Verwendung neuer kernjodierter Benzoesäurederivate als Röntgenkontrastmittel
Gegenstand der vorliegenden Erfindung ist die Verwendung neuer kernjodierter Benzoesäurederivate als Röntgenkontrastmittel. Es handelt sich dabei um Verbindungen der nachstehenden Formel I

Besonders bevorzugt sind dabei Verbindungen, in denen X den Methylen-, Äthylen, oder Methyläthylenrest darstellt.
Es wurde gefunden, dass die neuen Verbindungen der Formel I Röntgenkontrastmittel sind, die sich vor allem zur Darstellung des Gallensystems sowohl nach intravenöser, vor allem aber peroraler Applikation ausgezeichnet eignen. Für die perorale Verabreichung können die erfindungsgemäss zu verwendenden Röntgenkontrastmittel tablettiert, in Gelatinekapseln abgefüllt oder in Drageeform gebracht werden, wobei sie zweckmässig mit inerten Füll-und Bindemitteln, wie Stärke, Talkum usw. gemischt werden. Die bevorzugte Verab- reichungsform sind die Salze, z. B. das Natriumsalz.
Für die intravenöse Verabreichung dienen bevorzugt die wässrigen Lösungen der Salze, z. B. des Natrium-, des Methylglukamin-, oder Diäthanolaminisalzes.
Die neuen Röntgenkontrastmittel können auf einfache Weise durch Alkylierung, der in der österreichi- schen Patentschrift Nr. 224 264 beschriebenen, entsprechenden 3-Acylamino-2, 4, 6-trijodbenzoesäureamide an der Acylaminogruppe in 3-Stellung erhalten werden.
Als Alkylierungsmittel können dabei Alkyl-bzw. Alke nylester anorganischer Säuren, insbesondere Alkyl-bzw.
Alkenylhalogenide, Dialkylsulfate und Dialkenylsulfate dienen.
Besonders vorteilhaft gestaltet sich die Herstellung der Verbindungen der Formel I, wenn man die neuen Säurechloride der allgemeinen Formel II

in der R2 und X wie oben definiert sind oder mit deren Estern bei erhöhter Temperatur umsetzt, worauf vor handene Estergruppen gewünschtenfalls verseift werden können. Aus der so erhaltenen alkalischen Verseifungs- lösung können entweder direkt die Alkalisalze durch Fällen mit einem geeigneten organischen Lösungsmittel, wie z. B. Aceton, gewonnen werden oder aber die Verbindungen nach Ansäuern in Form der freien Säuren isoliert werden.
Die Umsetzung erfolgt vorzugsweise in einem indifferenten organischen Lösungsmittel, wie beispielsweise Chloroform, Tetrahydrofuran, Dioxan, Aceton, Methyl- äthylketon, Chlorbenzol, Toluol usw. Es ist aber auch die Durchfiihrung der Reaktion ohne Lösungsmittel möglich.
Die neuen Säurechloride der Formel II können entweder durch Alkylierung von 3-Acylamino-2, 4, 6-tri jodbenzoylchlorid mit Alkylierungsmitteln, wie Alkyl- halogenide oder Dialkylsulfate, oder durch Umsetzung der N-Alkyl-N-acyl-2, 4, 6-trijodbenzoesäuren mit Thionylchlorid, vorzugsweise in indifferemtem, organischem Medium erhalten werden.
Diese Herstellungverfahren werden anhand folgender Vorschriften näher erläutert :
18, 45 g 3-N-Ally1-N-acetyl-2, 4, 6-trijodbenzoylchlorid werden in 30 ml Chloroform gelöst und mit 8 g ss-Amino- propionsäureäthylester versetzt. Nach Abklingen der Reaktion wird das Gemisch noch 1 Stunde unter Rückfluss gekocht, dann wird abgekühlt und die Lö, sung mit Chloroform verdünnt. Nach Waschen derselben mit ver dünnter Salzsäure, Wasser, Natriumbicarbonatlösung und wiederum Wasser wird die Chloroformlosung ge- trocknet und eingedampft Der Eindampfrückstand wird in Alkohol gelöst und nach Zugabe von 50 ml ln Natronlauge 10 Minuten zum Sieden erhitzt.
Die so erhaltene alkalische Lösung wird mit Wasser verdünnt, mit Kohle filtriert und mit Salzsäure auf pH 1 angesäuert, wobei die rohe Säure ausfällt, die aus Essigester umkristallisiert wird. Man, erhält so 13, 5 g N- (3-N'-Acetyl-N'-allylamino-2, 4, 6-trijodbenzoyl) ss-aminopropionsäure vom Fp. 187 bis 193 C.
27,36 g N-(3-N'-Acetylamino-2,4,6-trijodbenzoyl) N-n-propyl-ss-amino-α-methylpropionsäure werden in 100 ml 2n Natronlauge gelöst und innerhalb von 10 Minuten bei 0 bis 10 C unter Kühlung mit einer Lösung von 12, 93 g Diäthylsulfat in 30 ml Aceton versetzt.
Nach zweistündiger Reaktionszeit wird das Aceton im Vakuum entfernt und die Lösung, die durch Filtration von unlöslichen Anteilen befreit wurde, mit Salzsäure auf pH 1 gebracht. Die entstandene Fällung wird isoliert, in 300 ml Essigester gelöst, die Lösung getrocknet und auf ein Volumen von 100 ml eingeengt, wobei die Säure auskristallisiert. Man erhält so 21 g N- (3-N'-Aceyl-N'-äthylamino-2, 4, 6-trijodlbenzoyl)- N-n-propyl-ss-amino-α-methylpropionsäure vom Fp. 170 bis 178 C.
In analoger Weise können alle anderen Röntgenkontrastmittel gemäss der Formel I erhalten werden.
Die Verbindungen der allgemeinen Formel I exi- stieren zum Teil in zwei geometrisch isomeren Formen, deren Auftrennung prinzipiell möglich ist.
Die Verbindungen der Formel I sind wertvolle Röntgenkontras. tmiMd, die vor allem für die Cholecysto- graphie und zum Teil auch die Cholangiographie geeignet sind. Unter diesen befinden sich Verbindungen, die auf Grund ihrer niedrigen Toxizität und guten Ver träglichkeit für die intravenöse Verabreichung in Frage kommen. Als Vertreter dieser Verbindungen ist bei- spielsweise die N- (3'-N'-Acetyl-N'-methylamino-2, 4, 6-trijodbenzoyl)- N-y-methoxypropyl-ss-aminopropionsäure zu nennen.
Besonders hervorzuheben ist jedoch, dass eine eihe der Verbindungen der Formel I aus dem Darm sehr rasch und vollständig resorbiert und schnell über die Galle ausgeschieden werden, wobei sehr hohe Jodkonzentrationen in der Galle erzielt werden. Diese Ver bindungen der Formel I, unter denen z. B. die
N-(3'-N'-Acetyl-N'-methylamino-2, 4, 6-trijodbenzoyl) N-allyl-ss-amino-α
-methylpropionsäure oder die N- (3-N'-Acetyl-N'-allylamino-2, 4, 6-trijodbenzoyl)-
N-methyl-ss-aminopropionsäure zu nennen sind, sind daher Mittel für die Schnellchole- cystographie nach oraler Verabreichung, wobei die Bilder der Gallenblase bereits nach 1-2 Std. sichtbar werden und nach 3 Stunden schon optimale Bilder erhalten wer- den können. Die üblichen ocalen Gallenkontrastmittel rufen dagegen erst nach etwa 14 Stunden eine Sichtbar machung der Gallenblase hervor. Die Verbindungen der Formel I zeichnen sich femer durch eine für orale Gallenkontrastmittel überraschend günstig liegende intravenöse, akute Toxizität aus.
Die Brauchbarkeit der Verbindungen der Formel I wurde in pharmakologischen Untersuchungen neben Toxizitätsuntersuchungen an Mäusen sowohl an Ausscheidungsversuchen an der Ratbe als auch an der Kontrastdarstellung an der Katze geprüft. Als Verbindungen, die sich bei diesen Untersuchungen bewährt haben, können genannt werden :

N-(3-N'-Acetyl-N'-methylamino-2, 4, 6-trijodbenzoyl) N-3-methoxypropyl-ss-aminopropionsäure werden in 2000 ml 0, 5n NaOH gelöst und mit Wasser auf 2856 ml aufgefüllt.
Man erhält so eine Lösung mit einem Gehalt von 25, 0 g Saure auf 100 ml Lösung.
Beispiel 2
668, 0 g N- (3-N'-Acetyl-N'-methylamino-2, 4, 6-trijodbenzoyl) N-allylaminoessigsäure werden in 500 ml 2n NaOH und 400 ml Wasser gelöst, dann 16, 7 g Polyvinylpyrrolidon in Lösung gebracht und mit Wasser auf ein Volumen von 1670 ml ergänzt. Die Lösung enthält pro 100 ml 40, 0 g Säure.
Beispiel 3
714, 1 g N- (3-N'-Acetyl-N'-methylaniino-2, 4, 6-trijodbenzoyl) N-3-metthoxypropyl-ss-aminopropionsäure werden mit 193, 20 g N-Methyl-d-glucosamin und 1500 ml Wasser in Lösung gebracht und, um eine Lösung mit 25, 0 g Säure pro 100 ml zu erhalten, auf 2856 ml verdünnt.
Die Lösungen der Beispiele 1 bis 3 sind für Injektionszwecke geeignet.
Beispiel 4
750, 0 g N- (3-N'-acetyl-N'-methylamino-2, 4, 6-trijodbenzoyl) N-n-propyl-ss-amino-a-methylpropionsaures Natrium werden mit 157, 0 g Sbärke, 26, 4 g Talkum und 6, 6 g Magnesiumstearat versetzt, homogenisiert und aus der Mischung Tabletten zu 940 mg gepresst. Die Tabletten enthalten 750 mg Wirkstoff.

Das fertige Granulat wird mit weiteren 100 g Maisstärke und 5 g Magnesiumstearat vermengt und zu Tabletten mit einem Wirkstoffgehalt von 500 mg verpresst.

N-(3-N'-Acetyl-N'-allylamino-2, 4, 6-trijodbenzoyl)
N-methyl-ss-aminopropionsäure-Natriumsalz 300 mg Erdnussöl und 50 mg Lezithin werden zu einer fliessfähigen Paste angerührt und in Gelatinekapseln abgefüllt.
Diese sind zur oralen Verabreichung geeignet.
Auch die übrigen genannten Verbindungen können auf analoge Weise in Tabletten, Kapseln oder Lösungen übergeführt werden.

The new X-ray contrast media can be obtained in a simple manner by alkylating the corresponding 3-acylamino-2,4,6-triiodobenzoic acid amides described in Austrian patent specification No. 224 264 on the acylamino group in the 3-position.

Some of the compounds of the general formula I exist in two geometrically isomeric forms, the separation of which is possible in principle.
The compounds of the formula I are valuable X-ray contrasts. tmiMd, which are particularly suitable for cholecystography and to some extent also for cholangiography. These include compounds which are suitable for intravenous administration because of their low toxicity and good tolerability. Examples of these compounds are N- (3'-N'-acetyl-N'-methylamino-2,4,6-triiodobenzoyl) - N-γ-methoxypropyl-β-aminopropionic acid.
It should be particularly emphasized, however, that a number of the compounds of the formula I are absorbed very quickly and completely from the intestine and rapidly excreted via the bile, with very high iodine concentrations being achieved in the bile. The compounds of the formula I are furthermore distinguished by an intravenous, acute toxicity which is surprisingly favorable for oral bile contrast media.
