Wie die Laktone der allgemeinen Formel I als auch die Säuren der allgemeinen Formel IV zeigen einen bedeutsamen therapeutischen Effekt bei Versuchstieren mit gewissen transplantierten Tumoren, und zwar nach peroraler bzw. parenteraler Verabreichung. Die Toxi- zität der Stoffe ist sehr niedrig, und es wurden auch bei langzeitiger Verabreichung keine unerwünschten Ne benwirkungen beobachtet.
Gemäss vorliegender Erfindung werden die gγAryl- aαss-dihalogencrotonlaktone der allgemeinen Formel dadurch erhalten, dass man eine Mukohalogensäure der allgemeinen Formel II verwendet 2

Diese Kondensierung kann man zweckmässig in Gegenwart von wasserfreiem Zinkchlorid oder in Gegen wart eines Gemisches von wasserfreiem Zinkchlorid mit Phosphorpentoxyd, bei einer Temperatur von 60 bis 85 C, vorteilhaft bei 75 bis 80 C, durchführen.
Die Anwendung von wasserfreiem Zinkchlorid (höchstens mit 5 % Wassergehalt), und zwar in einer halben bis gleichen Gewichtsmenge der als Ausgangs material dienenden Mukohalogensäure der allgemeinen Formel II oder von seinem Gemisch mit 5 bis 20 Phosphorpentoxyd, bei der Kondensierung der erwähn ten Säure mit dem substituierten Benzol der allgemeinen Formel III, bietet eine Reihe von Vorteilen im Ver gleich mit der bereits zu diesem Zwecke benützten Polyphosphorsäure.
Man umgeht die verhältnismässig anspruchsvolle Herstellung der erforderlichen grossen Menge dieser Säure, das Volumen des Reaktionsge misches wird kleiner, die Ausbeuten sind höher und gut reproduzierbar, die Ausgangskomponenten und das Re aktionsprodukt sind einem kleineren Zersetzungseinfluss ausgesetzt, darüber hinaus ist es auch möglich, einige Derivate, beispielsweise y-4-Hydroxy-2-methoxyphenyl- aαss-dibromcrotonlakton, herzustellen, welche unter Ver wendung der Polyphosphorsäure als Kondensations mittel nicht zu gewinnen sind.
Die optimale Reaktionsdauer hängt besonders vom Charakter des substitutierten Benzols der allgemeinen Formel III ab; sie kann von 15 Minuten bis 8 Stunden variieren und ihre weitere Verlängerung hat infolge der nicht aggressiven Natur des Kondensationsmittels in der Regel keinen merklichen Einfluss auf die Ausbeute des Endproduktes.
Die Isolierung der Laktone aus dem Reaktions gemisch ist sehr einfach, sie kann beispielsweise ent weder durch Verteilung zwischen Chloroform und Was ser, Verdampfen des organischen Anteils zur Trockne und Reinigung des Rückstandes durch Umkristallisieren aus einem geeigneten Lösungsmittel oder durch Chro matographie an einer Silikagelsäule und nachfolgendes Umkristallisieren des erhaltenen Produktes aus einem organischen Lösungsmittel verwirklicht werden.
Die erfindungsgemäss erhaltenen Crotonlaktone der Formel I können verwendet werden, um durch Hydro lyse die entsprechenden Akrylsäuren der Formel IV herzustellen. Die Hydrolyse der gγAryl-aαss-dihalogen- ^a,ss-crotonlaktone der allgemeinen Formel I führt man am besten durch Einwirkung von Wasser im Gemisch mit einem organischen, mit Wasser mischbaren Lösungs mittel, vorteilhaft Dioxan, in Gegenwart von Zinkoxyd, bei Siedetemperatur des Reaktionsgemisches durch, wobei man zur Hydrolyse Zinkoxyd zweckmässig in einer Menge von 1 bis 4 Mol, bezogen auf 1 Mol des Ausgangslaktons der allgemeinen Formel I, verwendet.
Unter diesen Bedingungen verläuft die Hydrolyse in sehr guter Ausbeute, in gewissen Fällen fast quantitativ. Das entstandene Zinksalz der ss-Aryl-ss-halogenakryl- säure ist im Vergleich beispielsweise mit dem Magne sium- oder Natriumsalz wesentlich stabiler und erleidet während des Reaktionsverfahrens keine Zersetzung. Der bei der Hydrolyse des Laktons entstandene Halogen wasserstoff kann in Form des entsprechenden Zinkhalo genids gebunden werden.
Die Anwendung des Zink oxyds ermöglicht, besonders infolge der hohen Stabilität des entstandenen Zinksalzes und ebenfalls infolge seines minimalen aggressiven Einflusses auf diese Salze, die Gewinnung von gewünschten Säuren in hohen Ausbeu ten, meistens über<B>90%,</B> und zwar in fast analytischem Reinheitsgrad.
Die ss-Aroyl-ss-halogenakrylsäuren der allgemeinen Formel IV kann man beispielsweise so isolieren, dass man nach Beendigung der Hydrolyse das Reaktions gemisch unter vermindertem Druck zur Trockene ver dampft, den Rückstand unter Zusatz von Natrium- hydrogencarbonat in Wasser löst, wonach man die ge wünschte Säure aus der Lösung ihres Natriumsalzes durch Ansäuern mit Salzsäure in Freiheit setzt. Man kann auch so vorgehen, dass man den Rückstand zwi schen verdünnte Salzsäure und Chloroform verteilt, den Chloroformextrakt zur Trockene verdampft und den Rückstand durch Kristallisation reinigt.
<I>Beispiel 1</I> -4-n-Hexyloxyphenyl-aαss-dibrom-^aαss-crotonlakton Ein Gemisch von 25,8g Mukobromsäure, 13,6 g wasserfreiem Zinkchlorid, 1,36 g Phosphorpentoxyd und 17,8 g n-Hexylphenyläther erhitzt man unter Aus schluss der Luftfeuchtigkeit, unter Rühren, 5 Stunden in einem bei 75 bis 80 C gehaltenen Bad.
Nach Abkühlen der Schmelze auf etwa 40 C gibt man 125 ml Chloro- form und 50 ml etwa 30 C warmen Wassers hinzu und verrührt gründlich die entstandene Mischung. Die Chloroformschicht trennt man ab, schüttelt sie wieder holt mit Wasser aus und trocknet sie über wasserfreiem Natriumsulfat. Nach Filtrieren entfernt man das Lö sungsmittel durch Destillation. Man erhält 36,1 g (86 %) viskoses Rohprodukt, das durch Chromatographie an einer Silikagelsäule, unter Verwendung von Benzol als Eluierungsmittel, den kristallinen Stoff in einer Aus beute von 34,4 g (82% mit F. 56 bis 58 C (Methanol) liefert.
<I>Beispiel 2</I> gγ4-Hydroxy-2-methoxyphenyl-a,ss-dibrom- ^aαss-crotonlakton Ein Gemisch von 25,8g Mukobromsäure, 12,4 g Resorcinmonomethyläther und 13,6 g wasserfreiem Zinkchlorid erhitzt man unter Ausschluss der Luft feuchtigkeit, unter Rühren, 3 Stunden in einem bei 75 bis 80 C gehaltenen Bad. Nach Aufarbeitung des Re aktionsgemisches wie im vorstehenden Beispiel (ohne Chromatographie) erhält man das Produkt in einer Ausbeute von 28,7 g (79 %).
Nach Umkristallisieren aus Methanol schmilzt der reine Stoff bei 155 bis 146 C. <I>Beispiel 3</I> γ-4-n-Pentoxyphenyl-aαss-dichlor-^aαss-crotonlakton Unter Verwendung von 16,9 g Mukochlorsäure, 16,4 g n-Pentylphenyläther, 13,6g wasserfreiem Zink chlorid und 1,36 g Phosphorpentoxyd erhält man durch analoges Verfahren wie in den vorstehenden Beispielen, das gewünschte Produkt in einer Ausbeute von 28,6 g (91 %); F. 58 bis 59 C (Methanol).
In analoger Weise, unter Verwendung von densel ben Molverhältnissen der entsprechenden Reaktions komponenten und derselben Menge des wasserfesten Zinkchlorids und Phosphorpentoxyds, wurden in Aus beuten von 80 bis 95 % folgende Verbindungen her gestellt:

Beispiel 4 ss-4-Äthoxybenzoyl-ss-bromakrylsäure Zum siedenden Gemisch von 240 ml Wasser und Dioxan (1 : 1) und 5 g Zinkoxyd tropft man innerhalb 20 Minuten eine Lösung von 22,2 g gγ4-Äthoxyphenyl- aαss-dibrom-^aαss-crotonlakton in 20 ml Dioxan zu, und zwar so schnell, dass das Reaktionsgemisch fortwährend siedet.
Nach weiteren 75 Minuten Sieden entfernt man aus dem Reaktionsgemisch die Lösungsmittel durch Destillation bei 40 C/14 Torr und extrahiert den Rück stand 1 Stunde mit einer 30 bis 35 -C warmen Lösung von 24g Natriumhydrogencarbonat in 240 ml Wasser. Die erhaltene Lösung des ss-4-Äthoxybenzoyl-brom- akrylsäurenatriumsalzes säuert man nach dem Filtrieren unter starkem Rühren mit konzentrierter Salzsäure auf pH 2 bis 3 an. Die ausgeschiedene Säure saugt man nach Abkühlen auf 5 C ab und trocknet sie an der Luft bei Zimmertemperatur. Ausbeute: 17,2 g (94%).
F. 149 bis 150 C (Benzol).
Das Reaktionsgemisch kann man nach dem Ab destillieren der flüchtigen Anteile zweckmässig auch so aufarbeiten, dass man den Rückstand mit Salzsäure und Chloroform durchschüttelt, in welchen die in Freiheit gesetzte ss-4-Athoxybenzoyl-ss-bromakrylsäure fast quan titativ übergeht.
