Verfahren zur Herstellung von 18-Jod-20-hydrogy-steroiden Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von 18-Jod-ste.roiden, wel che in 20-Stellung eine freie Hydroxygruppe aufwei sen.
18-Halogensteroide sind bisher nicht bekannt ge^ wegen, da eine direkte Halogenierung der Methyl- gruppe am quaternären Kohlenstoffatom 13 nicht durchgeführt werden kann.
Da sich ein Halogenatom in 18-Stellung, insbesondere unter Mitwirkung von Sauerstoffunktionen in 11ss- oder 20-Stellung leicht in eine 18-Sauerstoffunktion umwandeln lässt, be- sitzen 18-Halogensteroide als Ausgangsstoffe für die Herstellung 18-oxygenierter Steroide, insbesondere von solchen vom Typus des hochwirksamen, natür lichen Mineralcorticoids Aldosteron grosse Bedeu tung.
Bei dem im Schweizer Patent Nr. 407 111 be schriebenen Verfahren zur Herstellung von 18-Hy- droxy-18,20-oxido-pregnan-Verbindungen entstehen zwar als Zwischenprodukte auch äusserst labile 18 Jod-18,20-oxido-pregnane, welche alsJodwasserstoff- säurees.ter von Cyclohemiacetalen sehr leicht in die freien Cycloheniiacetale (18-Hydroxy-18,20-oxido- verbindungen) und Jodwasserstoffsäure zerfallen. Das im genannten Patent beschriebene Verfahren besteht darin,
dass man auf 20-Hydroxy-verbindungen der Pregnanreihe oxydierend wirkende Bleiacylate in Ge genwart von Jod einwirken lässt.
Es wurde nun überraschenderweise gefunden, dass die Hauptprodukte der Reaktion je nach der verwendeten Menge Jod verschieden sind. Reduziert man z.
B. in einer Versuchsreihe die Jodmenge von 2 Mol pro Mol 20-Hydroxyverbindung nach und nach immer weiter, so erhält man zunächst als Haupt produkt eine instabile 18-Jod-18,20-oxido-verbin- dung, dann zunehmende Mengen der relativ stabilen 18-Jod-20-hydroxy-verbindung, welche bei Mengen unter 1 Mol Jod zum Hauptprodukt werden, deren Ausbeute aber bei Verwendung von weniger als 0,
5 Mol Jod wieder abnimmt. Es entstehen dann schliess- lich in einer langsamen Reaktion die jodfreien 18,20- Äther; diese sind das Hauptprodukt, wenn die Blei- tetraacetat-Oxydation ohne Jod durchgeführt wird.
Das neue Verfahren der vorliegenden Erfindung besteht darin, dass man eine 18-unsubstituierte 20 Hydroxy-verbindung der Pregnanreihe oder 19-Nor- pregnanreihe mit einem oxydierend wirkenden Blei- acylat in Gegenwart von. weniger als einem Moläqui- valent Jod (bezogen auf die 20-Hydroxy-verbindung) umsetzt.
Für .die verfahrensgemässe Umsetzung verwendet man insbesondere Acylate des 4-wertigen Bleis, wie z. B. Dialkylbleidiacylate, besonders aber Bleitetra- acylate, wie Bleitetraacetat, Bleitetrapropionat, Blei tetrachloracetat usw., und setzt z.
B. 0,5-1 Moläqui- valente Jod. zu, bezogen auf die 20-Hydroxyverbin- dung. Da bei der Reaktionstemperatur das Jod mit dem Bleitetraacylat ebenfalls reagiert, verwendet man vorteilhaft mehr als die theoretisch notwendigeMenge von 0,5 Moläquivalenten Jod, jedoch nur soviel,
dass die überschüssige Jodmenge während der Reak tionszeit durch das Bleitetraacylat verbraucht wird. Diese Menge kann, falls nötig, in einem Blindversuch leicht festgestellt werden. Man verwendet vorteilhaft das Bleitetraacylat im Überschuss (d. h. ca. 2-5 Mol- äquivalente), da sobald kein Jod mehr vorhanden ist, weitere Reaktionen nur langsam ablaufen.
Man kann die Reaktion in einem, gegenüber denOxydationsmit- teln inerten Lösungsmittel durchführen, z. B. in einem gesättigten cyclischen Kohlenwasserstoff, wie Cyclo- hexen, Me#thylcyclohexan, Dimethylcyclohexan, doch können. auch aromatische Kohlenwasserstoffe, be- sonders Benzol,
verwendet werden. Man arbeitet vor- teilhaft bei Temperaturen über 50 , bei Normal druck oder bei erhöhtem Druck, wobei die Reak- tionsdauer von der Temperatur und der Art des ver wendeten Lösungsmittels abhängt. Beim Arbeiten in siedendem Cyclohexan und unter intensiver Belich tung ist die Reaktion in der Regel nach 20-40 Mi nuten beendet,
was leicht durch das vollständige Ver schwinden der Jodfarbe festgestellt werden kann. Zur Neutralisierung vorhandener oder sich bildender Säure kann gegebenenfalls eine schwache Base, z. B. Calcium-, Strontium- oder Bleicarbonat, zugesetzt werden.
Die Reaktion wird vorteilhaft durch Belich- tung eingeleitet und beschleunigt, wobei sich die Ben- strahlung mit Licht von der Wellenlänge der sichtba ren Jod Absorptionsbande besonders günstig aus wirkt.
Zur Aufarbeitung können aus der Reaktionsmi- schung die anorganischen Salze durch Filtration ent fernt werden.
Die erhaltenen 18-Jod-20-Hydroxy-verbindungen der Pregnan-Reihe können anschliessend mit hydro- lysierenden, insbesondere alkalischen Mitteln umge- setzt werden, wobei man 18,20-Oxido-verbindungen erhält. Für diese Umwandlung eignen sich sowohl anorganische, als auch organische Basen,
wie Hy- droxyde von Alkali- und Erdalkalimetallen, z. B. Natrium-, Kalium- oder Bariumhydroxyd, aber auch Silberoxyd, Alkalimetallcanbonaten, wie Natrium- oder Kaliumcarbonat, Triäthylamin, Methylpiperidin, Collidin, Pyridin usw.
Sogar Metallsalze von Carbon- säuren, wie Kaliumacetat, Natriumpropionat, Silber- acetet und dgl. lassen. sich zur Bildung der 18,20- Oxidoverbindungen verwenden. Man kann dabei mit wasserhaltigen oder wasserfreien Lösungsmitteln ar beiten, wie Alkoholen, z.
B. Methanol, Äthanol, Pro- panol oder Äthern, z. B. Tetrahydrofuran, Dioxan, Glykoldimethyläther, etc. oder in Pyridin, Dimethyl- formamid usw.
Anderseits lassen sich die verfahrensgemäss er haltenen 18-Jod-20-Hydroxy-verbindungen, insbeson- dere in saurer Lösung, zu 18-Jod-20-oxo-verbindun- gen oxydieren. Besonders geeignete Oxydationsmit- tel sind Verbindungen des 6-wertigen Chroms, z. B. Chromtrioxyd in Eisessig oder auch in Aceton unter Zusatz von Schwefelsäure.
In den so erhaltenen 18-Jod-20-oxo-verbindun- gen lässt sich das 18 Jodatom wiederum durch<B>Hy-</B> drolyse durch eine Sauerstoffunktion ersetzen. Man erhält so 20-Hydroxy-18,20-oxido@verbindungen, d.h. Hemiketale der 18-Hydroxy-20-oxo-verbindungen. Die Hydrolyse lässt sich, wie oben für die Umwand lung der 18-Jod-20-hydroxy-verbindungen zu 18,20- Äther beschrieben,
durchführen. Verwendet man als Lösungsmittel einen Alkohol, so erhält man sehr leicht die 20-Alkoxy-18,20-oxido-Verbindungen, wel che sich durch saure Hydrolyse, z. B. mit wässriger Mineralsäure in einem nicht alkoholischen Medium (z. B. in Dioxan) zu 20-Hydroxy-18,20-oxido@ver- bindungen spalten lassen.
Als Ausgangsstoffe für das vorliegende Verfah ren eignen sich sowohl 20a- wie auch 20ss-Hydroxy- verbindungen der 5a- und 5ss-Pregnanreihe und der 19-Nor-pregnanreihe, die im Ringsystem, insbeson dere in einer oder mehreren der Stellungen 1, 2, 3, 4, 5, 6, 7, 8, 9, 10, 11, 12, 14, 15, 16, 17, 21 weitere Substituenten aufweisen können,
wie funktionell ab gewandelte Hydroxygruppen oder freie oder funktio@ nell abgewandelte Oxogruppen, Alkyl- wie Methyl- gruppen, oder Halogenatome. Als funktionell abge wandelte Hydroxy- oder Oxogruppen kommen ver- esterte oder verätherte Hydroxygruppen bzw.
ketali- sierte Oxogruppen in Frage. Ausserdem können die Ausgangsstoffe auch Doppelbindungen oder Oxido- gruppen aufweisen, z. B. ausgehend vom Kohten- stoffatom 5 und/oder in 9,11-Stellung. Spezifische Ausgangsstoffe sind z.

ferner die durch Umsetzung der entsprechenden 20-Oxopre" gnan-Verbindungen mit einer Methylmetallverbin- dung erhaltenen 20-Methyl-pregnane. Die genannten Ausgangsstoffe werden vorteilhaft in Form ihrer 3- Mono- bzw. 3,11-Diester bzw. A5-3-Ketale angewen det.
überraschenderweise werden unter den verfah- rensgemässen Reaktionsbedingungen, im Gegensatz zur Behandlung mit Bleitetraacetat ohne Jod, Methy- lengruppen neben Ketonen nicht oder fast nicht an gegriffen, so dass auch Ausgangsstoffe mit freien RTI ID="0002.0231" WI="21" HE="4" LX="1152" LY="1820"> Ketogruppen, z.
B. 3-Ketone, 44-3-Ketone, 11-Ke- tone usw. verwendet werden können. Auch 41,4-3- Ketone bind unter den verfahrensgemässen Reak- tionsbedingungen weitgehend stabil.
Die Ausgangsstoffe werden z. B. durch Reduk tion der entsprechenden 20-Ketone mit Wasserstoff und Edelmetallkatalysatoren z.
B. Platin oder mit komplexen Metallhydriden, wie Natriumborhydrid, Lithium-tri-tertiär-butoxy-aluminium-hydrid ete. ge- wonnen. Ein besonders vorteilhaftes Verfahren für die Herstellung der Ausgangsstoffe mit einer d4-3- Oxo- oder A5-3-Ketalgruppierung ist im Schweizer Patent Nr. 394 170 beschrieben.
Auch die 19-Nor-20-hydroxy-verbindungem kön nen aus den entsprechenden 19-Nor-pregnan-20-ke- tonen hergestellt werden, insbesondere aus 19-Nor- progesteron und aus dem durch mikrobiologische 1 l a-Hydroxylierung daraus erhaltenen l 1 a-Hydroxy- 19-nor-pro@gesteron. Aus dieser Verbindung kann man gemäss dem Verfahren des
Schweizer Patents Nr. 394 170 die für das erfindungsgemässe Verfah- ren vorzugsweise verwendeten 20-Hydro@xy-19-nor- pregnan-Verbindungen gewinnen.
Die erhaltenen 18-Jod-20-hydroxy- und 18-Jod- 20-oxo-Verbindungen und ihre funktionellen Deri vate können entsprechend den oben genannten Aus- gangsstoffen im Ringsystem weitere Subs.tituenten und/oder Doppelbindungen aufweisen. Von diesen gesättigten und ungesättigten 18-Jod-20-hydroxy-und 18-Jod-20-oxo-Verbindungen und ihren funktionellen Derivaten sind
insbesondere solche Verbindungen, die in 17- und/oder 21-Stellung unsubstituiert sind oder freie, veresterte oder verätherte Hydroxylgrup- pen aufweisen, von Bedeutung.
In den Estern kommen als Säurereste in, Frage solche von niederen, aliphatischen Mono- oder Di- carbonsäuren mit 1-8 Kohlenstoffatomen, wie der Essigsäure, Propionsäure, Buttersäure, Trimethyl- essigsäure, Bernstems:
äure, Glutarsäure. Glycol- und Digtycolsäure., Trifluoressigsäure, oder einer aroma tischen Carbons.äure, vorzugsweise einer monocycli- schen Säure, wie der Benzoesäure, einer monocycli- schen cycloaliphatischen oder araliphatischen Säure, wie der Hexahydrophthalsäure, Tetrahydro#phthal- säure, Cyclohexansäure,
Zimtsäure, Phenylpropion- säure, einer aliphatischen oder aromatischen hetero,- cyclischen Säure, wie der Furancarbonsäure, Nico- tinsäure oder auch von Sulfonsäure, wie der Methan- sulfonsäure, Benzolsulfonsäure, p-Toluolsulfonsäure usw.
In den Äthern ist der Alkoholrest vorzugsweise ein niederer aliphatischer Alkoholrest, wie der des Methyl-, Äthyl-, Propyl-, Butyl-, Isopropyl-, Isobu- tylalkohols, oder eines araliphatischen Alkohols, wie eines monocyclischen niederen araliphatischen Alko hols, wie des Benzylalkohols,
oder eines heterocycli- schen Alkohols, wie Tetrahydropyranols usw. Für die Ketalisierung oder Acetalisierung von Oxogrup- pen können insbesondere niedere zweiwertige alipha- tische Alkohole, wie Äthylenglycol, Propylenglycol usw. verwendet werden.

Andere im Ring A ungesättigte Verbindungen lassen sich in analoger Weise umwandeln und nach Einführung einer 44-3-Oxa-gruppierung und gegeben nenfalls nach Hydroxylierung in 11-Stellung und Oxydation ebenfalls in Aldosteron überführen.
Aus den in 11-Stellung ursubstituierten Verfah rensprodukten lässt sich wie oben angegeben auch das bekannte 18 Hydroxy-progesteran herstellen. Die 11 a-hydroxylierten Verfahrensprodukte stellen auch bekannte Zwischenproduktes zur Herstellung von 9a- Fluor-18-hydroxy-corticosteron und 9a-Fluoraldcste- ron dar.
Die Temperaturen sind in den nachfolgenden Bei spielen in Celsiusgraden angegeben.
<I>Beispiel 1</I> Eine gerührte Suspension von 5,0g trockenem Calciumcarbonat und 15,0g vorgetrocknetem Blei- (IV)-acetat in 500 ml Cyclohexan wird 1 Stunde am Rückfluss gekocht, anschliessend mit 1,5 g Jod und 5,0g 3ss,lla-Diacetoxy-20ss hydroxy-5a-pregnan ver setzt und bis zur Entfärbung (18-30 Min.) unter Rühren weitergekocht.
Das auf Raumtemperatur ab- gekühlte Reaktionsgemisch wird nun durch Celit fil- triert, der Rückstand mit Cyclohexan nachgewaschen und die vereinigten. Filtrate mit einer Lösung von 25 g Natriumthiosulfat in. 100 ml Wasser, und drei mal mit Wasser gewaschen, getrocknet und einge dampft.
Die erhaltenen 6,55 g rohen 3ss,lla-Diacet- oxy-20-hydroxy-18 jodo-5a-pregnans löst man an- schliessend in 100 ml Aceton und versetzt unter Rüh ren bei 0 mit 5,9 ml einer Lösung von Chrom-(VI)- oxyd in Schwefelsäure [13,26 g Chrom-(VI)-oxyd in 20 ml Wasser gelöst, mit 11,5 ml konz. Schwefelsäure verdünnt und auf 50 ml mit Wasser aufgefüllt].
Nach 30 Min. bei gleicher Temperatur wird eine Lösung von 56 g krist. Natriumacetat in 100 ml Wasser zu gegeben und das Reaktionsgemisch mit Benzol extra hiert.
Die 3mal mit halbgesättigter Natriumchlorid- Lösung gewaschene organische Schicht liefert nach Trocknen und Eindampfen des Lösungsmittels im Vakuum 5,85 g eines öligen Produktes, das nach Umlösen aus Äther-Petroläther 4,117 g rohes 3ss,11 a Diacetoxy-18-jodo-20-oxo-5a-pregnan vom F.<B>118-</B> 121' (Zerr.) liefert.
<I>Beispiel 2</I> Eines Lösung von 4,20g 3ss,1 la-Dia.cetoxy-18- jodo-20-oxoo-5a-pregnan (amorphes Rohprodukt aus 3,30 g 3ss,lla Diacetoxy-20ss-hydroxy-5a-pregnan) in 400 ml Methanol wird nach Zugabe von 4,0g Silber acetat 3 Stunden am Rückfluss gekocht.
Das ausge fallene Silberjodid (Trockengewicht 1,210 g) wird zu sammen mit dem überschüssigen Silberacetat abfil- triert, das Filtrat im Vakuum eingedampft, der Rück stand in Äther aufgenommen, erneut filtriert und die Lösung im Vakuum eingedampft. Das so erhaltene Produkt wird direkt in 80 ml Eisessig gelöst und nach Zugabe von 16 ml Wasser 2 Stunden auf 85 erwärmt. Das Reaktionsgemisch dampft man dann im Vakuum auf ca. 10 ml ein, verdünnt den Rück stand mit Wasser und extrahiert mit Äther.
Die neu tral gewaschene und getrocknete Lösung liefert 3,30 g eines amorphes Rohproduktes., das in Benzol gelöst an neutralem Aluminiumoxyd (Akt. 11) chromatogra- phiert wird.
Mit Benzol - Essigester - (1:1) - gemische.n eluiert man 1,02 g 3ss,lla-Diacetoxy-18-hydroxy-20-oxo- 5a-pregnan, das in Form eines 18-20-Hemiketals vorliegt und nach zweimaligem Umkristallisieren das Methylenchlorid-Äther-Petroläther bei 160 bis<B>1611</B> schmilzt. [a]D = + 18,2 1 (c = 1,098 in Chloro- form). Im IR.-Spektrum der Verbindung treten u. a.
Absorptionsbanden bei 2,82,u, 5,82,u, 8,10 ,u, 9,73,u, 10,41 Ni und 11,23 ,u auf.
<I>Beispiel 3</I> Zu einer 1 Stunde unter Rühren gekochten Sus pension von 500 mg Calciumcarbonat und 1,50 g Blei-(IV)-acetat in 75 ml Cyclohexan werden 180 mg Jod und 500 mg 3-Äthylen-dioxy-20ss-hydroxy-ds- pregrien zugegeben. Nach weiteren 30 Min. bei 80 wird das Reaktionsgemisch auf Raumtemperatur ab gekühlt, durch Ce:lit filtriert, der Rückstand mit 120 ml Cyclohexan nachgewaschen und das Filtrat im Beispiel 1 beschrieben mit Natriumthiosulfat und Wasser gewaschen und im Vakuum eingedampft.
Die erhaltenen 610 mg rohes 3-Äthylendioxyd-18-jodo,- 20ss-hydroxy-d5-pregnen oxydiert man unter analo gen Bedingungen wie im Beispiel 1 beschrieben mit Chrom-(V1)-oxyd in Schwefelsäure, wobei man das 3-Äthylendioxy-18-jodo-20-oxo-A5-pregnen erhält. Dieses wird ohne Reinigung in 60m1 Methanol ge löst und mit 600 mg Silberacetat 4 Stunden am Rückfluss gekocht.
Nach Abfiltrieren von 198 mg Silberjodid dampft man die methanolische Lösung im Vakuum ein, nimmt den Rückstand in Äther auf, filtriert und dampft die Lösung erneut im Vakuum ein. Das kristalline Rohprodukt (450 mg) wird in 10 ml Eisessig gelöst, mit 2 ml Wasser versetzt und 1 Stunde auf 80-90 erwärmt.
Nach Zugabe von weiteren 3 ml Wasser wird die Reaktionsmischung im Vakuum bis auf ca. 3 ml eingeengt, erneut mit Wasser verdünnt und anschliessend mit Äther extra- hiert. Die nacheinander mit Wasser, Natriumhydro- gencarbonat-Lösung und Wasser gewaschene und mit Natriumsulfat getrocknete Lösung liefert nach Ein dampfen im Vakuum 365 mg eines gelblichen Schau mes.
Durch chromatographische Reinigung an neu tralem Aluminiumoxyd liefert dieser 82 mg kristal lines 18-Hydroxy-progesteron, das nach 2-maligem Umkristallisieren aus Methylenchlorid-Äther bei 154 bis 156 schmilzt. Das IR.-Spektrum des Präparates ist mit demjenigen des authentischen 18-Hydroxy-pro- gesterons vollkommen identisch.
<I>Beispiel 4</I> Analog zu den Beispielen 1, 2 und 3 wird auch das aus 3ss-Acetoxy-20-oxo-ds-pregnen durch Re= duktion mit Lithium-tritertiärbutoxy-aluminiumhydrid zugängliche 3ss-Acetoxy-20-hydroxy-dl-pregnen (5,0 g) durch nacheinanderfolgende Behandlung mit Blei- (IV)-acetat und Jod, Chrom-(VI)-oxyd in Schwefel säure, Silberacetat in Methanol und verdünnte Essig säure in die 18,
20-Cyclo-hemiketalForm des 3ss- Acetoxy-18-hydroxy-20-oxo-ds-pregnens übergeführt. Dieses schmilzt nach 3-maligem Umkristallisieren aus Äther-Hexan bei 158-161 .
Im IR-Spektrum der Verbindung treten u. a. Absorptionsbanden bei 2,80,u, <I>5,81</I> ,u, 8,10,u, 9,00,u, 9,66 ,u und:
11,25,u. <I>Beispiel 5</I> 780 mg Jod und 2,5 g 3-Äthylendioxy-11a-acet- oxy-20ss-hydroxy-d5-pregne#n werden zu einer, 45 Min. vorgekochten Suspension von 2,5 g Calcium- carbonat und 8,0g im Hochvakuum getrocknetem Blei-(IV)
-acetat in 250m1 Cyclohexan zugegeben und das Gemisch anschliessend bis zur Entfärbung bei 80 gerührt. Die auf 20 abgekühlte Mischung wird durch Celit filtriert und das Filtrat wie im Beispiel 1 angegeben mit Natriumthiosulfat-Lösung und Wasser gewaschen und eingedampft.
Das so er haltene 3-Äthylendioxy-lla-acetoxy-18-jodo-20-hy- droxy-d5-pregnen schmilzt nach 2-maligem Umkri- stallisieren aus Isopropanol unter Zersetzung bei <B>153'.</B>

The precipitated silver iodide (dry weight 1.210 g) is filtered off together with the excess silver acetate, the filtrate evaporated in vacuo, the residue taken up in ether, filtered again and the solution evaporated in vacuo. The product thus obtained is dissolved directly in 80 ml of glacial acetic acid and, after addition of 16 ml of water, heated to 85 for 2 hours. <I> Example 3 </I> To a suspension of 500 mg calcium carbonate and 1.50 g lead (IV) acetate in 75 ml cyclohexane, cooked for 1 hour with stirring, are added 180 mg iodine and 500 mg 3-ethylene-dioxy -20ss-hydroxy-ds- pregrien added. Chromatographic purification on neutral aluminum oxide gives 82 mg of crystalline lines 18-hydroxy-progesterone, which, after recrystallizing twice from methylene chloride-ether, melts at 154 to 156.
