Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung von neuen tricyclischen Verbindungen der allgemeinen Formel

Verbindungen der Formel II sind bekannt und können z. B. gemäss Chem. Ber., 84, 671-679 (1951) hergestellt werden. Verbindungen der Formel III sind ebenfalls bekannt.
Das erfindungsgemässe Verfahren wird zweckmässig in an sich bekannter Weise unter den für Grignard Reaktionen üblichen Bedingungen durchgeführt. Besonders günstige Ausbeuten werden erhalten, wenn X in der Formel III Chlor darstellt. Gemäss einer bevorzugten Ausführungsform wird lO-Brom- (oder Chlor-)-di benzo[a,d]cycloheptal[l,4,6]trien-5-on mit einem 3 Methoxypropylmagnesiumhalogenid, insbesondere dem Chlorid, umgesetzt.
Die Verfahrensprodukte sind wichtige Zwischenprodukte für die Herstellung von in 5-Stellung eine basisch substituierte Seitenkette tragenden Verbindungen, welche sich durch vielfältige Wirkungen auf das Nervensystem auszeichnen. Solche Verbindungen können z. B. hergestellt werden, indem man die Verbindungen der Formel I halogeniert und anschliessend mit einem Amin umsetzt.
Beispiel 1
In einem 2-Liter-Dreihalskolben mit Rührer, Tropftrichter und Kühlvorrichtung wurden 15 g Magnesiumspäne mit 50 ml Ather überdeckt und mit 0,5 ml Methyljodid versetzt. Nach dem Nachlassen der lebhaften Reaktion wurde eine Lösung von 54,6 g 1-Chlor-3methoxy-propan in 300 ml trockenem Ather derart zugetropft, dass das Reaktionsgemisch am Sieden gehalten wurde. Es wurde 3 Stunden bei 45 unter Rückfluss gekocht.
Dann wurde das Reaktionsgemisch mit Eiswasser gekühlt, eine Lösung von 69,2 g 10-Brom-dibenzo[a,d]- cyclohepta 1,4,6]trien-5-on in 500 ml Äther im Laufe einer Stunde zugetropft und das Ganze noch 16 Stunden bei 400 unter Rückfluss gerührt. Am folgenden Tag wurde nochmals mit Eiswasser gekühlt und die Reaktionsmischung mit einer kalt gesättigten Ammo niumchlondlösung versetzt. Die organische Schicht wurde dann abgetrennt, die wässrige Phase zweimal mit je 150 ml Äther ausgeschüttelt und die vereinigten Ätherextrakte über Natriumsulfat getrocknet und eingedampft.
Der Rückstand ergab, nach Umkristallisieren aus hochsiedendem Petroläther, farblose, bei 82840 schmelzende Kristalle von 1 0-Brom-5 -hydroxyl-5-(3'- methoxy-propyl)-dibenzo[a,djcyclohepta[1 ,4,6jtrien.
50 g des erhaltenen 10-Brom-5-hydroxy-5-(3'- methoxy-propyl)-dibenzo[a,dJ cyclohepta[ 1,4, 6]trien und 500 ml einer 3 siegen alkoholischen Chlorwasserstofflösung wurden 3 Stunden unter Rückfluss am Dampfbad erwärmt und dann der Alkohol unter vermindertem Druck abgedampft. Der Rückstand wurde in Äther aufgenommen, die ätherische Lösung mit Wasser, Natriumbicarbonatlösung und nochmals mit Wasser gewaschen, über Natriumsulfat getrocknet und eingedampft.
Durch Destillation des Rückstandes unter einem Druck von 0,05 mm Hg bei 1550 wurde das 10-Brom-5-(3' methoxypropyliden) - dibenzo[a,d]cyclohepta[l,4,6]trien als dickes, bald kristallisierendes Ö1 erhalten.
Beispiel 2
In einem 2-Liter-Dreihalskolben mit Rührer, Tropftrichter und Kühlvorrichtung wurden 15 g Magnesiumspäne mit 50 ml trockenem Äther überdeckt und mit 0,5 ml Methyljodid versetzt.
Nach dem Nachlassen der lebhaften Reaktion wurde eine Lösung von 54,6 g 1-Chlor-3-methoxy-propan in
300 ml Ather derart zugetropft, dass der Äther am Sieden gehalten wurde. Es wurde noch 3 Stunden unter Rückfluss gekocht.
Dann wurde das Reaktionsgemisch mit Eiswasser gekühlt, eine Lösung von 58,4 g 10-Chlor-dibenzo[a,d] cyclohepta[l,4,6]trien-5-on in 500 ml trockenem Äther im Laufe einer Stunde zugetropft und das Ganze noch
16 Stunden bei 400 unter Rückfluss gerührt. Am folgenden Tag wurde nochmals mit Eiswasser gekühlt und mit einer kaltgesättigten Ammoniumchloridlösung ver setzt. Die organische Schicht wurde dann abgetrennt, die wässrige Phase zweimal mit je 150 ml Äther ausgeschüttelt und die vereinigten Ätherextrakte über Natrium sulfat getrocknet und eingedampft. Der Rückstand ergab, nach Umkristallisieren aus hochsiedendem Petrol äther, farblose, bei 82-890 schmelzende Kristalle von 10-Chlor-5-hydroxy-5-(3'-methoxy-propyl) dibenzo[a,d] cyclohepta[ 1 ,4,6trien.
35 g 1 0-Chlor-5-hydroxy-5 -(3 -methoxy-propyl)-di- benzo[a,d]cyclohepta[l,4,6]trien und 350 ml einer 3 %gen, alkoholischen Chlorwasserstofflösung wurden 4 Stunden unter Rückfluss am Dampfbad erwärmt und dann der Alkohol unter vermindertem Druck abgedampft. Der Rückstand wurde in Äther aufgenommen, die ätherische Lösung mit Wasser, Natriumcarbonatlösung und nochmals mit Wasser gewaschen, über Natriumsulfat getrocknet und eingedampft. Durch Destillation des Rückstandes unter einem Druck von 0,1 mm Hg bei 1600 C wurde das lO;Chlor-5-(3'-methoxy-pro- pyliden)-dibenzo[a,d]cyclohepta[1,4,6]trien als dickes, bald erstarrendes öl erhalten.

The process according to the invention is expediently carried out in a manner known per se under the conditions customary for Grignard reactions.
