Verfahren zur Herstellung von 2-Sulfonamidopyrimidin-derivaten
Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung von 2-Sulfonamidopyrimidin-derivaten sowie deren Salzen. Die neuen Verbindungen, falls sie eine Hydroxy]gruppe enthalten, können auch durch Umsetzung mit niederen oder mittleren aliphatischen Mono- oder Dicarbonsäuren, aromatischen oder araliphatischen Carbonsäuren in die entsprechenden Ester überführt werden.
Die neuen Verbindungen werden durch die folgende Formel dargestellt:

in der X ein Sauerstoff- oder Schwefelatom oder eine direkte C-C-Bindung, Y ein Sauerstoffatom oder eine direkte C-C-Bindung, R und R' ein Wasserstoffatom oder einen niederen oder mittleren Alkylrest oder R und R' zusammen mit dem Stickstoffatom einen heterocyclischen Ring bedeuten und R" für einen unverzweigten oder verzweigten, gesättigten oder ungesättigten, gegebenenfalls durch ein oder mehrere Sauerstoffatome unterbrochenen 2wertigen Kohlenwasserstoffrest und Z für ein Wasserstoffatom oder eine Hydroxylgruppe steht.
Aus der belgischen Patentschrift Nr. 609 270 ist bekannt, dass u.a. 2-Benzolsulfonamido-5-nieder-alkoxy äthoxypyrimidine, die im Phenylrest durch niedere Alkyloder Alkoxygruppen und/oder Halogen substituiert sein können, blutzuckersenkende Eigenschaften besitzen.
Die erfindungsgemäss erhältlichen Verbindungen weisen ebenfalls hervorragende blutzuckersenkende Eigenschaften auf. Die Senkung des Blutzuckerspiegels wurde an Kaninchen bestimmt. Nach 24stündiger Nüchternheit wurde der Blutzuckerspiegel der Versuchstiere (Initialkonzentration) gemessen und anschliessend die wässrige Lösung des Natriumsalzes der Testsubstanz per os appliziert.
Die guten blutzuckersenkenden Eigenschaften der neuen Verbindungen werden bei spiel smässig durch die folgende Tabelle gezeigt.

Aus der Tabelle ist ersichtlich, dass die erfindungsgemäss erhältlichen Substanzen selbst in so geringen Mengen wie 8 bzw. 16 mg/kg eine beachtliche Wirkung zeigen.
Zum therapeutischen Gebrauch können die neuen Sulfonamide verabreicht werden als freie Verbindungen, als Salze mit physiologisch vertretbaren anorganischen und/oder organischen Basen, wie z.B. Natrium-, Lithium-, Calcium-, Ammoniumhydroxyd, Aminen wie Methylglukamin, Morpholin, Piperazin, Äthanolamin u.a., oder auch in Form von Mischungen der freien Sulfonamide mit einem geeigneten Alkalicarbonat bzw. -hydrogencarbonat.
Die Konfektionierung der Substanzen kann ohne oder mit den in der galenischen Pharmazie üblichen Zusätzen, Trägersubstanzen und Geschmackskorrigenzien erfolgen, und zwar beispielsweise in Pulverform, als Tabletten, Dragees, Kapseln, Pillen, in Form von Suspensionen oder Lösungen. Die Substanzen sollen gewöhnlich in einer Menge von 0,01 - 0,6 g/kg Körpergewicht verabfolgt werden. Die Sulfonamide sollen normalerweise oral und zu Testzwecken auch intravenös appliziert werden.

Die so erhaltenen Produkte können ohne Isolierung mit den entsprechenden Guanidinderivaten erfindungsgemäss kondensiert werden.
Die neuen Produkte können mit physiologisch vertretbaren anorganischen und/oder organischen Basen in die entsprechenden Salze überführt werden
Beispiel 1
2,5 g Natrium werden in 250 ml Methanol gelöst.
Dann werden 16 g a-n-Propoxy-p-dimethylaminoacrolein und 40 g 4-Dimethylaminocarbonylmethylenbenzolsul guanidin zugegeben. Das Gemisch wird 8 Stunden unter Rückfluss gekocht, anschliessend das Methanol abdestilliert und der Rückstand mit Wasser versetzt. Der Niederschlag von überschüssigem Sulfoguanidin wird abfiltriert und das Filtrat mit Salzsäure angesäuert. Zur Reinigung wird das ausgefällte Produkt nochmals in verdünntem Ammoniak gelöst, mit Kohle filtriert und mit Salzsäure wieder ausgefällt. Es werden so 30 g 4-(5-n-Propoxy pyrimidinyl -2) -sulf onamidophenylessigsäuredimethylamid vom Schmelzpunkt 1370C erhalten.

Beispiel 2
1 kg 4-(5-n-Propoxypyrimidinyl-2)-sulfonamido-phenylessigsäuredimethylamid, 50 g Polyvinylpyrrolidon, 50 g Kartoffelstärke und 5 g Magnesiumstearat werden homogen gemischt und zu Tabletten mit Bruchrille von 551 mg gepresst.
Die Tabletten enthalten 500 mg Wirkstoff.
Beispiel 3
1 kg 4-(5-n-Propoxypyrimidinyl-2)-sulfonamido-phenylessigsäurediäthylamid werden auf einer Strahlmühle zu Partikelchen von maximal 4 ,u Grösse vermahlen.
Nach Zumischen, zwecks besserer Fliesseigenschaft, von 1% kolloidaler Kieselsäure (AEROSIL'µ') wird in Hartgelatinekapseln (Grösse 1) gefüllt, dass in jeder Kapsel 100 mg Wirkstoff enthalten sind.
Beispiel 4
1 kg leicht wasserlösliches Natriumsalz von 4-(5-n -Propoxypyrimidinyl - 2)-sulfonamido-phenoxyessigsäurediäthylamid wird mit 450 ml Äthylalkohol verknetet, granuliert und über ein Wirbelstromtrockner bis zur Alkoholfreiheit geschickt. Dem Granulat mischt man 150 g Maisstärke, 26 g Talkum und 4 g Magnesiumstearat hinzu. Dann wird in stabförmige Tabletten (5,5 X 17,5 X 6,5 mm) gepresst. Gewicht der Tabletten 590 mg (500 mg Wirkstoff). Die Stabtabletten werden mit einer Lösung von 30 g Polyglykol 6000 in 70 ml Aceton in einem Dragierkessel überzogen und getrocknet.
Beispiel 5
Zur Herstellung von Injektionslösungen zur intravenösen Applikation des Natriumsalzes (Beispiel 4) werden 1 kg Salz in 19,339 Ltr. Ampullenwasser, welches 1 g äthylendiamintetraessigsaures Natrium enthält, gelöst.
D20 = 1,017, pH-Wert: 8,5 - 8,7. Nach Filtration über Seitz Filterschicht EKS II wird in Ampullen von 20 ml abgefüllt und 20 Minuten bei 1200C sterilisiert.
Beispiel 6
1 kg 4-(5-Methoxypyrimidinyl-2)-sulfonamido-phenoxyessigsäurediäthylamid wird mit 450 g pressfähigem Calciumcarbonat, 45 g Maisstärke und 5 g Magnesiumstearat gemischt und zu Tabletten, wie üblich von 150 mg Gewicht entsprechend 100 mg Wirkstoff, verpresst.
Beispiel 7
4 kg 4-(5-Hydroxyäthoxypyrimidinyl-2)-sulfonamido- -phenoxyessigsäurediäthylamid werden auf einer Strahlmühle bis auf Teilchengrösse von ca. 6 > vermahlen. Anschliessend wird jeweils eine Menge von 400 mg in Hartgelatinekapseln (Grösse 0) abgefüllt.
