Verbindungen der Formel 1 besitzen interessante therapeutisch auswertbare Eigenschaften, wie z. B.
Wirkungen auf das Herz ; u. a. bewirken sie eine Er höhung des Coronardurchflusses und eine Dilatation des Herzmuskels. Die in 1, 2- und/oder 6, 7, 8, 9-Stel- lung hydrierten Derivate können verwendet werden, indem man sie zu therapeutisch wirksamen Pyridocarbazolderivaten gemäss Formel 1 dehydriert.
Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass man ein gegebenenfalls 5, 6, 7, 8 tetrahydriertes Carbazolderivat der Formel

worin R7 Wasserstoff oder die Hydroxylgruppe bedeutet, durch Behandeln mit einem Kondensations- mittel zum Ringschluss bringt.
Ist R7 Wasserstoff, so erhält man bei der Kondensation die 1, 2-Dihydroderivate ; ist R7 die Hydroxylgruppe, so wird diese zusammen mit dem Wasserstoff des benachbarten. C-Atoms unter Bildung einer Doppelbindung als Wasser abgespalten. Geht man von einem im nichtalkyliertsn Benzolring tetrahydrierten Carbazolderivat aus, so erhält man im ersten Fall die 1, 2, 6, 8, 9-Hexahydroderivate, im zweiten Fall die 6, 7, 8, 9-Tetrahydroderivate.
Als Kondensationsmittel eignen sich besonders Polyphosphorsäure, Phosphorpentoxyd und Phos phoroxychlorid. Die Kohdensationsreaktion wird zweckmässig bei erhöhter Temperatur, z. B. zwischen 50 und 200 C, mit oder ohne Verwendung eines Lösungsmittels durchgeführt. Als Lösungsmittel eignen sich z. B. Benzol, Toluol, Xylol oder Nitrobenzol.
Zur Herstellung des Ausgangsmaterials kann man beispielsweise von einem 1-und/oder 4-alkylierten Carbazol oder 5, 6, 7, 8-Tetrahydrocarbazol ausgehen, welches in 2-Stellung eine Gruppe aufweist, welche der Kettenverlängerung unterworfen werden kann, beispielsweise eine Cyan-, Carboxyl-, Aldehyd-oder Acylgruppe. Diese Gruppen lassen sich in an sich bekannter Weise in die ss-Aminoäthylgruppe über- führen, welche dann acyliert wird.
Die in der genannten Weise erhaltenen, in 1, 2 und/oder 6, 7, 8, 9-Stellung hydrierten Derivate der Verbindungen gemäss Formel 1 lassen sich zu Pyrido carbazolderivaten der Formel 1 dehydrieren. Als Dehydrierungsmittel eignen sich besonders Platinkabalysatoren und andere Edelmetallkatalysatoren, Chinone und Selen. Die Dehydrierung kann in Gegenwart oder nicht eines Lösungsmittels, wie Tetralin oder Decalin, durchgeführt werden.
Im Falle der Hexyhydroderi- vate kann die Dehydrierung in zwei Stufen durchgeführt werden, wobei unter milderen Bedingungen zunächst die Doppelbindung in 1, 2-Stellung, unter schärferen Bedingungen nachher die beiden Doppelbindungen in 6, 7- und 8, 9-Stellung eingeführt werden.
Die erfindungsgemäss erhältlichen Pyridocarbazol- derivate gemäss Formel 1 sowie deren hydrierte Derivate können als freie Basen oder in Form ihrer Salze mit organischen oder anorganischen Säuren gewonnen werden. Besonders gut eignet sich hierzu Wein- säure.
Beispiel
4, 11-Dimethyl-10-H-pyrido-(4, 3-b)-carbazol
5 g 1-Methyl-2-(ss-acetamido-äthyl)-carbazol werden in 800 cm3 absolutem Toluol gelöst, mit 26, 5 cm3 Phosphoroxychlorid versetzt und ll/2 Stunden unter Rückfluss erhitzt. Man entfernt darauf das Lösungs- mittel unter Vakuum, löst den Rückstand in warmer, verdünnter, wässeriger Salzsäure und filtriert von wenig Unlöslichem ab. Den sauren Auszug stellt man darauf mit konzentrierter Ammoniaklösung alkalisch und schüttelt erschöpfend mit Chloroform aus. Der Chloroformrückstand kristallisiert aus Chloroform/ Aceton, wobei man 4, 1 g 1, 2-Dihydro-4, 11-dimethyl 10-H-pyrido- (4, 3-b)-carbazol erhält.
4 g dieser Substanz werden mit 3 g Palladiumkohle (5 %) in 300 cm3 Decalin 11/2 Stunden unter r Rückfluss erhitzt. Man lässt darauf abkühlen, kühlt weiter auf 0 C und nutscht den Katalysator und das auskristallisierte Endprodukt miteinander ab. Letzteres wird in Chloroform gelöst, durch Filtrieren vom Katalysator befreit und aus Chloroform/Aceton kristallisiert.
Man erhält so 3, 6 g derbe, hellgelbe, pris matische Kristalle vom Schmelzpunkt 319-324 C (Zers.). UV-Spektrum in Athanol :
