Verfahren zur Herstellung von C-Acylaminopenicillansäuren Im deutschen Patent Nr. 1 149 361 ist vorge schlagen worden, 6-Acylaminopenicillansäuren da durch herzustellen, dass man auf 6-Aminopenicillan- säure in Gegenwart von Carbonsäuren, deren Salzen oder Derivaten bei einem pH von höchstens 6,5, Bakterien bzw.
aus diesen gewonnene Extrakte, Enzyme oder Enzymanreicherungen einwirken lässt, die fähig sind, die Phenylessigsäure mit der Amino- gruppe der 6-Aminopenicillansäure amidartig zu ver knüpfen. Als geeignete Carbonsäurederivate hatten sich unter anderem Acylglycokolle oder Acylglutamin- säuren erwiesen.
Es wurde nun gefunden, dass man manche 6-Acylaminopenicillansäuren in wesentlich höherer Ausbeute erhält, wenn man 6-Aminopenicillansäure mit Carbonsäureestern von Hydroxy- oder Mercapto- verbindungen, insbesondere von Hydroxy- und Mercaptocarbonsäuren oder ihren Derivaten umsetzt, und zwar in Gegenwart von Bakterien bzw.
aus die sen gewonnenen Extrakten, Enzymen oder Enzym anreicherungen, die fähig sind, Phenylessigsäure mit der Aminogruppe der 6-Aminopenicillansäure amid- artig zu verknüpfen.
Als 6-Aminopenicillansäure können sowohl die synthetische oder die durch Abbau gewonnene, kristallisierte Substanz oder gegebenenfalls angerei cherte rohe Lösungen, wie sie aus Fermentationen oder andern Prozessen anfallen, benutzt werden.
Hydroxy- und Mercaptoverbindungen im Sinne der Erfindung sind Alkohole, Mercaptane, Phenole und Thiophenole. Beispiele für Alkohole sind Äthanol, Butanol, Dodecylalkohol, Cyclohexanol, Phenylmethylcarbinol und Glykolsäure. Geeignete Mercaptane sind Mercaptobenzthiazol, Thioglykol- säure,
2-Mercaptobuttersäure und Cysteamin.
Als Phenole oder Thiophenole kommen Phenol, p-Oxybenzoesäure, Thiophenol oder Thiokresol in Frage.
Beispiele brauchbarer Verbindungen sind Phenylessigsäurethiophenolester, Phenoxyessigsäuremethylester, a-Phenoxypropionylthioglykolsäure, o-Chlorphenoxyacetylthioglykolsäureamid, p-chlorphenoxyacetylthioglykolsaures Natrium. Besonders geeignet sind Carbonsäureester von sol chen Alkoholen und Mercaptanen, die wiederum Carboxylgruppen tragen, beispielsweise Carbonsäure- ester von Glykol- und Thioglykolsäure.
Die Gegenwart der genannten Enzyme ist erfor derlich, da die Carbonsäureester in wässrigem Milieu bei pH 4-8 allein nicht in der Lage sind, 6-Amino- penicillansäure zu den erwünschten 6-Acylderivaten zu acylieren. Vergleichsversuche haben ergeben, dass beispielsweise a-Phenoxypropionylthioglykolsäure,
bei pH 4-8 mit 6-Aminopenicillansäure behandelt, keine a-Phenoxyproponyl - 6 - aminopenicillansäure liefert, während der Zusatz einer Enzymlösung die ge wünschte Synthese schnell und in hoher Ausbeute bewirkt. Es ist nicht erforderlich, die synthese bewirkenden Enzyme zu isolieren. Vielmehr sind be reits Bakterienzellen, die diese Enzyme enthalten, oder ihre Lysate als reaktionsbedingende Agenzien verwendbar.
Ein besonderer Vorteil des vorliegenden Ver fahrens ist es ferner, solche 6-Acylaminopenicillan- säuren mit den wertvollen antibiotischen Eigenschaf- ten der Penicilline in hoher Ausbeute darzustellen, die durch Fermentation mit penicillinbildenden Pilzen in Gegenwart der entsprechenden Carbonsäuren (so genannten Präcursoren) nicht entstehen.
<I>Beispiel 1</I> 160 Liter 2 Vol. % ige Maisquellwasserlösung, enthaltend 0,2/10 Kaliumphenylacetat, werden mit Kaliumhydroxydlösung auf pH 7,0 eingestellt und 30 Minuten auf 120 C erhitzt. Nach Abkühlung wird die Lösung durch Zentrifugieren geklärt und 40 Minuten bei 110 C im Fermenter sterilisiert.
Diese Nährlösung wird nach Abkühlung mit 400 em3 einer 18stündigen Schüttelkultur von E. coli ATCC <B><I>11</I>105</B> beimpft. Der Ansatz wird dann mit 150 Liter Luft/Min. bei 150 Umdrehungen des Rührwerkes pro Minute belüftet und 17 Stunden bei 31 C ohne Überdruck kultiviert. Während der gesamten Wachs tumszeit werden durch eine von der Luftleitung des Fermenters getrennte Leitung 5 Liter Kohlendioxyd/ Minute in die Kultur eingeleitet.
Die Bakterienzellen werden aus der Kulturlösung abzentrifugiert, in 16 Liter 1/15 m Phosphatpufferlösung von pH 6,0 ge waschen und nach dem Abzentrifugieren zu einer dichten Suspension in 1/15 m Phosphatpufferlösung von pH 6,0 resuspendiert.
Dieser Suspension setzt man 0,125 % 6-Amino- penicillansäure, 0,5 % a-Phenoxypropionylthioglykol- säure und 0,1 % Toluol zu.
Das Reaktionsgemisch wird dann mit konzentrierter Sodalösung auf pH 6,5 eingestellt und eine Stunde bei 37 C aufbewahrt. Nach dieser Zeit enthält der Ansatz 884 Einheiten Penicillinaktivität pro cm3, gemessen an einem Peni- cifin-G-Standard im Plattentest Bac. subtilis ATCC 6633.
Diese Penicillinaktivität wird von der bei der enzymatischen Reaktion entstandenen a-Methyl- phenoxyacetyl-6-aminopenicillansäure bewirkt.
Herstellung der a - Phenoxypropionylthioglykol- säure: In 200 cm3 4,4 % ige Natronlauge werden 9,2 g Thioglykolsäure gelöst. Zu dieser Lösung gibt man unter Einleiten von Stickstoff und gutem Rühren bei 0 bis 5 C 18,4 g a-Phenoxypropion- säurechlorid innerhalb von 30 Minuten. Nach zwei stündigem Nachrühren bei 20 C wird mit Salzsäure angesäuert. Das abgeschiedene Öl wird mit Äther aufgenommen.
Nach dem Trocknen und Verdampfen des Äthers hinterbleiben 22,9 g a-Phenoxypropionyl- thioglykolsäure als viskoses Öl, das nach längerem Stehen kristallisiert. F. 52 .
<I>Beispiel 2</I> Einer wie in Beispiel 1 gewonnenen Bakterien suspension werden 0,025 % 6-Aminopenicillansäure, 0,5 % o-Chlorphenoxyacetylthioglykolsäure und 0,1 Toluol zugesetzt. Das Reaktionsgemisch wird dann mit Salzsäure auf pH 5,0 eingestellt und eine Stunde bei 37 C aufbewahrt. Nach dieser Zeit enthält der Ansatz 324 Einheiten Penicillinaktivität pro cm3, gemessen an einem Penicillin -G-Standard im Platten test mit Bac. subtilis ATCC 6633.
Diese Penicillin- aktivität wird von der bei der enzymatischen Reak tion entstandenen o - Chlorphenoxyacetyl - 6 - amino- penicillansäure bewirkt.
Die verwendete o-Chlorphenoxyacetylthioglykol- säure wird aus o-Chlorphenoxyacetylchlorid mit Thioglykolsäure nach Art der Phenoxypropionylthio- glykolsäure (siehe Beispiel 1) hergestellt. Sie bildet farblose Kristalle vom F. 120 .
<I>Beispiel 3</I> Einer wie in Beispiel 1 gewonnenen Bakterien suspension werden 0,125 % 6-Aminopenicillansäure, 0,5 % p-Methoxyphenoxyacetylthioglykolsäure und 0,1 % Toluol zugesetzt. Das Reaktionsgemisch wird dann mit konzentrierter Sodalösung auf pH 6,5 ein gestellt und eine Stunde bei 37 C aufbewahrt.
Nach dieser Zeit enthält der Ansatz 744 Einheiten Peni- cillinaktivität pro cm3, gemessen an einem Peni- cillin-G-Standard im Plattentest mit Bac. subtilis ATCC 6633. Diese Penicillinaktivität wird von der bei der enzymatischen Reaktion entstandenen p-Methoxyphenoxyacetyl-6-aminopenicillansäure be wirkt.
Die verwendete p-Methoxyphenoxyacetylthio- glykolsäure wird aus p-Methoxyphenoxyacetylchlorid mit Thioglykolsäure dargestellt (siehe Beispiel 1). Farblose Kristalle vom F. 85 .
<I>Beispiel 4</I> Einer wie in Beispiel 1 gewonnenen Bakterien suspension werden 0,125 % 6-Aminopenicillansäure, 1,0% 2,6-Dichlorphenoxyacetylthioglykolsäure und 0,1 % Toluol zugesetzt. Das Reaktionsgemisch wird dann mit Salzsäure auf pH 5,0 eingestellt und eine Stunde bei 37 C aufbewahrt.
Nach dieser Zeit ent hält der Ansatz 247 Einheiten Penicillinaktivität pro cm3, gemessen an einem Penicillin-G-Standard im Plattentest mit Bac. subtilis ATCC 6633. Diese Penicillinaktivität wird von der bei der enzyma tischen Reaktion entstandenen 2,6-Dichlorphenoxy- acetyl-6-aminopenicillansäure bewirkt.
Die verwendete 2,6-Dichlorphenoxyacetylthio- glykolsäure wird aus 2,6-Dichlorphenoxyacetylchlorid mit Thioglykolsäure hergestellt (siehe Beispiel 1). Sie bildet farblose Kristalle vom F. 60 .
<I>Beispiel S</I> Einer wie in Beispiel 1 gewonnenen Bakterien suspension werden<B>0,125%</B> 6-Aminopenicillansäure, 1,0% p-Nitrophenoxyacetylthioglykolsäure und 0,1 Toluol zugesetzt. Das Reaktionsgemisch wird dann mit Salzsäure auf pH 5,0 eingestellt und eine Stunde bei 37 C aufbewahrt. Nach dieser Zeit enthält der Ansatz 256 Einheiten Penicillinaktivtät pro cm3, ge messen an einem Penicillin-G-Standard im Platten test mit Bac. subtilis ATCC 6633.
Diese Penicillin aktivität wird von der bei der enzymatischen Reak, tion entstandenen p-Nitrophenoxyacetyl-6-amino- penicillansäure bewirkt. Die p-Nitrophenoxyacetyl- thioglykolsäure wird aus p-Nitrophenoxyacetylchlorid mit Thioglykolsäure hergestellt (siehe Beispiel 1). Sie bildet ein hellbraunes Pulver vom F.<B>101'.</B>

from these extracts, enzymes or enzyme enrichments which are able to link phenylacetic acid with the amino group of 6-aminopenicillanic acid in an amide-like manner.
The 6-aminopenicillanic acid used can be either the synthetic substance or the crystallized substance obtained by degradation or, if appropriate, enriched crude solutions, such as those obtained from fermentations or other processes.
Hydroxy and mercapto compounds in the context of the invention are alcohols, mercaptans, phenols and thiophenols. Examples of alcohols are ethanol, butanol, dodecyl alcohol, cyclohexanol, phenylmethylcarbinol and glycolic acid. Suitable mercaptans are mercaptobenzothiazole, thioglycolic acid,
2-mercaptobutyric acid and cysteamine.
Phenol, p-oxybenzoic acid, thiophenol or thiocresol are suitable as phenols or thiophenols.
Examples of useful compounds are phenylacetate, methyl phenoxyacetate, α-phenoxypropionylthioglycolic acid, o-chlorophenoxyacetylthioglycolic acid amide, sodium p-chlorophenoxyacetylthioglycolic acid. Carboxylic acid esters of such alcohols and mercaptans, which in turn carry carboxyl groups, for example carboxylic acid esters of glycolic and thioglycolic acid, are particularly suitable.
The presence of the enzymes mentioned is necessary since the carboxylic acid esters alone are not able to acylate 6-amino penicillanic acid to the desired 6-acyl derivatives in an aqueous medium at pH 4-8. Comparative tests have shown that, for example, a-phenoxypropionylthioglycolic acid,
Treated with 6-aminopenicillanic acid at pH 4-8, does not provide a-phenoxyproponyl - 6 - aminopenicillanic acid, while the addition of an enzyme solution causes the desired synthesis to be achieved quickly and in high yield. It is not necessary to isolate the synthesis-causing enzymes. Rather, bacterial cells which contain these enzymes or their lysates can already be used as reaction-causing agents.
A particular advantage of the present process is furthermore to produce in high yield those 6-acylaminopenicillanic acids with the valuable antibiotic properties of penicillins which are not produced by fermentation with penicillin-forming fungi in the presence of the corresponding carboxylic acids (so-called precursors).
<I> Example 1 </I> 160 liters of 2% by volume corn steep liquor solution, containing 0.2 / 10 potassium phenyl acetate, are adjusted to pH 7.0 with potassium hydroxide solution and heated to 120 ° C. for 30 minutes. After cooling, the solution is clarified by centrifugation and sterilized in a fermenter at 110 ° C. for 40 minutes.

The reaction mixture is then adjusted to pH 6.5 with concentrated soda solution and stored at 37 ° C. for one hour. After this time, the batch contains 884 units of penicillin activity per cm3, measured on a penicifin-G standard in the Bac plate test. subtilis ATCC 6633.

Production of a - phenoxypropionylthioglycolic acid: 9.2 g of thioglycolic acid are dissolved in 200 cm3 of 4.4% sodium hydroxide solution. 18.4 g of α-phenoxypropionic acid chloride are added to this solution at 0 ° to 5 ° C. while passing in nitrogen and stirring thoroughly over the course of 30 minutes. After stirring for two hours at 20 ° C., the mixture is acidified with hydrochloric acid. The separated oil is absorbed with ether.

<I> Example 3 </I> 0.125% 6-aminopenicillanic acid, 0.5% p-methoxyphenoxyacetylthioglycolic acid and 0.1% toluene are added to a bacterial suspension obtained as in Example 1. The reaction mixture is then adjusted to pH 6.5 with concentrated soda solution and stored at 37 ° C. for one hour.
