Verfahren zur Herstellung von Pyrazincarbonsäuren
Den Homologen der Pyrazincarbonsäure kommt in Wissenschaft und Technik eine stetig steigende Bedeutung zu.
Die bisher bekanntgewordenen Verfahren für die Herstellung derartiger Verbindungen sind jedoch sehr umständlich und teuer. Es sind daher nur wenige einschlägige Verbindungen hergestellt worden. Sie werden gewöhnlich erhalten durch Oxydationsprozesse. So ist die 5-Methyl-pyrazincarbonsäure von Stoehr, J. Prakt.
Chem. (2) 47. S. 480 (1893) erhalten worden durch Oxydation von 2, 5-Dimethyl-pyrazin. Die 6-Methylpyrazincarbonsäure wurde von Leonard & Spoerri, Journal of the Amer. Chem. Soc. 68. S. 526 (1946), hergestellt durch Oxydation von 2-Methyl-chinoxalin.
Bei diesen Oxydationsprozessen verwendet man gewöhnlich Kaliumpermanganat als Oxydationsmittel.
Für die Herstellung von 1 kg einer homologen Pyrazincarbonsäure werden gewöhnlich 10 kg Kaliumpermanganat und mehr benötigt.
Ein weiterer Nachteil der bekannten Verfahren besteht darin, dass sich bei der Oxydation von Polyalkyl-pyrazinen mit Kaliumpermanganat ausser den gewünschten Alkyl-pyrazincarbonsäuren verständlicherweise stets auch Pyrazinpolycarbonsäuren bilden. überdies greift der Oxydationsprozess auch immer das Pyrazinringskelett an und zerstört so einen nicht geringen Anteil des Produktes. Beides führt zu uneinheitlichen Produkten und zu geringen Ausbeuten.
Das bereits vorhandene Interesse wird sich noch stark erhöhen, sobald ein gutes Herstellverfahren zur Verfügung steht.

in einem Lösungsmittel unter alkalischen Bedingungen kondensiert und die dabei erhaltene Dihydro-pyrazincarbonsäure in diesem Lösungsmittel oxydiert.
Als Oxydationsmittel genügt bereits Luft.
Die Kondensation und anschliessende Oxydation muss in einem Lösungsmittel ausgeführt werden, wobei als bevorzugtes Lösungsmittel ein Alkohol oder ein Alkohol-Derivat verwendet wird. Besonders bewährt haben sich Methanol oder Alkoxyäthanole, wie z. B.
Methoxyäthanol.
Das Gelingen dieses Verfahrens ist für den Fachmann überraschend. Die Kondensation von Äthylen- diamin mit Glyoxal oder niedrigen a, ss-Diketonen oder a, ss-Ketoaldehyden ist bisher nicht gelungen. Es war demnach nicht anzunehmen, dass die analoge Kondensation von a, ss-Diaminopropionsäure mit den erwähnten Diketonen bzw. Ketoaldehyden gelingt.
Durch das erfindungsgemässe Verfahren werden die eingangs erwähnten Nachteile der vorbekannten Verfahren umgangen und es ist möglich mit guten Ausbeuten Alkyl-pyrazin-carbonsäuren zu erhalten, welche frei sind von Pyrazin-poly-carbonsäuren.
Beispiel 1
5, 6-Diphenyl-pyrazin-2-carbonsäure.
35 g a, ss-Diamina, pr. opionsäure-hydrochlorid wer- den in 2250 ccm Methanol, worin bereits 40 g Natriumhydroxyd aufgelöst sind, eingetragen.
In die erhaltene Lösung fügt man unter Rühren 52, 5 g Benzil. Die Reaktionsmischung wird während 20 Minuten am Rückflusskühler gekocht. Anschliessend wird während 40 Minuten Luft durch die Reaktionsmi- schung gesaugt oder geblasen, wodurch sich die Reaktionslösung aulhellt. Diese vird im Vakuum auf-l Volumen von ca. 350 ccm eingeengt, mit 300 ccm Diäthyläther versetzt und während 12 bis 16 Stunden bei 0 C stehengelassen. Das dabei ausgeschiedene Natrium-Salz von 5, 6-Diphenylpyrazin-2-carbonsäure wird abgenutscht und getrocknet.
Ausbeute : 68, 5 g.
Das so erhaltene rohe Salz wird in Wasser aufgelöst, die Lösung durch Zusatz von Salzsäure auf pH 6-6, 5 eingestellt, mit Aktivkohle entfärbt, biarauf durch Zusatz weiterer Salzsäure auf pH 2 eingestellt.
Die freie 5, 6-Diphenyl-pyxazin-2-carbonsäure beginnt nun auszukristallisieren. Sie wird durch Extralc- tion mit Äther vollständig abgetrennt. Nach dem Verdampfen der Ätherlösung und Umkristallisieren des Rückstandes aus ca. 30 /oigem Äthanol werden insgesamt 45, 3 g 5, 6-Diphenyl-pyrazin-2-carbonsäure vom Schmelzpunkt 174-179 erhalten.
Beispiel 2
5, 6-Dimssthyl-pyrazin-2- : carbonsäure.
28 g ass-Diaminopropionsäure-hydrochlorid werden in eine Lösung von 32 g Natriumhydroxyd in 1800 ccm Methanol eingetragen. In die erhaltene Lösung werden unter Rühren bei 20 C 17, 6 ccm Diacetyl eingetropft. Anschliessend wird noch während 3 Stunden Luft durch die Reaktionslösung gesogen oder geblasen. Die Lösung wird zur Trockene eingedampft, der Rückstand wird in Wasser aufgenommen und die wässrige Lösung durch Zusatz von Salzsäure auf pH 2 eingestellt. Allmählich scheidet sich die gebildete 5, 6 Dimethyl-pyrazin-2-carbonsäure aus. Die Kristallisation wird durch Impfen, Kratzen mit einem Glasstab und längeres Stehenlassen befördert.
Das so erhaltene Produkt wird zur Reinigung aus wenig Wasser umkristallisiert. Man erhält so 14, 1 g 5, 6- Dimethyl-pyrazin-2-carbonsäure vom Schmelzpunkt 180-181 C.
Beispiel 3 5-und 6-Phenyl-pyrazin-2-carbonsäure.
16 g Natrium-hydroxyd werden in 1000 ecm Methanol aufgelöst. Bei Raumtemperatur werden dieser Lösung unter Rühren 14 g a, ss-Diamino-propion- säure-hydrochlorid und nach deren Auflösung 17 g Phenyl-glyoxal-dihydrat zugefügt.
Die Reaktionslösung wird während 2 Stunden bei Raumtemperatur gerührt. Anschliessend wird noch während 4 Stunden trockene Luft durch die Lösung geblasen. Schliesslich wird im Vakuum zur Trockene eingedampft. Der : tückstand wird mit 100-200 ccm Wasser behandelt. Die erhaltene Suspension wird filtriert und das Filtrat mit Salzsäure angesäuert. Das dabei aus der Lösung sich ausscheidende Produkt wird abgenutscht und wiederholt aus 95 %igem Äthanol umkristallisiert. Man erhält so 4, 1 g einer Phenyl-pyra- zin-2-carbonsäure, welche bei 205 C schmilzt.
Der nach dem Eindampfen und Aufnehmen in Wasser erhaltene schwerer lösliche suspendierte Anteil wird durch Zusatz von viel Wasser (800 csm) X Lösung gebracht. Durch Ansäuern mit Salzsäure wird eine Fällung erzeugt. Diese wird abfiltriert, getrocknet und ebenfalls wiederholt aus 95 feigem Äthanol umkri- stallisiert. Man erhält so 3, 9 g einer reinen Phenylpyrazin-2-carbonsäure vom Schmelzpunkt 190 C.
Auf Grund der positiven Farbreaktion mit Nitroprussid-natirum, welche gewöhnlich nur von 2,6 substituierten Pyrazin-Derivaten erhalten wird, ist die bei 205 C schmelzende Verbindung als 6-Phenyl-pyrazin2-carbonsäure anzusprechen.
Die bei 190 C schmelzende isomere Form ist auf Grund der fehlenden Farbreaktion als 5-Phenyl-pyra zin-2-carbonsäure anzusprechen.
Beispiel 4
6-Methyl-pyrazin-2-carbonsäure.
16 g Natrium-hydroxyd werden in 1000 ccm Methanol aufgelöst. Die Lösung wird mit 14 g a, ss-Dia mino-propionsäure-hydro. chlorid versetzt. Nach Auflösung der leitzteren werden 21,8 g 33%iges methylglyoxal zugefügt. Die so erhaltene rote Lösung wird während 1l/2 Stunden bei Raumtemperatur gerührt.
Anschliessend wird während 3 Stunden Luft durch die Lösung perlen gelassen. Nach Stehenlassen über Nacht wird die Löisung zur Trockene verdampft. Der Eindampfrückstand wird in wenig Wasser aufgelöst, klarfiltriert und danach mit Salzsäure auf pH 1, 5 angesäu ert. Es entsteht eine geringe Ausfällung. Die Kristal lisation des Produktes wird-durch Stehenlassen über Nacht bei 0 C vervollständigt. Danbach wird genutscht und getrocknet. Nach Umkristallisieren aus wenig wasser erhält man 5,7 g einer bie 197 C schmelzenden Methyl-pyrazin-carbonsäure, Mikroanalyse beredhnet für C. H.
N (138, 13) berechnet : C : 52, 17 /o ; H : 4, 3 ; 8%; N: 20,28% ; gefunden :. C : 51, 68 /o ; H: 5,51 %; N : 20, 03 /R.
Dieses Produkt wird auf Groun seiner positiven Farbreaktion mit Nitroprussid-Natirum als 6-methyl-pyrazin-2-carbonsäure angesprochen.
Process for the preparation of pyrazine carboxylic acids
The homologues of pyrazine carboxylic acid are becoming increasingly important in science and technology.
The previously known methods for the production of such compounds are very complicated and expensive. Few related compounds have therefore been made. They are usually obtained through oxidation processes. The 5-methylpyrazine carboxylic acid from Stoehr, J. Prakt.
Chem. (2) 47, p. 480 (1893) obtained by oxidation of 2,5-dimethyl-pyrazine. The 6-methylpyrazine carboxylic acid was described by Leonard & Spoerri, Journal of the Amer. Chem. Soc. 68. p. 526 (1946), prepared by the oxidation of 2-methyl-quinoxaline.
In these oxidation processes, potassium permanganate is usually used as an oxidizing agent.
For the production of 1 kg of a homologous pyrazine carboxylic acid, 10 kg of potassium permanganate and more are usually required.
Another disadvantage of the known processes is that, when polyalkylpyrazines are oxidized with potassium permanganate, in addition to the desired alkylpyrazine carboxylic acids, pyrazine polycarboxylic acids are understandably also formed. In addition, the oxidation process always attacks the pyrazine ring skeleton and thus destroys a not inconsiderable proportion of the product. Both lead to inconsistent products and low yields.
The already existing interest will increase significantly as soon as a good manufacturing process is available.

Air is sufficient as an oxidizing agent.
The condensation and subsequent oxidation must be carried out in a solvent, an alcohol or an alcohol derivative being used as the preferred solvent. Methanol or alkoxyethanols, such as. B.

The success of this process is surprising to the person skilled in the art. The condensation of ethylene diamine with glyoxal or lower a, ss-diketones or a, ss-ketoaldehydes has not yet been successful. It was therefore not to be assumed that the analogous condensation of α, β-diaminopropionic acid with the diketones or ketoaldehydes mentioned would succeed.
The method according to the invention circumvents the disadvantages of the previously known methods mentioned at the outset and it is possible to obtain, with good yields, alkyl pyrazine carboxylic acids which are free from pyrazine polycarboxylic acids.
example 1
5, 6-diphenylpyrazine-2-carboxylic acid.
35 g a, ss-Diamina, pr. Opionic acid hydrochloride are added to 2250 cc of methanol, in which 40 g of sodium hydroxide have already been dissolved.
52.5 g of benzil are added to the solution obtained with stirring. The reaction mixture is refluxed for 20 minutes. Subsequently, air is sucked or blown through the reaction mixture for 40 minutes, as a result of which the reaction solution brightens. This is concentrated in vacuo to a volume of approx. 350 ccm, 300 cc of diethyl ether are added and the mixture is left to stand at 0 C for 12 to 16 hours. The sodium salt of 5, 6-diphenylpyrazine-2-carboxylic acid which separates out is filtered off with suction and dried.
Yield: 68.5 g.
The crude salt thus obtained is dissolved in water, the solution is adjusted to pH 6-6.5 by adding hydrochloric acid, decolorized with activated charcoal, then adjusted to pH 2 by adding more hydrochloric acid.
The free 5, 6-diphenyl-pyxazine-2-carboxylic acid now begins to crystallize out. It is completely separated off by extraction with ether. After evaporation of the ether solution and recrystallization of the residue from about 30% ethanol, a total of 45.3 g of 5,6-diphenyl-pyrazine-2-carboxylic acid with a melting point of 174-179 are obtained.
Example 2
5, 6-Dimsthyl-pyrazine-2-: carboxylic acid.
28 g of aqueous diaminopropionic acid hydrochloride are added to a solution of 32 g of sodium hydroxide in 1800 cc of methanol. 17.6 ccm of diacetyl are added dropwise to the solution obtained at 20 ° C. with stirring. Subsequently, air is sucked or blown through the reaction solution for a further 3 hours. The solution is evaporated to dryness, the residue is taken up in water and the aqueous solution is adjusted to pH 2 by adding hydrochloric acid. The 5, 6-dimethylpyrazine-2-carboxylic acid formed gradually separates out. Crystallization is promoted by seeding, scratching with a glass rod and allowing it to stand for a long time.
The product thus obtained is recrystallized from a little water for purification. 14.1 g of 5,6-dimethylpyrazine-2-carboxylic acid with a melting point of 180-181 ° C. are obtained in this way.
Example 3 5- and 6-phenyl-pyrazine-2-carboxylic acid.
16 g of sodium hydroxide are dissolved in 1000 ecm of methanol. At room temperature, 14 g of α, β-diamino-propionic acid hydrochloride and, after its dissolution, 17 g of phenyl-glyoxal dihydrate are added to this solution with stirring.
The reaction solution is stirred for 2 hours at room temperature. Dry air is then blown through the solution for a further 4 hours. Finally, it is evaporated to dryness in vacuo. The: residue is treated with 100-200 cc of water. The suspension obtained is filtered and the filtrate is acidified with hydrochloric acid. The product which separates out of the solution is suction filtered and repeatedly recrystallized from 95% ethanol. This gives 4.1 g of a phenylpyrazine-2-carboxylic acid which melts at 205.degree.
The sparingly soluble suspended portion obtained after evaporation and taking up in water is brought into solution by adding a lot of water (800 csm). Acidification with hydrochloric acid creates a precipitate. This is filtered off, dried and also repeatedly recrystallized from fig ethanol. This gives 3.9 g of a pure phenylpyrazine-2-carboxylic acid with a melting point of 190 C.
Due to the positive color reaction with nitroprusside natirum, which is usually only obtained from 2,6 substituted pyrazine derivatives, the compound which melts at 205 C is to be addressed as 6-phenyl-pyrazine-2-carboxylic acid.
The isomeric form, which melts at 190 C, is to be addressed as 5-phenyl-pyrazine-2-carboxylic acid due to the lack of a color reaction.
Example 4
6-methylpyrazine-2-carboxylic acid.
16 g of sodium hydroxide are dissolved in 1000 cc of methanol. The solution is with 14 g of a, ss-Dia mino-propionic acid-hydro. chloride added. After dissolution of the leading ones, 21.8 g of 33% strength methylglyoxal are added. The red solution obtained in this way is stirred at room temperature for 1/2 hour.
Then air is bubbled through the solution for 3 hours. After standing overnight, the solution is evaporated to dryness. The evaporation residue is dissolved in a little water, filtered clear and then acidified to pH 1.5 with hydrochloric acid. A slight precipitation occurs. The crystallization of the product is completed by leaving it to stand at 0 ° C. overnight. Danbach is sucked and dried. After recrystallization from a little water, 5.7 g of a methylpyrazine carboxylic acid melting at 197 ° C. are obtained, microanalysis discussed for C. H.

This product is referred to as 6-methyl-pyrazine-2-carboxylic acid because of its positive color reaction with nitroprusside sodium.
