Verfahren zur Herstellung von neuen Piperidinderivaten Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer Piperidinderivate mit wertvollen pharmakologischen Eigenschaften.
Es wurde überraschenderweise gefunden, dass Piperidinderivate der Formel I,

in welcher R1 einen Alkyl- oder Alkenylrest mit höchstens 8 Kohlenstoffatomen, den Cyclohexylrest oder einen Phenylalkylrest mit höchstens 9 Koh- lenstoffatomen, R2 Wasserstoff oder den Methylrest bedeutet, R3, R4, R,
und RE unabhängig voneinander Was serstoff oder Methylreste bedeuten und R,-CO- den Acylrest einer Carbonsäure mit höch stens 3 Kohlenstoffatomen bedeutet, und ihre Salze mit anorganischen und organischen Säuren wertvolle phar makologische Eigenschaften,
insbesondere antitussive sowie analgetische Wirksamkeit bei oraler wie parentera- ler Applikation besitzen und zugleich relativ wenig toxisch sind. Sie eignen sich deshalb zur Dämpfung des Hustenreizes sowie auch zur Linderung oder Behebung von Schmerzen verschiedener Genese.
In den Verbindungen der Formel I und den zuge hörigen, weiter unten genannten Ausgangsstoffen ist R1 beispielsweise durch Alkylreste wie den Methyl-, Äthyl-, n-Propyl-, Isopropyl-, n-Butyl-, n-Octyl-; durch Alkenyl- reste wie den Allylrest.
Der Rest R,-CO- ist z. B. dar Acylrest der Essig säure oder Propionsäure.
Zur Herstellung der Verbindungen der Formel I setzt man eine Verbindung der Formel II,

Dabei entstehen als unmittelbare Reaktionsprodukte Verbindungen der For mel II, in denen X durch -Mg-Halogen verkörpert ist.
Diese werden vorzugsweise direkt mit reaktionsfähigen funktionellen Derivaten von Carbonsäuren der Formel III umgesetzt, d. h. anstatt die erhaltene Lösung des Produktes der Grignard-Reaktion in Diäthyläther oder einem andern ätherartigen Lösungsmittel in üblicher Weise zu zersetzen, versetzt man sie mit einem reaktions- fähigen funktionellen Derivat einer Carbonsäure der Formel III,
das vorzugsweise in einem inerten, wasser freien organischen Lösungsmittel wie z. B. Benzol oder Tetrahydrofuran gelöst ist, und erwärmt das Gemisch, nötigenfalls unter Abdestilliexen eines niedrigsiedenden Lösungsmittels der Grignard-Reaktion, bis die Umset zung beendet ist.
Bei der Acylierung von freien Hydroxyverhindungen der Formel II können Anhydride und Halogenide nie derer Alkansäuren im überschuss zugleich als Reaktions medium dienen; andere Acylierungsmittel werden in inerten organischen Lösungsmittel wie z. B. Diäthyl- äther, Tetrahydrofuran oder Benzol angewendet. Ein säurebindendes Mittel, z.
B. eine tertiäre organische Base wie Pyridin oder Triäthylamin, kann gewünschtenfalls zugefügt werden, ist aber für eine vollständige Umset zung nicht unerlässlich. Die Acylierung von Verbindun gen der Formel II, in denen X durch Wasserstoff ver körpert ist, erfolgt mit Vorteil bei Temperaturen von 0 bis ca.
1000 , da es sich bei den betreffenden Ausgangs stoffen d-er Formel II um tertiäre Hydroxyverbindungen handelt, die mit Acylierungsmitteln bei noch höheren Temperaturen leicht unter Wasserabspaltung reagieren.
Die neuen Piperidinderivate werden, wie weiter vorne erwähnt, rektal oder parenteral verabreicht. Die täglichen Dosen der freien Basen oder von nichttoxischen Salzen derselben bewegen sich zwischen 1 und 800 mg für er wachsene Patienten. Geeignete Doseneinheitsformen, wie Dragees, Tabletten, Suppositorien oder Ampullen,
enthalten als Wirkstoffe vorzugsweise 1 bis 200 mg eines erfindungsgemäss herstellbaren Piperidinderivates oder eines nicht toxischen Salzes desselben.
Unter nichttoxischen Salzen der erfindungsgemäss erhaltenen Basen sind Salze mit solchen Säuren zu ver stehen, deren Anionen bei den in Frage kommenden Do- sierungen pharmakologisch annehmbar sind, d. h. keine toxischen Wirkungen ausüben. Ferner ist es von Vorteil, wenn die zu verwendenden Salze gut kristallisierbar und nicht oder wenig hygroskopisch sind. Als nichttoxische Salze kommen z.
B. die Salze mit der Chlorwasserstoff- säure, Bromwasserstoffsäure, Schwefelsäure, Phosphor säure, Methansulfonsäure, Äthansulfons:äure, ,ss Hydroxy- äthansulfonsäure, Essigsäure, Milchsäure, Oxalsäure;
Bernsteinsäure, Fumarsäure, Maleinsäure, Äpfelsäure, Weinsäure, Zitronensäure, Benzoesäure, Salicylsäure, Phenylessigsäure, Mandelsäure und Embonsäure in Be tracht.
Doseneinheitsformen für die orale Anwendung ent- halten als Wirkstoff vorzugsweise zwischen 1 % und 90 % eines Piperidinderivates der Formel I oder eines nichttoxischen Salzes eines solchen. Zu ihrer Herstel lung verarbeitet man den Wirkstoff z.
B. mit festen, pulverförmigen Trägerstoffen, wie Lactose, Saccharose, Sorbit, Mannit; Stärken, wie Kartoffelstärke, Maisstärke oder Amylopektin, ferner Laminariapulver oder Citrus- pulpenpulver;
Cellulosederivaten oder Gelatine, gegebe nenfalls unter Zusatz von Gleitmitteln, wie Magnesium- oder Calciumstearat oder Polyäthylenglykolen (Carbo- waxen) von geeigneten Molekulargewichten zu Tabletten oder zu Dragee-Kernen. Letztere überzieht man bei spielsweise mit konzentrierten Zuckerlösungen, welche z.
B. noch arabischen Gummi, Talk und/oder Titan dioxyd enthalten können, oder mit einem in leichtflüch tigen organischen Lösungsmitteln oder Lösungsmittel- gemischen gelösten Lack. Diesen Überzügen können Farbstoffe zugefügt werden, z. B. zur Kennzeichnung verschiedener Wirkstoffdosen.
Fernar kommen besonders für die Behandlung des Hustens z. B. auch Lutschtabletten sowie nicht-einzel- dosierte orale Applikationsformen, wie z. B. mit den üblichen Hilfsstoffen bereitete Hustensirups und Husten tropfen in Betracht.
Als Doseneinheitsformen für die rektale Anwendung kommen z. B. Suppositorien, welche aus einer Kom bination eines Piperidinderivates der Formel I oder eines geeigneten Salzes desselben mit einer Neutralfettgrund- lage bestehen, oder auch Gelatine-Rektalkapseln, welche eine Kombination des Wirkstoffes oder eines geeigneten Salzes desselben mit Polyäthylenglykolen (Carbowaxen)
von geeignetem Molekulargewicht enthalten, in Betracht.
Ampullen zur parenteralen, insbesondere intramus- kulären Verabreichung enthalten vorzugsweise ein was serlösliches Salz eines Piperidinderivates der Formel I in einer Konzentration von vorzugsweise 0,5 bis 5 )/o, gegebenenfalls zusammen mit geeigneten Stabilisierungs- mitteln und Puffersubstanzen,
in wässriger Lösung.
Die folgenden Vorschriften sollen die Herstellung von Tabletten und Dragees näher erläutern: a) 250 g 1-Methyl-4-allyl-4-acetoxy piperidin-Hydro- chlorid werden mit 175,80 g Lactose und 169,70 g Kar toffelstärke vermischt, die Mischung mit einer alkoholi schen Lösung von 10 g Stearinsäure befeuchtet und durch ein Sieb granuliert. Nach dem Trocknen mischt man 160g Kartoffelstärke, 200 g Talk, 2,
50g Magne- siumstearat und 32 g kolloidales Siliciumdioxyd zu und presst die Mischung zu 10 000 Tabletten von je 100 mg Gewicht und 25 mg Wirkstoffgehalt, die gewünschten falls mit Teilkerben zur feineren Anpassung der Dosie- rung versehen sein können.
b) Aus 250 g 1 Methyl-4-allyl-4.-acetoxy-piperidin- Hydrochlorid, 179,50 g Lactose und der alkoholischen Lösung von 10 g Stearinsäure stellt man ein Granulat her, das man nach dem Trocknen mit 56,60 g kolloida lem Siliciumdioxyd, 165g Talk, 20 g Kartoffelstärke und 2,50 g Magnesiumstearat mischt und zu 10 000 Dragee-Kernen presst.
Diese werden anschliessend mit einem konzentrierten Sirup aus 522,28 g krist. Saccha- rose, 6 g Schellack, 10 g arabischem Gummi,<B>215</B> g Talk, 15 g kolloidalem Siliciumdioxyd, 0,22 g Farbstoff und 1,5g Titandioxyd überzogen und getrocknet. Die er haltenen Dragees wiegen je 145 mg und enthalten je 25 mg Wirkstoff.
c) Zur Bereitung eines Hustensirups werden 20 g 1-Methyl-4-allyl-4-acetoxy-piperidin-Hydrochlorid, 42 g p-Hydroxy-benzoesäure-methylestex, 18 g p-Hydroxy- benzoesäure-propylester und 5000 g Kristallzucker so wie ein beliebiger Geschmackstoff in destilliertem Was ser zu 10 1 gelöst.
d) Zur Herstellung von Hustentropfen löst man 500 mg 1-Methyl-4-allyl-4-acetoxy-piperidin-Hydrochlo- rid, 10 g Ascorbinsäure, Süsstoff, z. B. 5 g Natrium- cyclamat, Geschmackstoff nach Belieben und 2500 g Sorbit (70o/oig) in destilliertem Wasser zu 101.
Die nachfolgenden Beispiele erläutern die Durchfüh- rung der erfindungsgemässen Herstellungsverfahren näher. Die Temperaturen sind in Celsiusgraden ange geben.
<I>Beispiel 1</I> In, einem 750 nzl Vierhalskolben werden 10,2 g Magnesiumspäne in 30 ml abs. Äther vorgelegt und unter Rühren wenig Allylbromid zugetropft. Sobald die Reaktion in Gang gekommen ist, werden 100 ml abs. Äther zugefügt und 50,8g Allylbromid derart zuge tropft, dass der Äther siedend bleibt (Rückfluss)
und anschliessend 10 Minuten weiter gerührt. Dann werden 28,2 g 1-Propyl-4-piperidon in 50 ml abs. Äther zu- getropft, 15 Minuten unter Rückfluss gekocht, die Hei zung entfernt und 109,2 g Propionsäureanhydrid in 70 ml abs. Benzol zugetropft. Die gelbliche Suspersion wird hierauf 6 Stunden unter Rückfluss erhitzt, auf Eis gegossen,
mit konz. Salzsäure sauer gestellt, und die ätherische Phase dreimal mit 6-n. Salzsäure extrahiert. Die vereinigten sauren Auszüge werden mit konz. Am moniak alkalisch gestellt, viermal mit Chloroform aus gezogen, getrocknet, eingedampft und destilliert. Das 1-Propyl-4-allyl-4-propionoxy-piperidin siedet bei 88 / 0,04 Torr. Daraus wird das Hydrochlorid in abs. Äther mit ätherischer Salzsäure hergestellt, Smp. 225-226 (aus Isopropanol-Äther)

1-(2'-Phenyl-äthyl)-4-allyl-4-propionoxy-piperidin, Kp. 152 /0,05 Torr, Hydrochlorid Smp. 228-230 . <I>Beispiel 2</I> In einem 1500 ml Vierhalskolben werden 14,58 g Magnesiumspäne in 25 ml abs. Äther vorgelegt und wenig Crotylbromid (1 Brom-2-buten) zugetropft. Ist die Reaktion in Gang gekommen,
werden 215 ml abs. Äther zugegeben und während 4-5 Stunden unter Rühren der Rest von 27g Crotylbromid langsam zugetropft. An- schliessend wird das Reaktionsgemisch weitere 30 Minu ten gerührt, 11,3 g 1-Methyl-4-piperidon in 100 ml abs. Äther in 10 Minuten zugegeben, das Gemisch 30 Minu ten unter Rückfluss gekocht,
50 ml Acetanhydrid zu gegeben und die gelbe Suspension noch 2 Stunden unter Rückfluss gekocht. Hierauf wird das Gemisch auf Eis gegossen, mit konz. Salzsäure sauer gestellt, und die ätherische Phase dreimal mit 6-n. Salzsäure extrahiert. Die vereinigten sauren Auszüge werden mit konz. Am moniak alkalisch gestellt, mit Chloroform viermal aus gezogen, die Chloroformlösung getrocknet und einge dampft und der Rückstand destilliert. Das 1-Methyl-4- (1'-methyl-allyl)-4-ace-toxy-piperidin siedet bei 59-65 / 0,05 Torr.
Das Hydrochlorid wird durch Versetzen der Lösung der Base in abs. Äther mit ätherischer Salzsäure herge stellt. Smp. 198 (Aceton).
In gleicher Weise wird das 1-Methyl-4-(1'-methyl- allyl)-4-propionoxy-piperidin hergestellt, dessen Hydro- chlorid bei 207 schmilzt (aus Aceton-Isopropanol). <I>Beispiel 3</I> a) 5,1g Magnesiumspäne werden in einem 350 ml Vierhalskolben in 15 ml Äther vorgelegt und wenig Allylbromid zugetropft. Ist die Reaktion in Gang ge kommen, werden 60 ml abs. Äther zugegeben und unter Rühren der Rest von 25,4 g Allylbromid so zu- getropft,
dass der ,Äther im Sieden bleibt (Rückfluss). Anschliessend rührt man noch 10 Minuten weiter, wor auf<B>11,3</B> g 1 Methyl-4-piperidon in 30 ml abs. Äther langsam zugegeben werden.
Nach 30 Minuten Kochen unter Rückfluss wird das Gemisch mit Eis zersetzt, mit konZ. Säure Sauer gestellt und die ätherische Phase dreimal mit 6-n.
Salzsäure extrahiert. Die vereinigten sauren Auszüge werden mit konz. Ammoniak alkalisch gestellt und mit Chloroform extrahiert, die Chloroform- lösung getrocknet und eingedampft und der Rückstand destilliert. Das 1 Methyl-4-allyl-4 piperidinol siedet bei 60-64 /0,0l Torr;
Citrat Smp. 95-97 (aus Methanol- Äther). Auf gleiche Weise wird das 1 Propyl-4-allyl-4- piperidinol, Kp. 69-76 /0,02 Torr, erhalten.
b) 7,6 g des Produktes a) werden mit 40 ml Acetan- hYdrid 3 Stunden unter Rückfluss gekocht. Hierauf wird die Reaktionslösung am Rotationsverdampfer einge dampft, der Rückstand mit Eis zersetzt, stark alkalisch gemacht und fünfmal mit Chloroform ausgezogen. Die Chloroformlösung wird getrocknet und eingedampft und der Rückstand destilliert.
Das 1-Methyl-4-allyl-4-acet- oxy-piperidin siedet bei 112-l15 /12 Torr, Hydrochlo- rid Smp. 167-168 (Isopropanol Äther).
Auf gleiche Weise wird das 1 Methyl-4-allyl-4-pro- pionoxy-piperidin hergestellt, Hydrochlorid Smp. 157- 158 (aus Isopropanol-Äther). <I>Beispiel 4</I> 3,5 g 1 Methyl-4-allyl-4-piperidinol-hydrochlorid und 250 m1 p-Toluolsulfonsäure werden in 150 ml Chlo roform gelöst.
Durch diese Lösung wird frisch aus Ace ton hergestelltes Keton geleitet (Apparatur nach Quad- beck, Angew. Chemie 68, 369; 1956).
Das Fortschrei ten der Reaktion wird mit Dünnschichtchromatographie verfolgt (Silicagel mit 0,5 %, Natronlauge, Eluiermittel: Methanol). Nach 15 Minuten kann kein Ausgangsmate rial mehr festgestellt werden. Während der Reaktion ver färbt sich die Lösung gelb und die Temperatur steigt auf ca.. 40 .
Die Lösung wird auf die Hälfte eingeengt und mit je 100 ml 2-n. Salzsäure zweimal extrahiert. Die saure Lösung wird mit Soda alkalisch gestellt, das ausge fallene Produkt dreimal mit je 100 ml Äther extrahiert und mit wasserfreiem Natriumsulfat getrocknet und ein gedampft.
Das ölige 1-Methyl-4-allyl-4-acetoxy-piperi- din wird ins Citrat übergeführt, Smp. <B>1819.</B> <I>Beispiel 5</I> In einer Grignardapparatur werden 5,64g Magne sium in 15 ml absolutem Äther vorgelegt und von 31,3 g Methallylbromid 2-3 ml zugegeben.
Sobald die Reak tion in Gang gekommen ist, wird 60 ml Äther zugefügt und anschliessend das restlicheMethallylbromid während 30 Minuten so zugetropft, dass die Lösung siedet. An schliessend wird 10 Minuten am Rückfluss erhitzt und das N-Methylpiperidon in. 40 ml Äther zugegeben. Nach dem die Reaktion nachgelassen hat, wird noch 30 Minu ten am Rückfluss erhitzt.
Hierauf werden 50) ml Propion- säureanhydrid in 50 ml absolutem Benzol zugefügt und 6 Stunden am Rückfluss erhitzt. Dann wird mit Eis und konz. Salzsäure zersetzt, die saure Phase abgetrennt und die organische Phase noch zweimal mit 2-n. Salzsäure ausgezogen.
Die wässrigen Auszüge werden vereint, mit konz. Ammoniak alkalisch gestellt, viermal mit Chloro form ausgezogen, getrocknet, verdampft und destilliert. Das 1-Methyl-4-methallyl-4-propionoxy-piperidin siedet unter 0,04 mm Quecksilber bei 60=65 . Daraus wird in üblicher Weise das Citrat hergestellt und aus Methanol umkristallisiert. Es schmilzt bei 177-178 .

These are preferably reacted directly with reactive functional derivatives of carboxylic acids of the formula III; H. instead of decomposing the resulting solution of the product of the Grignard reaction in diethyl ether or another ethereal solvent in the usual way, it is treated with a reactive functional derivative of a carboxylic acid of the formula III,

As mentioned above, the new piperidine derivatives are administered rectally or parenterally. The daily doses of the free bases or of non-toxic salts thereof are between 1 and 800 mg for adult patients. Suitable unit dosage forms, such as dragees, tablets, suppositories or ampoules,
preferably contain 1 to 200 mg of a piperidine derivative which can be prepared according to the invention or a non-toxic salt thereof as active ingredients.
Non-toxic salts of the bases obtained according to the invention are to be understood as meaning salts with acids whose anions are pharmacologically acceptable in the dosages in question, ie. H. have no toxic effects. It is also advantageous if the salts to be used are readily crystallizable and have little or no hygroscopic properties. The nontoxic salts are e.g.

of suitable molecular weight.
Ampoules for parenteral, especially intramuscular administration preferably contain a water-soluble salt of a piperidine derivative of the formula I in a concentration of preferably 0.5 to 5) / o, optionally together with suitable stabilizing agents and buffer substances,
in aqueous solution.
The following instructions are intended to explain the production of tablets and coated tablets in more detail: a) 250 g of 1-methyl-4-allyl-4-acetoxy piperidine hydrochloride are mixed with 175.80 g of lactose and 169.70 g of potato starch, the Mixture moistened with an alcoholic solution of 10 g of stearic acid and granulated through a sieve. After drying, mix 160g potato starch, 200 g talc, 2,
50 g of magnesium stearate and 32 g of colloidal silicon dioxide are added and the mixture is pressed into 10,000 tablets each weighing 100 mg and containing 25 mg of active ingredient, which, if desired, can be provided with partial notches for a more precise adjustment of the dosage.
b) From 250 g of 1-methyl-4-allyl-4.-acetoxy-piperidine hydrochloride, 179.50 g of lactose and the alcoholic solution of 10 g of stearic acid, granules are produced which, after drying, contain 56.60 g Mix colloidal silicon dioxide, 165 g talc, 20 g potato starch and 2.50 g magnesium stearate and press them to form 10,000 coated tablets.
These are then crystallized with a concentrated syrup of 522.28 g. Sucrose, 6 g shellac, 10 g gum arabic, 215 g talc, 15 g colloidal silicon dioxide, 0.22 g colorant and 1.5 g titanium dioxide coated and dried. The coated tablets each weigh 145 mg and each contain 25 mg of active ingredient.
c) To prepare a cough syrup, 20 g of 1-methyl-4-allyl-4-acetoxy-piperidine hydrochloride, 42 g of p-hydroxy-benzoic acid methylestex, 18 g of p-hydroxy-benzoic acid propyl ester and 5000 g of granulated sugar as Any flavoring substance dissolved in 10 l of distilled water.
d) To produce drops of cough, 500 mg of 1-methyl-4-allyl-4-acetoxypiperidine hydrochloride, 10 g of ascorbic acid, sweetener, e.g. B. 5 g sodium cyclamate, flavoring at will and 2500 g sorbitol (70%) in distilled water to 101.

1-Methyl-4- (1'-methyl-allyl) -4-propionoxy-piperidine, the hydrochloride of which melts at 207 (from acetone-isopropanol), is prepared in the same way. <I> Example 3 </I> a) 5.1 g of magnesium shavings are placed in a 350 ml four-necked flask in 15 ml of ether and a little allyl bromide is added dropwise. Once the reaction has started, 60 ml abs. Ether is added and the remainder of 25.4 g of allyl bromide is added dropwise while stirring,
that the, ether remains in the boil (reflux). The mixture is then stirred for a further 10 minutes, whereupon 11.3 g of 1-methyl-4-piperidone in 30 ml of abs. Ether can be added slowly.
After boiling under reflux for 30 minutes, the mixture is decomposed with ice, with conc. Acid acidified and the ethereal phase three times with 6-n.
Hydrochloric acid extracted. The combined sour extracts are concentrated with Made ammonia alkaline and extracted with chloroform, the chloroform solution dried and evaporated and the residue distilled. The 1-methyl-4-allyl-4-piperidinol boils at 60-64 / 0.0l Torr;
Citrate m.p. 95-97 (from methanol-ether). 1-propyl-4-allyl-4-piperidinol, b.p. 69-76 / 0.02 torr, is obtained in the same way.
b) 7.6 g of product a) are refluxed with 40 ml of acetanhydride for 3 hours. The reaction solution is then evaporated on a rotary evaporator, the residue is decomposed with ice, made strongly alkaline and extracted five times with chloroform. The chloroform solution is dried and evaporated and the residue is distilled.
The 1-methyl-4-allyl-4-acetoxy-piperidine boils at 112-115 / 12 torr, hydrochloride melting point 167-168 (isopropanol ether).
1-methyl-4-allyl-4-propionoxypiperidine, hydrochloride mp 157-158 (from isopropanol ether) is prepared in the same way. <I> Example 4 </I> 3.5 g of 1-methyl-4-allyl-4-piperidinol hydrochloride and 250 ml of p-toluenesulfonic acid are dissolved in 150 ml of chloroform.
Ketone freshly prepared from acetone is passed through this solution (apparatus according to Quadbeck, Angew. Chemie 68, 369; 1956).
The progress of the reaction is followed by thin layer chromatography (silica gel with 0.5%, sodium hydroxide solution, eluent: methanol). After 15 minutes, no more starting material can be determined. During the reaction the solution turns yellow and the temperature rises to approx. 40.
The solution is concentrated to half and with 100 ml of 2-n. Hydrochloric acid extracted twice. The acidic solution is made alkaline with soda, the product which has precipitated out is extracted three times with 100 ml of ether each time, dried with anhydrous sodium sulfate and evaporated.
The oily 1-methyl-4-allyl-4-acetoxy-piperidine is converted into citrate, melting point 1819. Example 5 In a Grignard apparatus, 5.64 g of magne are added sium presented in 15 ml of absolute ether and added 2-3 ml of 31.3 g of methallyl bromide.
As soon as the reaction has started, 60 ml of ether is added and then the remaining methallyl bromide is added dropwise over a period of 30 minutes so that the solution boils. The mixture is then refluxed for 10 minutes and the N-methylpiperidone in 40 ml of ether is added. After the reaction has subsided, reflux is continued for 30 minutes.
50 ml of propionic anhydride in 50 ml of absolute benzene are then added and the mixture is refluxed for 6 hours. Then with ice and conc. Hydrochloric acid decomposed, the acidic phase separated and the organic phase twice with 2-n. Extracted hydrochloric acid.
The aqueous extracts are combined with conc. Ammonia made alkaline, extracted four times with chloro form, dried, evaporated and distilled. The 1-methyl-4-methallyl-4-propionoxy-piperidine boils below 0.04 mm of mercury at 60 = 65. The citrate is produced from this in the usual way and recrystallized from methanol. It melts at 177-178.
