Als Alkylrest mit 1-4 C-Atomen kommt z. B.
Methyl, Äthyl oder Propyl in Frage. Als Acylreste R', R" und R"' können Alkanoyl, z. B. Acetyl, Propionyl oder Butyroyl, ferner Aroyl, z. B. Eenzoyl, Toluyl oder Xyloyl, oder Alkoxyaroyl, z. B. Methoxybenzoyl oder Athoxybenzoyl, oder Halogenaroyl, z. B. Chlorbenzoyl oder Brombenzoyl oder auch Nitrobenzoyl in Frage kommen.
Beispielsweise wird eine 5-O-Acyl-1 ,2-G4sopropyl- idin-D-erythro-3-pentulofuranose mit einem Grignardreagens zu einer 5-O-Acyl- 1 2-isopropylidin-3 -allcyl-D- ribofuranose umgesetzt. Hierauf führt man die erhaltene 3-Alkyl-D-ribofuranose mittels saurer Alkoholyse in ein Alkyl - 5 -0 - acyl - 3-alkyl-D-ribofuranosid über, welches dann in basischem Milieu zum Alkyl-2,3,5-tri-O- acyl-3-alkyl-D-ribofuranosid acyliert wird.
Das so erhaltene Ribofuranosid kann dann mittels basischer Solvolyse gefolgt von einer weitern Hydrolyse mit einer starken Säure in wässrigem Medium in den entsprechenden freien Zucker übergeführt oder es kann in den entsprechenden Halogenester der Formel

in der Xa Halogen und R, R', R" und R"' die angegebene Bedeutung haben, durch Halogenierung mit einem Wasserstoffhalogenid in einem geeigneten Lösungsmittel übergeführt werden
Gemäss einer besonderen Ausführungsform des erfindungsgemässen Verfahrens wird eine 5-O-Acyl-1,2-O- isopropylidin-D-erythro-3-pentulofuranose mit einem Grignardreagens der angegebenen Formel in nahezu stöchiometrischem Verhältnis bei einer Temperatur zwischen 5 und ungefähr 800 C während einigen Minuten bis ungefähr 5 Stunden umgesetzt,
wobei sich eine 5-0 Acyl-1,2-O-isopropylidin-3-alkyl-D-ribofuranose bildet.
Als Grignardreagens kommt dabei ein C-C4-Alkylma- gnesiumhalogenid in Frage. Beispiele solcher Grignardreagenzien sind: Methylmagnesiumbromid, Äthylmagne- siumbromid, Methylmagnesiumchlorid, Athylmagnesium chlorid, Methylmagnesiumjodid, Äthylmagaesiumj odid oder Propylmagnesiumbromid.
Die saure Alkoholyse kann z. B. mit Salzsäure, Bromwasserstoffsäure oder Schwefelsäure während einigen Minuten bis ungefähr 5 Stunden bei 5 bis ungefähr 600 C erfolgen, wobei ein Alkyl-5-O-acyl-3-alkyl bribofuranosid entsteht, welches dann unter basischen Bedingungen z. B. mit Acylhalogenid oder einem entsprechenden Anhydrid behandelt wird. Als Acylierungsmittel eignen sich insbesondere Benzoylchlorid, Benzoylbromid, Nitrobenzoylchlorid, Essigsäureanhydrid oder Propionsäureanhydrid. Die Acylierungsstufe wird zweckmässig bei Temperaturen, die zwischen ungefähr 20 und 1000 C liegen, durchgeführt und dauert ungefähr 2 bis 72 Stunden. Als Endprodukt fällt ein 2,3,5 - Tri-O-acyl-3-alkyl - D - ribofuranosid an.
Dieses kann dann gewünschtenfalls mittels basischer Solfolyse in den freien Zucker übergeführt werden, wobei man das erhaltene Ribofuranosid mit einem Alkohol mit 1-4 C-Atomen bei Temperaturen zwischen 15 und ungefähr 600 C während einigen Minuten bis einigen Stunden behandelt und dann mit einer starken Säure wie Salzsäure, Bromwasserstoffsäure oder Schwefelsäure in wässrigem Medium während ungefähr 2 bis 24 Stunden bei 5 bis ungefähr 500 C behandelt. Die Über- führung des Ribofuranosids in den entsprechenden halogenierten Zucker mittels eines entsprechenden Wasserstoffhalogenids in einem geeigneten Lösungsmittel wie Essigsäure, Methylenchlorid, Tetrachlor äthan oder Propionsäure erfolgt vorzugsweise bei Temperaturen von ungefähr 5 bis 300 C während einigen Minuten bis einigen Tagen.
Alle Umsetzungen werden zweckmässig in im wesentlichen stöchiometrischen Verhältnis der Reaktionsteilnehmer durchgeführt.
Beispiele für nach dem erfindungsgemässen Verfanren erhältliche Verbindungen sind: 2-O-Acetyl-3,5-di-O-benzoyl-3-methyl-D ribofuranosylchlorid, 2-Acetyl-3,5-di-O-benzoyl-3-methyl-D- ribofuranosylbromid, 2-O-Acetyl-3,5-di-O-benzoyl-3-äthyl-D- ribofuranosylchlorid, 2-O-Acetyl-3,5 -di-O-benzoyl-3 -äthyl-D- ribofuranosylbromid, 2,3.
,5-Tri-O-benzoyl-3-methyl-D-ribofuranosylchlorid, 2,3 ,5-Tri-O-benzoyl-3-methyl-D-ribofuranosylbromid, 2,3,5-Tri-O-benzoyl-3-äthyl-D-ribofuranosylchlorid, 2,3,5 -Tri-O-benzoyl-3-äthyl-D-ribofuranosylbromid, 2,3,5-Tri-O-benzoyl-3 -propyl-D-ribofuranosylchlorid, 2,3 ,5-Tri-O-benzoyt-3-propyl-D-ribofuranosylbromid, 2,3 -Di-O-acetyl-5-O-benzoyl-3 -methyl-D ribofuranosylchlorid, 2,3 -Di-O-acetyl-5-O-benzoyl-3 -methyl-D- ribofuranosylbromid, 2,3 -Di-O-acetyl-5-O-benzoyl-3 -äthyl-D ribofuranosylchlorid, 2,3-Di-O-acetyl-5-O-benzoyl-3-äthyl-D ribofuranosylbromid, 3 -Methyl-D-ribofuranose, 3 -Äthyl-D-ribofuranose, 3 -Propyl-Di-ribofuranose .
Beispiel I Herstellung von Methyl-2,3,5-Tri-O-benzoyl-3-methyl
D-ribofuranosid
Das Grignardreagens Methylmagnesiumjodid, hergestellt durch Umsetzung von 690 mg (28,4 mMol) Magnesium mit 3,85 g (27,5 mMol) Methyljodid in 32 ml trockenem Äther, wird unter Rühren zu einer Lösung von 1,0 g (3,43 mMol) 1,2-O-Isopropylidin 5-O-benzoyl-a-D-erythro-pentulofuranose in 100 ml trockenem Äther bei 50 C gegeben. Nach ungefähr 3 Stunden wird die Reaktionsmischung in eine Mischung von 50 g Ammoinumchlorid, 200 ml Eiswasser und 200 ml Äther gegossen. Die Schichten werden voneinander getrennt und die wässrige Phase zweimal mit je 150 ml Äther extrahiert. Die über Magnesiumsulfat getrocknete ätherische Lösung wird zur Trockne eingedampft und der Rückstand (1,24 g) aus Äther umkristallisiert.
Man erhält dabei im ganzen 524 mg
1, 2-O-Isopropylidin-5-O-benzoyl-3 -methyl-a-D;ribofura- nose.
Eine Lösung von 1,0 g (3,25 mMol) 1,2-O-Isopro pylidin-5-o-benzoyl-3-methyl-a-D-ribofuranose in 25 ml 3 %iger methanolischer Salzsäure wird während 75 Minuten auf 250 C gehalten. Die Reaktionsmischung wird dann neutralisiert durch portionenweise Zugabe von 2,5 g (30 mMol) Natriumbicarbonat. Die Mischung wird dann filtriert und der Filterrückstand mit Wasser gewaschen. Das Filtrat und die Waschlösung werden konzentriert und dreimal mit je 50 ml Methylenchlorid extrahiert. Die Methylenchloridlösung behandelt man dann zur Entfärbung mit wenig Aktivkohle, filtriert danach und konzentriert ein. Der Rückstand wird an 20 g Silicagel chromatographiert.
Mit einer Athylacetat/Chloroformmischung (1: 9) eluiert, erhält man 290 mg an rohem Methyl 5-O-Benzoyl 2,3-O-isopropylidin;3-methyl-,ss-D-ribofuranosid. Eine weitere Eluierungsfraktion mit einer Athylacetat/Chloroformmischung (1: 9) ergibt ungefähr 240 mg eines gemischten Produktes. Schlussendlich ergibt eine Eluierfraktion mit einer Athylacetat/Chloroformmischung (1:1) 420 mg (46 %) an Methyl-5-O-benzoyl-3-methyl- D-ribofuranosid in Form eines Öles.
Die 420 mg (1,49 mMol) des so erhaltenen Methyl 5 - 0 benzoyl - 3 - methyl-D-ribofuranosides werden in 7,5 ml trockenem Pyridin gelöst und in einem Eisbad gekühlt. Danach gibt man tropfenweise unter Rühren eine Lösung von 463 mg (3,3 mMol) Benzoylchlorid in 2,5 ml trockenem Chloroform zu. Die Reaktionsmischung wird dann während 24 Stunden auf 250 C belassen, worauf 0,5 ml Wasser hinzugegeben werden.
Nach 30 Minuten wird die Mischung dann in 30 ml einer Mischung von Eis und Wasser gegossen und dann dreimal mit je 30 ml Chloroform extrahiert. Die Chloroformlösung wäscht man dann so lange mit S 'iger kalter Salzsäure, bis die Waschlösung sauer reagiert und anschliessend mit einer Natriumchloridlösung. Dann trocknet man die Chloroformlösung über Magnesiumsulfat und dampft sie zur Trockne ein. Der Rückstand ist das Methyl- 2,5- di- O-benzoyl-3-methyl- D-ribofuranosid.
Eine Lösung von 230 mg (0,595 mMol) Methyl 2,5-di-O-benzoyl-3-methyl-D-ribofuranosid in 3 ml trockenem Pyridin wird mit einer Lösung von 90 ml (0,64 mMol) Benzoylchlorid in 1 ml trockenem Chloroform behandelt. Die Reaktionsmischung wird während 16 Stunden auf 1000 C erhitzt, dann auf 250 C gekühlt und mit 0,5 ml Wasser behandelt und schliesslich wieder auf 400 C erwärmt. Anschliessend kühlt man die Lösung ab und gibt sie zu einer EislWassermischung und extrahiert danach dreimal mit je 50 ml Chloroform. Die Chloroformlösung wäscht man dann so lange mit 10:0iger Salzsäure, bis die Waschlösung sauer reagiert und danach mit 10 einem Natriumbicarbonat.
Danach trocknet man die Chloroformlösung über Magnesiumsulfat und konzentriert sie ein. Der Rückstand (370 mg) wird an 8 g Silicagel chromatographiert. Nach Aufarbeitung des Eluates erhält man 280 mg (95 S) an Methyl-2,3,5-tri-O-benzoyl-3-methyl D-ribofuranosid.
Beispiel 2
Herstellung von Methyl-2,3,5-tri-O-benzoyl-3-äthyl-
D-ribofuranosid
Wenn an Stelle des mit Methyljodid gemäss Beispiel 1 hergestellten Grignardreagens ein solches, welches mit Äthylbromid hergestellt wird, mit 1,2-O-Iso propylidin - 5-O-benzoyl-cl-D-erythro-3 -pentulofuranose zur Reaktion gebracht wird, so erhält man als Endprodukt das Methyl-2,3,5-tri-O-benzoyl-3-äthyl-D-ribofu- ranosid.
Beispiel 3 Herstellung von Athyl-2,3-diO-acetyl-5-O-benzoyl-3- methyl-D-ribofuranosid
Das Grignardreagens Methylmagnesiumjodid, hergestellt durch Umsetzung von 690 mg (228,4 mMol) Magnesium mit 3,85 g (27,5 mMol) Methyljodid in 32 ml trockenem Äther, wird unter Rühren zu einer Lösung von 1,0 g (3,42 mMol) 1,2-O-Isopropylidin 5-O-benzoyl-a-D-erythro-pentulofuranose in 100 ml trockenem Äther bei 50 C gegeben. Nach ungefähr 3 Stunden wird die Reaktionsmischung in eine Mischung von 50 g Ammoniumchlorid, 200 ml Eiswasser und 200 ml Ather gegossen. Die Schichten werden voneinander getrennt und die wässrige Phase zweimal mit je 150 ml Äther extrahiert. Die über Magnesiumsulfat getrocknete ätherische Lösung wird zur Trockne eingedampft und der Rückstand (1,24 g) aus Äther umkristallisiert.
Man erhält dabei im ganzen 524 mg
1,2- O-Isopropylidin-5-O-benzoyl-3-methyl-a-D-ribofu- ranose. Eine Lösung von 1,0 g (3,25 mMol) 1,2-0 isopropylidin-5-O-benzoyl-3-methyl-a-D-ribofuranose in 25 ml äthanolischer Salzsäure wird während 75 Minuten auf 250 C gehalten. Die Reaktionsmischung wird dann mit festem Natriumbicarbonat neutralisiert. Danach filtriert man die Mischung und konzemriert das Filtrat ein, welches dann mit Methylenchlorid extrahiert wird. Das Methylenchloridextrakt seinerseits wird ebenfalls eingeengt und das übrig bleibende Ö1 am Silicagel chromatographiert. Danach eluiert man mit einer Athylacetat/Chloroformmischung. Nach Aufarbeitung des Eluates erhält man Athyl-5-O-benzoyl-3- methyl-D-ribofuranose in Form eines Öles.
Eine Lösung von 1,0 g (3,4 mMol) Äthyl-5-O benzoyl-3-methyl-D-ribofuranosid in 20 ml trockenem Pyridin wird gekühlt und dann mit 1,0 g (10 mMol) Essigsäureanhydrid behandelt. Nachdem die Reaktionsmischung während 1 Stunde auf 250 C gehalten wurde, wird sie während 20 Stunden auf 1000 C erhitzt, dann wieder auf 250 C abgekühlt, worauf 1 ml Wasser hinzugegeben wird. Nach diesem Arbeitsprozess giesst man die Mischung in ein Eiswassergemisch und extrahiert anschliessend dreimal mit je 100 ml Chloroform.
Die Chloroformlösung wird zur Entfernung eines Pyridinüberschusses mi. einer 10 %igen Salzsäurelösung und danach bis zur neutralen Reaktion mit verdünnter Natriumbicarbonatlösung gewaschen. Danach trocknet man die Chloroformlösung über Magnesiumsulfat, engt sie ein und erhält als Rückstand das Athyl-2,3-di-O- acetyl-5 -O-benzoyl-3-methyl-D-ribofuranosid .
Beispiel 4
Herstellung von 2,3,5-Tri-O-benzoyl-3-methyl-D ribofuranosylbromid
Eine Lösung von 2 g (4,04 mMol) Methyl-2,3,5 tri-O-benzoyl-3-methyl-D-ribofuranosid (hergestellt nach Beispiel 1) in 10 ml Essigsäure wird in einem Eisbad gekühlt und dann 1 ml Acetylbromid hinzugegeben, worauf noch 10 ml einer 33 %igen Lösung von Bromwasserstoff in Essigsäure addiert werden. Nachdem di Reaktionsmischung während 15 Minuten auf einer Temperatur von 0 bis 50 C gehalten wurde, wird sie während 35 Minuten auf einer Temperatur von 250 C belassen. Dann engt man die Reaktionsmischung ein.
Das resultierende Ö1 wird von den übrigbleibenden Spuren des Bromwasserstoffes befreit, indem man es mit 3 Teilen trockenem Toluol destilliert. Man erhält so das 2,3,5-Tri-O-benzoyl-3-methyl-D-ribofuranosyl- bromid.
Beispiel 5 Herstellung von 2,3-Di-O-acetyl-5-O-benzoyl-3 -methyl-
D-ribofuranosylbromid
Eine Lösung von 1,52 g (4,0 mMol) Athyl-2,3-di O-acetyl-5-O-benzoyl-3-methyl-D-ribofuranosid (herge stellt nach Beispiel 3) in 10 ml Essigsäure wird in einem Eisbad gekühlt und dann gibt man zuerst 1 ml Acetylbromid und dann 10 ml einer 33 %igen Lösung von Bromwasserstoff in Essigsäure hinzu. Nachdem die Lösung während 50 Minuten auf 250 C gehalten wurde, wird die Reaktionsmischung eingeengt und man erhält als Rückstand das 2,3-Di-O-acetyl-5-O-benzoyl 3-methyl-D-ribofuranosylbromid. Verbleibende Spuren von Bromwasserstoff und Essigsäure werden noch entfernt, indem man das Produkt mit 3 Teilen trockenem Toluol destilliert.
Beispiel 6
Herstellung von 2,3 ,5-Tri-O-benzoyl-3 -methyl-D- ribofuranosylchlorid
Eine Lösung von 2 g (4,0 mMol) Methyl-2,3,5 Tri - O - benzoyl - 3-methyl-D-ribofuranosid (hergestellt nach Beispiel 1) in 10 ml Essigsäure wird auf einem Eisbad gekühlt und dann zuerst 1 ml Acetylchlorid und anschliessend 10 ml einer 20 %igen Salzsäurelösung in Essigsäure hinzugegeben. Nachdem die Reaktionsmischung während 48 Stunden auf 250 C belassen wurde, wird sie eingeengt. Anschliessend destilliert man den Rückstand zur Entfernung von noch vorhandenen Spuren von Essigsäure und Salzsäure mit 3 Teilen trockenem Toluol und erhält schliesslich das 2,3,5-Tri O-benzoyl-3 -methyl-D-ribofuranosylchlorid.
Beispiel 7
Herstellung von 3-Methyl-D-ribofuranose
Eine Lösung von 2,0 > g (4,0 mMol) Methyl-2,3,5 Tri - O - benzoyl - 3-methyl-D-ribofuranosid (hergestellt nach Beispiel 1) in 50 ml Methanol wird mit 5 ml einer 0,5 -n methanolischen Bariummethoxydlösung behandelt. Nachdem diese während 30 min auf Rückfluss gekocht wurde, wird sie auf ein Volumen von ungefähr
10 ml eingeengt und dann mit 50 ml Wasser verdünnt.
Nach Zugabe von 5 ml 0,5 -n Schwefelsäure fällt Bariumsulfat aus, das abfiltriert wird. Das Filtrat wird bis zu einem pH-Wert von 2 mit Salzsäure angesäuert und während mehrerer Stunden auf einer Temperatur von 600 C gehalten. Die Aufarbeitung der wässrigen Lösung ergibt die 3 -Methyl-D-ribofuranose.
Beispiel 8
Herstellung von 2,3,5-Tri-O-benzoyl-3-äthyl-D ribofuranosylbromid
Eine Lösung von 2,0 g (4,0 mMol) des nach Beispiel 1 erhaltenen 2,3, 5-Tri-O-benzoyl-3 -methyl-D-ribo- furanosides in 10 ml Essigsäure wird in einem Eisbad gekühlt, worauf man zuerst 1 ml Acetylbromid und dann 10 ml einer 33 %igen Lösung von Bromwasserstoff in Essigsäure hinzugibt. Nachdem die Reaktion mischung während 15 Minuten auf einer Temperatur, die zwischen 0 bis 50 C liegt, gehalten wurde, wird sie während 35 Minuten auf 250 C belassen. Nach Einengung der Lösung erhält man ein Ö1, das von noch verbleibenden Spuren von Bromwasserstoff und Essigsäure befreit wird, indem man es mit 3 Teilen Toluol destilliert. Man erhält das 2,3,5-Tri-O-benzoyl-3-äthyl- D-ribofuranosylbromid.
Beispiel 9
Herstellung von 3 Äthyl-D-ribofuranose
Eine Lösung von 2,05 g (4,0 mMol) Methyl-2,3,5 tri-O-3 -äthyl-D-ribofuranosid (hergestellt nach Beispiel 2) in 50 ml Methanol wird mit 5 ml einer 0,5 -n methanolischen Bari ummethoxydlösung behandelt.
Nachdem die Lösung während 30 Minuten auf Rückfluss gekocht wurde, engt man sie auf ein Volumen von ungefähr 10 ml ein und verdünnt anschliessend mit 50 ml Wasser. Man gibt dann 5 ml 0,5 -n Schwefelsäure hinzu, welche die Ausfällung von Bariumsulfat bewerkstelligt, welches abfiltriert wird. Anschliessend säuert man das Filtrat bis zu einem pH-Wert von 2 mit Salzsäure an und belässt die Mischung während einiger Stunden auf einer Temperatur von 600 C. Nach Einengung der wässrigen Lösung wird als Rückstand die 3 -Äthyl-D-ribofuranose erhalten.
Die nachfolgenden zwei Beispiele 16 und 17 erläutern die Verwendung einiger erfindungsgemäss erhältlichen Verbindungen als Ausgangsmaterialien zur Herstellung von wichtigen Nucleosiden, wobei natürlich auch mit anderen erfindungsgemäss erhaltenen Produkten Nucleoside hergestellt werden können.
Beispiel 10
Herstellung von 9-(3-Methyl-D-ribofuranosyl)
6-chlorpurin
Ungefähr 100 ml Xylol werden aus einer Suspension von 6,55 g (16,8 mMol) Chlormercuri-6-chlorpurin in 460 ml Xylol derweise abdestilliert, dass die in der Suspension noch vorhandenen Wasserspuren entfernt werden. Anschliessend gibt man unter Rühren zu der Suspension bei 250 C eine Lösung von 8,25 g (16,8 mMol) 2,3 ,5-Tri-O-benzoyl-3 -methyi-D-n.bofuran- oxylbromid in 40 ml trockenem Xylol hinzu. Die Mischung wird dann während 2 Stunden auf Rückfluss gekocht und anschliessend zur Entfernung von unlöslichen Materialien heiss filtriert. Das Filtrat engt man dann auf ein Volumen von 150 ml ein und verdünnt es anschliessend mit 300 ml Petroläther. Dann belässt man die Reaktionsmischung während 1 Stunde auf einer Temperatur von 50 C und filtriert ab.
Der Filterrückstand wird dreimal mit je 20 ml Petroläther gewaschen und getrocknet. Das Rohprodukt wird danach in 300 ml heissem Chloroform gelöst, dann zweimal mit je 80 ml 30 einem Kaliumjodid und anschliessend zweimal mit je 80 ml Wasser gewaschen. Aus der über Magnesiumsulfat getrockneten Chloroformlösung erhält man nach Einengung des 9-(2,3,5-Tri-Obenzoyl- 3-methyl-D-ribofuranosyl)-6-chlorpurin, welches noch mittels Chromatographie an einer kurzen Tonerdekolonne und Eluierung mit Chloroform gereinigt werden kann.
Eine Lösung von 479 mg (1,0 mMol) 9-(2,3,5-Tri O-benzoyl - 3 - methyl-D-ribofuranosyl)-6-chlorpurin in 20 ml kaltem Methanol, welches ausserdem noch 2 g wasserfreies Ammoniak enthält, wird während 20 Stunden auf 50 C gehalten. Anschliessend engt man die Lösung unter reduziertem Druck bei einer Temperatur unterhalb 200 C ein. Der Rückstand aus Methanol umkristallisiert ergibt 9-(3-methyl-D-ribofuranosyl)-6 chlorpurin.
Beispiel 11
Herstellung von 3'-Methyladenosin
Eine siedende Mischung von 605 mg (2,0 mMol) 9-(3 -methyl-D-ribofuranosyl)-6-chlorpurin in 30 ml wasserfreiem Methanol wird mit 20 ml einer mit Methylmercaptan gesättigten 0,1 -n methanolischen Natrium-Methoxydlösung behandelt. Nachdem die Reaktionsmischung während ungefähr 30 Minuten auf Rückfluss gekocht wurde, wird sie gekühlt und zur Trockne eingedampft. Den Rückstand löst man danach in heissem Wasser, wobei nach Abkühlen der Lösung das 9 - (3 - Methyl - D-ribofuranosyl)-6-methylthiopurin ausfällt.
Eine Mischung von 1,26 g (1,8 mMol) 9-(2,3,5-Tri O -benzoyl-3-methyl-ss-D-ribofuranosyl)-6-benzimidopu- rin und 13 ml trockenem Methanol wird mit einer Natriummethoxydlösung hergestellt durch Umsatz von 70 mg (3 mMol) Natrium in 3ml Methanol behandelt.
Nachdem die Mischung während 2 Stunden auf Rückfluss gekocht wurde, wird sie eingeengt und dann mit 50 ml Wasser verdünnt. Der pH-Wert wird durch Zugabe von 5 Tropfen Essigsäure von 11,5 auf 5,2 eingestellt. Anschliessend extrahiert man die Lösung fünfmal mit je 20 ml Chloroform und filtriert dann die wässrige Schicht und engt sie zur Trockne ein. Der Rückstand wird danach in Methanol gelöst, worauf 430 mg eines unreinen. amorphen Produktes mit Äther ausgefällt und abfiltriert wird. UNTERANSPRÜCHE
1. Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass die saure Alkoholyse bei einer Temperatur zwischen 5 und 600 C während einigen Minuten bis etwa 5 Stunden und die Acylierung unter basischen Bedingungen bei einer Temperatur zwischen 20 und 1000 C während 2 bis 72 Stunden erfolgt.
2. Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass man 5-O-Acyl-1,2-O-isopropyliden- 3-alkyl-n.bofuranose der Formel III durch saure Alkoholyse in Alkyl-5-O-acyl-3-alkyl-D-ribofuranosid der Formel IV und dieses durch basische Acylierung unter basischen Bedingungen in Alkyl-2,3,5-tri-O-acyl3-alkyl-D-ribofuranosid der Formel V überführt.
3. Verfahren nach Unteranspruch 2, dadurch gekennzeichnet, dass man 1 ,2-O-Isopropyliden-5-O-benzoyl-3-methyl-a-D-ribofuranose mit methanolischer Salzsäure bei etwa 250 C während 75 Minuten behandelt und das erhaltene Methyl-5-O-benzoyl-3 -methyl-D-ribo- furanosid der Formel IV mit Benzoylchlorid in Gegenwart von Pyridin bei etwa 1000 C während etwa 16 Stunden zu Methyl-2,3 , 5-tri-O-benzoyl-3 -methyl-D-ribofu- ranosid acyliert wird.

Example I Preparation of methyl 2,3,5-tri-O-benzoyl-3-methyl
D-ribofuranoside
The Grignard reagent methyl magnesium iodide, prepared by reacting 690 mg (28.4 mmol) of magnesium with 3.85 g (27.5 mmol) of methyl iodide in 32 ml of dry ether, is stirred into a solution of 1.0 g (3.43 mmol) 1,2-O-isopropylidine 5-O-benzoyl-aD-erythro-pentulofuranose in 100 ml of dry ether at 50 C. A solution of 1.0 g (3.25 mmol) of 1,2-O-isopropylidine-5-o-benzoyl-3-methyl-aD-ribofuranose in 25 ml of 3% methanolic hydrochloric acid is heated to 250 ° C. for 75 minutes held. The 420 mg (1.49 mmol) of the methyl 5-0 benzoyl-3-methyl-D-ribofuranosides thus obtained are dissolved in 7.5 ml of dry pyridine and cooled in an ice bath. A solution of 463 mg (3.3 mmol) of benzoyl chloride in 2.5 ml of dry chloroform is then added dropwise with stirring. The reaction mixture is then left at 250 ° C. for 24 hours, whereupon 0.5 ml of water is added.
After 30 minutes, the mixture is then poured into 30 ml of a mixture of ice and water and then extracted three times with 30 ml of chloroform each time. A solution of 2 g (4.04 mmol) of methyl 2,3,5 tri-O-benzoyl-3-methyl-D-ribofuranoside (prepared according to Example 1) in 10 ml of acetic acid is cooled in an ice bath and then 1 ml Acetyl bromide is added, whereupon 10 ml of a 33% solution of hydrogen bromide in acetic acid are added. A mixture of 1.26 g (1.8 mmol) of 9- (2,3,5-tri O -benzoyl-3-methyl-ss-D-ribofuranosyl) -6-benzimidopurine and 13 ml of dry methanol is with a sodium methoxide solution prepared by reacting 70 mg (3 mmol) of sodium in 3 ml of methanol.
After the mixture has been refluxed for 2 hours, it is concentrated and then diluted with 50 ml of water. The pH is adjusted from 11.5 to 5.2 by adding 5 drops of acetic acid. The solution is then extracted five times with 20 ml of chloroform each time and the aqueous layer is then filtered and evaporated to dryness. The residue is then dissolved in methanol, whereupon 430 mg of an impure. amorphous product is precipitated with ether and filtered off. The filtrate is then evaporated to dryness and the residue is crystallized from water. After recrystallization from 0.7 ml of water, 126 mg of 3'-methyladenosine are obtained.

SUBCLAIMS
1. The method according to claim I, characterized in that the acidic alcoholysis takes place at a temperature between 5 and 600 C for a few minutes to about 5 hours and the acylation under basic conditions at a temperature between 20 and 1000 C for 2 to 72 hours.

The method according to claim I, characterized in that 5-O-acyl-1,2-O-isopropylidene-3-alkyl-n.bofuranose of the formula III by acidic alcoholysis in alkyl-5-O-acyl-3- alkyl-D-ribofuranoside of the formula IV and this converted into alkyl-2,3,5-tri-O-acyl3-alkyl-D-ribofuranoside of the formula V by basic acylation under basic conditions.

The method according to dependent claim 2, characterized in that 1, 2-O-isopropylidene-5-O-benzoyl-3-methyl-aD-ribofuranose is treated with methanolic hydrochloric acid at about 250 C for 75 minutes and the methyl-5 obtained -O-benzoyl-3-methyl-D-ribofuranoside of the formula IV with benzoyl chloride in the presence of pyridine at about 1000 C for about 16 hours to give methyl-2,3,5-tri-O-benzoyl-3-methyl- D-ribofuranoside is acylated.
4. The method according to claim I, characterized in that the ribofuranose derivative of the formula V obtained by basic solvolysis and acidic hydrolysis into the free sugar of the formula

5. The method according to dependent claim 4, characterized in that the basic solvolysis between 15 and 600 C is carried out for a few minutes to a few hours.
