Method and device for separating the components of a mixture
The invention relates to a method and a device for separating the components of a mixture by means of different phases, in particular for separating mixtures of substances by chromatography.
An object of the invention is to carry out the separation of a mixture by means of different phases in a circuit of finite length in such a way that an infinitely long circuit is used for the separation of a sample. The invention also relates to an improved process for the continuous separation of mixtures which has a better separation efficiency compared to the known methods.
It is already known to have a mixture of gases or liquids to be separated and a mobile one
Continuously driving phase through a column of solid phase in such a way that the opposite ends of the column are connected by a pump, which has the same effect as a separation column of infinite length. In such a process, however, at least part of the separation effect achieved with each passage of the mixture through the column is lost due to the remixing caused by the pump.
The invention eliminates this disadvantage in that the mobile phase is moved in the opposite direction to the stationary phase, namely at such a speed that the mixture to be separated remains within the column and only the molar phase is passed through the pump. For this purpose, the material of the stationary phase is introduced into a closed ring column through which the mobile phase is driven in a single direction.
This avoids remixing as in the known method and achieves a continuous separation effect, so that the separation efficiency achieved in a certain time is significantly greater.
In the continuous separation of mixtures by chlorombatography, it is known to circulate the stationary phase of a separating column of endless form through this column and to introduce a stream of carrier gas into this column so that the mobile and stationary phases flow in the same direction over a section of the column and over another section of the column, the two phases flow in countercurrent direction.
Since only the passage on the countercurrent principle is effective for the separation, either a considerable part of the introduced carrier gas is not used or a substantial part of the column length is not always available for the separation with the result that larger quantities and / or greater gas flow rates are required than the theoretical calculation results, and / or a much longer column than theoretically calculated is required for such a method.
In order to avoid these disadvantages, the method for separating the components of a mixture by chromatography by means of a closed, annular column-forming adsorbent through which a carrier flows, is characterized according to the invention in that the Ko ton moves in one direction of rotation, whereby it is not effective for separating the mixture over a certain distance of its orbit and at the same time is blocked from flowing through the carrier, so that the entirety of the carrier is forced to flow in the opposite direction to the direction of rotation over most of the ring through the column, which Part for the separation of the mixture is available.
To carry out the method, a device is used which is characterized by a rotatably mounted wheel with a tube forming a ring over its circumference for receiving the adsorbent, with organs for supplying and removing the carrier, the mixture or the fractionated components to or .
Sangearranged distributed by the ring, further by subdivisions made in the ring, which divide the ring into circular sections, the size of which is smaller than the distance between the access point and the removal point for the wearer, with adjacent circular sections being connected to each other by an overflow valve, and by means which control these overflow valves at least partially during their passage between the supply and extraction elements in the closed position, these means not influencing the overflow valves on their remaining movement path, d. H. leave it in the open position.
With a shorter column, continuous separation and thus greater separation efficiency can be achieved.
The drawing shows an exemplary embodiment of the subject matter of the invention, on the basis of which the method according to the invention is also explained.
Show it:
1 shows a perspective view with partial section of a device for the chromatographic separation method, which is suitable both for the separation of samples and for a continuous separation,
2 shows a cross section through part of the distribution ring,
3 shows a section through the lubricating device and
4 shows a combined device in view.
The device shown in FIGS. 1, 2 and 3 consists of the base plate 10 with the frame 11 on which the wheel 12 is rotatably mounted, the circumference of which is formed by a tube 13 running around it.
This tube is divided by subdivisions 14 into eight circular sections of equal size. Each section of a circle contains a spirally wound heat exchanger 15 and is made with a granular substance 16. Each circular section is connected to the adjacent section by an overflow valve 17, the lines 18 opening into openings 19 of the pipe on both sides of the partitions 14. Each overflow valve 17 has a control member 20 which slides on the stationary cam 21, whereby the opening and closing of the overflow valve is controlled during the rotational movement of the wheel, as described later.
The two ends of each heat exchanger 15 are connected to the inlet and outlet openings of liquid control valves 22 via lines 23.
From each valve 22 two inlet lines 24 and two outlet lines 25 lead to the central manifold 26, which is accommodated in the wheel hub, which manifold is in communication via the rotating sealing sleeves 27 and the lines 28 with two independent reservoirs of the heat exchange media. Each valve 22 has two control members 30 and 31, which are on the stationary cam rings 32, respectively. 33 slide, the cams being designed so that they control the control members in such a way that each valve 22 alternately connects the reservoir of one and the reservoir of the other heat exchange medium with the heat exchanger 15 assigned to it, as the wheel rotates.
The cam 21 and the Nooken rings 32 and 33 are combined to form a ring 34, which ring is attached to the non-rotating cover plate 36. The cam 21 is arranged on one side of the ring 34 and the cam rings 32 and 33 on the opposite side of this ring. The cover plate 36 forms the lateral end of the wheel 12, the ring 34 protruding into the wheel.
On the stationary cover plate 36, four connections 37A, 37B, 37C and 37D are also attached, each of which has a thumb 38 which, when the wheel 12 rotates, successively actuates the valve control members 39, which are located at closely spaced intervals on the outside of the pipe 13 are arranged. The connections 37 also have two openings which correspond to two circles of valve openings 40 on the front of the pipe.
The two interconnected openings of the connections 37 are advantageously circular arc-shaped slots that are long enough to bridge the distance between adjacent valve openings 40, so that between one or the other opening of the connection 37 and the interior of the tube 13 via a There is a permanent connection to the valve opening, the valve being temporarily moved into the open position by the thumb 38.
The four connections 37 are distributed around the wheel at intervals of approximately 900 and are used for introducing and removing the mixture to be separated, the separated mixture components and the carrier gas, depending on the working method of the device, as will be described later. For the rotation of the wheel clockwise at a certain speed, a variable speed drive (not shown) is provided so that the solid phase in the tube 13 - with respect to the stationary connections 37 - is constantly in motion in the clockwise direction.
If the device is used to separate a sample or a pattern, it works as follows: The supply line 41A of the connection 37A (on the right side of the wheel in FIG. 7) is connected to a pressurized carrier gas supply, and a similar supply line at the lowest connection 37D leads back to the carrier gas supply via a pump.
When the pump is in operation, a stream of carrier gas flows constantly through the pipe 13 between the points A and D. So that this gas stream only flows in the direction opposite to the direction of rotation of the wheel, the cam plate 21 closes each overflow valve 17 one after the other, when it passes by position A, the location of the gas-supplying connection 37A, and opens this valve again as soon as the next valve 17 that passes by position A has been closed or at the latest when the valve that was closed first is closed Point D is approaching.
Each valve 17 remains open for the remainder of each revolution of the wheel, so that only the short arc between points A and D is not traversed by carrier gas.
The sample to be separated or the mixture pattern to be separated is introduced into the gas stream in a known manner, namely before the entry of the carrier gas at point A. The speed of rotation of the wheel in relation to the flow rate of the carrier gas through the pipe 13 is regulated so that the sample is kept in the same angular position with respect to the wheel hub until the desired separation of the various components of the sample is complete.
The control of the position of the sample and the degree of separation is expediently carried out in the overflows with the help of the subdivisions 14 via the beäckead valves 17. A separation detector of a known type (not shown), consisting of an oscilloscope or chart recorder for the separation peaks and minima , which indicates the concentration of the various components, be arranged to detect the presence and separation of the sample when the overflow assumes the angular position at which the sample is to be held, whereby the rotational speed of the wheel and / or the flow rate of the carrier gas can be changed
if the detector indicates an undesired displacement of the sample, and the process is stopped when the detector detects a sufficient degree of separation.
During the process described with a sample, both supplies of heat exchange media, which can be water, for example, are kept at the same temperature so that the entire solid phase column in tube 13 is kept at the desired temperature and the action of valves 22 during the separation is almost irrelevant.
Under certain conditions, however, the temperature in the pipe sections can be increased as they pass in certain zones of the orbit in order to support the removal of the separated components from the column after separation. This temperature increase is achieved by the optional actuation of the valves 22, whereby the exchanger 15 is supplied with heat accumulation media of different temperatures.
In general, it is sufficient if the rotation of the wheel is interrupted and the connection 37D is connected to a suitable collecting device at point D and waiting until the carrier gas brings the separated components one after the other in this collecting direction.
One advantage of this chromatographic separation method is that the various components of the mixture sample injected into column i are carried along by the adsorbent in a clockwise direction of rotation and by the flow of the carrier gas in a counterclockwise direction, so that a spatial separation of the individual components is achieved over a certain distance of the column .
In the continuous process, as in the test process, the carrier gas is introduced at point A and sucked off at point D, but the mixture to be separated is fed in at point C and the connections at points D and B are each with separate collection facilities for the individual mixture fractions connected. These extraction devices are designed in such a way that the carrier gas can be separated out and returned to the circulation circuit.
The valves 22 serve via the rotating sealing sleeve on one side of the wheel hub as a connection to the supply of a relatively cold heat exchange medium and via the rotating sealing sleeve on the other side of the hub as a connection to the supply of a relatively hot heat exchange medium. The cam rings 31 and 32 influence the valves 22 in such a way that the hot medium flows through each exchanger 15 in sequence when it is in the section between B and A, and that on the other part of the orbit cold medium flows through these exchangers flows.
The speed of rotation of the wheel is adjusted in relation to the flow rate of the carrier gas through the pipe 13 so that the one, faster, component fraction of the mixture is carried by the carrier gas in the counterclockwise direction of rotation from C to D and is not carried sufficiently clockwise by the material of the solid phase that it could come out through the connection at point B,
while another, slower, component fraction is carried clockwise from C to B by the material of the solid phase and is not carried enough counterclockwise by the carrier gas that it could emerge through the connection at point D. The way in which the valves 17 are operated, which allows the gas flow to flow in only one direction, is the same as when using a sample.
In order to ensure that the desired component exits the connection at point B, the valves 22 cause the quarter circle between B and A to act as a skimming zone, whereby the hot medium is introduced into the exchange chunks 15 while they traverse this distance, This guarantees that the adsorbed component is separated from the material of the solid phase and carried along by the gas in order to exit at point B. The two desired fractions of the original mixture are continuously separated from the carrier gas in the collecting devices connected at B and D.
Instead of the heat exchanger 15, other means can be used for heating the separation zone. For example, inductive heat could be generated in this sector by installing a conductive substance in the pipe filling.
The heating of the exchangers could be supplemented by the additional heating of the carrier gas before it is introduced into the column. Likewise, the carrier gas could be passed through a heat exchanger at each of the overflow valves between the gas inlet and outlet points in order to regulate the temperature of the gas accordingly.
The choice of material for the filling or packing of the tube 13 is made with regard to the purposes to be achieved by the device. It has been found, however, that diatomaceous earth particles that are coated with a liquid with selective adsorption properties are particularly suitable for gas-liquid chromatography. Such a filling was used in the device described, the wheel diameter being 150 cm and the inner diameter of the pipe being about 38 mm. In this way, very remarkable separation capacities were achieved in comparison to the known apparatus. For gas-solid chromatography, an adsorbent, such as. B.
activated carbon or aluminum oxide, can be used as filling or packing of the tube.
Advantageously, the inner surface of the pipe 13 is thermally insulated, or the wall of the pipe consists of a thermally insulating material in order to make it easier to raise or lower the temperature in any desired zone within the separation column without excessive waste of heat on the adjacent column sections or on the apparatus itself, and to reduce the heat loss of the apparatus to the surrounding atmosphere.
In the continuous process, an additional port 37C 'can be provided in the vicinity of the port through which the G mixture is injected and on the side to which the slower fraction is to be carried, i. H. that is, just behind the mixture feed connection 37C in the direction of movement of the column 13.
The function of this additional port 37C 'is that normally a small amount of carrier gas can be injected into each valve 40 after the mixture has flowed through the valve so that it can be purged of the mixture before it can be used as an outlet valve for the separate component is used. Furthermore, separate valves 39 can be provided for the inlet and outlet in order to prevent contamination of the emerging fractions.
In the continuous process, the device can furthermore be equipped with a connection 37E between the connection provided for the outlet 37D of the faster fraction and the connection provided for the supply 37A of the carrier gas. Such an additional connection must be attached in such a way that there is always a closed valve 17 or other means to prevent the gas from flowing in the column between this connection and the connection 37A for the carrier gas feed.
This additional connection 37E is used for the re-injection of parts of a mixture of carrier gas and faster fraction, which are already from the connection; for the fast fraction, or for injecting some other mixture that is relatively rich in a fraction to enrich it.
The uniform distribution of the mixture to be separated or of the carrier over the cross section of the column 13 can be improved by attaching distributors (FIG. 2) in the form of tubes 40 'provided with holes, which are connected to the valves, which is made even easier by the fact that the filling 16 is stationary with respect to the wall surrounding it.
Means 42 are expediently provided for lubricating the valve control elements 39 arranged on the outside of the pipe 13. For this purpose, the element according to FIG. 3, consisting of a grease nipple 43, is used to inject grease into the space between the two parallel O-rings 44 and 45, through which the valves pass one after another, delivers.
It is possible to arrange several separation columns on a single axis and to use this combination for the separation of the components of complex mixtures. The combination of columns on a single axis can also serve to increase the absolute length of the column effective for the separation without increasing its cross-section.
4 shows such a combination, in which six wheels 112, 212, 312, 412, 512 and 612 are rotatably mounted on the same shaft 127. A circular tube 116, 416, 616 is arranged on its circumference.
The tubes 116 to 616 are connected to one another in series, with connections for the supply and removal of the carrier being arranged. 137A is a connection for the supply of carrier gas, 437C is a connection for the supply of the mixture that is to be separated into heavier and lighter fractions in the continuous process. The outlet connection 137B serves to discharge the slower fraction and part of the carrier gas and the connection 637D serves to discharge the carrier gas residue and the faster fraction.
