Verfahren zur Herstellung von N-distlbstituierten 3-Aniino-1,2-benzisothiazol-Derivaten
In der Literatur sind bisher nur N-monosubstituierte 3-Amino-1,2-benzisothiazole beschrieben worden, die nach der deutschen Auslegeschrift 1174783 durch Umsetzung von 3-Chlor-1,2-benzisothiazol mit basisch alkylierten Aminen gewonnen wurden. Ferner konnten N-monosubstituierte 3-Amino- 1,2-benzisothiazole durch Umsetzung von N-Alkyl- bzw. N-Aryl-3 -chlor- 1,2-benz- isothiazoliumhalogeniden mit Ammoniak dargestellt werden [Chem. Ber. 99, 2566, (1966)].
Gegenstand der Erfindung ist ein Verfahren neuartiger N-disubstituierter 3-Amino-l 2-benzisothiazole.
Es wurde gefunden, dass man neuartige N-disubstituierte 3-Amino-1,2-benzisothiazol-Derivate erhält, wenn man N-monosubstituierte 3-Amino- 2-benzisothiazole der allgemeinen Formel

worin R für Wasserstoff gleiche oder verschiedene Alkyl-, Alkoxy oder Halogen-Reste sowie ein Rest R auch für NO2 stehen kann, n eine ganze Zahl von 1 bis 3 bedeutet, und R1 einen gegebenenfalls substituierten aliphatischen, araliphatischen oder Aryl-Rest bedeutet, mit einem Alkylierungsmittel oder Aralkylierungsmittel der allgemeinen Formel R2-X worin R2 für einen Alkyl- oder Aralkylrest steht und X Chlor, Brom, Jod oder die Gruppierung -O-SO2-Phenyl bzw. -O-SO2-Tolyl bedeutet, umsetzt.
Als Alkyl- bzw. Alkoxy-Reste (R) seien vorzugsweise solche mit 1 bis 4 Kohlenstoffatomen, als Halogene (R) Fluor, Chlor, Brom, Jod, vorzugsweise jedoch Chlor und Brom genannt. Als gegebenenfalls substituierte aliphatische Reste (R1) sind geradkettige oder verzweigte, gegebenenfalls eine Doppelbindung enthaltende Alkylreste mit 1 bis 12, vorzugsweise mit 1 bis 4, Kohlenstoffatomen, cycloaliphatische Reste mit 5 bis 12, vorzugsweise mit 5 oder 6 Kohlenstoffatomen im Ring, geeignet. Als Substituenten seien niedere Alkoxyreste (vorzugsweise mit 1 bis 4 Kohlenstoffatomen) und im Falle des cycloaliphatischen Restes auch niedere Alkylreste mit vorzugsweise 1 bis 4 Kohlenstoffatomen genannt.
Araliphatische Reste enthalten im aliphatischen Teil zweckmässig 1 bis 4, vorzugsweise 1 oder 2, Kohlenstoffatome, wobei als aromatischer Rest der Phenyl- und Naphthylrest, vorzugsweise ersterer, genannt seien. Als Substituenten im aromatischen Teil seien niederer Alkyloder Alkoxyrest genannt. Aromatische Reste (R1) sind z.B. Phenyl, Naphthyl, vorzugsweise Phenyl, wobei die genannten Reste gegebenenfalls durch niedere Alkyloder Alkoxygruppen substituiert sein können.
Alkylreste (R2) sind z.B. solche mit 1 bis 8, vorzugsweise mit 1 bis 4, Kohlenstoffatomen. Die Aralkylreste R2 können gleich oder verschieden von den unter R1 genannten sein.
Das Verfahren sei an folgendem Beispiel erläutert:

Zur Durchführung des Verfahrens wird am besten 1 Mol N-monosubstituiertes 3-Amino-l ,2-benzisothiazol in überschüssigem Alkyl-halogenid, vorzugsweise Alkyljodid (2 bis 3 Mol) gelöst und die Lösung im Autoklaven 12 bis 18 Stunden auf etwa 60 bis 1200C erhitzt.
Anschliessend wird in üblicher Weise aufgearbeitet und das resultierende Rohprodukt sorgfältig im Vakuum fraktioniert.
Es ist aber auch möglich, die Umsetzung in einem Lösungsmittel, vorzugsweise Dimethylsulfoxid, unter Zusatz eines der üblichen Säureacceptoren wie z.B. (K5CO5) vorzunehmen. Die Umsetzungstemperatur liegt dabei zwischen etwa 50 und etwa 1000C.
Die neuen Verbindungen sind schwach gelbliche Öle.
Sie sind schwach basisch und bilden mit starken Säuren gut kristallisierende Salze. Die neuen Verbindungen entsprechen der allgemeinen Formel

worin R, n, R1 und R2 die angegebene Bedeutung haben.
Die erfindungsgemäss hergestellten N-disubstituierten 3-Amino-1,2-benzisothiazole sind ausgezeichnete Antimykotika mit einem sehr breiten Wirkungsspektrum.
Sie wirken in einer Konzentration von 4 y/ml Substrat fungizid auf Trichophyton mentagrophytes und Microsporium felinieum und bewirken in Konzentrationen von 10 bis 20 y/ml Substrat vollständige Wachstumshemmung bei Candida albicans, Penicillium commune und Aspergillus neigen Die neuen Vebindungen sind bei experimentellen Versuchstierinfektionen sowohl lokal als auch systemisch wirksam. Sie sollen als Antimykotika in der Human- und Veterinärmedizin und im Pflanzenschutz angewendet werden.
Beispiel I
50,0 g (0,28 Mol) 3-Äthylamino-l,2-benzisothiazol werden in 109 g (0,7 Mol) Äthyljodid gelöst und die Lösung im Autoklaven 18 Stunden auf 1000C erhitzt. Anschliessend wird das Reaktionsprodukt in Chloroform aufgenommen, mit verdünnter Natronlauge durchgeschüttelt und darauf mit Wasser gewaschen. Die Chloroformphase wird getrocknet, im Vakuum abgezogen und der zurückbleibende Sirup im Vakuum fraktioniert. Man erhält 30,5 g (52,5% d.Th.) 3-Diäthylamino-1,2-benzisothiazol als hellgelbes Öl vom Kp. 87 bis 95cm/0,01 Torr; nD20= 1,6071.
In analoger Weise wurden erhalten:
3-Dimethylamino- 1,2-benzisothiazol: gelbliches öl, Kp. 760C/0,01 Torr, nD20 = 1,6326; Ausbeute 35SO d. Th.
3-Methyl-äthylamino-1,2-benzisothiazol: hellgelbes Öl, Kp. 67 bis 73"C/0,01 Torr, nD20 = 1,6240; Ausbeute 42% d. Th.
In der prinzipiell gleichen Weise wurden die folgenden Verbindungen hergestellt: 3-Dipropylamino-1,2-benzisothiazol: gelbes öl, Ausbeute 53,5% der Theorie; Kp. 1080C/0,01 Torr, nD20 = 1,5852.
3-Äthyl-cyclohexylamino-1,2-benzisothiazol: blassgelbes öl; Ausbeute 43% der Theorie; Kp. 1510C/0,3 Torr; und20= 1,5998.
3-Diisobutylamino-1,2-benzisothiazol: blassgelbes öl; Ausbeute: 30% der Theorie; Kp. 108-110 C/0,01 Torr; nD20 = 1,5707.
3-Di-(2'.Äthylhexyl)-amino- 1,2- benzisothiazol: blassgelbes öl; Ausbeute: 63% der Theorie; Kp. 124-1280C/ 0,01 Torr.
3-Dibutylamino- 1,2-benzisothiazol: blassgelbes öl; Ausbeute; 32% der Theorie; Kp. 1280C/0,01 Torr; nD2" = 1,5681.
3-Diallylamino - 1,2 - benzisothiazol: blassgelbes Öl; Ausbeute: 26% der Theorie; Kp. 1250C/0,01 Torr.
Beispiel 2
35,6 g (0,20 Mol) 3-Äthylamino-1,2-benzisothiazol werden in 75 ml Dimethylsulfoxid gelöst und 13,8 g (0,1 Mol) Kaliumcarbonat (gepulvert) sowie 12,7 g (0,1 Mol) Benzylchlorid zugegeben. Die Mischung wird 18 Stunden bei 500C gerührt. Sodann wird die Reaktionslösung in 500 ml Wasser eingetragen und die Mischung mit Chloroform extrahiert. Der Extrakt wird getrocknet, eingedampft und im Vakuum fraktioniert. Zunächst erhält man als Vorlauf, Kp. 1160C/0,01 Torr 14,3 g unverändertes 3-Äthylamino-1,2-benzisothiazol. Das 3-Äthyl -benzylamino-1,2-benzisothiazol geht als gelbes, viskoses öl vom Kp. 1650C/0,01 Torr über; Ausbeute 6,6 g (25% d. Th.).
Das als Ausgangsverbindung verwendete 3 -Äthylami- no-1,2-benzisothiazol (F.: 760C) ist nach Chem. Ber. 99, 2566 (1966) erhalten worden.
Auch die anderen für die Durchführung des Verfahrens Verwendung findenden Ausgangsverbindungen können in analoger Weise erhalten werden.
Process for the preparation of N-disstituted 3-amino-1,2-benzisothiazole derivatives
In the literature so far only N-monosubstituted 3-amino-1,2-benzisothiazoles have been described, which were obtained according to the German Auslegeschrift 1174783 by reacting 3-chloro-1,2-benzisothiazole with basic alkylated amines. Furthermore, N-monosubstituted 3-amino-1,2-benzisothiazoles could be prepared by reacting N-alkyl or N-aryl-3-chloro-1,2-benz isothiazolium halides with ammonia [Chem. Ber. 99, 2566, (1966)].
The invention relates to a process for novel N-disubstituted 3-amino-l 2-benzisothiazoles.
It has been found that novel N-disubstituted 3-amino-1,2-benzisothiazole derivatives are obtained when N-monosubstituted 3-amino-2-benzisothiazoles of the general formula are obtained

where R represents hydrogen, identical or different alkyl, alkoxy or halogen radicals and one radical R can also represent NO2, n represents an integer from 1 to 3, and R1 represents an optionally substituted aliphatic, araliphatic or aryl radical, with an alkylating agent or aralkylating agent of the general formula R2-X in which R2 is an alkyl or aralkyl radical and X is chlorine, bromine, iodine or the grouping -O-SO2-phenyl or -O-SO2-tolyl.
Alkyl or alkoxy radicals (R) are preferably those with 1 to 4 carbon atoms, and halogens (R) are fluorine, chlorine, bromine, iodine, but preferably chlorine and bromine. Suitable optionally substituted aliphatic radicals (R1) are straight-chain or branched, optionally containing a double bond, alkyl radicals with 1 to 12, preferably with 1 to 4, carbon atoms, cycloaliphatic radicals with 5 to 12, preferably with 5 or 6 carbon atoms in the ring. Lower alkoxy radicals (preferably having 1 to 4 carbon atoms) and, in the case of the cycloaliphatic radical, also lower alkyl radicals having preferably 1 to 4 carbon atoms may be mentioned as substituents.
Araliphatic radicals in the aliphatic part suitably contain 1 to 4, preferably 1 or 2, carbon atoms, the phenyl and naphthyl radicals, preferably the former, being mentioned as aromatic radicals. Lower alkyl or alkoxy radicals may be mentioned as substituents in the aromatic part. Aromatic radicals (R1) are e.g. Phenyl, naphthyl, preferably phenyl, where the radicals mentioned can optionally be substituted by lower alkyl or alkoxy groups.
Alkyl radicals (R2) are e.g. those with 1 to 8, preferably with 1 to 4, carbon atoms. The aralkyl radicals R2 can be identical to or different from those mentioned under R1.

To carry out the process, 1 mol of N-monosubstituted 3-amino-1,2-benzisothiazole is best dissolved in excess alkyl halide, preferably alkyl iodide (2 to 3 mol) and the solution in the autoclave to about 60 to 1200C for 12 to 18 hours heated.
It is then worked up in the usual way and the resulting crude product is carefully fractionated in vacuo.
It is also possible, however, to carry out the reaction in a solvent, preferably dimethyl sulfoxide, with the addition of one of the customary acid acceptors such as e.g. (K5CO5). The reaction temperature is between about 50 and about 1000C.
The new compounds are pale yellowish oils.
They are weakly basic and form well-crystallizing salts with strong acids. The new compounds correspond to the general formula

The N-disubstituted 3-amino-1,2-benzisothiazoles prepared according to the invention are excellent antimycotics with a very broad spectrum of activity.
In a concentration of 4 μg / ml substrate they have a fungicidal effect on Trichophyton mentagrophytes and Microsporium felinieum and in concentrations of 10 to 20 μg / ml substrate they cause complete growth inhibition in Candida albicans, Penicillium commune and Aspergillus. The new compounds are both local in experimental animal infections as well as systemically effective. They are to be used as antimycotics in human and veterinary medicine and in plant protection.
Example I.
50.0 g (0.28 mol) of 3-ethylamino-1,2-benzisothiazole are dissolved in 109 g (0.7 mol) of ethyl iodide and the solution is heated to 1000C for 18 hours in the autoclave. The reaction product is then taken up in chloroform, shaken with dilute sodium hydroxide solution and then washed with water. The chloroform phase is dried, stripped off in vacuo and the remaining syrup is fractionated in vacuo. 30.5 g (52.5% of theory) of 3-diethylamino-1,2-benzisothiazole are obtained as a light yellow oil with a boiling point of 87 to 95 cm / 0.01 Torr; nD20 = 1.6071.
In a similar way the following were obtained:
3-dimethylamino-1,2-benzisothiazole: yellowish oil, boiling point 760C / 0.01 Torr, nD20 = 1.6326; Yield 35 SO d. Th.
3-methyl-ethylamino-1,2-benzisothiazole: light yellow oil, b.p. 67 to 73 "C / 0.01 Torr, nD20 = 1.6240; yield 42% of theory.
The following compounds were prepared in basically the same way: 3-Dipropylamino-1,2-benzisothiazole: yellow oil, yield 53.5% of theory; Bp 1080C / 0.01 Torr, nD20 = 1.5852.
3-ethyl-cyclohexylamino-1,2-benzisothiazole: pale yellow oil; Yield 43% of theory; Bp 1510C / 0.3 Torr; and 20 = 1.5998.
3-diisobutylamino-1,2-benzisothiazole: pale yellow oil; Yield: 30% of theory; Bp 108-110 C / 0.01 torr; nD20 = 1.5707.

Example 2
35.6 g (0.20 mol) of 3-ethylamino-1,2-benzisothiazole are dissolved in 75 ml of dimethyl sulfoxide and 13.8 g (0.1 mol) of potassium carbonate (powdered) and 12.7 g (0.1 mol ) Benzyl chloride added. The mixture is stirred at 50 ° C. for 18 hours. The reaction solution is then introduced into 500 ml of water and the mixture is extracted with chloroform. The extract is dried, evaporated and fractionated in vacuo. First of all, 14.3 g of unchanged 3-ethylamino-1,2-benzisothiazole are obtained as a forerun, b.p. The 3-ethylbenzylamino-1,2-benzisothiazole goes over as a yellow, viscous oil with a boiling point of 1650C / 0.01 Torr; Yield 6.6 g (25% of theory).
The 3-ethylamino-1,2-benzisothiazole (F .: 760C) used as the starting compound is according to Chem. Ber. 99, 2566 (1966).
The other starting compounds used for carrying out the process can also be obtained in an analogous manner.
