Procédé pour la préparation d'alcoxy-triphényl-méthanols substitués basiques
L'invention est relative à un procédé pour la préparation d'un nouveau groupe d'alcoxy-triphényl- méthanols substitués basiques dotés d'intéressantes propriétés physiologiques.
On peut considérer les composés en question comme des alcoxy-triphényl-méthanols substitués basiques de formule (I) :

Pour préparer les méthanols substitués en question, on fait réagir un halogénure de phénylmagné- sium substitué, tel qu'un bromure ou un chlorure ou un phényl lithium, un phényl sodium ou un phényl potassium, possédant sur le noyau phényle le substituant R4, avec une benzophénone de formule :

On peut obtenir la benzophénone substituée en faisant réagir un sel de métal alcalin d'une hydroxy benzophénone avec un halogénure tel qu'un bromure ou un chlorure de dialcoylaminoalcoyle selon l'équation chimique suivante :

Les composés obtenus selon le procédé de l'inven- tion possèdent une intéressante activité physiologique, qui se traduit notamment par un blocage de la formation du cholestérol dans le corps. Ils présentent donc un grand intérêt pour le traitement de l'hypercholesté- rolémie et des troubles cardio-vasculaires aggravés par de hautes teneurs de cholestérol dans le sang.
Ils sont efficaces par voies orale ou parentérale, et on peut les administrer à des doses comprises entre 25 mg et 4 grammes par jour selon la gravité des troubles et l'état du patient, que le médecin traitant doit apprécier. Lorsqu'ils sont administrés par voie orale, ils peuvent être présentés sous forme de capsules ou de comprimés qui peuvent, si on le désire, être kératinisés. On peut aussi préparer des suspensions ou des compositions liquides à administrer par voie orale. On peut préparer des solutions injectables, si on le désire, en utilisant, d'une manière classique, des solutions salines isotoniques.
Bien entendu, l'intérêt des composés en question ne dépend pas d'une quelconque présentation posologique, et l'homme de l'art peut établir des prépa- rations convenables en se conformant aux principes classiques de la pratique pharmaceutique.
Les composés en question possèdent aussi une activité endocrine, et ils sont intéressants en tant qu'agents antiinflammatoires et en tant qu'agents oestrogènes et antioestrogènes.
Certains des agents anticholestérol actuellement connus agissent en bloquant la formation du cho lestérol à un stade tardif de sa synthèse biochimique, probablement dans le foie. Ceci a toutefois pour résultat l'accumulation d'un autre stérol connu sous le nom de desmostérol. Les composés en question apparaissent comme capables de bloquer la formation du cholestérol par un autre mécanisme selon lequel la synthèse du cholestérol est bloquée à un stade précoce, avant la formation de la structure stérolique.
Les non-stérols qui résultent de ce processus de blocage sont bien plus facilement métabolisés et excrétés que ne le sont les stérols. Il s'agit là, bien entendu, d'une propriété inattendue et avantageuse des composés en question.
Exemple 1 Préparation du p- (-diéíhylaminoéíhoxy)-phényl-p- tolyl-p-chlorophenylmethanol
On ajoute une solution éthérée de 0, 08 mole de bromure de p-chlorophénylmagnsium à une solution éthérée de 15, 5 g (0, 05 mole) de p-(, 8-diéthyl- aminoéthoxy)-p'-méthylbenzophénone, et on chauffe deux heures à reflux le mélange résultant. On décom- pose le complexe de Grignard à l'aide d'une solution de chlorure d'ammonium ; on sépare la couche organique et on la sèche sur sulfate de magnésium.
On évapore le solvant, et il reste ainsi une huile que l'on convertit en citrate diacide dans de la butanone, en utilisant 9, 6 g (0, 05 mole) d'acide citrique.
Après deux recristallisations dans de la butanone, on obtient ainsi le citrate diacide de p-(-diéthyl- aminoéthoxy)-phényl-p-tolyl-p-chlorophénylméthanol,
P. F. 89-91 . Ce composé est un excellent agent anticholestérol, capable de bloquer la formation du cholestérol à un stade précoce de sa synthèse biochimique, et c'est, de ce point de vue, l'un des plus avantageux parmi les composés en question.
Pour préparer la p-(ss-diéthylaminoéthoxy)-p'- méthylbenzophénone, on opère de la façon suivante : on agite pendant 30 minutes un mélange de 210 g de p-hydroxy-p'-méthylbenzophénone, 55 g de mé- thoxyde de sodium en poudre et 400 ml d'éthanol.
On y ajoute ensuite une solution de 150 g de chlorure de (3-diethylaminoethyle dans 300 ml de toluène, et l'on chauffe quatre heures le mélange à reflux.
On élimine le solvant, on reprend le résidu par de l'éther, on extrait la solution éthérée à l'aide d'une solution de NaOH à 10 /o, puis deux fois à l'eau, on élimine l'éther, et on distille le résidu. On obtient le produit sous la forme d'une huile, P. E. environ 2320 sous 0, 6 mm de mercure.
Exemple 2
Préparation du p-(ss-détStylaminoéthoxy)-phényl-p- tolyl-p-trluorométhylphénylméthanol
Lorsqu'on met en oeuvre le mode opératoire de l'exemple 1 en utilisant du bromure de p-trifluorométhylphénylmgnésium à la place du bromure de p-chlorophénylmagnésium, on obtient le produit désiré (recristallisation dans un mélange d'éther et d'éther de pétrole), P. F. 104-106 .
Exemple 3 Préparation du p- (ss-diéthylaminoéthoxy)-phényl-p
tolyphénylméthanol
Lorsqu'on met en oeuvre le mode opératoire de l'exemple 1 en utilisant du bromure de phényl- magnésium à la place du bromure de p-chlorophényl- magnésium, on obtient le citrate diacide du composé désiré (recristallisation dans de la butanone), P. F.
89-91 .
Exemple 4
Préparation du p- (ss-diéthylaminoéthoxy)-phényl-p- methoxyphenyl-p-chlorophenylmethanol
Lorsqu'on met en oeuvre le mode opératoire de l'exemple 1 en utilisant de la p-hydroxy-p'-chlorobenzophénone à la place de la p-hydroxy-p'-méthyi- benzsophénone, on obtint, par recristallisation dans de l'éthanol, de la p-(ss-diéthylaminoéthoxy)-p'- chlorobenzophénone, P. F. 79-80 . Lorsqu'on traite cette cétone par du chlorure de p-méthoxyphénylmagnésium, on obtient le composé désiré, dont le citrate diacide fond à 84-86,,.
Exemple 5 Préparation du p- (ss-diéthylaminoéthoxy)-phényl-p
tolyl-p-méthoxyphénylméthanol
Lorsqu'on met en oeuvre le mode opératoire de l'exemple l en utilisant du bromure de p-méthoxyphénylmagnésium à la place du bromure de p-chlorophénylmagnésium, on obtient, par recristallisation dans de l'éther de pétrole, le composé désiré, P. F.

Exemple 6
Préparation du p-(ss-diméthylaminoéthoxy)-phényl-p-
tolyphénylméthanol
Lorsqu'on met en oeuvre le mode opératoire de l'exemple 1 en remplaçant la p- (ss-diéthylamino- éthoxy)-p'-méthylbenzophéhone et le bromure de p-chlorophénylagnésium respectivement par de la p-(ss-diméthylaminoéthoxy)-benzophénone et par du bromure de p-méthylphénylagnésium, on obtint, par recristallisation dans de l'éther de pétrole, le composé désiré, fondant à 69-71 .
Exemple 7 Préparation du p- (ri-diéthylaininoéthoxy)-phényl-p- fluorophényl-phérrylméthanol
Lorsqu'on met en oeuvre le mode opératoire de 1'exemple 1 en remplaçant la p- (ss-diéthylamino- éthoxy)-p'-méthylbenzophénone et le bromure de p-cilorophénylmagnésium respectivement par de la p-(-diéthylaminoéthoxy)-benzophénone et par du bromure de p-fluorophénylmagnésium, on obtint, par recristallisation dans de l'éther de pétrole, le composé désiré, P. F. 52-54o.
Exemple 8
Préparation du p-(ss-morpholinoéthoxy)-phényl-p- tolyl-p-chlorophehylmethanol
Lorsqu'on met en oeuvre le mode opératoire de l'exemple 1 en remplaçant la p- (ss-diéthylamino- éthoxy)-p'-méthylbenzophéhone. par de la p-(ss morpholinoéthoxy)-p'-méthylbenzophénone, on obtient, par recristallisation dans un mélange d'éthanol et d'éther de pétrole, le composé désiré, fondant à 111-1130.
Exemple 9
Préparation du p- (Y-pipéridinopropoxy)-phényl-p-
toyl-p-chlorophénylméthanol
Lorsqu'on met en oeuvre le mode opératoire de l'exemple 1 en remplaçant la p- (-diéthylamino- éthoxy)-p'-méthylbenzophénone par de la p- (y-pipé- ridinopropoxy)-p'-méthylbenzophénone, on obtint, par recristallisation dans de l'éthanol, le composé désiré, P. F. 146-1480.

Préparation du p-(ss-diéthylaminoéthoxy)-phényl-bi
(p-chlorophényl)-méthanol
Le citrate diacide de ce composé fond à 88-90o, et on prépare ce composé en mettant en oeuvre le mode opératoire de l'exemple 1 en utilisant de la p- (ss-diéthylaminoéthoxy)-p'-chlorobenzophénone et du bromure de p-chlorophénylmagnésium.
Exemple 11
Préparation du p-(ss-diéthylaminoéthoxy)-phényl-bis- (p-tri f luoromethylphenyl)-methanol
Ce composé fond à 90-92 . et on le prépare à partir de p- (ss-diéthylaminoéthoxy)-p'- (trifluoromé- thyl)-benzophéhone et de bromure de p-trifluoro méthylphénylmagnésium.

The compounds obtained according to the process of the invention have an interesting physiological activity, which is reflected in particular by blocking the formation of cholesterol in the body. They are therefore of great interest for the treatment of hypercholesterolemia and cardiovascular disorders aggravated by high levels of cholesterol in the blood.
They are effective orally or parenterally, and can be administered in doses of between 25 mg and 4 grams per day depending on the severity of the disorder and the condition of the patient, which the attending physician should assess. When administered orally, they can be presented in capsule or tablet form which can, if desired, be keratinized. It is also possible to prepare suspensions or liquid compositions for oral administration. Injectable solutions can be prepared, if desired, using isotonic saline solutions in a conventional manner.
Of course, the benefit of the compounds in question does not depend on any dosage form, and those skilled in the art can make suitable preparations according to conventional principles of pharmaceutical practice.
The compounds in question also possess endocrine activity, and they are useful as anti-inflammatory agents and as estrogenic and antiestrogenic agents.
Some of the currently known anti-cholesterol agents work by blocking the formation of cholesterol at a late stage of its biochemical synthesis, probably in the liver. This, however, results in the accumulation of another sterol known as desmosterol. The compounds in question appear to be capable of blocking the formation of cholesterol by another mechanism according to which the synthesis of cholesterol is blocked at an early stage, before the formation of the sterolic structure.
The non-sterols which result from this blocking process are much more easily metabolized and excreted than are sterols. This is, of course, an unexpected and advantageous property of the compounds in question.
Example 1 Preparation of p- (-dieíhylaminoéíhoxy) -phenyl-p-tolyl-p-chlorophenylmethanol
An ethereal solution of 0.08 moles of p-chlorophenylmagnsium bromide is added to an ethereal solution of 15.5 g (0.05 moles) of p- (, 8-diethyl-aminoethoxy) -p'-methylbenzophenone, and reflux the resulting mixture for two hours. The Grignard complex is decomposed using an ammonium chloride solution; the organic layer is separated and dried over magnesium sulfate.
The solvent is evaporated, and an oil remains which is converted to citrate diacid in butanone, using 9.6 g (0.05 mole) of citric acid.
After two recrystallizations from butanone, the diacid citrate of p - (- diethylaminoethoxy) -phenyl-p-tolyl-p-chlorophenylmethanol is thus obtained,
P. F. 89-91. This compound is an excellent anticholesterol agent, capable of blocking the formation of cholesterol at an early stage of its biochemical synthesis, and it is, from this point of view, one of the most advantageous among the compounds in question.
To prepare p- (ss-diethylaminoethoxy) -p'-methylbenzophenone, the procedure is as follows: a mixture of 210 g of p-hydroxy-p'-methylbenzophenone, 55 g of methoxide is stirred for 30 minutes. sodium powder and 400 ml of ethanol.
A solution of 150 g of (3-diethylaminoethyl chloride in 300 ml of toluene) is then added thereto, and the mixture is heated for four hours at reflux.
The solvent is removed, the residue is taken up in ether, the ethereal solution is extracted with a 10% NaOH solution, then twice with water, the ether is removed, and the residue is distilled. The product is obtained as an oil, P.E. about 2320 under 0.6 mm Hg.
Example 2
Preparation of p- (ss-detStylaminoethoxy) -phenyl-p- tolyl-p-trluoromethylphenylmethanol
When the procedure of Example 1 is carried out using p-trifluoromethylphenylmagnesium bromide instead of p-chlorophenylmagnesium bromide, the desired product is obtained (recrystallization from a mixture of ether and ether of petroleum), PF 104-106.

Preparation of p- (ss-diethylaminoethoxy) -phenyl-p- methoxyphenyl-p-chlorophenylmethanol
When the procedure of Example 1 is carried out using p-hydroxy-p'-chlorobenzophenone in place of p-hydroxy-p'-methylbenzsophenone, one obtains, by recrystallization from l ethanol, p- (ss-diethylaminoethoxy) -p'-chlorobenzophenone, mp 79-80. When this ketone is treated with p-methoxyphenylmagnesium chloride, the desired compound is obtained, the diacid citrate of which melts at 84-86 ,,.

When carrying out the procedure of Example 1 by replacing p- (ss-diethylaminoethoxy) -p'-methylbenzophenone and p-cilorophenylmagnesium bromide respectively by p - (- diethylaminoethoxy) -benzophenone and with p-fluorophenylmagnesium bromide, by recrystallization from petroleum ether, the desired compound was obtained, mp 52-540.
Example 8
