Abstract
Prodn. of amino acid salts of glutathione by reacting glutathione with arginine or histidine in aq. solvent, and adding alcohol to ppte the salt. The salts are pharmaceutically active. 3.07 g Glutathione dissolved in 3.0 ml water, 1.74 g L-arginine added with stirring to dissolve .20 ml EtOH added, ppte pulverised, filtered, and reputed from water-EtOH to give 4.8 g glutathione-L-arginine salt m.p. 200-205 deg.C (decomp.).
Description
Verfahren zum Herstellen von neuen SH- oder SS-Glutathionsalzen
Das Hauptpatent Nr. 478 764 betrifft ein Verfahren zum Herstellen von SH- oder SS-Glutathionsalzen, dadurch gekennzeichnet, dass man SH- oder SS-Glutathion und mindestens eine Aminosäure, welche aus der Gruppe basische Aminosäuren sowie Tryptophan und Citrullin gewählt wird, in stöchiometrischen Mengen umsetzt.
In weiterer Ausbildung des Verfahrens nach Hauptpatent wurde gefunden, dass man als basische Aminosäure auch Arginin oder Histidin einsetzen kann und diese Umsetzungsprodukte ebenfalls wertvolle pharmazeutische Eigenschaften besitzen.
Gegenstand der vorliegenden Erfindung ist ein Verfahren zum Herstellen von SH- oder SS-Glutathionsalzen nach Hauptpatent, dadurch gekennzeichnet, dass man als basische Aminosäure Arginin oder Histidin einsetzt.
Die Herstellung der neuen erfindungsgemässen SHoder SS-Glutathionsalze kann nach an sich bekannten Methoden erfolgen, beispielsweise durch Lösen des SH- oder SS-Glutathions und der basischen Aminosäure oder Aminosäuren in Wasser, wässrigen Lösungsmitteln oder anderen geeigneten Lösungsmitteln und Isolierung des gebildeten Salzes.
Die so erhaltenen SH- oder SS-Glutathion-Aminosäure-Salze erhöhen bemerkenswert den Blutspiegel des SH-Glutathions, selbst bei einer oralen Verabreichung, im Vergleich mit der Verabreichung des SHoder SS-Glutathions allein.
Die erfindungsgemäss hergestellten neuen SH- oder SS-Glutathionsalze können in üblicher Weise nach allen Methoden verabreicht werden und können zu Tabletten oder Kapseln, erforderlichenfalls durch Zugabe geeigneter Träger und dergleichen, verarbeitet werden und auch als injizierbare Präparate benutzt werden.
Die Erfindung wird durch die folgenden Beispiele verdeutlicht, jedoch sollen diese hierdurch nicht beschränkt werden.
Beispiel I
In 3,0 ml einer wässrigen Lösung von 3,07 g SH-Glutathion werden 1,74 g L-Arginin unter Umrühren aufgelöst und mit etwa 20 ml Äthanol versetzt.
Es fällt ein weisser Niederschlag aus, der nach kurzer Zeit fest wird. Der Niederschlag wird abfiltriert und aus dem Wasser-Äthanol umkristallisiert, um 4,8 g des farblosen hygroskopischen SH-Glutathion-L-Argininsalzes \ mit dem Schmelzpunkt 200 bis 2050 C (unter Zersetzung) zu erhalten.

Beispiel 2
In 3,0 ml einer wässrigen Lösung von 3,07 g SH Glutathion werden 1,55 g Histidin aufgelöst. Die erhaltene Lösung wird gemäss Beispiel 1 aufgearbeitet.
Man erhält 4,6 g des farblosen hygroskopischen SH Glutathion-L-Histidinsalzes mit dem Schmelzpunkt 197 bis 2020 C (unter Zersetzung).

PATENTANSPRUCH
Verfahren gemäss Patentanspruch des Hauptpatentes, dadurch gekennzeichnet, dass man als basische Aminosäure Arginin oder Histidin einsetzt.

1. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass man die Umsetzung in Wasser oder in wässrigen Lösungsmitteln durchführt.
2. Verfahren nach Patentanspruch oder Unteranspruch 1, dadurch gekennzeichnet, dass man das Arginin oder Histidin in der L-Form einsetzt.

The novel SH or SS glutathione salts according to the invention can be prepared by methods known per se, for example by dissolving the SH or SS glutathione and the basic amino acid or amino acids in water, aqueous solvents or other suitable solvents and isolating the salt formed.
The thus obtained SH or SS-glutathione amino acid salts remarkably increase the blood level of SH-glutathione even when administered orally as compared with the administration of SH or SS-glutathione alone.
The new SH or SS-glutathione salts prepared according to the invention can be administered in the usual way by all methods and can be processed into tablets or capsules, if necessary by adding suitable carriers and the like, and can also be used as injectable preparations.
The invention is illustrated by the following examples, but these are not intended to be restricted thereby.
Example I.
1.74 g of L-arginine are dissolved in 3.0 ml of an aqueous solution of 3.07 g of SH-glutathione while stirring, and about 20 ml of ethanol are added.
A white precipitate separates out and solidifies after a short time. The precipitate is filtered off and recrystallized from the water-ethanol in order to obtain 4.8 g of the colorless, hygroscopic SH-glutathione-L-arginine salt with a melting point of 200 to 2050 C (with decomposition).

Example 2
1.55 g of histidine are dissolved in 3.0 ml of an aqueous solution of 3.07 g of SH glutathione. The solution obtained is worked up according to Example 1.
4.6 g of the colorless, hygroscopic SH glutathione-L-histidine salt with a melting point of 197 ° to 2020 ° C. (with decomposition) are obtained.
