Verfahren zur Herstellung neuer 0-(2-Guanidino-äthyl)-oxime
Die Erfindung betrifft ein Verfahren zur Herstellung neuer O-(2-Guanidino-äthyl)-oxime und Salze derselben.
Die neuen Verbindungen haben die Formel

wobei R1 und R2 ein Wasserstoffatom, eine gegebenenfalls mit Alkyl-, Alkoxy- oder Halogen substituierte Alkyl-, Aryl-, Aryloxy-alkyl oder Aralkyl-Gruppe mit 1 bis 12 Kohlenstoffatomen oder gemeinsam eine gegebenenfalls ungesättigte, mit Alkyl oder Aryl substituierte, oder nicht substituierte mit einem oder zwei alicyclischen oder aromatischen Ringen kondensierte oder nicht kondensierte alicyclische Gruppe mit 4 bis 14 Kohlenstoffatomen bezeichnen.
Als Säuren, mit denen diese Verbindungen geeignete Additionsverbindungen bilden können, lassen sich z.B. erwähnen: Salzsäure, Salpetersäure, Schwefelsäure, Phosphorsäure, Sulfaminsäure, Essigsäure, Weinsteinsäure, Zitronensäure, Ascorbinsäure, Benzoesäure, p-Aminobenzoesäure, Mandelsäure und Salicylsäure.
Beispiele der erfindungsgemäss hergestellten Verbindungen und deren Salze unter anderem mit den erwähnten Säuren sind: O-(2-Guanidino-äthyI)-cyclohexanonoxim, O-(2-Ouanidino-äthyl)-3,3 ,5-tnmethylcydohexen-5- -onoxim, O-(2-Guanidino-äthyl)-cyclopentanonoxim, O-(2-Guanidino-äthyl)-cycloheptanonoxim, 0-(2-Guanidino-äthyl)-cyclooctanonoxim, 0-(2-Guanidino-äthyl)-4-methyl-cyclohexanonoxim, O-(2-Guanidino-äthyl)-phenoxyacetonoxim, O-(2-Guanidino-äthyl)-fluorenonoxim, O-(2-Ouanidino-äthyl)-2,4-dimethylpentanon-3-oxim, O-(2-Guanidino-äthyl)-2,6-dichlorbenzaldoxim, O-(2-Guanidino-äthyl)-oxim von Kampfer.
Die erfindungsgemäss erhaltenen Verbindungen zeigen eine interessante, sich wahrscheinlich auf Sperrung des Sympathious gründende, blutdrucksenkende Wirkung. Diese Wirkung wurde unter anderem bei Untersuchungen festgestellt, bei denen narkotisierten Hunden und Katzen intravenös eine isotonische Lösung einer erfindungsgemässen Verbindung in Wasser injiziert wurden.
Im Vergleich zu bekannten, blutdrucksenkenden Gua- nidin-Abkömmlingen, wie dem bekannten Guanitidin, der Formel

haben die erfindungsgemäss hergestellten Verbindungen den Vorteil, dass nach Verabreichung der Blutdruckerniedrigung nur eine sehr kurze Periode der Blutdruckzunahme vorangeht. Ein weiterer Vorteil ist, dass Diarrhöe ganz oder nahezu ganz unterbleibt.
Dank dieser Eigenschaften eignen sich die Verbindungen besonders gut zur Herstellung von pharmazeutischen Präparaten zur Behandlung von zu hohem Blutdruck.

Feste pharmazeutische Formstücke werden in üblicher Weise dadurch hergestellt, dass der wirksame Stoff in feste pharmazeutische Träger, wie Milchzucker, Puderzucker, Kartoffelstärke, Talk, Magnesiumstearat, Gummi-Arabicum, Gelatine, Calciumphosphat und/oder Titandioxyd aufgenommen wird, worauf das Gemisch zu Tabletten oder Dragees verarbeitet wird.
Beispiel a) 0-(2-Guanidino-äthyl)-hydroxylamin-2HCI.
Eine Suspension von 0,60 g 0-(2-Guanidino-äthyl)-3', 4'-dimethoxy-acetophenonoxim-HNO3 (erhalten aus 0 -(2-Aminoäthyl)-3'4'-dimethoxy-acetophenonoxim und Cyanamid) in 15 cm 0,40 N HC1 wurde während 7 Stunden kontinuierlich extrahiert mit Äther. Die Wasserschicht wurde konzentriert im Vakuum, verdünnt mit wasserfreiem Alkohol und wieder konzentriert. Der Rückstand wurde in Äthanol gelöst. Nachdem Äther hinzugefügt war, scheidete sich ein öl ab. Daraus wurde nach einiger Zeit eine kristallinische Substanz erhalten. b) O-(2-Guanidino-äthyl)-indanoiz-1 -oxin-HNO3.
0,08 g 0-(2-Guanidino-äthy0-hydroxylamin 2 HCl und 0,053 g Indanon-l wurden gelöst in 0,8 cm3 Äthanol.
Eine Lösung von 0,044 g Na Acetat in 0,6 cm3 Wasser wurde hinzugefügt. Das Gemisch wurde 2 Stunden bei Zimmertemperatur aufbewahrt. Dann wurde eine Lösung von 0,062 g NH4NO3 in 1,5 cm3 Wasser hinzugefügt. Das Gemisch wurde konzentriert im Vakuum. Es wurde 0,29 g einer kristallinischen Verbindung erhalten.
Die Substanz wurde gemischt mit Äther, der Äther wurde abfiltriert und die festen Stoffe wurden nochmals gewaschen mit Äther und anschliessend mit Wasser. Die hergestellte Substanz hat einen Schmelzpunkt von 150 bis 1540C (Zersetzung).

The compounds obtained according to the invention show an interesting antihypertensive effect, probably based on blocking the sympathetic. This effect was found, inter alia, in studies in which anesthetized dogs and cats were injected intravenously with an isotonic solution of a compound according to the invention in water.
