Die vorliegende Erfindung bezieht sich auf ein Verfahren zur fermentativen Herstellung von Streptomycin in Gegenwart von Ionenaustauschern.
Man kennt zahlreiche Verfahren zur Gewinnung von Streptomycin. Der grösste Teil derselben führt in einer ersten Phase die Vergärung und in einer zweiten gesonderten Phase die Extraktion und Reinigung durch.
Ferner ist auch die Möglichkeit der Gewinnung von Streptomycin (oder anderer Antibiotika) aus den Kulturflüssigkeiten bekannt, bei welchem Verfahren die Isolierung zu gleicher Zeit mit der Vergärung durch Einführung von Ionenaustauschern in das Kulturmittel erfolgt, in deren Gegenwart die Vergärung stattfindet und welche das Antibiotikum nach Massgabe seiner Bildung aufsaugen, sodass auf diese Weise die schwierige Operation einer Filtrierung vermieden wird.
Diese Verfahren sind einfacher und erlauben die Erzielung von um etwa 25 O/o höheren Ausbeuten als die Verfahren, welche in zwei gesonderten Phasen arbeiten.
Die vorliegende Erfindung betrifft ein Verfahren, welches zu höheren Ausbeuten und grösseren Reinheitsgraden führt und welches in der Anwendung eines von Calcium- und Magnesiumionen freien Kulturmediums sowie in der Einführung von Ionenaustauschern, insbesondere von Carboxylgruppen enthaltenden Kationenaustauschern, vor der Inokulation und vorzugsweise nach der Sterilisation im Kulturmedium besteht. Die Zugabe der Ionenaustauscher kann einmalig oder in Portionen erfolgen.
Die Abtrennung des Ionenaustauschers aus dem Kulturmedium nach Beendigung der Vergärung erfolgt durch Dekantieren und Waschen mit enthärtetem Wasser unter vorheriger oder ohne Änderung der Viskosität des Mediums durch Hinzufügung von Koagulationsmitteln und Verdünnung mit enthärtetem Wasser. Die Extraktion erfolgt durch Chromatographie, wobei der Anteil gesammelt wird, welcher Streptomycin enthält.
Anschliessend wird das Endprodukt, z. B. nach bekannten Verfahren, erhalten.
Der pH-Wert der Kulturflüssigkeit wird zweckmässigerweise während der ganzen Dauer der Vergärung durch Verwendung des teilweise regenerierten, d. h. beispielsweise in sein Natriumsalz gebrachten Ionenaustauschers, als Puffer auf einem Optimalwert (in Abhängigkeit von dem erzeugenden Stamm und dem benutzten Medium) gehalten. Im Medium des nachfolgenden Beispiels wird der pH-Wert von 6,8 durch Verwendung des Harzes AMBERLITE IRC-50 , 95 O/o Natriumform und 5 O/o Säureform, aufrechterhalten. Für andere pH Werte oder andere Medien kann das regenerierte Harz in anderen Verhältnissen verwendet werden.
Es wurde festgestellt, dass der Prozess der Biosynthese und der Reinigung des Streptomycins mit Ionenaustauschern im Kulturmedium zu besonderen Wirkungen führt infolge der positiven Beeinflussung des Gleichgewichts zwischen der Konzentration des Streptomycins auf der inneren Fläche der Zellmembran und derjenigen auf der Aussenfläche derselben.
Ferner wurde beobachtet, dass die Ausbeuten grösser sind, wenn der Ionenaustauscher in das Medium bei der Streptomycinherstellung eingeführt wird, da er aus der Lösung das Streptomycin nach Massgabe seines Erscheinens herausholt, unter Verlagerung des Gleichgewichts im Sinne der Streptomycinbildung und der Übertragung einer zusätzlichen Streptomycinmenge, welche che aus dem Medium ausgeschieden wird.
Die Vergärung wird in einem Kulturmedium durchgeführt, welches in der Regel die hauptsächlichsten, üblichen, energetischen und die Biosynthes stimulierenden Faktoren enthält, jedoch keine Calcium- oder Magnesiumsalze, die als Puffersubstanzen für pH-Wert in den konventionellen Medien benutzt werden, da sie keine Energiequellen darstellen und oftmals die Entwicklung hemmen und die Biosynthese herabsetzen. Die Rolle der Puffersubstanz wird zweckmässig in diesem Fall von dem zu diesem Zweck teilweise regenerierten Ionenaustauscher sichergestellt.
Beispiel
In ein 500-Liter Fermentationsgefäss werden 300 Liter eines Kulturmediums mit folgender Zusammensetzung eingeführt:
Sojamehl 7,5 kg (2,5 o/o)
Glukose (reduzierender Zucker) 9,0 kg (3,0 O/o)
Natriumchlorid 0,9 kg (0,3 O/o)
Ammoniumsulfat 1,6 kg (0,5 o/o) prim. Kaliumphosphat 0,075 kg (0,025 O/o)
Harnstoff 0,3 kg (0,1 O/o) enthärtetes Wasser bis zu 250 Liter
Das Medium wird 1 Stunde bei 120 OC sterilisiert.
Separat werden 3 kg AMBERLITE IRC-50 (saure Form) mit 4 0/o-igem Natriumhydroxyd im tSber- schuss behandelt, mit Wasser (enthärtet) bis zur Neutralität gewaschen und dann mit 15 Litern Salzsäure l-n behandelt. Es wird enthärtetes Wasser bis zur Vervollständigung des Volumens auf 50 Liter hinzugegeben, bei 120 "C für eine Stunde sterilisiert, worauf die Suspension steril in den Fermentationstank eingeführt wird.
Der auf 28 C abgekühlte Fermentator wird mit einer vegetativen Kultur von Streptomyces griseus geimpft, worauf die Gärung erfolgt.
Nach einer Gärdauer von 120 bis 140 Stunden, in welcher Zeit durch das Harz der pH-Wert auf 6,8 aufrechterhalten wird, scheidet sich das Harz aus; dieses wird mit enthärtetem Wasser gewaschen und in eine Säule aus Glas eingeführt, welche eine Höhe von 80 cm aufweist. Man eluiert die Säule mit Schwefelsäure von 0,5-1,4-n, wobei die Konzentration fortschreitend erhöht wird.
Die letzten gesammelten Fraktionen enthalten das Streptomycin, welches folgende Eigenschaften aufweist:

Die Ausbeute ist um 53 0/0 höher als bei dem Verfahren mit separaten Phasen von Gärung und Extraktion und um 15 0/0 höher als beim Verfahren, welches Ionenaustauscher im üblichen Kulturmedium benutzt. Zu gleicher Zeit kann man feststellen, dass die Reinheit des Eluats wesentlich höhere Werte aufweist.
Das Eluat wird mit 0,5 Olo entmineralisierter aktiver Kohle gereinigt. Es wird über sulfokationische Harze geleitet, um die Metallspuren zu entfernen, es folgt eine Eindampfung, Entfärbung und Zerstäubung oder Lyophilisierung.
Das Endprodukt hat mindestens 715 IE/mg.
Das erfindungsgemässe Verfahren weist folgende Vorteile auf: man erhält höhere Ausbeuten als bei den bekannten Verfahren; das Endprodukt weist einen hohen Reinheitsgrad auf; das Verfahren ist viel einfacher; die Menge biosynthetisierten Streptomycins wird vergrössert; es wird die Stabilität des Streptomycins nach Massgabe der Biosynthetisierung sichergestellt; es werden die Operationen zur Ausscheidung der Calcium- und Magnesiumionen ausgeschaltet; das Verfahren ist universeller anwendbar.

The present invention relates to a process for the fermentative production of streptomycin in the presence of ion exchangers.
Numerous processes are known for the production of streptomycin. Most of them carry out fermentation in a first phase and extraction and purification in a second separate phase.
Furthermore, the possibility of obtaining streptomycin (or other antibiotics) from the culture fluids is known, in which process the isolation takes place at the same time as the fermentation by introducing ion exchangers into the culture medium, in whose presence the fermentation takes place and which the antibiotic after Soak up according to its formation, so that the difficult operation of a filtration is avoided in this way.
These processes are simpler and permit about 25% higher yields to be achieved than the processes which operate in two separate phases.

The pH value of the culture liquid is expediently adjusted during the entire fermentation period by using the partially regenerated, i.e. H. for example, brought into its sodium salt ion exchanger, kept as a buffer at an optimal value (depending on the producing strain and the medium used). In the medium of the following example, the pH value of 6.8 is maintained by using the resin AMBERLITE IRC-50, 95% sodium form and 5% acid form. The regenerated resin can be used in other proportions for other pH values or other media.
It was found that the process of biosynthesis and purification of streptomycin with ion exchangers in the culture medium leads to particular effects due to the positive influence on the balance between the concentration of streptomycin on the inner surface of the cell membrane and that on the outer surface of the same.
It was also observed that the yields are greater when the ion exchanger is introduced into the medium during the streptomycin production, since it removes the streptomycin from the solution as it appears, shifting the equilibrium in terms of streptomycin formation and the transfer of an additional amount of streptomycin which surface is excreted from the medium.
The fermentation is carried out in a culture medium, which usually contains the main, usual, energetic and biosynthesis stimulating factors, but no calcium or magnesium salts, which are used as buffer substances for pH in conventional media, as they are not energy sources and often inhibit development and reduce biosynthesis. The role of the buffer substance is expediently ensured in this case by the ion exchanger, which is partially regenerated for this purpose.

The medium is sterilized at 120 ° C. for 1 hour.
Separately, 3 kg of AMBERLITE IRC-50 (acidic form) are treated with 40% sodium hydroxide in excess, washed with water (softened) until neutral and then treated with 15 liters of hydrochloric acid. Softened water is added to the completion of the volume to 50 liters, sterilized at 120 "C for one hour, after which the suspension is introduced into the fermentation tank in a sterile manner.
The fermenter, cooled to 28 C, is inoculated with a vegetative culture of Streptomyces griseus, whereupon fermentation takes place.
After a fermentation time of 120 to 140 hours, during which time the resin maintains the pH value at 6.8, the resin separates; this is washed with softened water and introduced into a column made of glass, which has a height of 80 cm. The column is eluted with sulfuric acid of 0.5-1.4-n, gradually increasing the concentration.
The last fractions collected contain streptomycin, which has the following properties:

The yield is 53% higher than in the process with separate phases of fermentation and extraction and 15% higher than in the process which uses ion exchangers in the usual culture medium. At the same time it can be seen that the purity of the eluate has significantly higher values.
The eluate is purified with 0.5% demineralized active charcoal. It is passed over sulfocationic resins to remove the traces of metal, followed by evaporation, decolorization and atomization or lyophilization.
The final product is at least 715 IU / mg.
The process according to the invention has the following advantages: higher yields are obtained than in the known processes; the end product has a high degree of purity; the process is much simpler; the amount of biosynthesized streptomycin is increased; the stability of the streptomycin is ensured in accordance with the biosynthesis; operations to eliminate calcium and magnesium ions are switched off; the procedure is more universally applicable.
