Verfahren zur Herstellung von substituierten Hydro-2-naphthalenonen
Die Erfindung betrifft ein Verfahren zur Herstellung von Verbindungen der Formel

worin Rl Wasserstoff oder die Gruppe COOR2 bedeutet und R nieder-Alkyl mit 1-6 Kohlenstoffatomen, insbesondere Methyl oder Äthyl darstellt.
Die Verbindungen der Formel I sind neu.
Die obige Formel I wie auch die im folgenden noch gezeichneten Formeln III bis VIII sollen auch die Diastereomeren umfassen, wobei unter Diastereomeren die durch die jeweils vorhandenen Asymmetriezentren der Verbindungen der allgemeinen Formeln I und III bis VIII möglichen Isomeren verstanden werden.
Das erfindungsgemässe Verfahren zur Herstellung von Verbindungen der Formel I ist dadurch gekennzeichnet, dass man 2-Formyl-2-methyl-4-isopropenyl- cyclohexanon der Formel

mit Aceton oder einem ssKetoester CHsCOCH2COOR2 umsetzt und eine im Umsetzungsprodukt gegebenenfalls vorhandene 3-Carboxygruppe wieder zu einer 3-Carbonieder-alkoxy-Gruppe verestert. Als Ausgangsstoff kann das 2a-, 2ss- oder ein Gemisch der 2a- und 2ss-Methyl Isomeren der Cyclohexanonkomponente eingesetzt werden.
Die Umsetzung von 2-Formyl-2-methyl-4-isopro- penylcyclohexanon mit Aceton bzw. dem genannten fi- Ketoester kann z. B. so vorgenommen werden, dass man eine Lösung des Cyclohexanons in Aceton bzw. dem ss-Ketoester unter Zusatz eines schwach basischen Katalysators, z. B. eines Amins, vorzugsweise eines cyclischen Amins, wie Pyrrolidin, oder, vorzugsweise, Piperidin, und unter Zusatz einer organischen Säure, z. B.
einer Carbonsäure, wie Essigsäure (vorzugsweise) oder Propionsäure, erwärmt, vorzugsweise auf Rückflusstemperatur der Reaktionslösung. Das so erhaltene primäre Reaktionsprodukt (das als Produkt der Kondensation der Formylgruppe der Cyclohexanonkomponente mit Aceton bzw. dem ss-Ketoester zu formulieren ist) kann hierauf, zweckmässig nach Isolierung aus der Reaktionslösung, mit einer starken Base, z. B. einem Alkalihydroxyd, wie Kalilauge, oder Natronlauge, in einem Alkohol, wie Methanol (bevorzugt) oder Äthanol, oder in Dioxan, Tetrahydrofuran oder einem andern mit Wasser mischbaren Lösungsmittel oder einem Alkoholat, wie ethylat oder Methylat, in einem Alkohol, wie Äthanol oder Methanol, bei erhöhter Temperatur, vorzugsweise bei der Rückflusstemperatur des Gemisches, zu einem Hydro-2-naphthalenon cyclisiert werden.
Wird anstelle von Aceton ein BKetoester als Ausgangsstoff verwendet, so erfolgt unter den alkalischen Bedingungen der Cyclisierungsreaktion eine mindestens teilweise Verseifung der Carbalkoxygruppe. Die Wiederveresterung kann nach an sich bekannten Methoden, vorzugsweise unter praktisch neutralen Bedingungen, beispielsweise mit Diazomethan, vorgenommen werden.
Wird als Ausgangsstoff ein Gemisch der 2a- und 2ss-Methyl-Isomeren der Cyclohexanonkomponente eingesetzt, entsteht ebenfalls ein Gemisch der 4aa- und 4 ass-isomeren Hydronaphthalenon-Verbindungen. Die Trennung in die Einzelkomponenten kann, wie z. B. im Falle des 4a-Methyl-613-isopropenyl-2,4a,5,ó,7,8-hexa- hydro-2-naphthalenons, durch fraktionierte Kristallisation erfolgen.
Das als Ausgangsstoff venvendete 2-Formyl-2-methyl-4-isopropenylcyclohexanon kann z. B. dadurch erhalten werden, dass 4-Isopropenylcyclohexanon mit Ameisensäureäthylester in Anwesenheit eines Alkoholats, z. B. eines Methylats, zur entsprechenden 2-Hydr oxymethylenverbindung umgesetzt wird und letztere mittels eines Methylierungsmittels in alkalischem Milieu, z. B. unter Zusatz eines Alkalicarbonats, zum 2-Formyl2-methyl-4-isopropenylcyclohexanon umgesetzt wird.
Das entstehende Gemisch von 2a-Formyl-2ss-methyl- verbindung und 2B-Formyl-2a-methylverbindung kann gewünschtenfalls, z. B. mittels präparativer Gaschromatographie, in die Einzelkomponenten aufgetrennt werden.
Die Verbindungen der Formel I zeichnen sich durch besondere Riechstoff- bzw. Aromaeigenschaften aus.
So weist z. B. das 4aa-Methyl-6ss-isopropenyl-2,4a,5,6,7, 8-hexahydro-2-naphthalenon einen Geruch nach Grapefruitschalen auf. Die Verbindungen der Formel I können dementsprechend als Riech- bzw. Aromastoffe Verwendung finden, z. B. zur Aromatisierung von Getränken.
Die Verbindungen der Formel I sind überdies wertvolle Zwischenprodukte zur Herstellung von Verbindungen der Formel

worin R1 obige Bedeutung hat und ---- eine C-C-Einfachbindung oder C=C-Doppelbindung darstellt.
Diese Verbindungen der Formel II sind, mit Ausnahme von 4eu-Methyl-4aoHmethyl-6ss-isopropenyl-
2,3,4,4a,5,6,7,8-octahydros2-naphthalenon (Nootkaton), ebenfalls neue Verbindungen. Sie weisen besondere wertvolle Riechstoff- bzw. Aromaeigenschaften auf. Das Nootkaton findet sich im Grapefruitschalenöl und ist eine der Komponenten, die der Grapefruit ihr charakteristisches Aroma verleihen.
4ss-Methyl-4aa-methyl-6ss-isopropenyl 2,3,4,4a, 5,6,7, 8-octahydro-2-naphthalenon kann als Träger von Holznoten Verwendung finden.
Die Überführung einer Verbindung der Formel I in verschiedene Verbindungstypen der Formel II (nämlich die Verbindungen der Formel III-VIII) kann nach folgendem Schema bewerkstelligt werden: Schema

Verbindungen der Formel III können erhalten werden, indem man eine Verbindung der Formel I hydrierend methyliert (das heisst in 4-Stellung eine Methylgruppe und in 3-Stellung ein Wasserstoffatom anlagert [eine Reaktion vom Typ der 1 4-Addinon]). Man geht dabei zweckmässig so vor, dass man eine Verbindung der Formel I mit Methyllithium oder einer Methylgrignardverbindung und einem Kupfersalz, wie Kupferjodid, Kupferacetat oder Kupferchlorid, vorzugsweise Kupferjodid, in einem Ather, z. B.
Diäthyläther, Dimethoxy äthan oder Dioxan, vorzugsweise Diäthyläther, bei Temperaturen unterhalb Raumtemperatur, vorzugsweise bei solchen um 0 C herum, umsetzt.
Die selektive Dehydrierung einer Verbindung der Formel III zu einer Verbindung der Formel IV kann beispielsweise unter der Einwirkung von Chinonen, z. B.
von 2,3-Dichlor-5,6-dicyano-1,4-benzochinon, vor sich gehen. Man arbeitet dazu in einem Lösungsmittel, wie Benzol, Toluol oder Dioxan, vorzugsweise Benzol, und bei erhöhter Temperatur, vorzugsweise bei der Rückflusstemperatur des Reaktionsgemisches, und vorzugsweise in Gegenwart einer Säure, z. B. einer organischen Säure, wie Essigsäure.
Die Hydrierung einer Verbindung der Formel IV, worin R1 die COOR2-Gruppe bedeutet, zu einer Verbindung der Formel V kann unter der Einwirkung von Natriumborhydrid in Anwesenheit von Stickstoff- oder Phosphorbasen, wie Triäthylamin, Pyridin, Piperidin, Collidin oder Triphenylphosphin vorzugsweise Pyridin, durchgeführt werden. Auch die Einwirkung von Alkylund Arylzinnhydrideln, vorzugsweise von Tri-n-butylzinnhydrid, in Anwesenheit von katalytischen Mengen eines Metallhalogenids, vorzugsweise Zinkchlorid, eignet sich zur Hydrierung. Es entstehen bei Verwendung dieser Hydrierungsmittel Verbindungen der Formel V, in denen die Methylgruppen in den Stellungen 4 und 4a cis Konfiguration aufweisen.
Die Hydrolyse der Esterbindung einer Verbindung der Formel V und die anschliessende Decarboxylierung der freien Säure kann in an sich bekannter Weise durchgeführt werden, z. B. hydrolysiert man alkalisch, vorzugsweise bei Zimmertemperatur. Die Decarboxylierung kann durch vorsichtiges Erhitzen vorgenommen werden.
Die Verbindung der Formel VII lässt sich aus einer Verbindung der Formel IV, worin R1 Wasserstoff bedeutet, durch Behandeln mit einem Alkalimetall, z. B.
Kalium, Natrium oder vorzugsweise Lithium, in Ammoniak erhalten. Man arbeitet zweckmässig in einem Lösungsmittel, z. B. einem Äther, z. B. Diäthyläther, Dioxan oder, vorzugsweise, Tetrahydrofuran und bei Temperaturen bis zum Siedepunkt des Ammoniaks. Nach erfolgter Reduktion wird mit Vorteil ein Protonendonator, z. B. Ammonchlorid (bevorzugt), Ammonsulfat oder verdünnte Salzsäure, addiert.
Die Verbindung der Formel VIII schliesslich kann ebenfalls aus einer Verbindung der Formel IV, worin R1 Wasserstoff bedeutet, durch Behandlung mit denselben Reduktionsmitteln, bei denselben Temperaturen und in denselben Lösungsmitteln erhalten werden, wie sie zur Herstellung der Verbindung der Formel VII angewandt werden, doch wird nun nach der Hydrierung der 1 (8a) Doppelbindung in der Verbindung der Formel IV zum Reaktionsgemisch ein Alkohol, z. B. Äthanol, Propanol oder Butanol, oder Wasser, vorzugsweise aber Äthanol, gegeben, worauf anschliessend die Doppelbindung in Stellung 3(4) ebenfalls noch hydriert wird.
In den nachfolgenden Beispielen sind die Temperaturen in Celsiusgraden angegeben.
Beispiel 1
19,7 g (109 mMol) 2-Formyl-2-methyl-4-isopropeny cyclohexanon werden in 172 ml reinem Aceton gelöst und unter Rühren nacheinander 11,1 g (131 mMol) reines Piperidin und 7,86 g (131 mMol) reiner Eisessig zugegeben. Die Lösung wird 64 Stunden unter Stickstoff am Rückfluss gehalten. Zur Aufarbeitung wird das Reaktionsgemisch in Äther aufgenommen, die organische Phase nacheinander mit verdünnter Salzsäure, mit Wasser, gesättigter Natriumbicarbonat-Lösung und nochmals mit Wasser gewaschen, getrocknet und das Lösung mittel abgesaugt. Das Rohprodukt (20,1 g) wird in
190 ml Methanol und 25 ml 50 % Kalilauge gelöst und 5 Stunden unter Stickstoff am Rückfluss gehalten.
Das Reaktionsgemisch wird in Äther aufgenommen, mit gesättigter Natriumdihydrogenphosphat-Lösung und mit Wasser gewaschen, getrocknet und das Lösungsmittel abgesaugt. Es resultieren 11,8 g eines Öls.
Drei solche Ansätze ergeben zusammen 35,96 g Ö1, das in einem Vigreux-Kolben destilliert wird. Die Fraktion mit dem Siedepunkt 122-1240/0,01 Torr (12,87 g) stellt ein Gemisch der 2 stereoisomeren 4asa; und 4ass-
Methyl-6ss-isopropenyl-2,4a,5,6,7,8-hexahydro -2 - naph- thalenone im Verhältnis 5 1 dar. Das Erstere wird durch Kristallisation aus Äther und Pentan rein isoliert.
Schmelzpunkt 39-400; UV-Spektrum: (Äthanol) Amax = 242 nm (e = 15 500); IR-Spektrum: (CC14) Banden bei
1665, 1635, 1608, 895 cml; NMR-Spektrum: (CDCl5) = 1,33 ppm (S, 3H).
Das als Ausgangsmaterial verwendete 2-Formyl-2- methyl-4-isopropenyl-cyclohexanon kann folgenderma ssen erhalten werden:
3,12 g (57,8 mMol) getrocknetes Natriummethylat werden in 58 ml absolutem Benzol suspendiert. Zu dieser Suspension werden unter starkem Rührem 4,27 g (57,8 mMol) reiner Ameisensäureäthylester und 28 ml absolutes Benzol gegeben, dann 30 Minuten weitergerührt. Nun tropft man 4,0 g (28,9 mMol) reines 4-Isopropenylcyclohexanon in 15 ml absolutem Benzol zu.
Die resultierende orangegefärbte Masse wird 16 Stunden im geschlossenen Kolben unter Stickstoff gerührt und hierauf wie folgt aufgearbeitet: Man nimmt in Äther auf und zieht 3 mal mit 2n Kalilauge aus. Die alkalischen Auszüge werden sofort bei 0 mit 2n Salzsäure angesäuert und mit Methylenchlorid extrahiert. Die Methylenchlorid-Extrakte werden getrocknet und das Lösungsmittel abgesaugt. Es resultieren 4,737 g 2-Hydroxymethylen4-isopropenylcyclohexanon. Siedepunkt: 55-60 /0,005 Torr; und: 1,5213; UV-Spektrum: (Äthanol) Amax 312nm (e. = 11 800); IR-Spektrum (CC1z): Banden bei
1660, 1620, 895 cm-1.
4,8 g (29 mMol) getrocknetes 2-Hydroxymethylen-4- isopropenyl-cyclohexanon werden in 41,6 ml reinem Aceton gelöst und 10,5 g (76,2 mMol) feinpulverisiertes und wasserfreies Kaliumcarbonat unter Rühren, hierauf 21 g (148 mMol) reines Methyljodid in 40 ml reinem Aceton addiert, der Kolben mit weiteren 21 ml reinem Aceton gespült und dieses ebenfalls addiert. Die resultierende Suspension wird, vor Licht geschützt, unter Stickstoff 24 Stunden gerührt. Das Kaliumcarbonat wird alsdann abfiltriert und das Filtrat in Äther aufgenommen. Die Ätherphasen werden 1 mal mit gesättigter Natriumchlorid-Lösung gewaschen, vereint und eingeengt. Es resultieren 4,92 g eines Produktes mit folgendem IR-Spektrum: (CCh) Banden bei 2725, 1740, 1715, 1695, 1650, 895 cm-1.
Aus dem Gaschromatogramm dieses Produktes ergibt sich die Anwesenheit von 3 Substanzen im Verhältnis von etwa 55%, 17%, 28S, die präparativ gaschromatographisch getrennt werden können:
Bei Peak 1 handelt es sich um 2a-Formyl-2ss-meth- yl-4ss-isopropenyl-cyclohexanon, Siedepunkt: 55 bis 650/0,01 Torr; IR-Spektrum:
Banden bei 2700, 1715, 1650, 895 cm-l; NMR: (in Cd13) < 3= 9,45 ppm (s, 1H), 1,23 ppm (s, 3H); Peak 2 stellt 2p-Formyl-2a- methyl-4ss-isopropenyl-cyclohexanon dar, Siedepunkt: 55-650/0,01 Torr, IR-Spektrum: (in CC Banden bei 2725, 173Q, 1710, 1645, 895 cm-1, NMR: (in CDC13) a = 9,76 ppm (s,1H),1,40 ppm (s, 311); Peak 3 repräsentiert 2-Methoxymethyien-4-isopropenyl-cyclohexanon, Siedepunkt: 65-700/0,01 Torr, IR-Spektrum:
(in CC Banden bei 1695, 1650, 1605, 1250, 895 cm-t, UV Spektrum: (Äthanol) Ämax: 275 nm (e = 5 540).
Beispiel 2
19,4 g (108 mMol) 2-Formyl-2-methyl-4-is opropenyl- cyclohexanon werden in 78,3 ml reinem Acetessigsäuremethylester gelöst und mit 11,05 g (130 mMol) reinem Piperidin und 7,72 g (129 mMol) reinem Eisessig versetzt. Die resultierende Lösung wird unter Stickstoff 65 Stunden bei 600 gerührt. Zur Aufarbeitung nimmt man in Äther auf und wäscht die ätherische Phase zweimal mit 2n Salzsäure, einmal mit Wasser, zweimal mit Natriumhydrogencarbonat und anschliessend mit Wasser bis neutral. Nach dem Trocknen über Natriumsulfat werden das Lösungsmittel und der überschüssige Acetessigsäuremethylester am Vakuum abgesaugt. Der Rückstand wird in 300 ml reinem Methanol und 40 ml einer 50 %igen Kaliumhydroxyd-Lösung gelöst und 5 Stunden in Stickstoff-Atmosphäre am Rückfluss gehalten.
Hierauf nimmt man in Äther auf und zieht die ätherische Phase dreimal mit 2n Kalilauge aus; die alkalischen Auszüge werden in der Kälte mit 2n Salzsäure angesäuert, und der resultierende Niederschlag wird in Methylenchlorid aufgenommen. Nach dem Neutralwaschen der Methylenchlorid-Phase und Trocknen derselben über Natriumsulfat wird das Lösungsmittel abgesaugt. Der verbleibende Rückstand wird in 200 ml absolutem Äther gelöst und mit 190 ml einer 0,4 molaren Diazomethan Lösung bei 0 während 15 Minuten behandelt. Die ätherische Lösung wird anschliessend mit wässrigem Natriumcarbonat und Wasser gewaschen und über Natriumsulfat getrocknet. Nach Entfernen des Lösungsmittels bleiben 16,5 g eines Öls zurück, welches hierauf an der 60fachen Menge Kieselgel chromatographiert wird.
Das erhaltene (5,14 g) 3-Carbomethoxy-4acFmethyl- 6ss-isopropenyl-2,4a,5,6,7,8-hexahydro -2 naphthalenon schmilzt bei 78-790; IR-Spektrum: (CC14) Banden bei 1760, 1730, 1680, 1655, 1440, 895 cm-t; UV-Spektrum: (Äthanol) max: 244 nm (e = 11 600); NMR Spektrum: (CDC13) Åa = 3,88 ppm (s, 3H); 1,38 ppm (s, 3H).

The above formula I as well as the formulas III to VIII shown below are also intended to include the diastereomers, diastereomers being understood as meaning the isomers possible due to the respective centers of asymmetry of the compounds of the general formulas I and III to VIII.
The process according to the invention for the preparation of compounds of the formula I is characterized in that 2-formyl-2-methyl-4-isopropenyl-cyclohexanone of the formula

with acetone or an ß-ketoester CHsCOCH2COOR2 and a 3-carboxy group which may be present in the reaction product is re-esterified to a 3-carbo-lower alkoxy group. The 2a-, 2ss- or a mixture of the 2a- and 2ss-methyl isomers of the cyclohexanone component can be used as the starting material.
The reaction of 2-formyl-2-methyl-4-isopropenylcyclohexanone with acetone or the mentioned fi-ketoester can, for. B. be made so that a solution of the cyclohexanone in acetone or the ss-ketoester with the addition of a weakly basic catalyst, eg. B. an amine, preferably a cyclic amine such as pyrrolidine, or, preferably, piperidine, and with the addition of an organic acid, e.g. B.
a carboxylic acid, such as acetic acid (preferably) or propionic acid, heated, preferably to the reflux temperature of the reaction solution. The primary reaction product obtained in this way (which is to be formulated as the product of the condensation of the formyl group of the cyclohexanone component with acetone or the β-keto ester) can then, advantageously after isolation from the reaction solution, with a strong base, e.g. B. an alkali hydroxide such as potassium hydroxide, or sodium hydroxide, in an alcohol such as methanol (preferred) or ethanol, or in dioxane, tetrahydrofuran or another water-miscible solvent or an alcoholate such as ethylate or methylate, in an alcohol such as ethanol or methanol can be cyclized to a hydro-2-naphthalenone at an elevated temperature, preferably at the reflux temperature of the mixture.
If a betoester is used as the starting material instead of acetone, an at least partial saponification of the carbalkoxy group takes place under the alkaline conditions of the cyclization reaction. The re-esterification can be carried out by methods known per se, preferably under practically neutral conditions, for example with diazomethane.
If a mixture of the 2a- and 2ss-methyl isomers of the cyclohexanone component is used as the starting material, a mixture of the 4aa and 4 ass-isomeric hydronaphthalenone compounds is also formed. The separation into the individual components can, such. B. in the case of 4a-methyl-613-isopropenyl-2,4a, 5, ó, 7,8-hexa-hydro-2-naphthalenone, done by fractional crystallization.
The 2-formyl-2-methyl-4-isopropenylcyclohexanone used as starting material can, for. B. obtained in that 4-isopropenylcyclohexanone with ethyl formate in the presence of an alcoholate, for. B. a methylate, is converted to the corresponding 2-Hydr oxymethylene compound and the latter by means of a methylating agent in an alkaline medium, for. B. with the addition of an alkali metal, is converted to 2-formyl2-methyl-4-isopropenylcyclohexanone.
The resulting mixture of 2a-formyl-2ss-methyl compound and 2B-formyl-2a-methyl compound can, if desired, for. B. by means of preparative gas chromatography, are separated into the individual components.
The compounds of the formula I are distinguished by special fragrance and aroma properties.
So z. B. the 4aa-methyl-6ss-isopropenyl-2,4a, 5,6,7, 8-hexahydro-2-naphthalenone a smell of grapefruit peel. The compounds of the formula I can accordingly be used as fragrances or aromatic substances, e.g. B. for flavoring beverages.
The compounds of the formula I are also valuable intermediates for the preparation of compounds of the formula

These compounds of the formula II are, with the exception of 4eu-methyl-4aoHmethyl-6ss-isopropenyl-
2,3,4,4a, 5,6,7,8-octahydros2-naphthalenone (nootkatone), also new compounds. They have particularly valuable fragrance and aroma properties. Nootkatone is found in grapefruit peel oil and is one of the components that give grapefruit its characteristic aroma.
4ss-methyl-4aa-methyl-6ss-isopropenyl 2,3,4,4a, 5,6,7,8-octahydro-2-naphthalenone can be used as a carrier for wood notes.
The conversion of a compound of the formula I into different types of compounds of the formula II (namely the compounds of the formula III-VIII) can be accomplished according to the following scheme: Scheme

Compounds of the formula III can be obtained by hydrotreating a compound of the formula I (that is, a methyl group is added in the 4-position and a hydrogen atom in the 3-position [a reaction of the 14-addinon type]). It is expedient to proceed in such a way that a compound of the formula I with methyllithium or a methyl Grignard compound and a copper salt such as copper iodide, copper acetate or copper chloride, preferably copper iodide, in an ether, e.g. B.
Diethyl ether, dimethoxy ethane or dioxane, preferably diethyl ether, is reacted at temperatures below room temperature, preferably at temperatures around 0 C.
The selective dehydrogenation of a compound of formula III to a compound of formula IV can, for example, under the action of quinones, e.g. B.
of 2,3-dichloro-5,6-dicyano-1,4-benzoquinone, go on. To do this, one works in a solvent such as benzene, toluene or dioxane, preferably benzene, and at elevated temperature, preferably at the reflux temperature of the reaction mixture, and preferably in the presence of an acid, e.g. B. an organic acid such as acetic acid.

The action of alkyl and aryl tin hydrides, preferably of tri-n-butyltin hydride, in the presence of catalytic amounts of a metal halide, preferably zinc chloride, is also suitable for the hydrogenation. The use of these hydrogenating agents results in compounds of the formula V in which the methyl groups in positions 4 and 4a have a cis configuration.
The hydrolysis of the ester bond of a compound of the formula V and the subsequent decarboxylation of the free acid can be carried out in a manner known per se, e.g. B. is hydrolyzed under alkaline conditions, preferably at room temperature. Decarboxylation can be done by careful heating.
The compound of the formula VII can be prepared from a compound of the formula IV in which R1 is hydrogen by treatment with an alkali metal, e.g. B.
Potassium, sodium or preferably lithium, obtained in ammonia. It is convenient to work in a solvent, for. B. an ether, e.g. B. diethyl ether, dioxane or, preferably, tetrahydrofuran and at temperatures up to the boiling point of ammonia. After the reduction has taken place, a proton donor, e.g. B. ammonium chloride (preferred), ammonium sulfate or dilute hydrochloric acid added.
Finally, the compound of the formula VIII can also be obtained from a compound of the formula IV in which R1 is hydrogen by treatment with the same reducing agents, at the same temperatures and in the same solvents as are used to prepare the compound of the formula VII now after the hydrogenation of the 1 (8a) double bond in the compound of formula IV to the reaction mixture, an alcohol, e.g. B. ethanol, propanol or butanol, or water, but preferably ethanol, is added, whereupon the double bond in position 3 (4) is also hydrogenated.

example 1
19.7 g (109 mmol) of 2-formyl-2-methyl-4-isopropeny cyclohexanone are dissolved in 172 ml of pure acetone and, while stirring, 11.1 g (131 mmol) of pure piperidine and 7.86 g (131 mmol) pure glacial acetic acid added. The solution is refluxed under nitrogen for 64 hours. For working up, the reaction mixture is taken up in ether, the organic phase is washed in succession with dilute hydrochloric acid, with water, saturated sodium bicarbonate solution and again with water, dried and the solution is filtered off with suction. The crude product (20.1 g) is in
190 ml of methanol and 25 ml of 50% potassium hydroxide solution and refluxed under nitrogen for 5 hours.
The reaction mixture is taken up in ether, washed with saturated sodium dihydrogen phosphate solution and with water, dried and the solvent is filtered off with suction. 11.8 g of an oil result.
Three such approaches together give 35.96 g of oil, which is distilled in a Vigreux flask. The fraction with the boiling point 122-1240 / 0.01 Torr (12.87 g) is a mixture of the 2 stereoisomeric 4asa; and 4ass-
Methyl-6ss-isopropenyl-2,4a, 5,6,7,8-hexahydro -2 - naphthalenone in the ratio 5: 1. The former is isolated in pure form by crystallization from ether and pentane.
Melting point 39-400; UV spectrum: (ethanol) Amax = 242 nm (e = 15,500); IR spectrum: (CC14) bands at
1665, 1635, 1608, 895 cm-1; Nuclear Magnetic Resonance Spectrum: (CDCl5) = 1.33 ppm (S, 3H).
The 2-formyl-2-methyl-4-isopropenyl-cyclohexanone used as starting material can be obtained as follows:
3.12 g (57.8 mmol) of dried sodium methylate are suspended in 58 ml of absolute benzene. 4.27 g (57.8 mmol) of pure ethyl formate and 28 ml of absolute benzene are added to this suspension with vigorous stirring, and stirring is continued for 30 minutes. 4.0 g (28.9 mmol) of pure 4-isopropenylcyclohexanone in 15 ml of absolute benzene are then added dropwise.
The resulting orange-colored mass is stirred in a closed flask under nitrogen for 16 hours and then worked up as follows: It is taken up in ether and extracted 3 times with 2N potassium hydroxide solution. The alkaline extracts are immediately acidified at 0 with 2N hydrochloric acid and extracted with methylene chloride. The methylene chloride extracts are dried and the solvent is filtered off with suction. 4.737 g of 2-hydroxymethylene-4-isopropenylcyclohexanone result. Boiling point: 55-60 / 0.005 torr; and: 1.5213; UV spectrum: (ethanol) Amax 312nm (e. = 11,800); IR spectrum (CC1z): bands at
1660, 1620, 895 cm-1.
4.8 g (29 mmol) of dried 2-hydroxymethylene-4-isopropenyl-cyclohexanone are dissolved in 41.6 ml of pure acetone and 10.5 g (76.2 mmol) of finely powdered and anhydrous potassium carbonate with stirring, then 21 g (148 mmol) of pure methyl iodide in 40 ml of pure acetone is added, the flask is rinsed with a further 21 ml of pure acetone and this is also added. The resulting suspension, protected from light, is stirred under nitrogen for 24 hours. The potassium carbonate is then filtered off and the filtrate is taken up in ether. The ether phases are washed once with saturated sodium chloride solution, combined and concentrated. 4.92 g of a product with the following IR spectrum result: (CCh) bands at 2725, 1740, 1715, 1695, 1650, 895 cm-1.
The gas chromatogram of this product shows the presence of 3 substances in a ratio of about 55%, 17%, 28S, which can be preparatively separated by gas chromatography:
Peak 1 is 2a-formyl-2ss-methyl-4ss-isopropenyl-cyclohexanone, boiling point: 55 to 650 / 0.01 Torr; IR spectrum:
Bands at 2700, 1715, 1650, 895 cm-1; NMR: (in Cd13) <3 = 9.45 ppm (s, 1H), 1.23 ppm (s, 3H); Peak 2 represents 2p-formyl-2a-methyl-4ss-isopropenyl-cyclohexanone, boiling point: 55-650 / 0.01 Torr, IR spectrum: (in CC bands at 2725, 173Q, 1710, 1645, 895 cm-1 , NMR: (in CDC13) a = 9.76 ppm (s, 1H), 1.40 ppm (s, 311); peak 3 represents 2-methoxymethylene-4-isopropenyl-cyclohexanone, boiling point: 65-700 / 0, 01 Torr, IR spectrum:
(in CC bands at 1695, 1650, 1605, 1250, 895 cm-t, UV spectrum: (ethanol) A max: 275 nm (e = 5 540).
Example 2
19.4 g (108 mmol) of 2-formyl-2-methyl-4-isopropenyl-cyclohexanone are dissolved in 78.3 ml of pure methyl acetoacetate and mixed with 11.05 g (130 mmol) of pure piperidine and 7.72 g (129 mmol) of pure glacial acetic acid. The resulting solution is stirred at 600 for 65 hours under nitrogen. For working up, it is taken up in ether and the ethereal phase is washed twice with 2N hydrochloric acid, once with water, twice with sodium hydrogen carbonate and then with water until neutral. After drying over sodium sulfate, the solvent and the excess methyl acetoacetate are suctioned off in vacuo. The residue is dissolved in 300 ml of pure methanol and 40 ml of a 50% strength potassium hydroxide solution and refluxed for 5 hours in a nitrogen atmosphere.
Then one takes up in ether and draws out the ethereal phase three times with 2N potassium hydroxide solution; the alkaline extracts are acidified in the cold with 2N hydrochloric acid, and the resulting precipitate is taken up in methylene chloride. After the methylene chloride phase has been washed neutral and dried over sodium sulfate, the solvent is filtered off with suction. The remaining residue is dissolved in 200 ml of absolute ether and treated with 190 ml of a 0.4 molar diazomethane solution at 0 for 15 minutes. The ethereal solution is then washed with aqueous sodium carbonate and water and dried over sodium sulfate. After removal of the solvent, 16.5 g of an oil remain, which is then chromatographed on 60 times the amount of silica gel.
The obtained (5.14 g) 3-carbomethoxy-4acFmethyl-6ss-isopropenyl-2,4a, 5,6,7,8-hexahydro -2 naphthalenone melts at 78-790; IR spectrum: (CC14) bands at 1760, 1730, 1680, 1655, 1440, 895 cm-t; UV spectrum: (ethanol) max: 244 nm (e = 11,600); Nuclear Magnetic Resonance Spectrum: (CDC13) Åa = 3.88 ppm (s, 3H); 1.38 ppm (s, 3H).
