Verfahren zur Herstellung des neuen Antibioticums SL <B>3042</B> Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung eines neuen Antibioticums, das nach stehend mit SL 3042 bezeichnet wird.
Erfindungsgemäss, gelangt man zu dem neuen Anti= bioticum SL 3042, indem man einen neuen Stamm der Pilzspecies Panus conchatus (Ball. ex. Fr.) Fr. in einer Nährlösung züchtet und das- Antibioticum aus der Fer- mentationsbrühe auf an sich bekannte Weise, z. B. durch Extraktion- oder Adsorption, isoliert und reinigt.
Der neue; erfindungsgemäss verwendete Stamm von Panus- conchatus (Bull. ex. Fr.)- Fr. wurde aus einem in Tirol (Oesierreich) gefundenen Fruchtkörper isoliert und eine Probe davon beim United States De= partment of Agricultare (Northem Utilization Research and Development Division), Peoria,
I11./USA, unter der Nummer NRRL 3253 deponiert.
Der neue Stamm Panus conchatus (Bull. ex. Fr.) Fries, NRRL 3253, wächst auf einem Malz-Hefeex- trakt-Agar bei 27 C mit einem, farblosen Substratmy- cel und einem lockeren weissen Luftmycel. Das Mycel zeigt die für Basidiomyceten typische Schnallenbildung. Der Durchmesser der hyalinen Hyphen variiert von 1,6 bis 4,1,u.
Auf seinem natürlichen- Substrat, an Stümpfen- und Ästen der Zitterpappel und Buche, fruktifiziert Panus conchatus entsprechend' der Beschreibung von R.
Küh ner und H. Romagnesi, Flore analytique des Champi gnons superieurs, Massen- et Cie., Paris 1953, S. 70, und derjenigen Beschreibung von R. Singer, The Aga- ricales in Modern Taxonomy, Weinheim, published by J. Cramer 1962,<B>9.172.</B>
Für das- erfindungsge sässe. Verfahren können auch Stämme verwendet werden; wie sie z- B. durch Selek tion oder Mutation unter Einwirkung von Ultraviolett- oder Röntgenstrahlen oder durch Anwendung anderer Massnahmen, wie z B-. durch. Behandlung von Labora- toriumskulturen- mit geeigneten Chemikalien aus dem neuen Stamm Panus eonehatus- (Bull. ex.
Fr.) Fries ge wonnen werden: können- Der, neue Stamm von Panus conehatus (Buil: ex. Fr.) Fries lässt sich auf vielerlei Nährböden; die die üblichen Nährstoffe enthalten, züchten.
So verwendet ein- solcher Stamm die für, kohlenstaffheterotrophen Organismen üblicherweise benutzten Nährstoffe, bei spielsweise Glucose, Stärke, Dextrin, Lactose, Rohr zucker usw. als Kohlenstoffquelle, organische und an organische stickstoffhaltige Verbindungen, wie Pepton, Hefe- oder Fleischextrakte, Ammoniumsulfat, Ammo- niumnitrat;
Aminosäuren usw. als Stickstoffquelle, sowie- die üblichen Mineralsalze und, Spurenelemente.
Eine Verfahrensform zur- Herstellung des neuen Antibioticums besteht darin, dass ein flüssiges Nährme dium mit Mycel eines neuen Stammes von- Panus con- chatus (Bull. ex. Fr.) Fries beimpft und die Kultur 5 bis 10 Tage bei 27 inkubiert wird.
Die Züchtung kann unter aeroben Bedingungen in: einer Oburflächenkultur oder submers unter Schütteln oder in Fermentern mit Begasung durch Luft oder Sauerstoff unter Rühren er folgen.
Sobald eine maximale Menge an Antibioticum produziert worden ist, wird filtriert und das Antibioti- cum durch extraktive oder adsorptive Arbeitsure- thoden- auf an sich bekannte Weise aus dem Filtrat ge wonnen.
Eine Methode, die sich als vorzugsweise geeignet erwiesen hat, ist die Extraktion des Filtrates mit Essig ester, jedoch können auch andere organische Lösungs mittel, wie z. B. Benzol, Chloroform, Butylacetat, Methylenehlorid oder Butanol, verwendet werden. An- schliessend werden die Extrakte vom Lösungsmittel befreit, z.
B. durch Destillation, und der Rückstand zur Isolierung des gewünschten Antibioticums auf chroma- tographischem Wege an adsorbierenden Mitteln, wie Tonerde, Kieselgel, Magnesiumsilicat und dgl. oder mittels-Gegenstromverteilung gereinigt.
Das neue Antibioticum SL 3042 kann wie folgt charakterisiert werden: SL 3042 ist ein leicht gelbliches, hochaktives, neutral reagierendes Öl der Mikroanalyse C=62,30/0, H=6,7%, 0=30,
3 % mit dem Refraktionsindex [n] D 20 = 1'5306 und der spezifischen Drehung [a] D20 =<B>-611</B> (c = 0,4 in CH2Cl2).
UV.-Spektrum: Maxima 242 nm [E] i em = 229 und 302,5 mg = 20,2, Schulter bei 320 mg = 15,5 (in Methanol). (Fig. 1) IR.-Spektrum:
Banden bei 3580, 3400, 2970, 2910, 1680, 1600, 1440, 1375, 1235, 1210, 1100, 1035, 990, 928, 875, 845, 815 cm-' (in Methylenchlorid). (Fig. 2) Das neue Antibioticum SL 3042 zeigt eine gute cytostatische Wirksamkeit und kann zur therapeuti schen Bekämpfung von Tumorerkrankungen an Mensch und Tier verwendet werden. Diese Wirkung wird durch die Hemmung der Vermehrung von Tumorzellen (Zellen des Mäuse-Mastocytoms P-815) bestimmt.
Im geeigneten Nährboden vermehren sich diese Tumorzellen innert 40 Stunden auf das vier- und fünffache der Ausgangszahl. Die DE-50 (Konzentra tion, welche die Vermehrung um 50 0/a hemmt) von SL 3042 ist 1gg/ml.
Das Antibioticum SL 3042 besitzt auch eine spezi fische antiprotozoale Wirkung, insbesondere gegen Trypanosoma cruzi mit der MIC (MIC = Dosis, bei der mehr als 50 % der Trypanosomen unbeweglich sind) 10 ,ug/ml.
Die akute Toxizität DL 50 von SL 3042 bei der weissen Maus ist 23 mg/kg i.p.
Das erfindungsgemäss hergestellte Antibioticum SL 3042 kann als Arzneimittel allein, und zwar sowohl in öliger Form als auch als Rohkonzentrat oder in ent sprechenden Arzneiformen für orale, enterale oder parenterale Verabreichung verwendet werden. Zwecks Herstellung geeigneter Arzneiformen wird dieses mit anorganischen oder organischen, pharmakologisch in differenten Hilfsstoffen verarbeitet. Als Hilfsstoffe wer den verwendet z. B.
Für Tabletten und Dragdes: Milchzucker, Stärke, Talk, Stearinsäure usw.
für Sirupe: Rohrzucker-, Invert- zucker-, Glucoselösungen <B>USW.</B>
für Injektionspräparate: Wasser, Alkohole, Gly- cerin, pflanzliche Öle und dergl.
für Suppositorien: natürliche oder gehärtete Öle, Wachse u. a. mehr. Zudem können die Zubereitungen geeignete Kon- servierungs-, Stabilisierungs-, Netzmittel, Lösungsver- mittler, Süss- und Farbstoffe, Aromantien usw. enthal ten.
In dem nachfolgenden Beispiel, welches die Aus führung des Verfahrens erläutert, den Umfang der Er findung aber in keiner Weise einschränken soll, erfol gen alle Temperaturangaben in Celsiusgraden. Die Schmelz- bzw. Zersetzungspunkte sind auf dem Kofler- Block bestimmt.
<I>Beispiel</I> In einem Fermenter werden 100 Liter einer Nähr lösung, die pro Liter 20 g Cerelose 2 g Malzextrakt (Schweiz.
Ferment AG) 2 g Bacto-Yeast 2 g Pepton 2 g KH2P04 2<B>9</B> M9S04. 7H20 und entmineralisiertes Wasser enthält, mit 10 Liter einer Vorkultur von Panus con- chatus (Bull. ex.
Fr.) Fries angeimpft und unter Belüf tung (1 L/Min.) und unter Rühren (300 Umdrehungen/ Min.) 88 Stunden bei 27 irrkubiert. Die Kulturbrühe wird filtriert und das Filtrat vom pH 4 bis 4,5 mit dreimal 50 Litern Essigester extrahiert. Der Essigester Extrakt wird einmal mit 5 Liter Wasser gewaschen, über MgS04 getrocknet und nach Filtration im Vakuum zur Trockne gebracht.
Der Rückstand wird an 2000 g Kieselgel (Merck 0,2-0,5 %) chromatogra- phiert. Zur Eluation wird zuerst Benzol-Äther (75:25) verwendet. Eluation mit Benzol-Äther (1:1) ergibt rei nes SL 3042.
