Procédé de préparation de nouaelles salicylamides La présente invention concerne un procédé de préparation de nouveaux composés analgésiques de la série des salicylamides.
On connait déjà par le brevet allemand No 1098001 publié le 26 janvier 1961, des com posés à action analgésique dont les deux seuls exem ples cités sont la N-(diéthyl-amino-éthyl)-salicylamide et la N-(diéthyl-amino-propyl)-salicylamide.
La titulaire a soumis à une étude systématique les composés de cette famille et en particulier ceux dont la chaîne latérale comporte un nombre croissant de groupes méthylène et ceux dont la fonction phénol est acylée.
Cette étude a été basée sur l'évaluation de l'in dex thérapeutique et de la toxicité de chaque com posé étudié.
L'index thérapeutique est, conune il est classique, le rapport entre la dose léthale 50 et la dose efficace 50, et donc d'autant plus favorable qu'il est plus élevé.
La toxicité a été déterminée par la méthode de Miller & Tainter sur des groupes homogènes de souris. Cette étude systématique a permis de faire une double constatation surprenante: les dérivés analo gues aux deux composés précités mais comportant un groupe méthylène de plus dans leur chaîne laté rale, ont un index thérapeutique très supérieur alors que ceux comportant un nombre encore plus élevé de groupes méthylène voient cet index redescendre, de sorte que l'on peut dire que sous un de ses aspects essentiels l'invention réside dans la découverte sur prenante d'un maximum d'efficacité thérapeutique de la N-(diéthyl-amino-butyl)
salicylamide.
La seconde constatation surprenante a été celle que l'acylation de la fonction phénol en position ortho par rapport à la chaîne latérale conduit à une augmentation de l'index thérapeutique et à une di minution de la toxicité, ces phénomènes étant très marqués pour le dérivé acétylé. En d'autres termes, sous un autre aspect l'invention a consisté dans la découverte de l'efficacité thérapeutique de l'ordre de 3 à 4 fois supérieure de la N-(diéthyl-amino-butyl)- acétylsalicylamide,
associée à une toxicité plus de deux fois inférieure de ce composé par rapport aux composés précités décrits dans le brevet allemand. On a réuni dans le tableau ci-après les valeurs de l'index thérapeutique et de la DL 50 pour les différents composés énumérés ci-après

<SEP> 15 <SEP> 976 Enfin, la titulaire a constaté que pour des rai sons pratiques, les deux dérivés à propriétés théra peutiques supérieures qu'elles a découverts, étaient avantageusement administrés sous forme de citrates. En effet, ces sels sont blancs et cristallisés, donc plus aisés à isoler, purifier et manipuler.
Il apparait clairement de ce tableau un maximum d'efficacité thérapeutique et un minimum de toxicité pour les dérivés butylés, que la fonction phénol soit ou non acylée, et plus encore quand la fonction phé nol est acétylée. Cette dernière constatation est d'au tant plus surprenante que l'acétylation de la fonc tion phénol dans les dérivés éthylés et propylés cor respondait au contraire à un abaissement de l'effi cacité et à un accroissement de la toxicité par rap port au composé correspondant non acétylé.
La chute des propriétés, quand on passe des dé rivés butylés aux dérivés pentylés, est également sur prenante, si l'on considère que la progression favo rable de ces propriétés se faisait régulièrement d'un homologue au suivant dans la même série, jusqu'au dérivé butylé.
II apparait donc que la titulaire a découvert, dans la série générale dont la préparation était décrite dans le brevet allemand précité, un composé particu lier et ses dérivés acylés, dont les propriétés se dis tinguent nettement, et de façon favorable, de celles des autres membres de cette famille. Dans la pratique, on pourra utiliser ces produits en thérapeutique humaine à des doses de l'ordre de 0,25 g (calculées en citrates), sous forme de compri més, de suppositoires ou de solutés injectables.
Les composés en question se préparent selon l'in vention, suivant un mode opératoire illustré par les deux exemples ci-après <I>Exemple 1</I> Fabrication du citrate de N-(4-diéthylaminobutyl)-salicylamide A une solution de 172,1 g (1,1 mole) de chlorure de l'acide salicylique, dissous dans 1 litre de ben zène anhydre, on ajoute lentement 144 g (1 mole) de 4-diéthylaminobutyl-amine (C.H;;) 2 N-CH@CH_- CH2-CH.,-NHz dissous dans 200 cm@ de benzène anhydre, en agitant et en maintenant la température entre 0 et 5a C.
Après fin d'addition, on continue l'agitation pendant 1 heure à la même température et ensuite 1 heure à la température d'ébullition du benzène. On décante ensuite le produit visqueux et on lave deux fois avec 250 cm, de benzène anhydre qu'on décante à chaque fois.
Le produit restant est traité avec 1,5 litre de solution de carbonate de so- dium à 20 % et laissé en contact avec agitation, le temps nécessaire pour que tout le produit devienne huileux (environ 2 heures). On extrait ensuite trois fois avec 500<I>ce</I> d'éther. Les couches éthérées réu- nies sont séchées sur sulfate de sodium.
On filtre ensuite, on lave le sulfate de sodium avec 350 cm3 d'éther et on distille l'éther. Les dernières traces d'éther sont chassées sous vide.
Le résidu est pesé et on calcule la quantité d'acide citrique C,;Hs0,0 nécessaire. Pour<B>264g</B> (1 mole) de résidu, qui est la N-(4-diéthylaminobu- tyl)-salicylamide, il faut 22.0 g (1,05 mol.) d'acide ci trique à une molécule d'eau. Le résidu est dissous dans environ 700 cm 2 d'alcool alcalin (pour 264 g). L'acide citrique est dissous dans environ 600 cm;, d'alcool absolu pour 220 g. Les deux solutions sont mélangées et on distille sous pression réduite de fa çon à éliminer la totalité de l'alcool.
Le résidu est ensuite recristallisé dans 1,2 litre d'alcool isopropyli- que. On laisse recristalliser ensuite pendant 72 heu res à 0-51, C. On filtre ensuite rapidement sur Buch ner et on obtient un produit blanc, pâteux,
plus ou moins cristallisé qu'on sèche sous pression réduite à 350 environ. Le rendement est de 060 à 70 %. <I>Exemple 2</I> Fabrication du B 862 (citrate de N-(4-diéthylaminobutyl)-acétylsalicylamide A une solution de 218,4 g (1,1 mole) de chlorure de l'acide acétylsalicylique dissous dans environ 1 litre d'éther anhydre, on ajoute lentement 144 g (1 mole)
de 4-diéthylaminobutyl amine dissous dans 150 cm@' d'éther anhydre, en agitant et en maintenant la tem pérature entre 0 et 5 C. Après fin d'addition on continue l'agitation pendant 1 heure à la même tem pérature et ensuite pendant 1 heure à ébullition à l'éther. On décante ensuite l'éther et le produit vis queux restant est lavé deux fois avec 250 cm$ d'éther anhydre qu'on décante à chaque fois.
Le produit res tant est traité avec un mélange de 1,4 litre de solu tion de carbonate de sodium à 20 10/0 et<B>55</B> crW de solution de soude caustique à 10 D/o. On laisse en contact en agitant le temps nécessaire pour que tout le produit devienne huileux (environ 2 heures). En suite on extrait trois fois avec 500 cm',' d'éther. Les couches éthérées réunies sont séchées sur sulfate de sodium. On filtre ensuite, lave le sulfate de sodium avec 350 ce d'éther anhydre, et on distille l'éther. Les dernières traces d'éther sont chassées sous pres sion réduite.
Le résidu est percé et on calcule la quantité d'acide citrique C6H80,. H\,0 nécessaire ; pour 306 g (1 mole) de résidu, qui est la N-(4-di- éthylaminobutyl)-acétylsalicylamide, il faut 220 g (1,05 mole) d'acide citrique à une molécule d'eau. Le résidu est dissous dans environ 750 cm3 d'alcool absolu (pour 306 g de résidu). L'acide citrique est dissous dans environ 600 cm$ d'alcool absolu (pour 220 g). Les deux solutions sont mélangées, et on dis tille sous pression réduite jusqu'à élimination totale de l'alcool.
Le résidu est ensuite recristallisé dans 1,2 litre d'alcool isopropylique. On laisse cristalliser ensuite pendant 72. heures à 0-50 C. On filtre ensuite rapidement sur Buchner. On obtient un produit blanc, pâteux plus ou moins cristallisé, qu'on sèche sous pression réduite à 350 C environ.
Le rendement est de 60 à 70,%.
