit 1 bis 6 Kohlenstoffatomen.
Es wurde festgestellt, dass die Verbindungen der For mel I und deren pharmazeutisch annehmbare Salze, im allgemeinen psychotrope und neurotrope Wirkungen aus üben, wie z.B. beruhigende Wirkungen, entiemetische
Wirkungen und spasmolytische Wirkungen, dass sie fer ner eine verlängerte Schlafdauer bewirken, dass sie über dies eine antagonistische Wirkung gegen Apomorphin, eine Hemmwirkung gegen eine bedingte Ausweichreak tion, eine antagonistische Wirkung gegen Methamphetamin, eine Katalepsie und Ptose hervorrufende Wirkung, eine antagonistische Wirkung gegen Adrenalin und eine antagonistische Wirkung gegen Histamin ausüben, wobei die Wirkung als Tranquilizer besonders bemerkenswert ist.
Demzufolge bezieht sich die vorliegende Erfindung auf ein Verfahren zur Herstellung der Verbindungen der Formel I und deren Salze, welche wertvolle psychotrope und neurotrope Mittel, z.B. Tranquillizer, sind. Die Verbindungen der allgemeinen Formel I können erfindungsgemäss nach verschiedenen Methoden hergestellt werden.
Das erfindungsgemässe Verfahren besteht in der Umsetzung einer Verbindung der Formel II' mit einem se kundären Amin der Formel 1V wie durch das folgende Reaktionsschema wiedergegeben:

Die Ausgangsverbindungen der Formel II' lassen sich dadurch herstellen, dass man eine Verbindung der Formel

handelt. Das organische tertiäre Amin kann ebenfalls selbst als Lösungsmittel dienen. Die Reaktionstemperatur hängt vom Ausgangsmaterial der Formel (II') ab, sowie vom verwendeten basischen Kondensationsmittel und dem verwendeten Lösungsmittel, wobei die Umsetzung im allgemeinen im Bereiche des Siedepunktes des Lösungsmittels durchgeführt wird.
Gewünschtenfalls kann man erhaltene Verbindungen der Formel I' nach folgendem Reaktionsschema in Verbindungen der Formel II" überführen.
mit einer Verbindung der Formel: Y-A-Y', worin Y' einen sauren Rest, z.B. ein Halogen, wie z.B. Chlor, Brom oder Jod, einen Schwefelsäurerest oder einen Sulfonsäurerest, z.B. einen Methansulfonyloxy-, einen Toluol sulfonyloxy-, einen Benzolsulfonyloxyrest, bedeutet, je doch reaktionsfähiger als der Rest Y ist, in Gegenwart eines basischen Kondensationsmittels, wie z.B. Natriumhydrid, in einem polaren Lösungsmittel, wie z.B. Dimethylformamid, behandelt.
Die als Ausgangsmaterialien verwendeten Verbindungen II können in an sich bekannter Weise hergestellt werden [vgl. z.B. Chemical Abstracts, 62, 16215 - 16216 (1965); ibid., 63, 2952 (1966); J. Pharm. Soc. Japan, 74, 426 (1954)] oder nach einer analogen Methode.
So kann man beispielsweise lO-(3-Chlorpropyl)-oxy- dibenzo-[b,f]-thiepin, eine der Ausgangsverbindungen der Formel II', dadurch herstellen, dass man Dibenzo-[b,f] -thiepin-lO(l lH)-on mit l-Brom-3-chlorpropan in Gegenwart von Natriumhydrid in einer Mischung von Benzol und Dimethylformamid umsetzt.
Die Umsetzung erfolgt vorzugsweise dadurch, dass man die Verbindungen der Formel II' mit'einem Amin der Formel IV in Gegenwart eines basischen Kondensationsmittels, wie z.B. einem Alkalimetallhydroxyd, z.B.
Natriumhydroxyd oder Kaliumhydroxyd, einem Alkalimetallcarbonat, wie z.B. Natriumcarbonat oder Kaliumcarbonat, einem Alkalimetallbicarbonat, wie z.B. Die Umsetzung erfolgt im allgemeinen unter Erwärmen auf eine in der Nähe des Siedepunktes des Lösungsmittels liegende Temperatur.
Man kann aber auch erhaltene Verbindungen der Formel I" nach der folgenden Gleichung in Verbindungen der Formel I"' überführen:

Calcium, deren Hydroxyde, z.B. Natriumhydroxyd oder Kaliumhydroxyd, Amide, z.B. Natriumamid, Alkanolate, z.B. Natriumäthylat, Carbonate, wie z.B. Natriumcarbonat oder Kaliumcarbonat, oder Bicarbonate, wie z.B. Natriumbicarbonat oder Kaliumbicarbonat, oder einem organischen tertiären Amin, wie z. B. Pyridin, Triäthylamin, Picolin oder Lutidin, vorzugsweise in einem Lösungsmittel, wie z. B. Methanol, Äthanol, Äther, Benzol, Aceton, Dimethylformamid oder Dimethylsulfoxyd, behandelt.
Die Reaktionstemperatur hängt vom Ausgangsmaterial der Formel (II'), vom basischen Kondensationsmittel und vom verwendeten Lösungsmittel ab, wobei die Umsetzung vorzugsweise innerhalb des Siedebereiches des Lösungsmittels erfolgt.
Noch eine weitere mögliche, zusätzliche Massnahme wird durch die folgende Gleichung gezeigt:

Die Umsetzung erfolgt dadurch, dass man die Verbindung der Formel I" mit einem Reaktionsmittel der Formel VI, im allgemeinen in einem Lösungsmittel, wie z.B.
Methanol, Äthanol, Aceton, Chloroform, Dioxan, n Hexan, Benzol, Toluol oder Xylol, behandelt. Das Reaktionsprodukt besteht aus der Verbindung der Formel I4, welches mit der Verbindung I5 verunreinigt ist und sich leicht in an sich bekannter Weise in die einzelnen Komponenten trennen lässt.
Man kann auch erhaltene Verfahrensprodukte gemäss folgendem Reaktionsschema weiter umsetzen:

Die Umsetzung erfolgt dadurch, dass man die Verbindung der Formel I14 mit der Carbonsäure der Formel VII oder einem reaktionsfähigen Derivat derselben in einem Lösungsmittel, wie z.B. Äther. Aceton, Dioxan, Acetonitril, Chloroform, Äthylenchlorid, Tetrahydrofuran, Äthylacetat oder Pyridin, nötigenfalls in Gegenwart eines basischen Materials, wie z.B. eines Alkalihydroxyds, beispielsweise Natriumhydroxyd oder Kaliumhydroxyd, eines Alkalicarbonats, z.B. Erhaltene Verbindungen der Formel I8 oder I10 können zur Herstellung weiterer Verbindungen der Formel 1' oder I1l gemäss folgendem Reaktionsschema verwendet werden:

Gewünschtenfalls kann man die erhaltenen Verbindungen der Formel I in ihre pharmazeutisch annehmbaren Salze nach an sich bekannten Methoden überführen.
Beispiele von Salzen mit Mineralsäuren, sind die Chlorhydrate, Sulfate, Nitrate, Phosphate, mit organischen Säuren, die Acetate, Propionate, Succinate, Oxalate, Maleate, Tartrate, Fumarate, Citrate oder Picrate.
Wie bereits erwähnt, besitzen die Verbindungen der Formel I und deren Salze psychotrope und neurotrope Wirkungen. So besitzen sie eine schlafverlängernde Wirkung, eine antagonistische Wirkung gegen Apomorphin, eine Hemmwirkung gegen eine bedingte Ausweichreaktion, eine antagonistische Wirkung gegen Methamphetamin, eine Katalepsie und Ptose bewirkende Wirkung, eine antagonistische Wirkung gegen Adrenalin, eine antagonistische Wirkung gegen Histamin und eine antagonistische Wirkung gegen Acetylcholin.
Zufolge der vorerwähnten Wirkungen sind die Verbindungen der Formel I und deren nicht toxische Salze wertvolle Tranquillizer.
Die Verbindungen der Formel I und deren nicht toxische Salze können nach an sich bekannten Methoden verabreicht werden. Dabei kann man die üblichen Einheitsdosierungen und die üblichen pharmazeutischen Trägerstoffe verwenden, um eine beruhigende Wirkung für den Menschen zu erzielen.
So kann man sie in Form von pharmazeutischen Präparaten verwenden, welche diese Wirksubstanzen in Beimischung mit einem pharmazeutischen, organischen oder anorganischen Trägermaterial, welche sich für die enterale oder parenterale Verabreichung eignet, enthalten. Die orale Verabreichung in Form von Tabletten, Kapseln oder in Form einer Flüssigkeit, z.B. Suspensionen, Lösungen oder Emulsionen, ist besonders vorteilhaft. Werden Tabletten geformt, so kann man die für therapeutische Einheitsdosierungen üblichen Bindemittel und desintegrierende Mittel verwenden. Beispiele von Bindemitteln sind Glucose, Lactose, Pflanzengummi, Akaziengummi, Gelatine, Mannitol, Stärkepaste, Magnesiumtrisilikat und Talg. Als desintegrierende Mittel kann man z.B. Maisstärke, Keratin, kolloidales Siliciumdioxyd und Kartoffelstärke verwenden.
Werden die Präparate in Form von Flüssigkeiten verabreicht, so kann man die üblichen Trägermittel für Flüssigkeiten verwenden.
Die Dosierung bzw. therapeutisch wirksame Menge der Verbindungen der Formel I und der nicht toxischen Salze davon für den Menschen kann innerhalb weiter Grenzen, z.B. von ca. 30 mg bis ca. 1000 mg pro Tag und pro Erwachsener, schwanken. Der obere Grenzwert wird lediglich durch das Ausmass der erwünschten Wirkung und durch wirtschaftliche Erwägungen begrenzt. Für die orale Verabreichung wird man im allgemeinen ca. 10 mg bis ca. 300 mg des therapeutischen Mittels pro Einheitsdosierung verwenden. Natürlich kann die Dosierung des zu verwendenden, bestimmten therapeutischen Mittels sehr stark schwanken und dies je nach dem Alter des Patienten und nach dem Grad der gewünschten therapeutischen Wirkung. Jede Einheitsdosierung der neuen therapeutischen Verbindungen kann ca. 5 Gew.-% bis ca.
95 Gew.-O neues therapeutisches Mittel, bezogen auf das Gewicht des gesamten Präparates, enthalten, wobei der Rest aus üblichen pharmazeutischen Trägermitteln besteht. Der Ausdruck pharmazeutische Trägermittel soll nicht therapeutische Materialien umfassen, welche üblicherweise bei Einheitsdosierungen Verwendung finden, wie z.B. Füllstoffe, Verdünnungsmittel, Bindemittel, Schmiermittel, desintegrierende Mittel und Lösungsmittel.
Selbstverständlich kann man die neuen therapeutischen Mittel auch in Form der reinen Verbindungen ohne Verwendung eines pharmazeutischen Trägermittels anwenden.
Die Erfindung sei durch die folgenden Beispiele er läutert.
Beispiel I [A] in Eine Suspension von 0,97 g Natriumhydrid in einer Mischung von Benzol und Dimethylformamid gibt man unter Kühlen mit Eis tropfenweise und unter Rüh ren eine Lösung von 4,52 g 10,11 -Dihydro- 10-oxodibenzo- -[b,f]-thiepin in Dimethvlformamid hinzu. Das erhaltene
Gemisch wird dann ca. während 1 Stunde bei 50 C ge rührt. Nach der tropfenweisen Zugabe einer Lösung von
3,2 g 1-Brom-3-chlorpropan in Dimethylformamid wird das erzielte Gemisch während 48 Stunden bei 50 C ge rührt. Das Reaktionsgemisch wird in Wasser gegossen und mit Benzol extrahiert. Der Benzolextrakt wird mit
Wasser gewaschen, über Magnesiumsulfat getrocknet und eingeengt, wobei man 5,7 g eines roten Öls erhält.
Das öl wird einer Siliciumdioxydgelchromatographie unter- worfen und mit Benzol eluiert. Das Eluat wird eingeengt und aus Äthanol umkristallisiert, wobei man 3,1 g 10-(3 -Chlorpropyl)-oxydibenzo-[b,f]-thiepin in Form von gel ben Körnern vom Schmelzpunkt 125 bis 126 C erhält.
Eine Mischung von 270 g 10-(3-Chlorpropyl)-oxydi- benzo-[b,f]-thiepin und 3,0 g 1-Methylpiperazin wird während 19 Stunden bei 120 bis 125 C gerührt. Das Re aktionsaemisch wird in Wasser gegossen und die Lösung mit Äther extrahiert. Die Atherschicht wird nach dem Waschen mit Wasser mit 10NOiger Salzsäure extrahiert.
Die wässrige Schicht wird mit Äther gewaschen, dann mit 10%iger Natriumhydroxydlösung alkalisch gestellt und hierauf mit Äther extrahiert. Die ätherische Schicht wird mit Wasser gewaschen und der Äther abdestilliert, wobei man 03 g eines öls erhält. Dieses öl wird mittels Maleinsäure in 0,32 g 10-[3-(4-Methyl-l-piperazinyl)-pro- pyl]-oxydibenzo-[b,f]-thiepin übergeführt, das man in Form eines farblosen Pulvers erhält. Dieses Pulver wird aus wässrigem Äthanol umkristallisiert, wobei man farb lose Nadeln vom Schmelzpunkt 195 bis 197 C erhält.
[B] Eine Mischung von 0,3 g 10-(3-Chlorpropyl)-oxy- dibenzo-[b.f]-thiepin und 3,0 g 1-(2-Hydroxyäthyl)-piper- azin wird während 17 Stunden bei 130 C gerührt. Das Reaktionsgemisch wird in der gleichen Weise wie in Ab satz [A] behandelt, wobei man 0.15 g 10-[3-(4-(2-Hy- droxyäthyl)-l - piperazinyl)-propyl] - oxydibenzo-[b,f]-thie- pin erhält. Das Maleat liefert farblose Nadeln vom Smp.
169 bis 170 C.

Beispiel 2 [A] Eine Lösung von 0,9 g 8-Chlor-10-[2-(4-äthoxy- carbonyl-1-piperazinyl)-äthyl]-oxydibenzo-[b,f]-thiepin in 40 cm3 Tetrahydrofuran wird tropfenweise zu einer Lö sung von 0,5 g Lithium-Aluminiumhydrid in 5 cm3 Tetrahydrofuran zugesetzt, worauf man das Gemisch während 17 Stunden unter Rückfluss erhitzt. Dem Reaktionsgemisch gibt man nach dem Kühlen 50 cm3 Äther, 2 cm3 Äthylacetat, dann nach weiteren 30 Minuten 50 cm3 Äther und hierauf 5 cm3 Wasser hinzu. Die ätherische Schicht wird nach dem Waschen mit Wasser mittels 10%iger Salzsäure extrahiert. Die wässrige Schicht wird mit 100/,iger Natriumhydroxydlösung neutral gestellt, mit Äther extrahiert und der Extrakt über Magnesiumsulfat getrocknet und eingeengt.
Der Rückstand wird in einer kleinen Menge Äthylacetat gelöst, worauf man die Lösung durch eine mit Aluminiumoxyd und Äthylacetat beschickte Kolonne leitet. Das erste Eluat wird eingeengt, wobei man 0,4 g 8-Chlor-l0-[2-(4-methyl-l-piperazinyl)- -äthyl]-oxydibenzo-[b,f]-thiepin in Form eines gelben Öls erhält. Dieses Öl wird in das entsprechende Maleat vom Schmelzpunkt 178 bis 1 800C übergeführt, [B] In der gleichen Weise wie im Absatz [A] werden 1,2 g 8 - Chlor -10-[2-(4-äthoxycarbonyl-1-piperazinyl) -äthyl]-oxydibenzo-[b.f]-oxepin behandelt, um 8-Chlor - 1 0-[2-(4 - methyl - 1 -piperazinyl) - äthyl]-oxydibenzo-[b,f] -oxepin in Form eines gelben Öles zu erhalten.

Bemerkung: entspricht dem Verfahren (I') < (II").
Eine Lösung von 3,0 g Natriumhydroxyd in 3 cm3 Wasser wird zu 3.3 g 8-Chlor-10-[2-(4-formyl-1-piperazi- nyl)-äthyl]-oxydibenzo-[b,f]-thiepin, gelöst in einer Mischung von 50 cm3 99%igem Äthanol und 10 cm3 Wasser, hinzugegeben und das Gemisch während 3 Stunden unter Rückfluss erhitzt. Die Hälfte des Volumens an Äthanol im Reaktionsgemisch wird durch Verdampfen entfernt. Der Rückstand wird mit Wasser versetzt und die wässerige Lösung mit Äthylacetat wiederholt extrahiert. Die Äthylacetatschicht wird getrocknet und hierauf eingedampft, um das Äthylacetat zu beseitigen, wobei man 1,5 g 8-Chlor-10-[2-(1-piperazinyl)-äthyl]-oxydiben- zo-[b,f]-thiepin in Form eines gelben Öles erhält, welches in das entsprechende Maleat umgewandelt wird.
Dieses wird aus einer Mischung von Äthanol und Äther umkristallisiert, wobei man 8-Chlor-10-[2-(1-piperazinyl)- -äthyl]-oxydibenzo-[b,f]-thiepin-maleat vom Smp. 1531560C (Zers.) erhält.
Beispiel 4
Bemerkung: entspricht dem Verfahren [I"] < f14] und [I].
Zu einer Lösung von 0,1 g 8-Chlor-10-[2-(1-piperazi nyl)-äthyl]-oxydibenzo- [b,f]-thiepin in 15 cm3 Methanol wird ein Überschuss an Propylenoxyd hinzugegeben und das Gemisch während 1 Stunde auf dem Wasserbade unter Rückfluss erhitzt. Das Methanol wird durch Verdampfen entfernt und der Rückstand mit Wasser versetzt. Die Lösung wird mittels Äthylacetat extrahiert. Die Äthylacetatschicht wird getrocknet und hierauf eingeengt, wobei man ein Öl erhält.
Dieses öl wird in das entsprechende Maleat in an sich bekannter Weise übergeführt, wobei aus Methanol umkristallisiert, 0,1 g blassgelbe Kristalle vom Smp. 162- 1640C (Zers.) erhalten werden, welche aus 8 - Chlor - 10 - [4 - (2 - hydroxypropyl) -1-piperazinyl- -äthyl]-oxydibenzo-[b.f]-thiepin-maleat, verunreinigt mit 8-Chlor- 10 - {2 - [4 - ( I -methyl-2-hydroxyäthyl)- 1 -piperazi nyl]-äthyl) -oxydibenzo-[b,f] -thiepin-maleat, bestehen.

Beispiel 5
Bemerkung: entspricht dem Verfahren [110] < [P1].
[A] Zu einer Lösung von 0,15 g 8-Chlor-10-[2-(1 -piperazinyl)-äthyl]-oxydibenzo-[b,f]-thiepin in 15 cm3 Dimethylacetoamid gibt man 0,3 g Äthylbromacetat und 5 cm3 Triäthylamin hinzu. Das Gemisch wird während ] Stunde bei 90 bis 950C gerührt. Dann wird das Reaktionsgemisch in einen Überschuss an Wasser gegossen und die Lösung mittels Äthylacetat wiederholt extrahiert. Die Äthylacetatschicht wird getrocknet und das Äthylacetat durch Verdampfen enfernt, wobei man 8-Chlor-10-[2-(4 -(äthoxycarbonylmethyl- 1 -piperazinyl)-äthyl]-oxydibenzo- -[b,f]-thiepin in Form eines Öls erhält, welches man in das entsprechende Maleat in an sich bekannter Weise überführt.
Dieses letztere wird aus Äthanol umkristallisiert, wobei man 8-Chlor-10-[2-(4-äthoxycarbonylmethyl - 11 piperazinyl)-äthyl] - oxydibenzo - [b,f] -thiepin- maleat vom Smp. l65-l660Cerhält.
[B] Zu einer Lösung von 0,25 g 8-Chlor-10-[2-(1 -piperazinyl)-äthyl]-oxydibenzo-[b,f] -thiepln in 20 cm3 Dimethylacetoamid gibt man 0,5 g Äthyl-a-brompropionat hinzu und rührt das Gemisch während 1 Stunde bei 90-950C. Das Reaktionsgemisch wird in einen Überschuss von Wasser gegossen und die Lösung mit Äthylacetat extrahiert. Die Äthylacetatschicht wird mit Wasser gewaschen, getrocknet und mit Aktivkohle behandelt, worauf man das Äthylacetat durch Eindampfen entfernt.
Auf diese Weise erhält man 0,2 g 8-Chlor-10-t2-[4-(1-äth- oxycarbonyläthyl)- 1 -piperazinyl]-äthyl )-oxydibenzo- [b,f]- -thiepin in Form eines Öls, das man in das entsprechende Maleat überführt. Dies letztere wird aus Äthanol umkristallisiert, wobei man 8-Chlor- 10- {2-[4-(1 -äthoxycarbo- nyläthyl)-1 - piperazinyl]-äthyl} -oxydibenzo-[b,f]-thiepin -maleat vom Smp. 153-1540C erhält.
Andere Verbindungen lassen sich in ähnlicher Weise herstellen, beispielsweise: 1 8-Chlor- 10- { 2- [4 - (2-acetoxyäthyl) - 1 piperazinyl] -äthyl}oxydibenzo[b,f]thiepin, Smp. 174-1760C (Maleat); 2 8-Chlor- 1 0-[2-(4-methyl- 1 -piperazinyl)äthyl]oxydiben zo[b,f]oxepin, Smp. 178-1800C (Maleat); 3 1 0-[3 - (4-Methyl- 1 -piperazinyl)propyl]oxydibenzo[b,f] - -thiepin, Smp. 195-1970C (Maleat).
Beispiel 6
Eine Lösung von 0,5 g Acetylchlorid in 10 cm3 Äther wird tropfenweise 0,3 g 8-Chlor-lO- (2-[4-(2-hydroxy- äthyl)-l-piperazinyl]-äthyl}-oxydibenzo-[b,f]-thiepin, gelöst in einer Mischung von 30 cm3 Tetrahydrofuran und 5 cm3 Triäthylamin, zugesetzt. Dieses Gemisch wird während 2 Stunden bei Zimmertemperatur stehengelassen und dann während 2 Stunden bei 400C umgesetzt. Das Reaktionsgemisch wird filtriert, das Filtrat unter vermindertem Druck eingeengt und der Rückstand mit Wasser versetzt. Dann wird die Lösung mit einer Mischung von Benzol und Äther extrahiert. Der Extrakt wird mit Wasser gewaschen, getrocknet und hierauf die Lösungsmittel verdampft, wobei man 0,2 g 8-Chlor-10-(2-[4-(2-acetoxy- äthyl) - 1 - piperazinyl] - äthyl} oxydibenzo-[b,f]-thiepin in Form eines Öles erhält.
Dieses öl wird in an sich bekannter Weise in das entsprechende Maleat übergeführt. Dieses letztere wird aus 99%igem Äthanol umkristallisiert, wobei man 8-Chlor- 10- {2-[4-(2-acetyloxyäthyl) - 1 -piper azinyl]-äthyl } -oxydibenzo-[b,f] -thiepin-maleat vom Smp.
174-1760C (Zers.) erhält.
Beispiel 7 [A] 0,1 g 8-Chlor- 1 0-[2-(4-äthoxycarbonylmethyl- 1 - -piperazinyl)-äthyl]-oxydibenzo-[b,f]-thiepin, gelöst in 20 cm3 trockenem Äther, wird tropfenweise bei Zimmertemperatur einer Suspension von 200 mg Lithium -Aluminiumhydrid in 20 cm3 Äther hinzugegeben.
Das Gemisch wird während 1 Stunde gerührt und hierauf während 1 Stunde unter Rückfluss erhitzt.
Hierauf wird das überschüssige Volumen an Lithium -Aluminiumhydrid im Reaktionsgemisch durch Zugabe einer kleinen Menge von Äthylacetat und hierauf von 5 cm3 Wasser zersetzt. Die ätherische Schicht wird abgetrennt und getrocknet. Der Äther wird durch Verdampfen entfernt, wobei man 8-Chlor-10-{2-[4-(2-hydroxy äthyl)-l - piperazinyl] - äthyl}-oxydibenzo-[b,f]-thiepin in Form eines öles erhält, welches man in das entsprechende Maleat umwandelt. Dabei erhält man Kristalle vom Smp. 147- 147,50C (Zers.).
[B] 0,2 g 8-Chlor-10-{2-[4-(1-äthoxycarbonyläthyl)- - 1 -piperazinyl]-äthyl}-oxydibenzo-[b,f]-thiepin, gelöst in einer Mischung von 20 cm3 Äther und 20 cm3 Tetrahydrofuran, wird tropfenweise bei Zimmertemperatur einer Suspension von 0,2 g Lithium-Aluminiumhydrid in 25 cm3 Äther hinzugegeben, worauf man das Gemisch in der gleichen Weise wie sub [A] behandelt. Auf diese Weise erhält man 0,1 g 8-Chlor-10-{2-[4-(1-methyl-2-hy- droxyäthyl)-1-piperazinyl]-äthyl}-oxydibenzo-[b,f]-thie- pin-maleat vom Smp. 150-1510C (Zers.).
Andere Verbindungen lassen sich in an sich ähnlicher Weise herstellen. Es handelt sich beispielsweise um die folgenden Verbindungen: 10- {3-[4-(2-Hydroxy-äthyl) - 1- -piperazinyl] - propyl)- oxydibenzo - [b,f] - thiepin- maleat, farblose Nadeln vom Smp. 169-1700C; 5-Methyl-5H-10 - {3 -(4-(2-hydroxyäthyl)- 1 -piperazinyl]-propyl} - oxydibenzo-[b,f]-azepin, usw.

The compounds of the formula I and their non-toxic salts can be administered according to methods known per se. The usual unit doses and the usual pharmaceutical excipients can be used to achieve a calming effect for humans.
They can thus be used in the form of pharmaceutical preparations which contain these active substances in admixture with a pharmaceutical, organic or inorganic carrier material which is suitable for enteral or parenteral administration. Oral administration in the form of tablets, capsules or in the form of a liquid, e.g. Suspensions, solutions or emulsions, is particularly advantageous. When tablets are formed, the binders and disintegrating agents customary for therapeutic unit doses can be used. Examples of binders are glucose, lactose, vegetable gum, acacia gum, gelatin, mannitol, starch paste, magnesium trisilicate and tallow. Example 7 [A] 0.1 g of 8-chloro-1 0- [2- (4-ethoxycarbonylmethyl- 1 - -piperazinyl) -ethyl] -oxydibenzo [b, f] -thiepin, dissolved in 20 cm3 of dry ether, is added dropwise at room temperature to a suspension of 200 mg lithium aluminum hydride in 20 cm3 ether.
The mixture is stirred for 1 hour and then refluxed for 1 hour.
