worin R eine niedere Alkylgruppe bedeutet, zur entsprechenden Phenanthren-2-carbonsäure oxydiert, diese verestert und den Ester mit einem komplexen Metallhydrid zum primären Alkohol reduziert.
Erhaltene Verbindungen der Formel I mit den Atomgruppierungen A1 oder A2 lassen sich in entsprechende Verbindungen der Formel I mit den Atomgruppierungen A2 bzw. As überführen. So kann eine Verbindung der Formel I mit der Atomgruppierung Ar durch Behandlung mit Lithium in flüssigem Ammoniak und anschliessende Hydrolyse des Reduktionsproduktes in eine Verbindung mit der Atomgruppierung A2 umgewandelt werden.
Eine Verbindung der Formel I mit der Atomgruppierung A2 kann z. B. mit Lithium in flüssigem Ammoniak zu einer Verbindung mit der Atomgruppierung A3 reduziert werden.
Zur Herstellung einer Verbindung der Formel III

geht man zweckmässig von 2-Acetyl-7-methoxy- 1,2,3 4,4a,9, 10,1 Oa-octahydro-phenanthren aus und oxydiert dieses zur 7-Methoxy-1, 2, 3, 4, 4a, 9, 10, Oa-octa- hydro-phenanthren-2-carbonsäure. Geeignete Oxydationsmittel sind z. B. Natriumhypobromit, Kaliumhypochlorit, Alkalimetallpermanganat und Chromsäure. Hierauf wird die erhaltene Phenanthren-2-carbonsäure verestert, z. B. mit Diazomethan oder einem niedrigen Alkanol. Die Estergruppe wird dann, wie gesagt, mit einem komplexen Metallhydrid zur -CH2OH Gruppe reduziert, zweckmässig mit Lithium-Aluminiumhydrid oder Natrium-Borhydrid.

wird vorteilhaft aus der entsprechenden Verbindung der Formel IV durch selektive Hydrierung der Ringdoppelbindung hergestellt, z. B. mit Lithium und Ammoniumchlorid in flüssigem Ammoniak.
Die nach dem erfindungsgemässen Verfahren erhältlichen Polyhydrophenanthrenverbindungen enthalten asymmetrische Kohlenstoffatome und treten demgemäss in verschiedenen stereoisomeren Formen auf.
Die Verfahrensprodukte wirken cholesterinspiegelsenkend und/oder hormonal bzw. antihormonal.
Sie können als Heilmittel mit cholesterinspiegelsenkender, antigonadotroper, antiandrogener, östrogener oder anabolischer Wirkung in Form pharmazeutischer Präparate verwendet werden, welche sie in Mischung mit einem für die enterale oder parenterale Applikation geeigneten pharmazeutischen, organischen oder anorganischen inerten Trägermaterial enthalten. Die pharmazeutischen Präparate können in fester oder in flüssiger Form, z. B. als Lösungen, Suspensionen oder Emulsionen, vorliegen. Sie können auch noch andere therapeutisch wertvolle Stoffe enthalten.
Beispiel 1
Ein Gemisch von 35,74 g 2-Acetyl-7-methoxy-l,2, 3, 4, 9, 10-hexahydroWphenanthren, 17 ml Äthylenglykol und 0,25 g p-Toluol-sulfonsäuremonohydrat in 240 ml Benzol wird 5 Stunden unter Abscheidung des Wassers zum Rückfluss erhitzt. Die Lösung wird zweimal mit 50 ml Natriumcarbonat und zweimal mit 50 ml Wasser gewaschen. Nach Trocknen mit Natriumsulfat wird das Lösungsmittel im Vakuum abgedampft.
Das im Rückstand verbleibende Ö1, das cyclische Äthylenketal von 2-Acetyl-7-methoxy-l ,2,3,4,9,10- hexahydro-phenanthren konnte nicht kristallisiert werden.
Eine Lösung von 30,1 g des erhaltenen Äthylenketals in 300 ml wasserfreiem Äther wird unter Rühren zu einer Lösung von 13,8 g Kalium in 900 ml flüssigem Ammoniak bei 400 zugefügt. Man rührt die Lösung 45 Minuten, fügt dann 23 g Ammoniumchlorid zu und lässt das Ammoniak verdampfen. Der Rückstand wird mit Eis, Wasser und 200 ml Ather behandelt. Die Ätherschicht wird abgetrennt und mit Wasser gewaschen. Die Wasserschichten werden mit 100 ml Äther gewaschen. Die Ätherauszüge werden vereinigt, mit Natriumsulfat getrocknet und dann wird das Lösungsmittel im Vakuum entfernt.
Man löst den Rückstand in 300 ml Methanol, fügt 7,5 g Oxalsäure und 75 ml Wasser zu und hält die Mischung eine Stunde unter Rückfluss. Das Me thanol wird im Vakuum abgedampft, der Rückstand mit 100 ml Wasser verdünnt und mit 300 ml Äther extrahiert. Die organische Schicht wird abgetrennt und mit Wasser gewaschen. Die wässrigen Schichten werden mit 100 ml Äther gewaschen. Man trocknet die vereinigten Ätherauszüge mit Natriumsulfat und entfernt das Lösungsmittel im Vakuum. Man erhält so 2-Acetyl-7-methoxy-l,2,3,4,4a,9, 10, I0a--octahydro- phenanthren (Ringe B/C trans); Schmelzpunkt 83-8 5 0.
78 g dieses Octahydrophenanthrens (gelöst in
1500 ml Dioxan) werden unter Rühren zu einer kalten Lösung von Natriumhypobromit (hergestellt aus
109 g Natriumhydroxyd, 160 g Brom und 1500 ml Wasser) so zugefügt, dass die Temperatur nicht über
100 steigt. Hierauf wird die Lösung noch eine Stunde bei Raumtemperatur gerührt. Die Lösung wird dann im Vakuum auf ungefähr l/4 eingeengt und dann mit 3 Liter Wasser verdünnt. Diese wässrige Lösung wird zweimal mit Äther extrahiert, auf 0 abgekühlt und dann mit 600 ml 3 n Salzsäure auf kongorot angesäuert. Der sich bildende Niederschlag wird abfiltriert, mit Wasser gewaschen und dann getrocknet.
Das erhaltene Produkt (78,2 g) wird in einem Liter warmem Chloroform gelöst, durch Cellit filtriert und das Filtrat unter Zugabe von Aceton auf ungefähr 250 ml eingeengt. Man erhält so 65,8 g 7-Me thoxy- 1,2,3 ,4,4a, 9,101,1 0a - octahydro-phenanthren-2- carbonsäure (Ringe B/C trans), Schmelzpunkt 2392420. Durch Einengen der Mutterlaugen erhält man weitere 6,47 g Substanz mit dem Schmelzpunkt 2352400; Ausbeute total 74,27 g (92 /o).
Eine Lösung von 63,25 g dieser Säure in 500 ml kaltem Methanol wird auf übliche Art mit einer Lösung von Diazomethan in Äther verestert. Man erhält 66 g rohen Ester, Schmelzpunkt 78,5-81,5". Nach zweimaliger Umkristallisation aus Hexan erhält man den Ester in Form farbloser Nadelnmit dem Schmelz punkt 85,5-860.
Eine Lösung von 61 g des Esters in 1000 ml Äther wird bei Raumtemperatur zu einer Lösung von 20 g Lithiumaluminiumhydrid in 350ml wasserfreiem Äther zugefügt. Nach beendigter Zugabe (30 Minuten) wird das Reaktionsgemisch während 30 Minuten unter Rückfluss erhitzt und dann über Nacht bei
Raumtemperatur stehen gelassen. Das überschüssige
Lithiumaluminiumhydrid wird dann mit überschüssi gem Äthylacetat zersetzt und verdünnte Schwefelsäure zugegeben bis sich der weisse Niederschlag gelöst hat.
Die Mischung wird zweimal mit Äther extrahiert und die Ätherlösung mit Wasser und dann mit 50/oiger Natriumbicarbonatlösung gewaschen. Nach Entfer nung des Lösungsmittels und Umkristallisation aus MethylenchioridlÄther erhält man 46,4 g reines 2-Hy droxymethyl-7-methoxy-1,2,3,4,4a,9, 10, 10a-octahy dro-phenanthren (Ringe B/C trans), Schmelzpunkt 94 bis 960. Aus der Mutterlauge erhält man weitere 3,88 g Substanz mit dem Schmelzpunkt 92-940; Ausbeute total 50,28 g (92 /o). Zwei weitere Umkristallisatio nen aus Methylenchlorid-Äther ergeben ein Produkt mit dem Schmelzpunkt 94,5".
Diese Verbindung wirkt stark antigonadotrop und hypocholesterinämisch.
Beispiel 2
Man gibt eine Lösung von 8 g 2-Hydroxymethyl 7-methoxy -.1,2,3,4,4a,9, 10, 10a- octahydro phenan- thren (Ringe B/C trans [hergestellt nach Beispiel 1]) in 600 ml Dimethoxyäthan und 150 ml Äthanol im Verlaufe von 20 Minuten zu einer Lösung von 30 g Lithium in 3 Liter flüssigem Ammoniak. Man rührt während 20 Minuten, vbt dann 420 ml Äthanol zu und lässt das Ammoniak verdampfen. Die überschüssigen Lösungsmittel werden im Vakuum abgedampft.
Der Rückstand wird mit Wasser verdünnt und mit Äther extrahiert. Der Ätherextrakt wird zweimal mit Wasser gewaschen und dann über wasserfreiem Natriumsulfat getrocknet. Nach Entfernungdes Lösungs- mittels und Umkristallisation aus Methylenchlorid Äther erhält man 5,41 g 2-Hydroxymethyl-7-methoxy
1,2,3,4,4a,5,8,9, 10, 10a-decahydro-phenanthren (Rin- ge B/C trans) mit dem Schmelzpunkt 108-110 . Aus den Mutterlaugen erhält man noch 0,87 g Substanz vom Schmelzpunkt 104-107 . Die Ausbeute beträgt total 6,28 g (78 /o). Nach weiteren zwei Umkristallisationen aus Methylenchlorid-Sither beträgt der Schmelzpunkt 111-113 .
Eine Lösung des erhaltenen Decahydro-phenanthrens in 620 ml Aceton wird eine Stunde mit 150 ml 3n Salzsäure in Stickstoffatmosphäre unter Rückfluss erhitzt. Die Lösung wird dann im Vakuum auf ungefähr / eingeengt und dann mit gesättigter Natrium chloridlösung verdünnt. Diese Mischung wird mit Äther extrahiert, der Ätherextrakt mit gesättigter Natriumchloridlösung, 5 0/obiger Natriumbicarbonatlösung und mit Wasser gewaschen.
Nach Entfernung des Lö- sungsmittels und Umkristallisation aus Methylenchlorid-Aethererhältman 4,42 g 2-Hydroxymethyl-7-oxo
1,2,3,4,4a,4b, 5,6,7,9,10,1 0a- dodecahydro -phenan - thren (RingeA/B/Canti-trans) mit dem Schmelzpunkt 108-109 . Aus den Mutterlaugen erhält man weitere 0,78 g Substanz mit dem Schmelzpunkt 104-1070.
Ausbeute total 5,2 g (88 O/o). Die reine Substanz schmilzt bei 108-1090 (aus Methylenchlorid-Äther und Sither-Hexan).
Die erhaltene Verbindung wirkt androgen.
