(Zusatzpatent zum Hauptpatent 428 097) Verfahren zur Herstellung eines neuen Antilbiotieums Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung der bisher unbekannten Verbindung Mono- desacetyl-Anguidin.
Diese Verbindung kann erfindungsgemäss durch Züchtung von Stämmen der Pilzgattung Fusarium Link ex Fr. (Fungi imperfecti) oder ihrer Mutanten in einer Nährlösung und anschliessende Isolierung aus dem Kul turfiltrat erhalten werden.
Zur Gewinnung von Monode- sacetyl-Anguidin auf mikrobiologischem Wege eignen sich Stämme der Pilzgattung Fusarium, beispielsweise Fu- sarium anguioides Sherb., Stamm NRRL 3020 (Northern Regional Research Laboratories, Peoria, Blinois, USA).
Es können aber auch andere Stämme verwendet werden, wie sie zum Beispiel durch Selektion oder Mutation unter der Einwirkung von Ultraviolett- oder Röntgenstrahlen oder durch Anwendung anderer bekannter Methoden, z.B. durch den Einfluss von Chemikalien, erhalten werden.
Die Züchtung kann entweder in ruhender Oberflä chenkultur oder submers unter Schütteln bzw. in Fer- mentern unter Rühren mit Begasung durch Luft oder Sauerstoff erfolgen, bei Temperaturen zwischen 12 und 30 C. Fusarium-Stämme lassen sich auf vielerlei Nähr boden, die die üblichen Nährstoffe enthalten, züchten.
So verwerten solche Stämme die für kohlenstoffheterotrophe Organismen üblicherweise benutzten Nährstoffe, bei spielsweise Glucose, Stärke, Dextrin, Lactose, Rohrzuk- ker usw. als Kohlenstoffquelle, organische und anorgani sche, stickstoffhaltige Verbindungen, wie Pepton, Hefe- oder Fleischextrakte, Ammoniumsulfat, Ammoniumni- trat, Aminosäuren usw.
als Stickstoffquelle, sowie die üb lichen Mineralsalze und Spurenelemente.
Es wurde nun gefunden, dass sich aus Kulturlösungen einer Reihe von Stämmen verschiedener Arten der Pilz gattung Fusarium die neue, antibiotisch wirksame Ver bindung Monodesacetyl-An,uidin in reiner Form isolie ren lässt.
Eine Form des erfindungsgemässen Verfahrens zur Herstellung von Monodesacetyl-Anguidin besteht darin, dass ein flüssiges Nährmedium mit einer Kultur eines Fusarienstammes beimpft und drei bis 25 Tage inkubiert wird.
Sobald die Menge an Monodesacetyl-Anguidin in der Kulturlösung ihren höchsten Wert erreicht hat, wird diese Lösung filtriert und das Monodesacetyl-Anguidin aus dem Filtrat nach an sich bekannten Methoden aus dem Filtrat isoliert, so z.B. durch Extraktion und/oder Adsorption. Für die Produktion im technischen Massstab wird die Extraktion bevorzugt, da sie weniger zeitraubend und billiger ist.
Zur Extraktion eignen sich mit Wasser nicht mischbare Lösungsmittel, wie chlorierte Kohlen wasserstoffe, z.B. Methylenchlorid, Äthylenchlorid, Chlo roform und dgl., ferner Alkohole mit geringer Wasser löslichkeit, wie Butanol, Amylalkohol und dgl., Alkylester von Fettsäuren, wie Äthylacetat, Propylacetat, Butylace- tat, Amylacetat und dgl., wenig wasserlösliche Ketone, wie Methylisobutylketon,
Methylamylketon und dgl. Der Extrakt der Kulturflüssigkeit kann vorteilhafterweise im Vakuum zur Trockne eingedampft werden, wobei man das Antibioticum in roher Form erhält.
Monodesacetyl-Anguidin kristallisiert aus Äther und hat einen Schmelzpunkt von 164-167 C. Die spezifische Drehung beträgt [2]25n = -25 (c = 0,48 in Chloroform).
Die Elementaranalysen ergeben folgende Werte: C 63,0% H 7,5 j, Das Antibioticum enthält weder N, noch S, noch Ha logen..
Die berechneten Werte für C"H2405 sind: C 62,95% H 7,46 j, Das IR.-Spektrum einer Lösung der Substanz in Me- thylenchlorid zeigt Banden bei 3590, 3440, 2940, 1730, 1670, 1450-1430, 1390-1365, 1240, 1165,<B>1</B>105, 1073, 1040, 988, 960, 935-920 cm-' (Fig. 1).
Die Testreaktionen nach Benedict, Fehling, Tollens, Millon, sowie mit Eisenchlorid und Kaliumpermanganat in Aceton sind negativ. Brom in Tetrachlorkohlenstoff wird langsam entfärbt.
Monodesacetyl-Anguidin ist lös lich in Aceton, Äthanol, Dioxan, Chloroform, Methylen- chlorid, Pyridin, Benzol, Toluol, schwerlöslich in Wasser, Diäthyläther, Diisopropyläther, Pentan, Hexan und Hep- tan.
Die pharmakodynamischen Eigenschaften von Mono- desacetyl-Anguidin decken sich weitgehend mit denen des Anguidins, so dass es für dieselben therapeutischen Zwecke verwendet werden kann, z.B. als Medikament für die Behandlung von Infektionskrankheiten. Ausserdem besitzt die neue Verbindung antimitotische Wirksamkeit und kann daher zur medikamentösen Bekämpfung von Tumoren dienen. In dem nachfolgenden Beispiel, wel ches die Ausführung des Verfahrens erläutert, den Um fang der Erfindung aber in keiner Weise einschränken soll, erfolgen alle Temperaturangaben in Celsiusgraden.
Die Schmelzpunkte sind unkorrigiert.
Beispiel <I>1</I> Mit einer Kultur von Fusarium anguioides (Stamm NRRL 3020) werden in einem Fermenter 10 Liter einer Nährlösung, die pro Liter 30 g Glucose, (Cerelose San- doz), 2 g Malz-Extrakt (Schweiz.
Ferment AG), 2 g Bac- to-yeast Extrakt (Difco), 4 g Ammoniumnitrat p.a., 2 g Kaliumhydrogenphosphat p.a., 2 g Magnesiumphosphat x 7H20 p.a. und entmineralisiertes Wasser ad 1 Liter enthält, 135 Stunden lang bei 27 unter Rühren (1450 U/Min.) und Belüftung (5 Liter Luft pro Minute)
in- kubiert. Die Kulturbrühe wird in einer Becherzentrifuge in einen festen Schlamm und eine schwachtrübe Lösung getrennt. 7,0 Liter der überstehenden Lösung werden zweimal mit dem gleichen Volumen Essigester und an- schliessend dreimal mit demselben Volumen n-Butanol extrahiert. Die organischen Lösungsmittel werden je zweimal mit Wasser gewaschen und einzeln im Umlauf verdampfer am Wasserstrahlvakuum eingeengt.
Der aus dem Essigester-Extrakt nach gründlicher Trocknung im Hochvakuum verbleibende Rückstand wird zweimal zwi schen je 100 ml Petroläther und 100 ml 90a/oigem wäss- rigem Methanol verteilt.
Der nach Einengen und Trock nen des Methanol-Wasser-Extraktes resultierende Rück stand wird in der üblichen Weise an Kieselgel chroma- tographiert. Die mit Chloroform-Methanol (199:1) eluierten Fraktionen enthalten Anguidin, aus zwei Fak toren, die mit Chloroform-Methanol (49: 1) eluiert wer den, wird nach Kristallisation aus Äther reines krist. Mo- nodesacetyl-Anguidin gewonnen.
Beispiel <I>2</I> Mit einer Kultur von Fusarium sambucinum, Stamm ATCC 11 852, werden in einem Fermenter 10 Liter einer Nährlösung, die pro Liter 20 g Glucose (Cerelose San- doz), 2 g Pepton (Cudahy), 2 g Malzextrakt (Schweiz.
Ferment A.G.), 2 g Bacto-yeast Extract (Difco), 2 g Ka- liumdihydrogenphosphat, 2 g Magnesiumsulfat X 7H20, und entmineralisiertes Wasser ad 1 Liter enthält, 145 Std. lang bei 27 unter Rühren (450 U/Min.) und Belüftung (5 Liter Luft/Min.) inkubiert. Die dunkelrote Kulturbrühe wird filtriert und das Filtrat der Reihe nach je drei Mal mit 10 Litern Benzin, 10 Litern Äthylacetat und 10 Litern n-Butanol extrahiert.
Der Äthylacetatextrakt wird mit 2 Litern Wasser nach gewaschen, über Natriumsulfat getrocknet und am Va kuum eingeengt. Der Eindampfrückstand wird in Chlo- roform gelöst und an 4,18 g Kieselgel chromatographiert. Die mit Chloroform eluierten Anteile werden verworfen.
Die mit Chloroform-Methanol-(99:1) eluierten Frak tionen geben aus Äther-Hexan kristallines Anguidin. In den mit Chloroform-Methanol-(1 : 1) eluierten Fraktio nen kann Monodesacetyl-Anguidin dünnschichtchroma- tographisch nachgewiesen werden.
Beispiel <I>3</I> Eine Kultur von Fusarium scirpi (Stamm S 1497) wird in 500 ml-Erlenweyerkolben mit je 100 ml der glei chen Nährlösung wie bei Beispiel 1, jedoch ohne Bacto yeast extract (Difco) 25 Tage lang bei 27 inkubiert. 37 solcher Kulturen werden durch Filtration vom Mycel getrennt und das Kulturfiltrat mit 3 Mal je 2 Litern Äthylacetat extrahiert, die Extrakte mit wenig Wasser nachgewaschen, über Natriumsulfat getrocknet und am Vakuum eingeengt.
Der erhalten Rückstand wird an 30 g Kieselgel chromatographiert. Die mit Benzol und Chloroform eluierten Teile werden verworfen. Die mit Chloroform-Methanol-(99: 1) eluierten Fraktionen geben aus Äther-Hexan kristallisiertes Anguidin vom Schmelz punkt 155-162 . In den mit Chloroform-Methanol-(99:1y eluierten Fraktionen kann Monodesacetyl-Anguidin dünnschichtchromatographisch nachgewiesen werden.

The cultivation can be done either in dormant surface culture or submerged with shaking or in fermenters with stirring with aeration by air or oxygen, at temperatures between 12 and 30 C. Fusarium strains can be grown on many different nutrient media, the usual nutrients contain, breed.
Such strains utilize the nutrients usually used for carbon-heterotrophic organisms, for example glucose, starch, dextrin, lactose, cane sugar, etc. as a carbon source, organic and inorganic compounds containing nitrogen, such as peptone, yeast or meat extracts, ammonium sulfate, ammonium nickel stepped, amino acids, etc.

It has now been found that the new, antibiotic compound monodesacetyl-an, uidine can be isolated in pure form from culture solutions of a number of strains of different species of the fungal genus Fusarium.
One form of the process according to the invention for the production of monodesacetyl anguidin consists in inoculating a liquid nutrient medium with a culture of a Fusarium strain and incubating it for three to 25 days.
As soon as the amount of monodesacetyl anguidine in the culture solution has reached its highest value, this solution is filtered and the monodesacetyl anguidine is isolated from the filtrate by methods known per se, e.g. by extraction and / or adsorption. For industrial scale production, extraction is preferred because it is less time consuming and cheaper.
Solvents which are immiscible with water, such as chlorinated hydrocarbons, e.g. Methylene chloride, ethylene chloride, chloroform and the like, also alcohols with low water solubility, such as butanol, amyl alcohol and the like., Alkyl esters of fatty acids such as ethyl acetate, propyl acetate, butyl acetate, amyl acetate and the like., Little water-soluble ketones such as methyl isobutyl ketone,

Monodesacetyl-anguidine is soluble in acetone, ethanol, dioxane, chloroform, methylene chloride, pyridine, benzene, toluene, and is sparingly soluble in water, diethyl ether, diisopropyl ether, pentane, hexane and hepatane.
The pharmacodynamic properties of mono-deacetyl-anguidine are largely identical to those of anguidine, so that it can be used for the same therapeutic purposes, e.g. as a drug for the treatment of infectious diseases. In addition, the new compound has antimitotic activity and can therefore be used to combat tumors with drugs. In the following example, which explains the implementation of the method but is not intended to limit the scope of the invention in any way, all temperatures are given in degrees Celsius.

The culture broth is separated into a solid sludge and a slightly cloudy solution in a cup centrifuge. 7.0 liters of the supernatant solution are extracted twice with the same volume of ethyl acetate and then three times with the same volume of n-butanol. The organic solvents are each washed twice with water and individually concentrated in a circulating evaporator in a water-jet vacuum.
The residue remaining from the ethyl acetate extract after thorough drying in a high vacuum is divided twice between 100 ml petroleum ether and 100 ml 90% aqueous methanol each time.
