Bei einer Katalysator-Konzentration von 0, 5 Mol/l verläuft die Reaktion bei Raumtemperatur innerhalb 24 - 48 h bis zur vollständigen Umsetzung, ohne dass grössere Mengen störender Nebenprodukte auftreten und ohne dass die Katalysator-Konzentration wesentlich abnimmt. Bei kleinerer Alkoholat-Konzentration muss die Reaktionszeit entsprechend verlängert und bzw. oder die Reaktionstemperatur erhöht werden. Umgekehrt kann bei höherer Katalysatorkonzentration die Reaktionszeit abgekürzt werden.
Nach beendeter Umesterung wird das Reaktionsprodukt in bekannter Weise isoliert. Da das 16a-Me- thyl-allopregnan-3 ss, l1a, 17 a-triol-20-on in den gebräuchlichen Lösungsmitteln schwer löslich ist, ist die Reinigung durch Umkristallisieren in technischem Massstab schwierig und kostspielig. Es wurde nun aber gefunden, dass das aus der Umesterung gewonnene Rohprodukt in äusserst einfacher Weise gereinigt werden kann, wenn man das rohe 16a-Methyl-allopregnan-3ss, llct, 17ct-triol-20-on mit einem niedrigen aliphatischen Keton, z. B. mit Aceton, Methyläthylketon, Diäthylketon usw., behandelt. Diese Behandlung erfolgt z.
B. durch Austeigen oder Aufschlämmen des Rohprodukts in diesem Lösungsmittel, wobei alle Nebenprodukte und Verunreinigungen (ebenso wie nicht völlig verseifte Anteile) gelöst werden. Durch einfaches Absaugen erhält man so das 16α-Methyl-allopregnan-3ss, 11α, 17α-triol-20-on in völlig reiner Form.
Das Verfahrensprodukt ist ein äusserst wichtiges und wertvolles Zwischenprodukt für die Herstellung

-Methyl-9a-fluor-predni-- triacetoxy-allopregnen ist aus Hecogenin aus Sisal-Agaven leicht herstellbar. Das vorliegende Verfahren stellt deshalb eine wichtige Stufe Jer Synthese der oben genannten wertvollen 16a-Methyl-hormone aus leicht zugänglichen und in grossen Mengen anfallenden Rohstoffen dar.

tigter Natriumhydrogencarbonatlösung, dreimal mit 20 cm3 n-Natronlauge, zweimal mit 20 cm3 Wasser, einmal mit 20 cm3 0, ln-Natriumthiosulfat-Lösung und zweimal mit 20 cm3 Wasser ausgeschüttelt. Die wässerigen Lösungen extrahiert man noch zweimal mit 120 cm3 Äther, worauf die ätherischen Lösungen vereint, getrocknet und eingedampft werden.

Stickstoff 40 h bei Raumtemperatur stehen gelassen, dann gibt man 1, 5 cm3 Eisessig zu, dampft im Wasserstrahlvakuum zur Trockne ein, schüttelt den Rückstand mit 25 cm3 Wasser gut durch, saugt ab und wäscht den Filterrückstand mit kaltem Aceton und mit Aceton-Äther-Gemisch.
Man erhält 2, 782g reines 16α-Methyl-allopregnan-3ss, 11α, 17α-triol-20-on vom Smp. 244 - 2480 (Sintern ab 2350).
Das Filtrat wird zur Trockne eingedampft, der Rückstand in Methylenchlorid aufgenommen und die Lösung mit Wasser gewaschen, getrocknet und nochmals eingedampft. Durch Acetylierung mit 2, 5 cm3 Acetanhydrid und 2, 5 cm3 Pyridin erhält man nach üblicher Aufarbeitung durch Kristallisation aus ÄtherHexan noch 780 mg 16α-Methyl-3ss, 11α-diacetoxy-allopregnan-17α-ol-20-on vom Smp. 168-174 , welches wiederum wie oben angegeben mit Natriummethylat verseift werden kann.
Das als Ausgangsmaterial verwendete A'-Enolacetat wird wie folgt hergestellt :
Zu einer aus 1 g Magnesium hergestellten Lösung von Methylmagnesiumjodid in 80 cm3 Äther gibt man 190 cm3 absolutes Tetrahydrofuran und destilliert anschliessend innerhalb 1/2 h 150 cm3 Lösungs-

setzt. Die Temperatur steigt dabei auf 29 und das Reaktionsgemisch färbt sich gelb. Nach 30minutigem Rühren lässt man wieder unter Kühlung mit einem Bad von 200 innerhalb 1 3/4 min eine Mischung von 3 cm3 Acetylchlorid und 25 cm3 Tetrahydrofuran zutropfen, wobei die Temperatur auf 280 steigt und die Farbe von Gelb nach Grau umschlägt.
Dann lässt man 40 min bei Zimmertemperatur rühren, kühlt auf 50

ab und lässt nacheinander 30 cm3 gesättigte Ammoniumchlorid-Lösung, 50 cm3 Äther und 30 cm3 Wasser zufliessen. Der aus zwei klaren Schichten bestehende Kolbeninhalt wird mit 100 cm3 Äther in einen
Scheidetrichter gespült, worauf man gut durchschüttelt, die wässerige Phase abtrennt und diese erneut mit 75 cm3 Äther extrahiert.
Die organischen Phasen werden nacheinander zweimal mit 50 cm3 Imol. i Natriumthiosulfat-Lösung, einer Mischung von 50 cm3 gesättigter Kochsalzlösung und 15 cm3 gesättigter
Natriumhydrogencarbonatlösung und zweimal mit 50 cm3 gesättigter Kochsalzlösung ausgeschüttelt, mit
Magnesiumsulfat getrocknet und zunächst bei Normaldruck und dann bei Wasserstrahlvakuum einge- dampft. Den Rückstand löst man in Xylol, dampft im Vakuum ein und wiederholt diese Operation noch einmal. Die Lösung des so erhaltenen Öles in 50 cm3 Hexan wird durch 8 g Aluminiumoxyd (Aktivität III) filtriert. Man wäscht mit 250 cm3 Hexan nach, dampft das Eluat bei Wasserstrahlvakuum ein und trock- net den Rückstand während l 1/2 h bei 800 und 0, 05 mm Druck im Rotationsverdampfer.
Dabei destillie- ren erhebliche Mengen eines wohlriechenden Öles ab. Der verbliebene, praktisch farblose Lack, dessen
Gewicht 8, 6 g beträgt, wird in 10 cm3 Pentan gelöst und mehrere Tage bei -150 stehen gelassen. Dann wird von den ausgeschiedenen Kristallen abfiltriert, mit kaltem Pentan gewaschen und bei 800 und 0, 05 mm Druck während 4 h getrocknet. Man erhält 4, 7 g #17(2)-16α-Methyl-3ss, 11α,20-triacetoxy- - allopregnen als Stereoisomerengemisch vom Smp. 123, 5-1290. In Folge der leichten Löslichkeit be- finden sich in der Mutterlauge noch erhebliche Mengen des genannten Enolacetates.
Das in Methylen- chlorid aufgenommene IR.-Spektrum des kristallisierten Enolacetats zeigt unter anderem folgende cha- rakteristische Banden : bei 5, 78/l mit Inflexion bei 5, 73 but und schwacher Schulter bei 5, 86 bol (Acetate

Extrakte werden mit Essigester nachextrahiert und die vereinigten organischen Lösungen getrocknet und im Wasserstrahlvakuum eingedampft. Man löst den Rückstand, welcher das rohe 16ct-Methyl-30, ll < a, 20- - triacetoxy-17, 20a-oxido-allopregnan darstellt, in 315 cm3 abs. Methanol und gibt unter Stickstoff 325 cm3 l. On-Natriummethylat in abs. Methanol zu und lässt 46 h bei Raumtemperatur stehen.
Anschlie- ssend gibt man 18 cm3 Eisessig zu und engt das Reaktionsgemisch auf ein Volumen von 240 cm3 ein, kühlt ab und fällt das Reaktionsprodukt durch Zugabe von 140 cm3 Wasser aus. Der Niederschlag wird abgesaugt, gut mit Wasser und schliesslich mit 150 cm3 Aceton gewaschen und getrocknet. Man erhält 28 g reines 16α-Methyl-allopregnan-3ss, 11α, 17α-triol-20-on vom Smp. 242 - 2440.
Aus dem Filtrat lässt sich wie in Beispiel 1 beschrieben nach Acetylierung noch eine weitere Menge des obigen Triols in Form des Diacetats isolieren.
Das in diesem Beispiel verwendete rohe Enolacetat wird wie folgt hergestellt :
Zu einer Mischung von 16, 3 cm3 Methyljodid in 120 cm3 Äther gibt man unter Rühren und unter Stickstoff in kleinen Portionen 5, 6 g Magnesiumspäne und kocht nach beendeter Reaktion noch 1 h unter Rückfluss. Dann kühlt man auf-100 ab und gibt 150 cm3 Tetrahydrofuran und 1, 85 g Cuprochlorid und schliesslich innerhalb 1 h eine Lösung von 50 g #16-3ss,11α-Diacetoxy-allopregnen-20-on in 150 cm3 Tetrahydrofuran zu. Nach beendeter Zugabe lässt man noch 1 h bei-5 bis 00 rühren und lässt dann unter Kühlen innerhalb 1 h 17 cm3 Acetylchlorid zutropfen.
Nach einer weiteren halben Stunde zersetzt man mit einer Lösung von 90 g Ammoniumchlorid, 5 g Natriumacetat und 5 g Natriumthiosulfat und extrahiert mehrmals mit Petroläther. Die Petroläther-Extrakte werden mit Natriumsulfat getrocknet und am Wasserstrahlvakuum eingedampft. Zur weiteren Reinigung wird das rohe Enolacetat in 600 cm3 Petrol- äther gelöst und mit 60 g Aluminiumoxyd (Akt. II) während 1 h gerührt. Dann wird abgesaugt und das
