Verfahren zur Herstellung von Östradiendionen
Die Erfindung betrifft ein Verfahren zur Herstellung von 4'9-Östradien-3 1 7-dionen.
Ein typisches Erzeugnis des erfindungsgemässen Verfahrens hat die Formel:

Diese Verbindung kann auf bekannte Weise in das entsprechende 1 7a-Äthinyl-1 7fi-hydroxy-4,9-östradien- 3-on oder 17-Chloräthinyl-1 7ss-hydroxy-4, 9-östradien-3- on umgewandelt werden, welche progestational aktiv sind. Die Äthinylverbindung kann z. B. durch Reaktion mit einem Metallacetylid in flüssigem Ammoniak erhalten werden. Ein Alkalimetall oder Erdalkalimetallacetylid, insbesondere Natrium-, Kalium- oder Calciumacetylid, sind hiefür geeignet.
Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass man ein 3-Hydroxy-östra-1,3,5(10), 9(1 1)-tetraen-17-on herstellt, welches man als das 9(11)-Östron bezeichnen kann, mit einem 2,2-Dialkyldioxolan umsetzt und in der entstandenen 17-Alkylendioxy-verbindung die 3-Hydroxygruppe veräthert und das erhaltene Produkt mit einer Persäure umsetzt und danach die entstandene 9,1 1-Epoxy-verbindung mit einem Alkalimetall in flüssigem Ammoniak umsetzt und die so erhaltene Verbindung mit einem Phosphoroxyhalogenid behandelt und das so erhaltene 2,5(10),9(11)- Östratrien mit einer Säure zur Reaktion bringt.
Manche nach dem erfindungsgemässen Verfahren hergestellte Verbindungen sind wertvoll als Progestationalia. Andere wiederum finden Verwendung als Zwischenprodukte, welche man zu Verbindungen mit Progestationalwirkung umsetzen kann.
Wie gesagt, wird das 9(1 1)-Östron in das entsprechende 3-Äther-17-ketal umgewandelt. Die in 3-Stellung befindliche Gruppe kann eine Aralkoxy- oder Niederalkoxygruppe sein, zweckmässigerweise eine Benzyloxy- oder Äthoxygruppe. Das Ketal bei C-17 kann z. B. Äthylendioxy sein. Das 3,17-geschützt-9(1 1)- Östron wird mit einer Persäure als Epoxydierungsmittel behandelt, um das 9(11)-Epoxyd zu erhalten. Perameisensäure, Peressigsäure, Perbenzoesäure oder Perphthalsäure, werden besonders bevorzugt. Dieses 9(11)-Epoxyd wird mit einem Alkalimetall, z. B.
Lithium oder Natrium in flüssigem Ammoniak, zweckmässig in Gegenwart eines niedrigen Alkanols, geeigneterweise n-Butanol oder Athanol, behandelt, um das entsprechende 2,5(10) Östradien-1 1-ol-3-äther-17-ketal zu erhalten. Die in 11 Stellung befindliche Hydroxylgruppe wird mit einem Phosphoroxyhalogenid als Dehydratisierungsmittel behandelt, beispielsweise in Gegenwart eines tertiären Amins, vorzugsweise Phosphoroxychlorid in Gegenwart von Pyridin, um das Östra-2, 5 (10), 9 (11)-trien-3-äther- 1 7-ketal zu erhalten, welches dann mit einer Säure hydrolysiert und gleichzeitig umgelagert wird, um das Östra-4,9-dien-3,17-dion zu erhalten. Diese Behandlung mit einer Säure kann z. B. mit einer Mineralsäure, z. BF Salzsäure, erfolgen.
Diese 1 7-Ketoverbindung ist ein wertvolles Zwischenprodukt zur Herstellung von Verbindungen, welche für ihre hohe Progestationalwirkung bekannt sind, beispielsweise 17B-Hydroxy-l 7a-äthinyl-östra-4,9-dien- 3-on und 17/,'-Hydroxy-17a-chloräthinyl-östra-4,9 3-on. Die 17a-Halogenäthinylverbindungen können beispielsweise dadurch hergestellt werden, dass man das entsprechende 17-Ketosteroid mit dem entsprechenden Athinylmetallderivat, beispielsweise einem Alkalimetallacetylid, geeigneterweise Kaliumacetylid, behandelt.
Ein acetylenisches Grignard-Reagens, beispielsweise ein Äthinyl- oder Halogenäthinyl-magnesiumhalogenid, wie Äthinyl-oder Chloräthinylmagnesiumbromid, kann ebenfalls verwendet werden.

Hierin bedeutet R einen Alkyl-, Allyl oder Aralkylrest, und die Wellenlinie bedeutet eine a- oder ss Konfiguration.
Im einzelnen kann wie folgt vorgegangen werden: Das im ersten Arbeitsschritt als Ausgangsmaterial verwendete 9(11)-Östron wird in einem 2,2-Dialkyl-dioxolan, vorzugsweise 2-Äthyl-2-methyl-1, 3-dioxolan aufgenommen und in Gegenwart eines sauren Katalysators, z. B. einer Sulfonsäure, z.B. p-Toluolsulfonsäure, erhitzt. Freigesetztes Alkanol, z. B. Butanon, wird gemeinsam mit dem Lösungsmittel langsam innerhalb von 4 bis 6 Stunden abdestilliert.
Das Produkt kann durch irgendeine herkömmliche Methode gewonnen werden, z.B. durch Verdünnung des abgekühlten Reaktionsgemisches mit einem mit Wasser nichtmischbarem Kohlenwasserstoff, wie Benzol, sodann durch Waschung der Mischung mit verdünnter wässriger Natriumbicarbonatlösung, Trocknen über einem Trocknungsmittel, z.B. wasserfreiem Natriumsulfat, und Entfernung des Lösungsmittels, so dass man das angestrebte 1 7-Ketal erhält.
Um den 3-Äther zu bilden, wird das 17-Ketal in einem im wesentlichen nichtpolaren Lösungsmittel gelöst, und hierauf wird eine schwache Base, z.B. ein Alkalimetallcarbonat und vorzugsweise Kaliumcarbonat hinzugegeben. Es wird ein Alkyl- oder ein Aralkylhalogenid, beispielsweise Methyljodid oder Benzyljodid hinzugegeben, worauf man bei Zimmertemperatur, beispielsweise zwischen 20 und 300 C, während einer Zeit von 12 bis 18 Stunden in einem zugestopften Gefäss stehenlässt. Das anorganische Material wird dann abfiltriert, und vom Filtrat das Lösungsmittel abgedampft, so dass das erwünschte Produkt als Rückstand verbleibt.
Das Produkt kann auf irgendeine bequeme Art und Weise gereinigt werden. Vorzugsweise wird der Rückstand in einer Mischung von Wasser und einem mit Wasser nichtmischbaren organischen Lösungsmittel, beispielsweise Äther, Benzol oder Äthylacetat, aufgenommen. Die so erhaltene organische Schicht wird mit Wasser gewaschen, über einem Trocknungsmittel, beispielsweise wasserfreiem Natriumsulfat, getrocknet, filtriert und das Lösungsmittel abgehoben. Rekristallisation des Rückstandes ergibt das praktisch reine Östra-1,3,5(10), 9(1 1)-tetraen-3-äther-1 7-ketal.
Das 9(1 l)-Östron-3-äther-17-ketal wird in einem gegenüber der vorzunehmenden Reaktion inerten organischen Lösungsmittel, beispielsweise einem Kohlenwasserstoff, vorzugsweise Benzol aufgenommen, und darin mit einer Persäure als Epoxydierungsmittel, geeigneterweise mit Perbenzoesäure, behandelt. Das Reaktionsgemisch wird bei einer Temperatur von 0 C bis zu etwa 100 C während einer Zeit von etwa 5 bis 7 Stunden gehalten.
Das Produkt kann auf irgendeine geeignete Weise gewonnen werden. Beispielsweise mischt man das Reaktionsgemisch mit kaltem Wasser, welches 2 bis 5 Gew. % Natriumsulfit enthält. Die organische Schicht wird dann abgetrennt, über einem Trocknungsmittel, z. B. Natriumsulfat, getrocknet, filtriert und das Lösungsmittel abgehoben. Man erhält so das rohe 9,11-Epoxy-Produkt, welches gewünschtenfalls durch Chromatographie gereinigt werden kann. Man kann dieses Produkt beispielsweise in Methylenchlorid auflösen und auf Magnesiumsilikat chromatographieren. Die Chromatographiekolonne wird dann mit Petroläther eluiert, welcher 2 bis 15 Gew. % Aceton enthält, worauf man nach Entfernung des Lösungsmittels praktisch reines 9,11-Epoxy- steroid erhält.
Das wie oben hergestellte 9,11-Epoxy-östron-3- äther-17-ketal löst man in einem gegenüber der Reaktion inerten Lösungsmittel, welches mit flüssigem Ammoniak mischbar ist, beispielsweise mit Diäthyläther oder mit Dioxan, und man gibt diese Lösung zu einer vorher bereiteten Lösung eines Alkalimetalls, geeigneter weise Lithium oder Natrium in flüssigem Ammoniak.
Ein niedriges Alkanol, beispielsweise n-Butanol oder Äthanol, wird zur Reaktionsmischung gegeben und dann abgedampft. Die Reaktion ist im allgemeinen in weniger als 60 Minuten beendet und es wird nur selten erforderlich sein, sie während längerer Zeit fortzuführen.
Selbst eine Zeit von weniger als 5 Minuten reicht häufig aus. Man lässt das flüssige Ammoniak bei Umgebungstemperatur verdampfen. Man kann aber gewünschtenfalls die Verdampfung des flüssigen Ammoniaks dadurch beschleunigen, dass man das Reaktionsgefäss von aussen auf Temperaturen von nicht über 500 C erwärmt.
Ein Strom von gegenüber der Reaktion inertem Gas, beispielsweise Stickstoff, kann über die Oberfläche des flüssigen Ammoniaks geführt werden. Dadurch wird die Neigung zu Nebenreaktionen herabgesetzt. Diese Massnahme ist jedoch nicht wichtig.
Das Produkt kann aus dem Rückstand auf jede geeignete Weise gewonnen werden. Beispielsweise nimmt man den Rückstand in Wasser auf und extrahiert die wässrige Lösung mit einem mit Wasser nicht mischbaren organischen Lösungsmittel, wie z. B. einem Kohlenwasserstoff oder einem im wesentlichen nichtpolaren Kohlenwasserstoff, geeigneterweise mit Benzol, Diäthyläther oder Äthylacetat Der organische Extrakt wird abgetrennt, mit wässriger Natriumbicarbonatlösung gewaschen, über einem Trocknungsmittel, z.B. wasserfreiem Natriumsulfat, getrocknet, filtriert und das Lö sungsmittel vom Filtrat abgehoben, so dass man Östra 2,5(10)-dien-1 1-ol-3-äther-17-ketal erhält,
welches in Form einer Mischung von 1 1a- und llss-Epimeren anfällt, welche durch Chromatographie getrennt werden kann.
Die Rerstellung der 9(11)-Dehydro-Verbindung, der 11-Hydroxy-Verbindung, entweder in Form einer Mischung oder in Form der reinen Verbindungen, kann dadurch erfolgen, dass die obgenannte reine Verbindung in einem tertiären Amin, vorzugsweise in Pyridin aufgenommen wird, worauf man sie mit einem Phosphoroxyhalogenid, z.B. Phosphoroxychlorid, behandelt. Man lässt die Mischung bei Umgebungstemperatur, vorzugsweise bei 20 bis 300 C während einer Zeit von 12 bis 18 Stunden stehen. Die Mischung wird gekühlt, d. h. bei einer Temperatur von 0 bis 50 C gehalten, und mit einer äquivalenten volumenmässigen Menge Wasser versetzt.
Beim Stehenlassen kristallisiert das angestrebte Östra 2,5(10),9(1 1)-trien und kann durch Filtration gewonnen werden.
Das so hergestellte Östratrien wird in einem gegen über der Reaktion inerten organischen Lösungsmittel aufgenommen. Ein im wesentlichen nichtpolares Lösungsmittel, beispielsweise Diäthyläther oder 2ithylace- tat, kann hiezu Verwendung finden. Die Mischung wird zu einem sauren Medium, beispielsweise der Lösung einer Mineralsäure in einem Alkohol, vorzugsweise wasserfreier Salzsäure in Methanol gegeben. Man lässt die Mischung bei einer Temperatur von 20 bis 300 C während 12 bis 18 Stunden stehen. Das Produkt, welches ein Östra-4,9-dien-3-on ist, kann nach irgendeiner bekannten Methode gewonnen werden, beispielsweise dadurch, dass man das Lösungsmittel entfernt, vorzugsweise im Vakuum, bei einer Temperatur von 0 bis 200 C.
Beispiel a) Eine Mischung von 1 g 3-Hydroxy-östra-1,3, 5(1 0),9( 1 1)-tetraen-1 7-on, 100 mg p-Toluolsulfonsäuremonohydrat und 20 ml 2-Äthyl-2-methyl-1,3-dioxolan lässt man während 5 Stunden rückfliessen, während das sich bildende Butan zusammen mit dem nichtumgesetzten Dioxolan gleichzeitig langsam durch eine kleine Claisen-Vigreux -Kolonne bei Normaldruck abdestillisiert wird. Das abgekühlte Reaktionsgemisch wird mit Benzol verdünnt, nacheinander mit 5 % iger wässriger Natriumbicarbonatlösung und mit Wasser gewaschen, über Natriumsulfat getrocknet und zur Trockenheit konzentriert, so dass man das gewünschte Produkt als Rückstand erhält.
Dieses Produkt kann durch Kristallisation gereinigt werden, worauf man praktisch reines 17 Athylendioxy-östra-1, 3, 5 (10), 9 (11)-tetraen-3-ol erhält. b) Zu 1 g des obgenannten Produktes, gelöst in 25 ml Aceton, gibt man 3 g wasserfreies Kaliumcarbonat. Man lässt 15 Minuten stehen, worauf 1 g thyl- jodid hinzugegeben wird und die Mischung bei Zimmertemperatur über Nacht in einem verschlossenen Gefäss stehengelassen wird. Der Niederschlag wird durch Filtration entfernt und aus s dem Filtrat das Lösungsmit- tel im Vakuum abgedampft. Der Rückstand wird in einer Mischung von Äthylacetat und Wasser aufgenommen, worauf die wässrige Schicht entfernt wird.
Der Athylacetat-Extrakt wird mit Wasser gewaschen, über wasserfreiem Natriumsulfat getrocknet, filtriert und das Lösungsmittel abgedampft, so dass das erwünschte Produkt zurückbleibt. Dieses Produkt kann durch Rekristallisation gereinigt werden, worauf man praktisch reines 3-Athoxy-17-äthylendioxy-östra-1, 1,3,5(10),9(11)- tetraen erhält.
Geht man wie oben beschrieben vor, verwendet aber Methyljodid oder Benzyljodid anstelle von Athyljodid, dann erhält man die entsprechenden 3-Benzyloxy- oder 3-Methoxy-steroide. c) 18 g des obgenannten Produktes löst man in 50 ml Benzol und kühlt auf etwa 100 C. Nun werden 9 g Perbenzoesäure in Benzol hinzugegeben und man hält diese Reaktionsmischung während etwa 6 Stunden bei einer Temperatur von etwa 80 C. Das Reaktionsgemisch wird dann mit kaltem Wasser, welches 5 Gew. % Natriumsulfit enthält, gewaschen. Die benzolische Lösung, welche das Produkt enthält, wird über wasserfreiem Natriumsulfat getrocknet, filtriert und im Vakuum abgedampft, so dass man das rohe 9,11-Epoxyd erhält.
Dieses Produkt kann gewünschtenfalls gereinigt werden.
Man kann es hiezu beispielsweise in 50 ml Methylenchlorid lösen, und diese Lösung auf 900 g synthetischem Magnesiumsilikat chromatographieren, worauf man die Kolonne mit 1000 ml Fraktionen entwickelt. Als Eluent verwendet man Petroläther, welcher Aceton enthält, wobei die Acetonbeigaben von 2 zu 15 % gesteigert werden. Die Abdampfung der späteren Fraktionen ergibt 3-Äthoxy- 1 7-äthylendioxy-9, 11 -epoxy-östra - 1,3,5(10) - trien.
Geht man wie soeben beschrieben vor, verwendet aber Perameisensäure, Peressigsäure oder Perphthalsäure anstelle von Perbenzoesäure, so erhält man die entsprechenden Produkte. d) Zu einer Lösung von 10 g Lithium in 1000 ml trockenem flüssigem Ammoniak gibt man einer Lösung von 10 g 3-Äthoxy-l7-äthylendioxy-9,l 1-epoxy- östra-1,3,5(10)-trien in 30 ml wasserfreiem Diäthyl äther und rührt diese Mischung während 30 Minuten.
Man gibt genügend n-Butanol hinzu um die blaue Farbe zu entfernen, welche Zugabe innerhalb von 20 Minuten erfolgt, nach welcher Zeit das Ammoniak abge dampft wird und der Rückstand mit Wasser verdünnt wird. Der auf solche Weise verdünnte Rückstand wird mit Diäthyläther extrahiert. Der Ätherextrakt wird mit Wasser gewaschen, dann mit einer gesättigten Natriumbicarbonatlösung wiederum gewaschen, über wasserfreiem Natriumsulfat getrocknet, filtriert und trockengedampft, so dass man eine Mischung von 11 a- und -Epimeren von 3-Äthoxy- 17 - äthylendioxy - östra - 2,5(1O)-dien-l 1-ol erhält, wobei man die Epimeren durch Chromatographie trennt.
In Übereinstimmung mit der soeben beschriebenen Verfahrensweise, jedoch unter Verwendung von Natrium oder Kalium anstelle von Lithium, erhält man die gleichen Produkte. e) 10 g 3-Äthoxy- 1 7-äthylendioxy-östra-2,5 (10)- dien-lla-ol löst man in 100 ml Pyridin und behandelt diese Lösung mit 11 ml Phosphoroxychlorid. Man lässt diese Mischung während etwa 18 Stunden bei einer Temperatur von etwa 250 C stehen, und gibt dann unter Eiskühlung Wasser hinzu. Es bildet sich nun ein Öl, welches beim Stehenlassen kristallisiert, welches darauf abfiltriert und getrocknet wird, so dass man 3 Äthoxy- 1 7-äthylendioxy-östra-2,5 (1 0),9 (11 )-trien erhält.
Aus dem Ild-Epimeren erhält man dasselbe Produkt, genauso wie man aus einer Mischung der beiden Epimeren das gleiche Produkt erhalten kann. f) Zu 500 ml einer gesättigten Lösung von wasserfreiem Chlorwasserstoff in wasserfreiem Äthanol gibt man eine Lösung von 5 g 3-Athoxy-l 7-äthylendioxy- östra-2.5(10),9(1 1)-trien in 50 ml Diäthyläther. Man rührt diese Reaktionsmischung bei einer Temperatur von etwa 250 C während 18 Stunden. Nun wird bei einer Temperatur von 0 C und bei Unterdruck abgedampft, so dass man das Östra-4,9-dien-3,17-dion erhält.

In detail, the procedure is as follows: The 9 (11) oestrone used as starting material in the first step is taken up in a 2,2-dialkyl-dioxolane, preferably 2-ethyl-2-methyl-1,3-dioxolane, and in the presence an acidic catalyst, e.g. A sulphonic acid, e.g. p-toluenesulfonic acid, heated. Liberated alkanol, e.g. B. butanone is slowly distilled off together with the solvent within 4 to 6 hours.
The product can be recovered by any conventional method, e.g. by diluting the cooled reaction mixture with a water-immiscible hydrocarbon such as benzene, then washing the mixture with dilute aqueous sodium bicarbonate solution, drying over a drying agent, e.g. anhydrous sodium sulfate, and removing the solvent to give the desired 17-ketal.
To form the 3-ether, the 17-ketal is dissolved in an essentially non-polar solvent and a weak base, e.g. an alkali metal carbonate and preferably potassium carbonate are added. An alkyl or an aralkyl halide, for example methyl iodide or benzyl iodide, is added, whereupon the mixture is left to stand in a closed vessel at room temperature, for example between 20 and 300 ° C., for a period of 12 to 18 hours. The inorganic material is then filtered off and the solvent is evaporated off from the filtrate, so that the desired product remains as a residue.
The product can be cleaned in any convenient way. The residue is preferably taken up in a mixture of water and a water-immiscible organic solvent, for example ether, benzene or ethyl acetate. The organic layer thus obtained is washed with water, dried over a drying agent such as anhydrous sodium sulfate, filtered, and the solvent is removed. Recrystallization of the residue gives the practically pure oestra-1,3,5 (10), 9 (11) -tetraen-3-ether-1 7-ketal.
The 9 (1 l) -estrone-3-ether-17-ketal is taken up in an organic solvent which is inert to the reaction to be carried out, for example a hydrocarbon, preferably benzene, and is treated therein with a peracid as an epoxidizing agent, suitably with perbenzoic acid. The reaction mixture is held at a temperature from 0 ° C. up to about 100 ° C. for a time from about 5 to 7 hours.
The product can be recovered in any suitable manner. For example, the reaction mixture is mixed with cold water which contains 2 to 5% by weight of sodium sulfite. The organic layer is then separated, over a drying agent, e.g. B. sodium sulfate, dried, filtered and the solvent lifted off. This gives the crude 9,11-epoxy product which, if desired, can be purified by chromatography. This product can, for example, be dissolved in methylene chloride and chromatographed on magnesium silicate. The chromatography column is then eluted with petroleum ether which contains 2 to 15% by weight of acetone, whereupon, after removal of the solvent, practically pure 9,11-epoxy steroid is obtained.
The 9,11-epoxy-estrone-3-ether-17-ketal prepared as above is dissolved in a solvent which is inert to the reaction and which is miscible with liquid ammonia, for example with diethyl ether or with dioxane, and this solution is added to a previously prepared solution of an alkali metal, suitably lithium or sodium in liquid ammonia.
A lower alkanol such as n-butanol or ethanol is added to the reaction mixture and then evaporated. The reaction is generally complete in less than 60 minutes and it will rarely be necessary to continue it for an extended period of time.
Even a time of less than 5 minutes is often sufficient. The liquid ammonia is allowed to evaporate at ambient temperature. However, if desired, the evaporation of the liquid ammonia can be accelerated by heating the reaction vessel from the outside to temperatures not exceeding 500.degree.
A stream of gas which is inert to the reaction, for example nitrogen, can be passed over the surface of the liquid ammonia. This reduces the tendency to side reactions. However, this measure is not important.
The product can be recovered from the residue in any suitable manner. For example, the residue is taken up in water and the aqueous solution is extracted with a water-immiscible organic solvent, such as. A hydrocarbon or a substantially non-polar hydrocarbon, suitably with benzene, diethyl ether or ethyl acetate. The organic extract is separated, washed with aqueous sodium bicarbonate solution, dried over a drying agent, e.g. anhydrous sodium sulfate, dried, filtered and the solvent lifted from the filtrate, so that Östra 2,5 (10) -dien-1 1-ol-3-ether-17-ketal is obtained,
which is obtained in the form of a mixture of 11a and 11ss epimers, which can be separated by chromatography.
The preparation of the 9 (11) -dehydro compound, the 11-hydroxy compound, either in the form of a mixture or in the form of the pure compounds, can take place in that the above-mentioned pure compound is taken up in a tertiary amine, preferably in pyridine , whereupon they are treated with a phosphorus oxyhalide, e.g. Phosphorus oxychloride treated. The mixture is left to stand at ambient temperature, preferably at 20 to 300 ° C., for a period of 12 to 18 hours. The mixture is cooled; H. kept at a temperature of 0 to 50 C, and treated with an equivalent volume of water.
When left to stand, the desired oestra 2.5 (10), 9 (11) -triene crystallizes and can be obtained by filtration.
The estatriene thus produced is taken up in an organic solvent which is inert towards the reaction. An essentially non-polar solvent, for example diethyl ether or 2ithylacetate, can be used for this purpose. The mixture is added to an acidic medium, for example the solution of a mineral acid in an alcohol, preferably anhydrous hydrochloric acid in methanol. The mixture is left to stand at a temperature of 20 to 300 ° C. for 12 to 18 hours. The product, which is an oestra-4,9-dien-3-one, can be obtained by any known method, for example by removing the solvent, preferably in vacuo, at a temperature of 0 to 200 C.
Example a) A mixture of 1 g of 3-hydroxy-oestra-1,3, 5 (1 0), 9 (1 1) -tetraen-1 7-one, 100 mg of p-toluenesulfonic acid monohydrate and 20 ml of 2-ethyl-2 Methyl-1,3-dioxolane is allowed to reflux for 5 hours, while the butane which forms, together with the unreacted dioxolane, is slowly distilled off at the same time through a small Claisen-Vigreux column at atmospheric pressure. The cooled reaction mixture is diluted with benzene, washed successively with 5% aqueous sodium bicarbonate solution and with water, dried over sodium sulfate and concentrated to dryness, so that the desired product is obtained as a residue.
This product can be purified by crystallization, whereupon practically pure 17 ethylenedioxy-oestra-1, 3, 5 (10), 9 (11) -tetraen-3-ol is obtained. b) 3 g of anhydrous potassium carbonate are added to 1 g of the above product, dissolved in 25 ml of acetone. The mixture is left to stand for 15 minutes, whereupon 1 g of ethyl iodide is added and the mixture is left to stand at room temperature overnight in a closed vessel. The precipitate is removed by filtration and the solvent is evaporated from the filtrate in vacuo. The residue is taken up in a mixture of ethyl acetate and water and the aqueous layer is removed.
The ethyl acetate extract is washed with water, dried over anhydrous sodium sulfate, filtered and the solvent evaporated to leave the desired product. This product can be purified by recrystallization, whereupon practically pure 3-ethoxy-17-ethylenedioxy-oestra-1, 1,3,5 (10), 9 (11) tetraene is obtained.
If one proceeds as described above, but uses methyl iodide or benzyl iodide instead of ethyl iodide, then one obtains the corresponding 3-benzyloxy- or 3-methoxy-steroids. c) 18 g of the above product are dissolved in 50 ml of benzene and cooled to about 100 ° C. 9 g of perbenzoic acid in benzene are now added and this reaction mixture is kept at a temperature of about 80 ° C. for about 6 hours cold water which contains 5% by weight sodium sulfite. The benzene solution containing the product is dried over anhydrous sodium sulfate, filtered and evaporated in vacuo to give the crude 9,11-epoxide.
This product can be cleaned if desired.
It can be dissolved in 50 ml of methylene chloride, for example, and this solution chromatographed on 900 g of synthetic magnesium silicate, whereupon the column is developed with 1000 ml of fractions. Petroleum ether, which contains acetone, is used as eluent, the acetone additions being increased from 2 to 15%. Evaporation of the later fractions gives 3-ethoxy-1 7-ethylenedioxy-9, 11 -epoxy-oestra-1,3,5 (10) -triene.
Proceeding as just described, but using performic acid, peracetic acid or perphthalic acid instead of perbenzoic acid, the corresponding products are obtained. d) To a solution of 10 g of lithium in 1000 ml of dry liquid ammonia is added to a solution of 10 g of 3-ethoxy-l7-ethylenedioxy-9, l 1-epoxy-oestra-1,3,5 (10) -triene in 30 ml of anhydrous diethyl ether and stir this mixture for 30 minutes.
Sufficient n-butanol is added to remove the blue color, which is added within 20 minutes, after which time the ammonia is evaporated and the residue is diluted with water. The residue diluted in this way is extracted with diethyl ether. The ether extract is washed with water, then washed again with a saturated sodium bicarbonate solution, dried over anhydrous sodium sulfate, filtered and evaporated dry, so that a mixture of 11 a- and epimers of 3-ethoxy-17-äthylendioxy-östra-2.5 (1O) -dien-l 1-ol is obtained, the epimers being separated by chromatography.
In accordance with the procedure just described, but using sodium or potassium instead of lithium, the same products are obtained. e) 10 g of 3-ethoxy-17-ethylenedioxy-oestra-2,5 (10) -dien-lla-ol are dissolved in 100 ml of pyridine and this solution is treated with 11 ml of phosphorus oxychloride. This mixture is left to stand for about 18 hours at a temperature of about 250 ° C. and then water is added while cooling with ice. An oil is now formed which crystallizes on standing, which is then filtered off and dried, so that 3 ethoxy-17-ethylenedioxy-oestra-2,5 (10), 9 (11) -triene is obtained.
The same product is obtained from the Ild epimers, just as the same product can be obtained from a mixture of the two epimers. f) To 500 ml of a saturated solution of anhydrous hydrogen chloride in anhydrous ethanol is added a solution of 5 g of 3-ethoxy-l 7-ethylenedioxy-oestra-2.5 (10), 9 (11) -triene in 50 ml of diethyl ether. This reaction mixture is stirred at a temperature of about 250 ° C. for 18 hours. It is now evaporated at a temperature of 0 C and under reduced pressure, so that the estra-4,9-diene-3,17-dione is obtained.
