In dem Schweizer Patent Nr. 410 958 ist ein Verfahren zur Herstellung von in 7-Stellung durch Halogen substituierten 10-Azaphenthiazinen der allgemeinen Formel

beschrieben. Hiernach werden Verbindungen der allgemeinen Formel

mit Hilfe von Oxydationsmitteln, z. B. mit Wasserstoffperoxyd, in die entsprechenden Sulfoxyde übergeführt. Diese verwandelt man dann durch Umsetzen mit Chlorwasserstoff in die entsprechenden in der 7-Stellung halogensubstituierten Verbindungen.
Bei der weiteren Ausbildung dieses Verfahrens wurde gefunden, dass man auf diese Art ein Brom in der 7-Stellung nicht nur bei basisch substituierten Azaphenthiazinen einführen kann, sondern auch in die Azaphenthiazinbase selbst. Man kann also als Ausgangssboff anstelle der basisch substituierten Derivate das sekundäre Azaphenthiazin verwenden.
Bei der Herstellung der Sulfoxyde arbeitet man ebenso wie beim Verfahren des erwähnten Patentes zweckmässig in Gegenwart von Lösungsmitteln, wie niederen aliphatischen Alkoholen, z. B. Äthanol oder l-Propanol. Hierbei lässt man die Umsetzung vorteilhaft in Gegenwart solcher Säuremengen stattfinden, dass die basischen Stickstoffgruppen die entsprechenden Additionssalze mit der Säure bilden. Man kann aber auch im wässrigen Medium arbeiten.
Die nach dem Verfahren der Erfindung erhaltenen Stoffe sind brauchbare Anthelmica und können ausserdem als Zwischenprodukte für die Herstellung der in der erwähnten Patentschrift beschriebenen Verbindungen verwendet werden.
Beispiel 1 : a) Herstellung des Sulfoxyds:
1000 g 4-Azaphenthiazin werden in 5 1 Äthylalkohol heiss gelöst und auf etwa 350 abgekühlt. Unter Rühren setzt man innerhalb von 20 Minuten 620 ml Wassersboffsuperoxyd von 30 Gew.-O/o zu.
Nach Stehen über Nacht erhitzt man 2 Stunden am Rückfluss. Der nach dem Abkühlen entstandene Kristallbrei wird abgesaut, mit Methanol ausgewaschen und getrocknet. Man erhält 801 g 4-Azaphenthiazin-9-oxyd vom Fp 236 bis 241. Durch Einengen der Mutterlauge auf etwa 1,5 1 und Abkühlen werden weitere 177 g vom Fp 239 bis 2420 C erhalten. b) Behandlung mit Bromwasserstoffsäure
43,2 g 4-Azaphenthiazin-9-oxyd werden mit 100 ml kalt gesättigter wässriger Bromwasserstoffsäure unter Zusatz von 1 g fein verteiltem rotem Phosphor versetzt und die Mischung auf 1000 erwärmt. Nach dem Stehen über Nacht saugt man das ausgefallene rohe Salz ab und trägt es unter Rühren in Ammoniakwasser ein.
Nach Absaugen, Auswaschen mit Wasser und Trocknen erhält man 51 g eines Rohproduktes, das nach Umkristallisation aus Dimethylformamid 31 g 7-Brom-4-azaphenthiazin vom Fp 207 bis 210 ergibt.
Beispiel 2:
50 Teile 4-Azaphenthiazin werden in 200 Teilen 480/oiger Bromwasserstoffsäure gelöst und bei 50 mit 13,3 Vol.-Teilen Brom, gelöst in 200 Teilen 48 0loiger Bromwasserstoffsäure, versetzt. Dabei spielen sich die Oxydation zum 9-Sulfoxyd und die Anlagerung von Brom in der 7-Stellung im gleichen Reaktionsansatz ab, ohne dass das Sulfoxyd aufgearbeitet werden muss. Nach einstündigem Erwärmen auf dem Wasserbad lässt man erkalten und giesst in 800 Teile 60/oiges Ammoniak-Wasser. Das ausgefallene 7-Brom-4-Azaphenthiazin wird mit Wasser gewaschen und getrocknet. Ausbeute 57 Teile vom Fp 198-200 . Nach Umkristallisation aus Toluol erhält man die Substanz rein. Sie hat einen Fp von 2120.

with the help of oxidizing agents, e.g. B. with hydrogen peroxide, converted into the corresponding sulfoxides. These are then converted into the corresponding compounds substituted by halogen in the 7-position by reaction with hydrogen chloride.
In the further development of this process it was found that a bromine in the 7-position can be introduced in this way not only in the case of base-substituted azaphenthiazines, but also into the azaphenthiazine base itself use.
In the preparation of the sulfoxides, as in the process of the patent mentioned, it is expedient in the presence of solvents such as lower aliphatic alcohols, e.g. B. ethanol or l-propanol. Here, the reaction is advantageously allowed to take place in the presence of such amounts of acid that the basic nitrogen groups form the corresponding addition salts with the acid. But you can also work in an aqueous medium.
The substances obtained by the process of the invention are useful anthelmics and can also be used as intermediates for the preparation of the compounds described in the patent mentioned.
Example 1: a) Production of the sulfoxide:
1000 g of 4-azaphenthiazine are dissolved in 5 liters of hot ethyl alcohol and cooled to about 350. With stirring, 620 ml of hydrogen peroxide of 30% by weight are added over the course of 20 minutes.
After standing overnight, the mixture is refluxed for 2 hours. The crystal sludge formed after cooling is drained, washed out with methanol and dried. 801 g of 4-azaphenthiazine-9-oxide with a melting point of 236 to 241 are obtained. 43.2 g of 4-azaphenthiazine-9-oxide are mixed with 100 ml of cold-saturated aqueous hydrobromic acid with the addition of 1 g of finely divided red phosphorus and the mixture is heated to 1000. After standing overnight, the precipitated crude salt is suctioned off and it is introduced into ammonia water with stirring.
After filtering off with suction, washing with water and drying, 51 g of a crude product are obtained which, after recrystallization from dimethylformamide, give 31 g of 7-bromo-4-azaphenthiazine of melting point 207 to 210.
Example 2:
50 parts of 4-azaphenthiazine are dissolved in 200 parts of 480% hydrobromic acid and, at 50, 13.3 parts by volume of bromine dissolved in 200 parts of 48% hydrobromic acid are added. The oxidation to the 9-sulfoxide and the addition of bromine in the 7-position take place in the same reaction mixture without the sulfoxide having to be worked up. After one hour of warming on the water bath, it is allowed to cool and poured into 800 parts of 60% ammonia water. The precipitated 7-bromo-4-azaphenthiazine is washed with water and dried.
