Procédé de préparation d'acides acéthydroxamiques substitués La présente invention est relative à un procédé de préparation de nouveaux acides arylacéthydroxamiques.

formule dans laquelle R représente un radical alkyle, cycloalkyle, alkyloxy, alkényloxy, alkylthio, cycloalkyl- oxy, cycloalkylalkyloxy, cycloalkylthio, arylalkyloxy ou arylalkylthio, le ou les substituants R' représentent un radical de même nature, identique ou différent, ou de l'hydrogène.
La titulaire a découvert que ces composés ont une activité thérapeutique en médecine humaine et vétéri naire, et en phytopharmacie, notamment une activité antalgique, antipyrétique, antiphlogistique, tranquillisan te et hypocholestérolémiante. La toxicité des composés étudiés s'est révélée faible.
Les composés suivants constituent quelques exem ples de produits actifs répondant à la formule géné rale (1).

<tb> acide <SEP> p-isobutyloxy-m,o-diméthylphényl acéthydroxamique Le procédé selon l'invention est caractérisé en ce qu'on fait réagir un arylacétate d'alkyle, substitué en position para par rapport au groupement carboxy par un radical alkyle, cycloalkyle, alkyloxy, alkényloxy,
alkylthio, cycloalkyloxy, cycloalkylalkyloxy, cycloalkyl- thio, arylalkyloxy ou arylalkylthio, avec de l'hydroxyl- amine pour obtenir l'acide arylacéthydroxamique corres pondant.
L'arylacétate d'alkyle de départ peut être obtenu en traitant par le soufre et une base secondaire, telle que la morpholine, une acétophénone substituée en position para par rapport au groupement cétone par un radical alkyle, cycloalkyle, alkyloxy, alkényloxy, alkylthio, cy- cloalkyloxy, cycloalkylalkyloxy, cycloalkylthio, aryl- alkyloxy ou arylalkylthio, en hydrolisant le complexe résultant de manière à obtenir l'acide aryl-acétique cor respondant et en estérifiant celui-ci.
L'acétophénone substituée peut elle-même être pré parée en partant d'une p-hydroxyacétophénone ou d'un de ses homologues ou analogues, que l'on condense avec un halogénure d'alkyle ou encore en partant de phénol que l'on condense avec un halogénure d'alkyle pour obtenir un alkoxybenzène que l'on traite avec de l'anhydride acétique en présence d'un acide de Lewis pour obtenir l'acétophénone désirée.
L'arylacétate d'alkyle de départ peut encore être obtenu en condensant un halogénure de p-alkyl-, p- cycloalkyl-, p-alkyloxy-, p-alkényloxy, p-alkylthio-, p- cycloalkyloxy-, p-cycloalkylalkyloxy-, p-cycloalkylthio-, arylalkylthio- ou arylalkyloxybenzyle avec un cyanure alcalin, ce qui donne un arylacétonitrile que l'on traite par un acide et un alcool pour obtenir l'ester désiré. <I>Exemple I</I> <I>Préparation de l'acide</I> p-terbutylphénylacéthydroxamique On dissout à part 5 g de Na dans<B>150</B> ml de métha nol absolu et 7 g de NH2HO-HCl dans 100 ml de méthanol absolu. On mélange les deux solutions et on filtre le précipité de NaCl formé.
Au filtrat obtenu, on ajoute du p-tertbutylphénylacétate de méthyle brut et on chauffe à reflux pendant 1 heure. Après refroidisse ment, on acidifie par HCl à 20 % et on recueille le pré cipité d'acide p-tertbutylphénylacéthydroxamique formé. On recristallise dans l'acétone et on recueille le produit blanc d'un point de fusion de 115-1180. Le produit dis sous dans le méthanol donne une coloration rouge vin avec le chlorure ferrique (test des acides hydroxami- ques).

<SEP> 6,76 Le p-tertbutylphénylacétate de méthyle peut être obtenu de la façon suivante A une solution de 15,5 g de NaCN dans 15 cc d'eau, on ajoute en 30 minutes une solution de 45 g de chlo rure de p-tertbutylbenzyle (point d'ébullition (3mm/Hg); 88-89 ; obtenu par chlorométhylation du tertbutyl- benzène) dans 45 cc d'éthanol.
Le mélange ainsi obtenu est chauffé au reflux pen dant 4 heures, puis refroidi ; le précipité inorganique est filtré et lavé avec un peu d'alcool. L'alcool du filtrat est alors distillé, le résidu repris dans l'éther est lavé à l'eau et séché sur du sulfate de magnésium. Après évaporation de l'éther, on distille sous pression réduite et on recueille le p-tertbutylphénylacétonitrile, point d'ébullition (16 mm) : 149-152".
On mélange sous agitation et en refroidissant 20 g (25 ml) de méthanol et 20 g (10,9 ml) d'acide sulfurique concentré. On place ensuite 17,3 g de p-tertbutylphényl- acétonitrile et on chauffe le tout à reflux pendant 10 heu res. On refroidit et on verse le mélange réactionnel dans un mélange eau-glace. On sépare la couche organique, on la sèche sur du MgS04 et on distille le méthanol. On obtient ainsi à l'état brut le p-tertbutylphénylacétate de méthyle qu'on n'isole pas, mais qu'on fait réagir avec l'hydroxylamine comme décrit ci-dessus.

On ajoute une solution de 5 g de sodium dans 150 cm3 de méthanol et on filtre le sel formé. On ajoute 22 g de p-n-butyloxyphénylacétate d'éthyle et on chauffe pendant 1 heure au reflux. Le mélange est refroidi et acidifié avec de l'acide chlorhy drique à 20 0/0. On obtient ainsi 14,7 g d'acide p-n butyloxyphénylacéthydroxamique avec un rendement de 710/0 (P. F. : 153-155).
Le p-n-butyloxyphénylacétate d'éthyle peut être obtenu de la façon suivante On mélange 136 g de p-hydroxyacétophénone, 140 g de bromure de butyle, 152 g de carbonate de potassium, 17 g d'iodure de potassium et 275 cm3 d'éthanol. On chauffe pendant 48 heures au reflux. Le mélange réac tionnel est refroidi, diulé à l'eau, puis extrait à l'éther. La phase éthérée est lavée avec une solution de soude à 10 0/0, puis à l'eau<B>;</B> on sèche, on évapore l'éther et on distille le produit sous pression réduite. On obtient 168 g de p-butyloxyacétophénone avec un rendement de 87 0/0 (160-162 C à 1 mm de Hg).
On mélange 192 g de p-butyloxyacétophénone, 42 g de soufre et 130 g de morpholine. On chauffe pendant 14 heures au reflux. On verse la solution résultante dans l'eau et on agite jusqu'à cristallisation du com plexe soufré. Celui-ci est filtré, lavé à l'eau et séché.
Production<B>:</B> 270 g (rendement de 88 0/0).
On dissout 200 g de soude dans 1500 cms d'éthanol. On ajoute 293 g du complexe soufré précédent. Le mé lange est chauffé au reflux durant une nuit. On distille ensuite le maximum d'alcool, puis on dilue avec de l'eau. La solution résultante est acidifiée par l'acide chlorhydrique, puis extraite à l'éther. La phase éthérée est lavée à l'eau, puis extraite avec une solution de car bonate de sodium à 10 0/0. La solution carbonée est aci difiée par de l'acide chlorhydrique à 50 0/0. Le précipi té résultant est filtré et séché. On obtient 110 g d'acide p-n-butyloxyphénylacétique (rendement de 70 0l0).
On chauffe pendant 5 heures au reflux 208 g d'acide p-n-butyloxyphénylacétique, 368 g d'éthanol et 18 cm3 d'acide sulfurique. On dilue le mélange avec de l'eau, puis on extrait à l'éther. La phase éthérée est lavée successivement à l'eau, au carbonate, puis à l'eau, et elle est ensuite séchée et évaporée. L'ester est distillé cous pression réduite. On obtient ainsi 200 g de p-n- butyloxyphénylacétate d'éthyle avec un rendement de 61 % (186 C sous 8 mm de Hg), que l'on fait réagir comme décrit ci-dessus.
<I>Exemple 4</I> Acide p-cyclopentyloxyphénylacéthydroxamique On traite 134 g de p-cyclopentyloxyphénylacétate d'éthyle par une solution de 27 g de sodium dans 810 cm3 de méthanol en mélange avec une solution de 33 g de chlorhydrate d'hydroxylamine dans 540 cm3 de méthanol. On chauffe pendant une heure au reflux. Après acidification par de l'acide chlorhydrique à 20 0/0, on obtient 87 g d'acide p-cyclopentyloxyphénylacét hydroxamique avec un rendement de 71 % (P. F.: 122-124).
Le p-cyclopentyloxyphénylacétate d'éthyle de départ peut être préparé de la façon suivante On mélange 200 g de bromure de cyclopentyle avec 181 g de p-hydroxyacétophénone, 23 g d'iodure de po tassium et 203 g de carbonate de potassium dans 370 cm3 d'éthanol. On chauffe pendant 48 heures au reflux. Le traitement poursuivi comme dans le cas de l'exemple 3 donne 195 g de p-cyclopentyloxyacétophé- none avec un rendement de 68 0/0.
On chauffe au reflux pendant une nuit 195 g de p- cyclopentyloxyacétophénone, 45 g de soufre et 117 g de morpholine. Le mélange résultant est hydrolysé par 188 g de soude dans 1900 cm3 d'éthanol. Le traitement habituel donne 180 g d'acide p-cyclopentyloxyphényl- acétique (rendement de 85 %).
180 g d'acide p-cyclopentyloxyphénylacétique sont estérifiés par 296 g d'éthanol absolu et 15 cm3 d'acide sulfurique. On obtient 152 g de p-cyclopentyloxyphényl- acétate d'éthyle avec un rendement de 76 % (180 C sous 7 mm de Hg), que l'on fait réagir comme décrit ci-dessus. <I>Exemple 5</I> Acide p-phényléthyloxyphénylacéthydroxamique On traite 43 g de p-phényléthyloxyphénylacétate d'éthyle par une solution de 5 g de sodium dans 150 cm3 de méthanol en mélange à une solution de 7 g de chlor hydrate d'hydroxylamine dans 100 cm3 de méthanol. On chauffe au reflux pendant une heure.
Après acidifica- tion par de l'acide chlorhydrique à 20 0/0, on obtient 26 g d'acide p-phényléthyloxyphénylacéthydroxamique avec un rendement de 62 0/0 (P. F. : 1540-156o).
L'ester de départ peut être obtenu de la façon sui vante On mélange 130 g de 1-bromo-2-phényl éthane avec 95g de p-hydroxyacétophénone, 12 g d'iodure de potas sium et 106 g de carbonate de potassium dans 200 cm3 d'éthanol. On chauffe pendant 48 heures au reflux. Le traitement habituel donne 110 g de p-phényléthyloxy- acétophénone avec un rendement de 65 %.
On chauffe 84 g de p-phényléthyloxyacétophénone, 17 g de soufre et 46 g de morpholine au reflux pendant une nuit. Le mélange résultant est hydrolysé par 66 g de soude dans 400 cm3 d'éthanol. Le traitement habituel donne 70 g d'acide p-phényléthyloxyphénylacétique avec un rendement de 72 %.
On estérifie 120 g de l'acide ainsi obtenu avec 184 g d'éthanol absolu et 9,2 cm3 d'acide sulfurique: On ob tient 101 g de p-phényléthyloxyphénylacétate d'éthyle avec un rendement de 68 % (204 C sous 2 mm de Hg), que l'on fait réagir comme décrit ci-dessus.
En utilisant la même technique de préparation, on a préparé les acides énumérés ci-après.

Acide p-isoamyloxyphénylacéthydroxamique coloration rouge vin avec FeCl3 ; P. F. : 148-1500. Acide p-sec-butyloxyphénylacéthydroxamique coloration rouge vin avec FeCl3 ; P. F. : 86,5-88,51. Acide p-isopropyloxyphénylacéthydroxamique coloration rouge vin avec FeCl3 ; P. F. : 117-1190.
Acide p-n-propyloxyphénylacéthydroxamique coloration rouge vin avec FeCl3 ; P. F. : 158-1600. Acide p-allyloxyphénylacéthydroxamique coloration rouge vin avec FeCl3 ; P. F. : 148-1500. Acide p-n-butyloxy-m-isopropyl-ortho-méthylphényl acéthydroxamique coloration rouge vin avec FeCl3 ; P. F. : 127-128 . Acide p-n-butyloxy-m-méthyl-o-méthylphénylacéthydro oxamique coloration rouge vin avec FeCl3 ; P. F. : 178-180 . Exempe 6: Acide p-n-butyloxy-m-méthylphénylacéthydroxamique Avec une solution de 15,5 g de sodium dans 495 cm3 de méthanol, on mélange 23,1 g de chlorhydrate d'hydroxylamine dans 330 cm3 de méthanol. A la solu tion filtrée, on additionne goutte à goutte, en agitant, 83 g de p-n-butyloxy-m-méthylphénylacétate d'éthyle. L'agitation est poursuivie durant une heure. On verse le mélange sur de la glace, puis on acidifie par du HCl à 20 0/0.
On isole finalement, après traitement à l'éther de pétrole, 52 g d'acide p-n-butyloxy-m-méthylphényl acéthydroxamique. Rendement: 65 % (P. F. : 110-111 ).
Le p-n-butyloxy-m-méthylphénylacétate d'éthyle de départ peut être préparé de la façon suivante On mélange 216 g d'orthocrésol dans 500 cm3 d'étha nol, 140 g de potasse dans 100 cm3 d'eau et 300 g de bromure de butyle. On soumet à reflux pendant 2 heu res, puis on distille l'alcool du mélange réactionnel. Le résidu est traité à l'eau, puis extrait à l'éther ou au tétrachlorure de carbone, on lave au carbonate et à l'eau, puis on sèche la solution. Le produit est distillé. On a 278 g d'ortho-méthylbutyloxybenzène (point d'ébullition<B>:</B> 2220 C/760 mm Hg). Rendement<B>:</B> 85 0/0.
On traite 278 g d'ortho-méthylbutyloxybenzène avec 260 g d'anhydride acétique et 19 g de chlorure de zinc. Le mélange est soumis à reflux pendant une heure en agitant. On dilue à l'eau, puis on extrait à l'éther. La solution éthérée est lavée à l'eau, au carbonate et à l'eau, puis séchée sur du sulfate de magnésium. Le pro- duit est distillé sous vide (150 C/2 mm). On a 254 g de p-n-butyloxy-m-méthylacétophénone. Rendement: 73 %.
On mélange 254g de p-n-butyloxy-m-méthylacéto- phénone, 58 g de soufre et 156 g de morpholine. Le mé lange est soumis au reflux durant une nuit. On ajoute 240 g de soude et 1200 cm3 d'éthanol et on reflue le mélange à nouveau durant une nuit. L'acide est isolé suivant le procédé habituel. On a 218 g d'acide p-n- butyloxy-m-méthylphénylacétique. Rendement: 78 0/0.
A 112g d'acide p-n-butyloxy-m-méthylphénylacéti- que, on mélange 185 g d'éthanol et 9,2 cm3 d'acide sul furique concentré. On chauffe à reflux pendant 2 heu res. On isole par distillation sous vide 83 g de p-n-butyl- oxy-m-méthylphénylacétate d'éthyle (162-163 C/3 mm). Rendement: 66 0/0. On traite cet ester comme décrit précédemment. <I>Exemple 7</I> Acide p-isobutyloxy-m-méthylphénylacéthydroxamique On procède comme à l'exemple 6, en remplaçant le bromure de butyle par du bromure d'isobutyle.
On isole finalement, à partir de 70 g d'ester p-isobu tyloxy-m-méthylphénylacétate d'éthyle, 48 g d'acide p isobutyloxy-m-méthylphénylacéthydroxamique. Rende ment: 72 %. (P. F. : 117,5 -118,5 ).
Aucun de ces acides hydroxamides n'a été décrit jusqu'à présent dans la littérature. Ces acides sont des tinés à être utilisés seuls ou en mélange avec d'autres substances dans la préparation de produits pharmaceu tiques.
On donne au tableau suivant divers résultats phar macologiques obtenus.

The licensee has discovered that these compounds have therapeutic activity in human and veterinary medicine, and in phytopharmaceuticals, in particular analgesic, antipyretic, antiphlogistic, tranquilizer and hypocholesterolemic activity. The toxicity of the compounds studied was found to be low.

The starting alkyl arylacetate can be obtained by treating with sulfur and a secondary base, such as morpholine, an acetophenone substituted in the para position relative to the ketone group by an alkyl, cycloalkyl, alkyloxy, alkenyloxy, alkylthio, cycloalkyloxy, cycloalkylalkyloxy, cycloalkylthio, arylalkyloxy or arylalkylthio, hydrolizing the resulting complex so as to obtain the corresponding arylacetic acid and esterifying it.
The substituted acetophenone can itself be prepared starting from a p-hydroxyacetophenone or one of its homologues or analogues, which is condensed with an alkyl halide or alternatively starting from phenol which is condenses with an alkyl halide to obtain an alkoxybenzene which is treated with acetic anhydride in the presence of a Lewis acid to obtain the desired acetophenone.
The starting alkyl arylacetate can also be obtained by condensing a halide of p-alkyl-, p- cycloalkyl-, p-alkyloxy-, p-alkenyloxy, p-alkylthio-, p- cycloalkyloxy-, p-cycloalkylalkyloxy-. , p-cycloalkylthio-, arylalkylthio- or arylalkyloxybenzyl with an alkali cyanide, which gives an arylacetonitrile which is treated with an acid and an alcohol to obtain the desired ester. <I> Example I </I> <I> Preparation of the acid </I> p-terbutylphenylacethydroxamic acid 5 g of Na are dissolved separately in <B> 150 </B> ml of absolute methanol and 7 g of NH2HO-HCl in 100 ml of absolute methanol. The two solutions are mixed and the NaCl precipitate formed is filtered off.
To the filtrate obtained, crude methyl p-tertbutylphenylacetate is added and the mixture is heated under reflux for 1 hour. After cooling, the mixture is acidified with 20% HCl and the precipitate of p-tertbutylphenylacethydroxamic acid formed is collected. Recrystallized from acetone and the white product with a melting point of 115-1180 is collected. The product dissolved in methanol gives a wine red color with ferric chloride (test for hydroxamic acids).

20 g (25 ml) of methanol and 20 g (10.9 ml) of concentrated sulfuric acid are mixed with stirring and cooling. Then 17.3 g of p-tertbutylphenylacetonitrile are placed and the whole is heated under reflux for 10 hours. Cool and pour the reaction mixture into a water-ice mixture. The organic layer is separated, dried over MgSO4 and the methanol distilled off. The maximum amount of alcohol is then distilled off, then diluted with water. The resulting solution is acidified with hydrochloric acid, then extracted with ether. The ethereal phase is washed with water and then extracted with a 10% sodium carbonate solution. The carbonaceous solution is acidified with 50% hydrochloric acid. The resulting precipitate is filtered off and dried. 110 g of p-n-butyloxyphenylacetic acid (yield 7010) are obtained.
208 g of p-n-butyloxyphenylacetic acid, 368 g of ethanol and 18 cm3 of sulfuric acid are heated for 5 hours at reflux. The mixture is diluted with water, then extracted with ether. The ethereal phase is washed successively with water, with carbonate, then with water, and it is then dried and evaporated. The ester is distilled under reduced pressure. 200 g of ethyl p-n-butyloxyphenylacetate are thus obtained with a yield of 61% (186 ° C. under 8 mm Hg), which is reacted as described above.
<I> Example 4 </I> p-Cyclopentyloxyphenylacethydroxamic acid 134 g of ethyl p-cyclopentyloxyphenylacetate are treated with a solution of 27 g of sodium in 810 cm3 of methanol mixed with a solution of 33 g of hydroxylamine hydrochloride in 540 cm3 of methanol. The mixture is heated for one hour under reflux. After acidification with 20% hydrochloric acid, 87 g of p-cyclopentyloxyphenylacet hydroxamic acid are obtained with a yield of 71% (m.p .: 122-124).
The starting ethyl p-cyclopentyloxyphenylacetate can be prepared as follows 200 g of cyclopentyl bromide are mixed with 181 g of p-hydroxyacetophenone, 23 g of potassium iodide and 203 g of potassium carbonate in 370 cm3 ethanol. Heated for 48 hours at reflux. The treatment continued as in the case of Example 3 gives 195 g of p-cyclopentyloxyacetophenone with a yield of 68%.
195 g of p-cyclopentyloxyacetophenone, 45 g of sulfur and 117 g of morpholine are heated at reflux overnight. The resulting mixture is hydrolyzed with 188 g of sodium hydroxide in 1900 cm3 of ethanol. The usual work-up gives 180 g of p-cyclopentyloxyphenylacetic acid (yield 85%).
180 g of p-cyclopentyloxyphenylacetic acid are esterified with 296 g of absolute ethanol and 15 cm3 of sulfuric acid. 152 g of ethyl p-cyclopentyloxyphenylacetate are obtained with a yield of 76% (180 ° C. under 7 mm Hg), which is reacted as described above. <I> Example 5 </I> p-Phenylethyloxyphenylacethydroxamic acid 43 g of ethyl p-phenylethyloxyphenylacetate are treated with a solution of 5 g of sodium in 150 cm3 of methanol mixed with a solution of 7 g of hydrochloride. hydroxylamine in 100 cm3 of methanol. It is heated under reflux for one hour.
After acidification with 20% hydrochloric acid, 26 g of p-phenylethyloxyphenylacethydroxamic acid are obtained with a yield of 62% (m.p .: 1540-156o).
The starting ester can be obtained as follows: 130 g of 1-bromo-2-phenyl ethane are mixed with 95 g of p-hydroxyacetophenone, 12 g of potassium iodide and 106 g of potassium carbonate in 200 cm3 of ethanol. Heated for 48 hours at reflux. The usual work-up gives 110 g of p-phenylethyloxyacetophenone with a yield of 65%.
84 g of p-phenylethyloxyacetophenone, 17 g of sulfur and 46 g of morpholine are heated at reflux overnight. The resulting mixture is hydrolyzed with 66 g of sodium hydroxide in 400 cm3 of ethanol. The usual work-up gives 70 g of p-phenylethyloxyphenylacetic acid with a yield of 72%.
120 g of the acid thus obtained are esterified with 184 g of absolute ethanol and 9.2 cm3 of sulfuric acid: 101 g of ethyl p-phenylethyloxyphenylacetate are obtained with a yield of 68% (204 C under 2 mm Hg), which is reacted as described above.

The starting ethyl pn-butyloxy-m-methylphenylacetate can be prepared as follows: 216 g of orthocresol are mixed in 500 cm3 of ethanol, 140 g of potassium hydroxide in 100 cm3 of water and 300 g of bromide of butyl. The mixture is refluxed for 2 hours, then the alcohol is distilled from the reaction mixture. The residue is treated with water, then extracted with ether or with carbon tetrachloride, washed with carbonate and with water, then the solution is dried. The product is distilled. We have 278 g of ortho-methylbutyloxybenzene (boiling point <B>: </B> 2220 C / 760 mm Hg). Yield <B>: </B> 85 0/0.
278 g of ortho-methylbutyloxybenzene are treated with 260 g of acetic anhydride and 19 g of zinc chloride. The mixture is refluxed for one hour with stirring. Diluted with water, then extracted with ether. The ethereal solution is washed with water, with carbonate and with water, then dried over magnesium sulfate. The product is distilled under vacuum (150 C / 2 mm). We have 254 g of p-n-butyloxy-m-methylacetophenone. Yield: 73%.
254 g of p-n-butyloxy-m-methylacetophenone, 58 g of sulfur and 156 g of morpholine are mixed. The mixture is refluxed overnight. 240 g of sodium hydroxide and 1200 cm3 of ethanol are added and the mixture is refluxed again overnight. The acid is isolated according to the usual method. There is 218 g of p-n-butyloxy-m-methylphenylacetic acid. Yield: 78%.
185 g of ethanol and 9.2 cm3 of concentrated sulphuric acid are mixed with 112 g of p-n-butyloxy-m-methylphenylacetic acid. The mixture is heated at reflux for 2 hours. 83 g of ethyl p-n-butyl-oxy-m-methylphenylacetate (162-163 C / 3 mm) are isolated by vacuum distillation. Yield: 66%. This ester is treated as described above. <I> Example 7 </I> p-isobutyloxy-m-methylphenylacethydroxamic acid The procedure is as in Example 6, replacing the butyl bromide with isobutyl bromide.
Finally, from 70 g of p-isobutyloxy-m-methylphenylacetate ester, 48 g of p-isobutyloxy-m-methylphenylacethydroxamic acid are isolated. Yield: 72%. (P.F .: 117.5 -118.5).
None of these hydroxamide acids has been described so far in the literature. These acids are intended to be used alone or in admixture with other substances in the preparation of pharmaceutical products.
