Verfahren zur Herstellung von Calcium-leucovorin
Gegenstand der vorliegenden Erfindung ist ein in technischem Massstabe gut durchführbares, vorteilhaftes und besonders ergiebiges Verfahren zur Herstellung von reinem Calcium-leucovorin mit höchster biologischer Aktivität.
Leucovorin fS-Formyl-5,6,7,8-tetrahydrn-folsäurej ist der spezifische Wachstumsfaktor von Leuconostoc citrovorum. Es ist eine Schlüsselsubstanz des Stoffwechsels. Als aEinkohlenstoff-überträger)y bei zahlreichen Formylierungs- respektive Transmethylierungs-Reaktio- nen zum Aufbau zellwichtiger Substanzen, insbesondere auch der Desoxiribonukleinsäure, spielt diese Verbindung eine wichtige Rolle und ist daher für den Zellstoffwechsel und die Zellteilung von Bedeutung.
Calcium-leucovorin - die am besten haltbare Form des Leucovorin - findet demgemäss Verwendung zur Behandlung besonders der perniziösen Leukämie, von megaloblastären Anämien bei der Schwangerschaft und von Leberparenchymschäden. Käufliches Calcium-leucovorin ist bis anhin stets unrein. Seine biologische Aktivität beträgt nur etwa zwei Drittel bis drei Viertel der chemisch ganz reinen Verbindung und das Papierchromatogramm eines käuflichen Produktes zeigt dementsprechend immer mehrere deutlich erkennbare Flecken.
Dies ist keineswegs verwunderlich, da die Synthese von Leucovorin viele Reaktionsstufen erfordert, die teilweise in saurem und teilweise in stark alkalischem Milieu durchgeführt werden müssen. Sowohl die Zwischenprodukte als auch das Endprodukt sind jedoch vorzugsweise in saurer und teilweise auch in stark alkalischer Lösung instabil. Vgl. dazu: Jukes, Science 120, Seite 324 (1954). Ganz besonders empfindlich ist das essentielle Zwischenprodukt, die 10-Formyl-tetrahydro-folsäure, welche durch Luftsauerstoff rasch zerstört wird. May et al., J. Amer chem. Soc. 73, 3067 (1951).
Aber auch dessen Reaktionsfolgeprodukt, das Anhydro-leucovorin, d.i. die 5,10-Anhydro-formyl- bzw.
5.10-Methenyl-tetrahydrofo]säure, ist - vor allem in Lösung - recht instabil.

Zwischenprodukte werden dabei nicht isoliert, allenfalls werden sie durch Eindampfen der Reaktionslösung konzentriert.
Bei den bisher bekannten Verfahren kann bei der Isolierung und Reinigung des Leucovorins auf dessen erhebliche Instabilität im sauren Gebiet nicht Rücksicht genommen werden.

Da infolge der Instabilität besonders der Zwischenprodukte in Lösung stets in erheblichem Masse mit Nebenreaktionen gerechnet werden muss und anderseits kein Zwischenprodukt in fester Form isoliert und damit von den Begleitstoffen und Nebenprodukten befreit wird, ist die im besten Fall erzielbare Ausbeute und Reinheit sehr begrenzt.
Es wurde nun gemäss der vorliegenden Erfindung gefunden, dass die Herstellung von Calcium-leucovorin mit vorher unerreicht hoher Ausbeute und Reinheit gelingt, wenn man das bisher nie isolierte Anhydro-leucovorin (5,10-Anhydroformyl- oder -Methenyl-tetrahydro- folsäure) aus dem Reaktionsgemisch, welches durch Anhydrisierung von mittels Hydrierung und Formylierung aus Folsäure gewonnener 10-Formyl-tetrahydro-folsäure erhältlich ist, durch Fällen mit einem Keton isoliert, damit von zersetzenden Reagenzien - insbesondere von Ameisensäure - und von Nebenprodukten rasch abtrennt,
und dass man danach das aus dem isolierten Anhydro-leucovorin durch Erwärmen mit verdünntem Alkalihydroxid erhaltene Alkalisalz von 5-Formyl-tetrahydro-folsäure unter strikter Vermeidung eines sauren Reaktionsmilieu's durch Zusatz eines Calciumsalzes direkt in das Calcium-leucovorin überführt.
Die Isolierung von Anhydro-Leucovorin erfolgt dabei vorzugsweise durch Fällen mit Methyl-isobutyl-keton.
Das aus dem isolierten Anhydro-leucovorin durch Erhitzen, vorzugsweise mit Natronlauge, gewonnene Natriumsalz von 5-Formyl-tetrahydro-folsäure wird in alkalischem Milieu direkt durch Zusatz eines leicht wasserlöslichen Calciumsalzes einer starken Säure, insbesondere von Calciumchlorid, in das stabile Calcium-leucovorin übergeführt.
Ketone sind als Fällungsmittel für Anhydro-leucovorin spezifisch geeignet. Anhydro-leucovorin wird nämlich durch ein Keton in einer besonders gut und sehr schnell filtrierbaren resp. zentrifugierbaren reinen Form ausgefällt und kann daher rasch weiter umgesetzt werden.
Bei Verwendung beispielsweise eines Alkohols, wie etwa von Isopropanol, als Fällungsmittel wird das Anhydro-leucovorin in sehr feiner, äusserst schwierig filtrierbarer Form erhalten.
Das bisher nicht angewandte strikte Vermeiden eines sauren Reaktionsmilieu's bei der Bildung des Calciumsalzes von Leucovorin hat den Zweck, die teilweise Zer störung der in saurem Gebiet sehr instabilen 5-Formyl -tetrahydro-folsäure zu vermeiden, verhindert also die Bildung von Nebenprodukten und führt damit ebenfalls zu einer Verbesserung der Reinheit und auch der Ausbeute.
Calcium-leucovorin wird nach dem erfindungsgemässen Verfahren mit einer Ausbeute von mindestens 30%, bezogen auf die eingesetzte Folsäure, erhalten. Diese Ausbeute übertrifft diejenige der vorbekannten Methoden etwa um das Doppelte.
Zudem weist das erfindungsgemäss erhaltene Verfahrensprodukt eine bisher unerreichte Reinheit auf.
A usführungsbeispiel
850 g Folsäure (92%ig) werden nach bekannter Methode durch ca. einstündiges Erwärmen auf 500 C formyliert und nach Zusatz von Platinoxid und Ascorbinsäure hydriert.
Nun wird durch halbstündiges Erhitzen auf 90 bis 950 C das 5-Formyltetrahydro-natrium-float gebildet. Die erhaltene Reaktionslösung wird vorsichtig, unter strikter Vermeidung eines auch nur teilweise sauren Milieu's auf einen pH-Wert von 7,5 eingestellt, mit einem Adsorbens aus synthetischem Magnesiumsilikat versetzt, zentrifugiert, im Vakuum bei 300 C auf ein Volumen von etwa 5 bis 6 Liter eingedampft, mit 1,3 Liter einer 20%igen (g/v) wässerigen Lösung von Calciumchlorid versetzt und durch Zusetzen von ca. 30 Liter Äthanol das rohe Calcium-leucovorin ausgefällt. Dieses wird bei 50 C abzentrifugiert, mit Äthanol gewaschen und getrocknet.
Die Feinreinigung erfolgt durch Chromatographieren an einer mit synthetischem Magnesiumsilikat beschickten Säule.
Ausbeute: 325 g reines Calcium-leucovorin 5 H20 [5-Formyl-d.1 -5,6,7,8.tetrahydro-pteroyl-L( + ) - glutamin säure. d.s. 30,5% bezogen auf die eingesetzte Polsäure.

Die entstandene 10-Formyl-tetrahydro-folsäure wird anhydrisiert durch 2stündiges Erwärmen auf 50 bis 520 C in einer Stickstoffatmosphäre.
Die ameisensaure Reaktionslösung, welche ein Volumen von 7 bis 8 Liter aufweist, wird in 60 Liter Methyl-isobutyl-keton eingerührt; das dabei ausfallende Anhydro-leucovorin wird nach 10minütigem Stehenlassen abzentrifugiert, mit 3 Liter Aceton gewaschen und sofort in verdünnte, eiskalte Natronlauge eingetragen.
Das Fällen, Zentrifugieren und Eintragen in die Natronlauge soll weniger als eine Stunde beanspruchen.
Das nach dem erfindungsgemässen Verfahren in höchster Ausbeute erhaltene Ca-Leucovorin ist demnach bei weitem reiner als das analog deklarierte Vergleichs- präparat des Handels.
Process for the manufacture of calcium leucovorin

Leucovorin fS-formyl-5,6,7,8-tetrahydric folic acid is the specific growth factor of Leuconostoc citrovorum. It is a key metabolic substance. Up to now, commercially available calcium leucovorin has always been impure. Its biological activity is only about two-thirds to three-quarters of that of the chemically pure compound and the paper chromatogram of a commercially available product accordingly always shows several clearly recognizable spots.
This is by no means surprising, since the synthesis of leucovorin requires many reaction steps, some of which have to be carried out in an acidic and some in a strongly alkaline medium. Both the intermediate products and the end product, however, are preferably unstable in acidic and sometimes also in strongly alkaline solution. See: Jukes, Science 120, page 324 (1954). The essential intermediate, 10-formyl-tetrahydro-folic acid, which is quickly destroyed by atmospheric oxygen, is particularly sensitive. May et al., J. Amer chem. Soc. 73, 3067 (1951).
But also its reaction product, the anhydro-leucovorin, i.e. the 5,10-anhydro-formyl or
5,10-methenyl-tetrahydrofo] acid is - especially in solution - quite unstable.

Intermediate products are not isolated, at best they are concentrated by evaporating the reaction solution.
In the processes known up to now, in the isolation and purification of leucovorin, its considerable instability in the acidic region cannot be taken into account.

Since, due to the instability of the intermediates in solution in particular, side reactions must always be expected to a considerable extent and, on the other hand, no intermediate product is isolated in solid form and thus freed from accompanying substances and by-products, the yield and purity that can be achieved in the best case are very limited.
It has now been found according to the present invention that the production of calcium-leucovorin with previously unattainable high yield and purity succeeds if the anhydro-leucovorin (5,10-anhydroformyl- or -methenyl-tetrahydro- folic acid), which has never been isolated, is successful the reaction mixture, which can be obtained by anhydration of 10-formyl-tetrahydrofolic acid obtained from folic acid by means of hydrogenation and formylation, isolated by precipitation with a ketone, so that it is quickly separated from decomposing reagents - in particular formic acid - and by-products,
and that the alkali salt of 5-formyl-tetrahydro-folic acid obtained from the isolated anhydroleucovorin by heating with dilute alkali hydroxide is then converted directly into the calcium leucovorin by strictly avoiding an acidic reaction medium by adding a calcium salt.
Anhydro-leucovorin is preferably isolated by precipitating it with methyl isobutyl ketone.
The sodium salt of 5-formyl-tetrahydro-folic acid obtained from the isolated anhydro-leucovorin by heating, preferably with sodium hydroxide solution, is converted into the stable calcium leucovorin in an alkaline medium by adding a readily water-soluble calcium salt of a strong acid, in particular calcium chloride .
Ketones are specifically suitable as precipitants for anhydro-leucovorin. Anhydro-leucovorin is namely by a ketone in a particularly good and very quickly filterable resp. centrifugable pure form precipitated and can therefore be quickly implemented.
If, for example, an alcohol such as isopropanol is used as the precipitating agent, the anhydro-leucovorin is obtained in a very fine, extremely difficult-to-filter form.
The so far not applied strict avoidance of an acidic reaction medium in the formation of the calcium salt of leucovorin has the purpose of avoiding the partial destruction of the 5-formyl-tetrahydrofoic acid, which is very unstable in acidic areas, thus prevents the formation of by-products and thus also leads to it to improve the purity and also the yield.
Calcium leucovorin is obtained by the process according to the invention with a yield of at least 30%, based on the folic acid used. This yield roughly doubles that of the previously known methods.
In addition, the process product obtained according to the invention has a purity that has not been achieved before.
Embodiment example
850 g of folic acid (92%) are formylated by a known method by heating to 500 ° C. for about one hour and, after adding platinum oxide and ascorbic acid, hydrogenated.
The 5-formyltetrahydro-sodium float is now formed by heating at 90 to 950 C for half an hour. The reaction solution obtained is carefully adjusted to a pH of 7.5, strictly avoiding an even partially acidic environment, mixed with an adsorbent made of synthetic magnesium silicate, centrifuged, in vacuo at 300 ° C. to a volume of about 5 to 6 liters evaporated, 1.3 liters of a 20% (w / v) aqueous solution of calcium chloride were added and the crude calcium leucovorin was precipitated by adding about 30 liters of ethanol. This is centrifuged off at 50 C, washed with ethanol and dried.
The fine purification is carried out by chromatography on a column loaded with synthetic magnesium silicate.
Yield: 325 g of pure calcium leucovorin 5 H20 [5-formyl-d.1 -5,6,7,8.tetrahydro-pteroyl-L (+) - glutamic acid. d.s. 30.5% based on the poly acid used.

The resulting 10-formyl-tetrahydrofolic acid is anhydrated by heating for 2 hours at 50 to 520 ° C. in a nitrogen atmosphere.
The acidic reaction solution, which has a volume of 7 to 8 liters, is stirred into 60 liters of methyl isobutyl ketone; the anhydro-leucovorin which precipitates is centrifuged off after standing for 10 minutes, washed with 3 liters of acetone and immediately added to dilute, ice-cold sodium hydroxide solution.
Felling, centrifuging and adding to the sodium hydroxide solution should take less than an hour.
The Ca-leucovorin obtained in the highest yield by the process according to the invention is accordingly by far purer than the commercially available comparative preparation declared analogously.
