Verfahren zur Herstellung von neuen Furoxanderivaten
Die Erfindung betrifft ein Verfahren zur Herstellung von neuen Furoxanderivaten.

in welcher R1 ein Halogenatom, die Nitro- oder die Trifluormethyl gruppe, eine niedere Alkoxy- oder Alkylthiogruppe oder in o- oder m- Stellung eine niedere Alkylgruppe, R Wasserstoff, eine niedere Alkyl- oder Alkoxygruppe und Ra Wasserstoff oder eine niedere Alkoxygruppe bedeutet, sind bisher nicht bekannt geworden.
Wie nun gefunden wurde, besitzen solche Verbindungen, insbesondere das 3-(o-Tolyl)-4-amino-furoxan, wertvolle pharmakologische Eigenschaften. Sie sind zentrale Muskelrelaxantien mit guter antikonvulsiver Wirkung. Diese Wirkungsqualitäten werden durch ausgewählte Standardversuche [vgl. W. Theobald und H.A.
Kunz, Arzneimittel-Forsch. 13, 122-125 (1963)] nachgewiesen. Die neuen Verbindungen können zur Behandlung von pathologisch erhöhtem Muskeltonus, z.B. bei rheumatischen Erkrankungen, Fibrositis, Bursitis, Myositis, Torticollis, Spondylitis und Discopathien verwendet werden. Man kann sie auch mit Antirheumatica oder Analgetica kombinieren.
In den Verbindungen der allgemeinen Formel I können R1, R2 und R3 die o-, m- oder p-Stellung einnehmen.
R1 kann als Halogenatom das Chlor-, Fluor- oder Bromatom bedeuten und als niedere Alkylthiogruppe z.B. die Methylthio-, Äthylthio-, Propylthio-, Isopropylthio-, Butylthio-, Isobutylthio-, sek. Butylthio-, tert. Butylthio-, Pentylthio-, Isopentylthio- sowie die 2,2-Dimethyl-propylthiogruppe. R1 und R2 können als niedere Alkylgruppe beispielsweise die Methyl-, Äthyl-, Propyl-, Isopropyl-, Butyl-, Isobutyl-, sek. Butyl-, tert. Butyl-, Pentyl-, Isopentyl- oder 2,2-Dimethyl-propylgruppe und R1, R2 oder R3 als niedere Alkoxygruppe beispielsweise die Methoxy-, Äthoxy-, Propoxy-, Isopropoxy-, Butoxy-, Isobutoxy-, sek. Butoxy-, tert.Butoxy-, Pentoxy-, Isopentoxysowie die 2,2-Dimethyl-propoxygruppe bedeuten.
Nach dem erfindungsgemässen Verfahren setzt man eine Verbindung der allgemeinen Formel II, (11)

in welcher R1, R2 und Ra die unter Formel I angegebene Bedeutung haben, mit 1 Moläquivalent Ammoniak in Gegenwart eines Oxydationsmittels um.
Als Oxydationsmittel kann Kaliumferricyanid verwendet werden. Die Reaktion wird vorzugsweise in einem Lösungsmittel durchgeführt. Geeignete Lösungsmittel sind z.B. hydroxylgruppenhaltige Lösungsmittel, wie Wasser. Es ist vorteilhaft, äquivalente Mengen des Oxydationsmittels zu verwenden.
Ausgangsstoffe der allgemeinen Formel II sind z.B.
Verbindungen, deren Reste R1, R2 und R3 mit den Gruppen übereinstimmen, die anschliessend an Formel I genannt sind. Solche Verbindungen kann man z.B. herstellen, wenn man von Phenylacetophenonen ausgeht, die im Benzolkern durch die Reste R1, R2 und R5 substituiert sind, und diese Verbindungen mit n-Butyl- oder Isopropylnitrit in Äthanol in Gegenwart von Natrium äthylat zu entsprechenden Phenylglyoxal-aldoximen umsetzt und diese mit 1 Moläquivalent Hydroxylamin-by- drochlorid in Pyridin kondensiert.
Die täglichen Dosen der Wirksubstanzen bewegen sich zwischen 45 und 6000 mg für erwachsene Personen von normalem Körpergewicht. Für Kinder gilt der ent sprechend dem Körpergewicht verminderte Dosierungsbereich. Geeignete Doseneinheitsformen, wie Dragees, Tabletten, Kapseln, Suppositorien und Ampullen, enthalten vorzugsweise 15-1000 mg der Verbindungen.
Die nachfolgenden Beispiele erläutern die Herstellung der neuen Verbindungen der allgemeinen Formel I und von bisher nicht beschriebenen Zwischenprodukten näner, sollen jedoch den Umfang der Erfindung in keiner Weise beschränken. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel 1
Eine Lösung von 19,8 g (0,1 Mol) p-Chlor-phenyl) -glyoxim (Gemisch der a- und 5-Form) [vgl. L. Avogadro, Gazz. chim. ital. 53, 698 (1923); C.A. 18, 661(1924)] in 180 ml konz. Ammoniak und 350 ml Wasser wird unter Rühren und Eiskühlung mit einer Lösung von 135 g (0,4 Mol) Kaliumferricyanid in 400 ml Wasser so langsam versetzt, dass die Temperatur nicht über 300 steigt.
Man rührt das Reaktionsgemisch noch 30 Minuten und filtriert die Suspension. Der Filterrückstand wird mit Wasser gewaschen und aus Benzol sowie Cyclohexan umkristallisiert. Man trocknet die erhaltenen Kristalle 12 Stunden bei 700C, wonach man das 3-(p-Chlor-phenyl)-4-amino-furoxan vom Smp. 1670 erhält.

in which R1 is a halogen atom, the nitro or trifluoromethyl group, a lower alkoxy or alkylthio group or, in the o- or m- position, a lower alkyl group, R is hydrogen, a lower alkyl or alkoxy group and Ra is hydrogen or a lower alkoxy group, have not yet become known.
As has now been found, such compounds, in particular 3- (o-tolyl) -4-amino-furoxane, have valuable pharmacological properties. They are central muscle relaxants with good anticonvulsant effects. These effectiveness qualities are determined by selected standard tests [cf. W. Theobald and H.A.
Kunz, Pharmaceutical Research. 13, 122-125 (1963)]. The new compounds can be used to treat pathologically increased muscle tone, e.g. be used in rheumatic diseases, fibrositis, bursitis, myositis, torticollis, spondylitis and discopathies. They can also be combined with anti-inflammatory drugs or analgesic drugs.
In the compounds of the general formula I, R1, R2 and R3 can occupy the o-, m- or p-position.
As a halogen atom, R1 can represent the chlorine, fluorine or bromine atom and, as a lower alkylthio group, e.g. the methylthio, ethylthio, propylthio, isopropylthio, butylthio, isobutylthio, sec. Butylthio, tert. Butylthio, pentylthio, isopentylthio and the 2,2-dimethylpropylthio group. As a lower alkyl group, R1 and R2 can, for example, be methyl, ethyl, propyl, isopropyl, butyl, isobutyl, sec. Butyl, tert. Butyl, pentyl, isopentyl or 2,2-dimethylpropyl group and R1, R2 or R3 as a lower alkoxy group, for example methoxy, ethoxy, propoxy, isopropoxy, butoxy, isobutoxy, sec. Butoxy, tert-butoxy, pentoxy, isopentoxy and the 2,2-dimethylpropoxy group.

in which R1, R2 and Ra have the meaning given under formula I, with 1 molar equivalent of ammonia in the presence of an oxidizing agent.
Potassium ferricyanide can be used as an oxidizing agent. The reaction is preferably carried out in a solvent. Suitable solvents are e.g. solvents containing hydroxyl groups, such as water. It is advantageous to use equivalent amounts of the oxidizing agent.
Starting materials of the general formula II are e.g.
Compounds whose radicals R1, R2 and R3 correspond to the groups which are named after formula I. Such compounds can e.g. produce when starting from phenylacetophenones, which are substituted in the benzene nucleus by the radicals R1, R2 and R5, and these compounds with n-butyl or isopropyl nitrite in ethanol in the presence of sodium ethylate to corresponding phenylglyoxal aldoximes and these with 1 molar equivalent Hydroxylamine hydrochloride condensed in pyridine.
The daily doses of the active substances range between 45 and 6000 mg for adults of normal body weight. For children, the dosage range reduced in accordance with their body weight applies. Suitable unit dosage forms, such as dragees, tablets, capsules, suppositories and ampoules, preferably contain 15-1000 mg of the compounds.
The following examples illustrate the preparation of the new compounds of the general formula I and of intermediates not previously described, but are not intended to restrict the scope of the invention in any way. The temperatures are given in degrees Celsius.

A solution of 19.8 g (0.1 mol) of p-chloro-phenyl) -glyoxime (mixture of the a- and 5-form) [cf. L. Avogadro, Gazz. chim. ital. 53, 698 (1923); C.A. 18, 661 (1924)] in 180 ml conc. Ammonia and 350 ml of water are mixed with a solution of 135 g (0.4 mol) of potassium ferricyanide in 400 ml of water while stirring and with ice cooling so slowly that the temperature does not rise above 300.
The reaction mixture is stirred for a further 30 minutes and the suspension is filtered. The filter residue is washed with water and recrystallized from benzene and cyclohexane. The crystals obtained are dried at 70 ° C. for 12 hours, after which the 3- (p-chloro-phenyl) -4-aminofuroxane with a melting point of 1670 is obtained.
