Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer, kondensierter heterocyclischer Verbindungen.
Es wurde überraschenderweise gefunden, dass man. hetercycolische Verbindungen der Formel I

Die nach diesem Verfahren in einfacher Weise herstellbaren neuen Verbindungen zeichnen sich über raschenderweise durch wertvolle pharmakologische
Eigenschaften aus. Insbesondere besitzen solche Ver bindungen entzündungshemmende, narkosepotenzierende und antikonvulsive Wirksamkeit und zugleich eine rel'a tiv geringe Toxizität. Ihre therapeutische Anwendung erfolgt hauptsächlich oral oder rektal; sie können je doch in Form von wässrigen, mit Hilfe von Lösungs vermittern und/oder Emulgiermitteln hergestellten Di spersionen auch parenteral verabreicht werden. Die Verbindungen der Formel I können ausserdem als Zwischenprodukte, z. B. zur Herstellung weiterer pharmakologisch wertvoller Stoffe, verwendet werden.
In den Verbindungen der Formel I und in den entsprechenden Ausgangsstoffen kann R z. B. Methyl, Äthyl, Propyl, Isopropyl, Butyl, Isobutyl, Amyl, Hexyl sein. A ist z. B. Äthylen, Trimethylen, Methyl-äthylen, 1,2-Dimethyl-äthylen, 1,1-Dimethyl-äthylen, Äthyl äthylen, o-Phenylen, 4-Methyl-o-phenylen, 4-Chlor-o- phenylen oder 4-Methoxy-o-phenylen. Die nichtsymmetrischen der unter A aufgezählten Reste können im Molekül des Endstoffes grundsätzlich in den verschiedenen möglichen Anordnungen vorliegen.
Viele der als Ausgangsstoffe in Frage kommenden Verbindungen der Formeln II, III und IV sind bekannt oder lassen sich nach allgemein bekannten Verfahren herstellen. Als Beispiele von Ausgangsstoffen der Formeln II und III seien die nachstehenden Verbindungen genannt: o-Acetyl-benzoesäure, o-Propionyl-benzoesäure, o-Butyryl-benzoesäure, o-Isobutyryl-benzoesäure, o-Valeroyl-benzoesäure, o-Isovaleroyl-benzoesäure,
3-Chlor-3-methyl-phthalid, o-Propionyl-benzoesäure-methylester,
3-Acetoxy-3-methyl-phthalid,
Als Ausgangsstoffe der Formel IV seien erwähnt:
2-Amino-äthanthiol,
1-Amino-2-propanthiol, 2-Amino-l-propanthiol,
1 -Aminc2-methyl-propanthiol,
1-Amino-2-butanthioi,
2-Amino-3-butanthiol, o-Amino-benzolthiol, 3-Amino-l-propanthiol und 2,2-Dimethyl-3 -amino-1-prop anthiol.
Zur Herstellung der erfindungsgemässen, Verbindungen lässt man ein Aminothiol der Formel IV in An- oder Abwesenheit eines Lösungsmittels, wie z. B. Toluol, Chlorbenzol, Xylol, o-Chlortoluol, o-Dichlorbenzol, Amylalkohol, bei Temperaturen von etwa 100-250 auf eine Ketocarbonsäure oder ein Derivat derselben entsprechend den Formeln II und/oder III einwirken. Die erfindungsgemässe Kondensation erfolgt unter Abspaltung der äquimolaren Menge der Verbindung H-X und Wasser und kann gegebenenfalls durch wasserabspaltende Mittel, wie z. B. Zinkchlorid, oder durch Abdestillieren des Reaktionswassers bzw. des freigesetzten Alkanols oder der freigesetzten Essigsäure, gefördert werden.
In manchen Fällen lassen sich bei der Durchfüh- rung des erfindungsgemässen Verfahrens zwei Reaktions- stufen feststellen. Zunächst entsteht unter Abspaltung einer Verbindung X-H, das heisst von Wasser, Halogenwasserstoff, niederem Alkanol oder niederer Alkansäure, ein Zwischenprodukt der Formel V,

welches in manchen Fällen ganz oder teilweise in der tautomeren Form entsprechend der Formel VI,

in welchen Formeln R und A die unter der Formel I angegebene Bedeutung haben, vorliegt. Durch Wasserabspaltung unter den vorgenannten Reaktionsbedingungen entsteht daraus das erfindungsgemässe Endprodukt der Formel I. Die vorliegende Erfindung umfasst entsprechend dem Vorstehenden auch die Herstellung dieser Endprodukte aus Zwischenprodukten der Formeln V und/oder VI.
Das betreffende Verfahren ist dadurch gekennzeichnet, dass man Verbindungen dieser allgemeinen Formeln wasserabspaltenden Bedingungen, am einfachsten Erhitzen in einem mit Wasser azeotrop destillierenden Lösungsmittel, wie z. B. Chlorbenzol oder oDichiorbenzol, oder Erhitzen in An- oder Abwesenheit eines wasserabspaltenden Mittels, wie z. B.
Zinkchlorid oder Magnesiumperchlorat, aussetzt.
Die nachfolgenden Beispiele sollen das erfindungs- gemässe Verfahren näher erläutern. Die Temperaturen sind in Celsiusgraden angegeben. Teile bedeuten Ge wichtsteiie; diese verhalten sich zu Volumteilen wie g zu-cmS.
Beispiel 1 1o,4 Teile o-Acetyl-benzoesäure, 12,5 Teile 2-Amino-benzolthiol (o-Amino-thiophenol) und 200 Teile Chlorbenzol werden so erhitzt, dass ein Gemisch von Chlorbenzol und Wasser während einer Stunde abdestiliert. Dann wird der Rest des Chlorbenzols im Vakuum abgedampft. Man löst den erhaltenen Rückstand in heissem Benzin und giesst von einem unlöslichen Rest ab. Beim Einengen erhält man Kristalle, weiche durch Umkristallisieren aus Benzin gereinige werden. Das reine 4b-Methyl-isoindolo-[1,2-b]-benzo- thiazol-l 1-(4bH)-on der Formel
