Verfahren zur Herstellung von peripher gefässerweiternd wirksamen Salzen des 2-[Pyridyl-(3')]-thiazolin-A2 Die Erfindung beschreibt ein Verfahren zur Herstel lung von Salzen des 2-[Pyridyl-(3')]-thiazolin-A2 der allgemeinen Formel

wobei n = 1 oder 2, R = H oder niedere, geradkettige oder verzweigte Alkylreste mit 1-4 C-Atomen und A = Anionen von anorganischen oder organischen Säuren bedeuten.
Die Herstellung dieser neuen Salze gelingt nach an sich für analoge Verbindungen bekannten Me thoden unter Verwendung von 2-[Pyridyl-(3')]-thi- azolin-A2 als Ausgangsprodukt, das auf verschiedene Weisen zugänglich ist. Eines der möglichen Darstel- lungsverfahren von 2-[Pyridyl-(3')]-thiazolin-A2 ist in der Literatur erwähnt (R. Kuhn und F. Drawert, Lie bigs Ann. Chem. 590, 68 [1954]).
Dagegen sind weder tertiäre noch quartäre Salze von 2-[Pyridyl-(3')]-thi- azolin-A2 bekannt.
Die Umwandlung der Base 2-[Pyridyl-(3')]-thi- azolin-A2 in die entsprechenden Salze geht in der Weise vor sich, dass man die frisch destillierte Base in Abwe senheit oder auch Gegenwart von Wasser mit den ent sprechenden anorganischen oder organischen Säuren umsetzt. Für die salzbildende Reaktion kann man auch niedermolekulare geradkettige oder verzweigte Alkanole, Ketone und andere organische Lösungsmittel benützen oder auch ohne Lösungsmittel arbeiten.
Während 2-[Pyridyl-(3')]-thiazolin-A2 selbst in Was ser sehr schwer löslich ist, gelingt es durch Salzbildung mit anorganischen oder organischen Säuren, sehr gut wasserlösliche Verbindungen zu erhalten.
Durch geeignete Auswahl der Anionen kann man die Löslichkeit der 2-[Pyridyl-(3')]-thiazolin-A2-Salze in einem vorgegebenen Lösungsmittel im gewünschten Sinne verändern. Für wässrige Lösungen der Salze von 2-[Pyridyl-(3')]-thiazolin-A2 erweist sich der Umstand günstig, dass die Hydrolyse von 2-substituierten A2-Thi- azolinen in saurer Lösung gehemmt wird (vgl. die Un tersuchungen von R.
B. Martin u. Mitarbeitern, J. Amer. chem. Soc. 81, 5089 [1959]; 83, 4830 [1961]). Wäh rend sich die UV-Spektren von 2-[Pyridyl-(3')l-thi- azolin-A2 in neutraler und alkalischer Lösung nur wenig unterscheiden, ist in saurer Lösung eine Verände- rung der Lage der Maxima wie auch der Extinktion zu beobachten.
Die quaternären Salze von 2-[Pyridyl-(3')]-thi- azolin-A2 erhält man durch Reaktion der Base mit Alkylierungsmitteln wie Alkylhalogeniden und Dialkyl- sulfaten in geeigneten Lösungsmitteln wie Benzol und Acetonitril oder auch ohne Lösungsmittel bei Zimmer temperatur oder durch Erwärmen. Die quartären Jodide stellen im Gegensatz zu den farblosen tertiären Salzen gelb gefärbte Verbindungen dar.
Die Salze von 2-[Pyridyl-(3')]-thiazolin-A2 zeich nen sich pharmakologisch durch eine starke und sehr lang anhaltende periphere Gefässerweiterungen aus. Bei Durchblutungsmessungen mit dem Bubble-flow-Meter im Gefässgebiet der vena femoralis des Hundes bewirken 20 mg/kg eine Zunahme der Durchblutung um das 2,1- fache. Die Wirkungsdauer beträgt ca. 30 Minuten.
Im Vergleich dazu. haben 0,1 biol. Einheiten Kali krein (ein biologisch gewonnenes Polypeptid, das bisher übliche Therapeutikum auf diesem Gebiet) eine unge fähr gleich starke, aber nur bis zu 5 Minuten anhaltende Wirkung, die mit einer deutlichen Blutdurcksenkung ein hergeht.
Die Salze von 2-[Pyridyl-(3')]-thiazolin-A2 bewir ken dagegen eine periphere Gefässerweiterung ohne Sen kung des mittleren Blutdruckes.
Ein weiterer Vorteil der erfindungsgemässen Verbin dungen besteht in der guten Wirksamkeit peroraler Applikation. Die Salzbildung von 2-[Pyridyl-(3')]-thi- azolin-A2 wird an den folgenden bevorzugte Ausfüh rungsformen zeigenden Beispielen veranschaulicht.

These new salts can be prepared by methods known per se for analogous compounds using 2- [pyridyl- (3 ')] -thiazoline-A2 as the starting product, which is accessible in various ways. One of the possible methods of preparing 2- [pyridyl- (3 ')] -thiazoline-A2 is mentioned in the literature (R. Kuhn and F. Drawert, Lie bigs Ann. Chem. 590, 68 [1954]).
In contrast, neither tertiary nor quaternary salts of 2- [pyridyl- (3 ')] -thiazoline-A2 are known.
The conversion of the base 2- [pyridyl- (3 ')] - thiazoline-A2 into the corresponding salts proceeds in such a way that the freshly distilled base is mixed with the corresponding inorganic in the absence or presence of water or organic acids. For the salt-forming reaction, it is also possible to use low molecular weight straight-chain or branched alkanols, ketones and other organic solvents or to work without a solvent.
While 2- [pyridyl- (3 ')] - thiazoline-A2 is very sparingly soluble even in what water, it is possible to obtain compounds that are very soluble in water by forming salts with inorganic or organic acids.
The solubility of the 2- [pyridyl- (3 ')] -thiazoline-A2 salts in a given solvent can be changed in the desired sense by suitable selection of the anions. For aqueous solutions of the salts of 2- [pyridyl- (3 ')] - thiazoline-A2, the fact that the hydrolysis of 2-substituted A2-thiazolines is inhibited in acidic solution has proven to be favorable (cf. the investigations of R.
B. Martin et al. Coworkers, J. Amer. chem. Soc. 81, 5089 [1959]; 83, 4830 [1961]). While the UV spectra of 2- [pyridyl- (3 ') l-thiazoline-A2 differ only slightly in neutral and alkaline solution, the position of the maxima as well as the extinction change in acidic solution observe.
The quaternary salts of 2- [pyridyl- (3 ')] -thiazoline-A2 are obtained by reacting the base with alkylating agents such as alkyl halides and dialkyl sulfates in suitable solvents such as benzene and acetonitrile or without a solvent at room temperature or by Heat. In contrast to the colorless tertiary salts, the quaternary iodides are yellow-colored compounds.
The salts of 2- [pyridyl- (3 ')] - thiazoline-A2 are pharmacologically distinguished by a strong and very long-lasting peripheral vasodilatation.
