Verfahren zur Herstellung von 4, 5-Dialkenyloxy-2-imidazolidonen
Die vorliegelnde Erfindung bezieht sich auf ein Ver- fahren zur Herstellung von 4, 5-Dialkenyloxy-2-imidazolidonen der Formel

worin R ein Alkenylradikal mit 3 bis 6 Kohlenstoffato- men bedeutet.
Eine Reihe von diesen Verbindungen sind neue Substanzen, die noch niemals beschrieben worden sind. Sie sin, d wegen ihrer pharmakologischen Wirksamkeit wert- voH.
Bisher wurden 4, 5-DiaJ!koxy-2-imidazoMotne'durch eine Kondensationsreaktion, von 4, 5-Dihydroxy-2-imid- azolidon mit emem gesättigten aliphatischen Alkohol in einer wässrigen anorganischen oder organischen Säure- losung synthetisiert. Wenn an SteRe des gesättigten a) i- phatischen Alkohols ein ungesÏttigter alipathischer Alkohol verwendet wurde, war die Ausbeute aal 4, 5-Dialkenyloxy-2-imidazolidon ausserordentlich niedrig. Im Gegensatz dazu gestattet das Verfahren der vorliegenden Erfindung die Gewinnung von 4, 5-Dialkenyloxy-2-imid azoMofnsnmhoherAusbeuteumdingEOssarReitiheiit.
Nach dem Verfahren der Erfiadung kam man eiime Reilhe neuer Verbindungen herstellen, die als Medika memte nützlich sind, insbesondere als Beruhtgungsmittel f r das Zentralnervensystem. Diese Verbindungen werden auf einem Weg gewonnen, der technische'und wirt- sohaidicbe Vorteile biatet. Man kann auch mit ihrer Hilfe pharmakologisch wirksame PrÏparate erzeugen, welche als Nervenberuhigungsmittel benutzt werden k¯nnen
Das crfindungsgemässe Verfahren ist dadurch ge- kennzeichnet, dass man 4,
5-Dihydroxy-2-imidazolidon mit emem Alkohol der Formel ROH in Gegenwart oines feston Dehydratationsmittels erhitzt.
Die Reaktion vollzieht sich nach folge-ndem Schema :

Bei dieser Reaktion ist es nicht immer notwendig, ein Lösungsmittel zu benutzen, es ist aber zweckmassig, ein solches zuzufügen, um den Reaktionsverlauf glatt zu gestalten. Ein geeignetes Lösungsmittel st schon das Alkemol am sich, das zu den Ausgangsstoffen des Verfahrens gehört. Gewöhnlich wird em solcher Alkohol m einer Menge von 2-10 Mol, vorzugsweise von etwa 5 Mol, pro Mol 4,5-Dihydroxy-2-imidazolidon angewandt.
Andere geeignete L¯sungsmittel sind verschiedene fl ssige alipathisc'he und aromatische Kohlenwasserstoffe, wie Pentan, Hexan, Heptan, Cyclohexan Benzol, Toluol und dergleichen, Aceton, Chloroform, Dimethylform- amid, Dimethylacetamid usw. Solche organische Lösungs- mittel können, wenn gewünscht, zusammen mit dem vorerwähnten Alkohol Verwendung finden, ohne dass dadurch die Reaktion gest¯rt wird. Die gemeinsame Verwendung ist vorzuziehen, weil dadurch die Isolierung und Abtrennung des Produkts erleichtert wird. SelbstverstÏndlich muss das benutzte L¯sungsmittel wasserfrei sein.
Alkohole, die für die Zwecke des neuen Verfahrens Verwendung finden, sind Alkohole mit 3-6 KoMenstoff- atomen wie Allylalkohol, Crotylalkohol, 3-Buten-l-ol, 4-Penten-l-ol, 3-Penten-l-ol, 4-Penten-2-ol, 4-Hexen- 1-ol, 4-Hexen-2-ol und dergleichen.
Geeignete feste Dehydrienmgsmittel smd z. B. Zinkchlorid, wasserfreies Kupfersulfat, getrocknetes Kupfersulfat (Kupfersulfatmonohydrat), wasserfreies Kalzium- sulfat, wasserfreies Magnesiumsulfat usw. Zinkchlorid ist besonders zu empfehlen. Die Menge des zu verwenden Dehydrierungsmittels, welche benutzt werden soll, hÏngt von seiner EntwÏsserungsfÏhigkeit ab und betrÏgt im allgemeinen 1-2 Mol pro Mol 4, 5-Dihydroxy-2-imid- azolidon.
Die Reaktionstemperatur liegt zweckmÏssig zwischen 0¯ C und dem Siedepunkt des Ausgangsalkohols oder des zugesetzten Lösungsmittels.
Die Dauer der Reaktion kann von der Art des verwendeten Entwässerungsmitbels und des Alkenols ab- hÏngen. Der Fortschritt der Reaktion kann an der Umwandlung einer weissen Suspension des 4,5-Dihydroxy 2-imidazolidon und des Alkenols in eine klare L¯sung erkannt werden.
Die Verbindungen, welche auf dem Wege des er- findungsgemässen Verfahrens erhalten werden, sind im allgemeinen farblose kristalline feste Körper. Sie sind in organischen Lösungsmitteln löslich, wenig in Wasser.
Diese Verbindungen sind als milde, lang anhaltende Beruhigungsmittel wegen ihrer Einwirkung auf das Zentralnervensystem n tzlich. Sie besitzen nur geringe ToxizitÏt. Diejenigen Verbindungen der allgemeinen Formel, in denen R gleich CH2 = CH-CH2-, CH3-CH = CH-(CH2)3- oder CH3-CH = CH-(CH2)2- ist zeigen eine milde Beruhigungswirkung, solche mit R gleich CH3-CH = CH-CH2-, CH2 = CH-(CH2)3- oder CH2 = CH-(CH2)2- weisen neben der Beruhigungswirkung auch eine hypnotische Aktion auf. Die Verbindungen der allgemeinen Formel sind daher als lang wirkende Seda- tive, Beruhiger und Hypnotica, mit geringer ToxizitÏt gegenüber Säugetieren, wertvol.
Beispiel 1 59,1 g (0,5 Mol) 4,5-Dihydroxy-2-imidazolidon, 145,2 g (2,5 Mol) Allylakohol und 136,3 g (1,0 Mol) Zinkchlorid werden in 200 ml Chloroform auf 55-60¯C erwärmt. Nach kurzer Zeit wird das Reaktionsgemisch eine klare, L¯sung. Nach 10 Minuten langem R hren unter Aufrechterhaltung der Temperatur ist die Reaktion beendet. Nach Abkühlung wird die Lösung mit Wasser gewaschen und dann wird das Lösungsmittel unter vermindertem Druck durch Destillation entfernt.
Man erhält 54, 0 g 4, 5-Diallyloxy-2-imidazolidon und die Ausbeute betrÏgt 54,5 % der Theorie. Die Verbindung beginnt bei 55¯ C zu sintern und schmilzt bei 61 C. Die Elementaranalyse ergibt ±ür C9H14O3N2: gefunden : C 54, 26 H 7, 15 N 14, 35 berechnet C 54, 46 H 7, 12 N 14, 13
Beispiel 2
23, 6 g (0, 2 Mol) 4, 5-Dihydroxy-2-imidazolidon werden in einer flüssigen Mischung von 72, 1 g (1, 0 Mol) Crotylalkohol und 30 ml Chloroform suspendiert. Dann werden der Suspension 54, 6 g (0, 4 Mol) ZmkcMorid zu- gesetzt und die Mischung wird im Wasserbad auf 60 bis 65 C erhitzt. Nach kurzer Zeit bildet sich eine tief gr n gefärbte Lösung.
Unter Aufrechterhaltung der Temperatur und 25 Minuten langem Rühren wird die Reaktion vollendet. Man setzt dem Reaktionsgemisdh nun weitere 400 ml Chloroform zu, wäscht mit Wasser und destilliert unter vermindertem Druck, um das Lösungsmittel zu entfernen. Durch Umkristallisieren des Rückstandes aus Aceton werden 33, 4 g 4, 5-Dicrotyloxy-2-imidazolidon erhalten. Die Ausbeute beträgt 74, 0 % der Theorie. Die Verbindung schmilzt bei 117-119 C.

Beispiel 3
23, 6 g (0, 2 Mol) 4, 5-Dihydroxy-2-imidazolidon,
86, 1 g (1, 0 Mol) 4-Penten-l-ol und 54, 6 g (0, 4 Mol) Zinkchlorid in 30 ml Chloroform werden in gleicher Weise zur Reaktion gebracht, wie im Beispiel 2. Nach Aufarbeitung des Gemisches und Umkristallisieren wie im Beispiel 2 erhält man 36, 8 g 4, 5-Di (4'-pentenyloxy)- 2-imidazolidon. Die Ausbeute ist 72, 5 I/o der Theorie.
Die Verbindung hat den Schmelzpunkt 68-70 C.

Beispiel 4
23, 6 g (0, 2 Mal) 4, 5-Dihydroxy-2-imidazolidon, 100 g (1, 0 Mol) 4-Hexen-l-ol und 54, 6 g (0, 4 Mol) Zinkchlorid in 50 ml Chloroform werden in derselben Weise umgesetzt wie im Beispiel 2. Man erhält 36, 6 g 4, 5-Di (4'-hexenyloxy)-2-imidazolidon. Ausbeute 65, 3 % der Theorie. Schmelzpunkt der Verbindung 109-110 C.

23, 6 g (0, 2 Mol) 4, 5-Dihydroxy-2-imidazolidon, 72, 1 g (1, 0 Mol) 3-Buten-l-ol und 54, 6 g (0, 4 Mol) Zinkchlorid in 50 ml Chloroform werden in gleicher Weise umgesetzt wie im Beispiel 2. Man erhält 29, 8 g 4, 5-Di (3'-butenyloxy)-2-imidazolidon. Ausbeute 65, 9 0/'o der Theorie.

Beispiel 6
23,6 g (0,2 Mol) 4,5-Dihydroxy-2-imidazolidon, 86,1 g (1 Mol) 3-Penten-1-ol und 54,6 g (0,4 Mol) Zinkchlorid in 50 ml Chloroform lÏsst man reagieren wie in Beispiel 2. Es werden 34,5 g 4,5-Di(3'-pentenyloxy)-2-imidazolidon mit einem Schmelzpunkt von 136-138¯ erhalten. Ausbeute 67,9 % der Theorie.

Beispiel 7
23. 6 g (0, 2 Mol) 4, 5-Dihydroxy-2-imidazolidon, 72, 1 g (1, 0 Mol) Crotylalkohol und 30 g (0, 188 Mol) wasserfreies Kupfersulfat in 100 ml Chloroform werden während 4-6 Stunden auf 65-70 C erhitzt.
Nach dem Abkühlen setzt man 300 ml Chloroform zu und filtriert das ungelöste Kupfersulfat ab. Das Filtrat wird mit Wasser gewaschen und zur Entfernung des L¯sungsmittels destilliert. Durch Umkristallisieren aus Aoeton ediäll man 29, 8 g 4, 5-Diccrotyloxy-2-imidazol- idon mit einer Ausbeute von 65, 9 % der Theorie.
Beispiel 8
23, 6 g (0, 2 Mol) 4, 5-Dihydroxy-2-imidazolidon, 72, 1 g (1, 0 Mol) Crotylikohol und 20 g (0, 166 Mol) wasserfreies Magnesiumsulfat in 100 ml Chloroform werden 3-5 Stunden auf 65-70¯ C erhitzt.
Nach dem Aufarbeiten und Umkristallisieren wie in Beispiel 7 werden 28, 3 g 4, 5-Dicrotyloxy-2-imidazol- idon mit einer Ausbeute von 62, 6 % der Theorie erhalten.

It is not always necessary to use a solvent in this reaction, but it is useful to add one in order to make the reaction proceed smoothly. A suitable solvent is the alkemol itself, which is one of the starting materials of the process. Usually em such alcohol is used in an amount of 2-10 moles, preferably about 5 moles, per mole of 4,5-dihydroxy-2-imidazolidone.

The reaction temperature is expediently between 0¯ C and the boiling point of the starting alcohol or the added solvent.

These compounds are useful as mild, long-lasting sedatives because of their action on the central nervous system. They have only low toxicity. Those compounds of the general formula in which R is CH2 = CH-CH2-, CH3-CH = CH- (CH2) 3- or CH3-CH = CH- (CH2) 2- show a mild sedative effect, those with R the same CH3-CH = CH-CH2-, CH2 = CH- (CH2) 3- or CH2 = CH- (CH2) 2- have a calming effect and a hypnotic action. The compounds of the general formula are therefore valuable as long-acting sedatives, calmers and hypnotics, with low toxicity to mammals.

23.6 g (0.2 mol) of 4,5-dihydroxy-2-imidazolidone are suspended in a liquid mixture of 72.1 g (1.0 mol) of crotyl alcohol and 30 ml of chloroform. 54.6 g (0.4 mol) of ZmkcMorid are then added to the suspension and the mixture is heated to 60 to 65 ° C. in a water bath. After a short time a deep green colored solution forms.
