Verfahren zur Herstellung von 1,1-Diphenyl-1-methoxy -3benzylaminopropan und dessen Salzen
Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung des neuen l, l-Diphenyl-l-metho- xy-3-benzylaminopropans sowie von dessen Salzen.
Diese neue Verbindung zeigt interessante pharmakologische Eigenschaften. Sie besitzt eine analgetische Wirkung, die nicht so stark ist, wie bei Morphium oder ähnlichen Verbindungen, die nur unter strenger ärztlicher Kontrolle verabreicht werden können, sondern es handelt sich um ein Analgeticum mit schwächerer, nicht Sucht erzeugender Wirkung, das sich besonders zur Behandlung von Schmerzzuständen aller Art im täglichen Leben und aussrhalb der Kontrolle des Arztes eignet. Dadurch unterscheidet sich die erfindungsgemässe Substanz von den bekannten Analgeticis aus der Diphenylmethanreihe. Verabreicht man die Verbindung in einer Einzeldosis von 15 mg an Menschen, so kommt es zu einer 3 bis 4 Stunden andauernden, guten analgetischen Wirkung. Bei stärkeren Schmerzen kann die Dosis unbedenklich auf 20 mg erhöht werden.
Ein nachteiliger Einfluss auf Blutbild und Hambefund konnte auch bei längerer Verabreichung bisher nicht festgestellt werden.
Das Verfahren zur Herstellung des 1,1-Diphenyl-1- methoxy-3-benzylaminopropan und von dessen Salzen ist dadurch gekennzeichnet, dass Diphenylmethanderiva- te der Formel I,

in der A die Gruppierung -N = CH- oder -NH-CO- bedeutet. reduziert, und bei der Reaktion anfallende Basen im Salze übergeführt bzw. solche aus Salzen in Freiheit gesetzt werden. Die Reduktion kann beispielsweise sehr vorteilhaft mit Hilfe von komplexen Metallhydriden, wie beispielsweise Natriumborhydrid, Kaliumborhydrid oder Lithiumaluminiumhydrid vorgenommen werden, wobei die üblichen Reaktionsbedingungen eingehalten werden können.
Vorteilhafterweise arbeitet man in einem Lösungsmittel wie beispielsweise Methanol, Äther, Dioxan, und bei einer Temperatur, die zwischen Raumtemperatur und dem Siedepunkt des verwendeten Lösungsmittels liegt. Die Isolierung des gebildeten 1,1 - Diphenyl-l-methoxy-3 ; benzylaminopropans gelingt nach
Zersetzung des überschüssigen Metallhydrids mit Wasser am besten durch Säurebehandlung.
Die Verbindungen der Formel I können aber auch durch katalytische Hydrierung unter Aufnahme von 1
Moläquivalent Wasserstoff mittels Metallkatalysatoren vor allem auf Platin- oder Palladiumbasis oder auch mittels Raney-Nickel reduziert werden. Diese Methode hat sich vor allem bei Verbindungen der Formel I bewährt, in denen A die Gruppierung -N=CH- bedeutet.
Im Falle, dass als Katalysator Palladiumkohle dient, hat sich überraschenderweise gezeigt, dass die in diesem Falle zu erwartende hydrogenolytisohe Abspaltung des Benzylrestes so viel langsamer verläuft, als die Reduk tion der Gruppierung A, dass auch mit diesem Katalysator die gewünschte Endverbindung in sehr guter Ausbeu te erhalten wird.
Bei der Reaktion anfallende Basen können durch
Umsetzung mit den entsprechenden Säuren in Salze übergeführt werden. Fallen bei der Reaktion direkt
Salze, z. B. halogenwasserstoffsaure Salze an, so können daraus die freien Basen oder auch Salze mit anderen
Säuren gewonnen werden. Als solche können z. B.
Sulfate, Nitrate, Succinate, Toiuolsulfonate, Mandelate, Malate, Tartrate und Cyclohexylsulfamate genannt werden.
Die zur Reduktion gelangenden Verbindungen der allgemeinen Formel I sind neu. Sie können erhalten werden, indem 1, 1-Diphenyl-1-methoxy-3-aminopropan mit Benzoldehyd und/oder Derivaten der Benzoesäure wie Benzoylchlorid oder Benzoesäureanhydrid zur Reaktion gebracht wird. Das 1,1 -Diphenyl-1-methoxy-3 -ami- nopropan kann erhalten werden, indem Benzhydrylmethyläther mit ss-Chloräthylamin in flüssigem Ammoniak mittels Natrium-oder Kaliumamid kondensiert wird.
Die Isolierung des Verfahrensproduktes kann nach üblichen Methoden geschehen, vorzugsweise durch Eindampfen der Reaktionslösung oder dadurch, dass man es mittels Mineralsäuren dem Reaktionsgemisch entzieht.
Beispiel 1
2,0 g Lithiumaluminiumhydrid und 150 ccm absolu- ter Äther werden in einen Reaktionskolben gebracht, der mit einem Extraktionsapparat nach Soxhlet versehen ist und dessen Filterhülse mit 5,0 g N-(3,3-Diphenyl-3methoxypropyl)-benzamid gefüllt wurde. Durch Sieden des Athers wird allmählich die Substanz gelöst und in das Reaktionsgefäss gespült. Es wird anschliessend noch fünf Stunden lang gekocht, das Reaktionsgemisch mit Wasser zersetzt und nach Abtrennen der Lithiumsalze mit Salzsäure ausgezogen. Aus der sauren Lösung kristallisiert das Hydrochlorid des 1,1-Diphenyl-1-me- thoxy-3-benzylaminopropans. Es werden 4,9 g erhalten, dies entspricht 92 /o der Theorie.
Nach dem Umkristallisieren aus Alkohol war der Schmelzpunkt 186-188 OC.
Daraus wird die freie Base durch Lösung in heissem Wasser und Alkalisieren mit Natronlauge erhalten. Sie hat den Schmelzpunkt 68-69 OC.
Das N-(3 ,3 -Diphenyl-3-methoxypropyl)enzamid wird durch Umsetzung von 1,1-Diphenyl-1-methoxy-3- aminopropan (FP 66-68 "C) mit Benzoesäureanhydrid und Triäthylamin erhalten. Es besitzt nach Umkristallisieren aus Toluol einen FP von 157-158 OC.
Aus der Base können Salze hergestellt werden, indem die ätherischen Lösungen der Base und der entsprechenden Säure vereinigt werden, wobei das Salz nach einigem Stehen ausfällt. Auf diese Weise wurden folgende Salze erhalten: Nitrat FP 133 - 134,5 OC D-Tartrat FP 150 - 151 OC D,L-Malat FP 153 - 155 OC Succinat FP 142-1440C Cyclohexylsulfamat FP 127-129 OC L-Mandelat FP 134-135,50C
Beispiel 2 13,0 g N-(3, 3-Diphenyl-3-methoxypropyi)-benzaldi- min werden in 200 ccm Alkohol mit 1,0 g Palladium- kohle bei Raumtemperatur hydriert.
Nach Aufnahme von 1 Moläquivalent Wasserstoff wird die Hydrierung abgebrochen, der Katalysator abgetrennt und die Lösung eingedampft. Es werden 12,5 g 1,1-Diphenyl-1-metho- xy-3zbenzylaminopropan erhalten, was einer Ausbeute von 96 O/o entspricht. Nach dem Umkristallisieren aus Alkohol war der Schmelzpunkt 66-69 OC. Das N < 3, 3- Diphenyl-3-methoxypropyl)-benzaldimin wird auch durch Umsatz von 1,1 -Diphenyl-1 -methoxy-3 -aminopro- pan mit Benzaldehyd in Alkohol erhalten. Es besitzt einen FP von 138-140 OC.
Beispiel 3
7,0 g N-(3, 3 Diphenyl-3-methoxypropyl)-benzaldi- min werden in 200 com Methanol suspendiert und eine Lösung von 0,6 g Natriumborhydrid in 20 ccm Methanol zugegeben. Das Reaktionsgemisch wird zunächst solange gerührt, bis alle Festanteile vollkommen gelöst sind, dann wird die Reaktion bei 50 "C unter Rühren noch eine halbe Stunde vollenden Gelassen. Nach beendeter Reaktion wird mit Salzsäure angesäuert, die Lösung eingedampft, der Eindampfrückstand alkalisiert und mit Äther ausgezogen. Nach Abdampfen des ethers werden 6,4 g 1,1 -Diphenyl-1 -methoxy-3-benzylamino- propan erhalten, das entspricht einer Ausbeute von 91 O/o der Theorie.
