Verfahren zur Herstellung von chlorsubstituierten stickstoffhaltigen Heterocyclen
Die Erfindung betrifft ein Verfahren zur Herstellung von chlorhaltigen Verbindungen der 1,3 ,4-Thia- diazol-, 1,3-Benzthiazol-, 1,3-Benzoxazol- und Pyrimidinreihe, deren Chloratom bzw. -atome in Nachbarstellung zum Ringstickstoff steht.
Stickstoffhaltige Heterocyclen, welche in a-Stellung zum Ringstickstoff ein Chloratom enthalten, wurden bisher aus den entsprechenden Hydroxyverbindungen mit starken Chlorierungsmitteln, wie Phosphorpentachlorid, Phosphoroxychlorid oder Phosphortrichlorid, hergestellt. Ferner ist es bekannt, sie aus entsprechenden Aminoverbindungen durch Einwirkung von salpetriger Säure in Gegenwart von Chlorionen zu erhalten. Diese Hydroxy- bzw. Aminoverbindungen sind aber nicht sehr einfach zugänglich; ausserdem sind die bisherigen Verfahren umständlich und geben die Halogenverbindungen nur in mässigen Ausbeuten.
Es wurde gefunden, dass man chlorhaltige Verbindungen der 1,3, 4-Thiadiazol-, 1, 3-Benzthiazol-, 1,3-Benzoxazol- und der Pyrimidinreihe, deren Chloratom bzw. -atome dem Ringstickstoff benachbart ist (sind), erhält, wenn man die entsprechenden Mercaptoverbindungen mit Phosgen umsetzt.
Der Verlauf der Reaktion war überraschend, da es bekannt ist, dass man bei der Einwirkung von Phosgen auf Mercaptoverbindungen der Imidazolreihe Diimidazoldisulfide erhält.
Die für das Verfahren als Ausgangsstoffe dienenden Verbindungen sind Mercaptoverbindungen der 1,3,4-Thiadiazol-, 1,3-Benzthiazol-, 1, 3-Benzoxazolund Pyrimidinreihe, deren Mercaptogruppe in a-Stellung zu einem Ringstickstoff steht, die also ringförmig gebunden die Gruppierung

einmal oder zweimal enthalten. Verbindungen dieser Art sind z. B.
2-Mercaptothiadiazol-(1,3,4),

Diese Mercaptoverbindungen werden vorteilhaft in Gegenwart von Lösungs- oder Verdünnungsmitteln mit Phosgen umgesetzt. Als Lösungs- oder Verdünnungsmittel kommen inerte organische Medien in Betracht, vorzugsweise aliphatische oder aromatische Kohlenwasserstoffe, aliphatische oder aromatische Halogenkohlenwasserstoffe oder Äther, beispielsweise Benzol, Toluol, Xylol, Ligroin, Chlorbenzol, Chloroform, Tetrahydrofuran, Dioxan. Der Siedebereich dieser Lösungsmittel soll zwischen etwa 35 und 2500 C liegen. Die Mengenverhältnisse zwischen Lösungsmittel und Mercaptoverbindung können in weiten Grenzen schwanken, z. B. zwischen 2:1 und 15 : 1.
Die Umsetzung kann bei Raumtemperatur oder bei erhöhter Temperatur, z. B. bei Siedetemperatur des gewählten Lösungsmittels, vorgenommen werden.
Mercaptoverbindung und Phosgen werden zweckmässig in etwa äquivalenten Mengen angewendet; im allgemeinen wird ein geringer Phosgenüberschuss verwendet.
Die Mercaptoverbindung bildet mit Phosgen zunächst ein loses (leicht zurückspaltbares) Addukt, das in manchen Fällen isoliert werden kann, dessen Isolierung aber weder nötig noch vorteilhaft ist. Dieses Zwischenprodukt zerfällt dann unter Abgabe von Kohlenoxysulfid und Chlorwasserstoff in die gewünschte Chlorverbindung. Der Zerfall des Zwischenproduktes wird zweckmässig durch Katalysatoren undfoder durch Erwärmen beschleunigt; beispielsweise sind als Katalysatoren N-substituierte Säureamide niederer Carbonsäuren, wie Dimethylformamid, N,N'-Di-methylacetamid, oder N-substituierte cyclische Carbonsäureamide, wie N-Methylpyrrolidon, wirksam. Die zugrunde liegenden Carbonsäuren enthalten im allgemeinen 1 bis 6 C-Atome. Diese Katalysatoren werden in der Regel in einer Menge von etwa 0,2 bis 0,5 Ges. % zugegeben.
Eine grössere Menge schadet jedoch nicht.
Die Reaktion kann für das Beispiel des 2-Mercaptobenzthiazols wie folgt formuliert werden:

Die gemäss der Erfindung erhältlichen Verbindungen sind wertvolle Zwischenprodukte, z. B. für die Herstellung von Farbstoffen.
Zur Durchführung des Verfahrens kann man Phosgen auf die Mercaptoverbindung leiten und einwirken lassen; zweckmässig ist es jedoch, das Phosgen in eine Suspension oder Lösung der Mercaptoverbindung einzuleiten oder in die Mercaptoverbindung in eine Phosgenlösung einzutragen. Wenn man das Phosgen in die Lösung oder Suspension der Mercaptoverbindung einleitet, so ist es vorteilhaft, die Umsetzung bei der Siedetemperatur des betreffenden Lösungsmittels durchzuführen. Wenn man dagegen das Phosgen im Lösungsmittel vorgibt und die Mercaptoverbindung einträgt, ist es zweckmässig, zunächst bei Raumtemperatur oder tiefer, z. B. bei - 100 C, die Reaktionsteilnehmer zu mischen und anschliessend auf Siedetemperatur des Gemisches zu erhitzen, wobei man gegebenenfalls noch weiter Phosgen einleitet.
Eine Erwärmung auf mehr als 2000 C ist jedoch nicht erforderlich. Die Reaktion läuft aber auch, allerdings langsam, in der Kälte, das heisst im Bereich von 0 bis 30" C, ab.
Im allgemeinen wird das Verfahren unter Atmosphärendruck ausgeführt. Man kann das Phosgen aber auch unter höherem Druck, z. B. 2 bis 3 atü, aufpressen oder die Reaktionsteilnehmer bei Raumtemperatur oder bei tieferer Temperatur mischen und dann etwa im Autoklaven erhitzen, wobei sich ein höherer Druck einstellt, der sich aus dem Dampfdruck des evtl. Lösungsmittels, des Phosgens und des als Umsetzungsprodukt entstehenden Kohlenoxysulfids ergibt. Da es für den Ablauf der Reaktion günstig ist, gebildetes Kohlenoxysulfid aus dem Reaktionsgemisch zu entfernen, wird die drucklose Ausführung bevorzugt.
Wenn man niedere N-Alkylcarbonsäureamide als Katalysator zur Beschleunigung der Kohlenoxysulfidabspaltung zusetzt, so kann man diese katalytisch wirksamen Stoffe bereits von Anfang an zugeben; vielfach ist es jedoch vorteilhaft, zunächst die Reaktionsteilnehmer, das heisst die Mercaptoverbindung und das Phosgen, gegebenenfalls in einem Lösungsmittel, in der Kälte zu mischen und erst dann die katalytisch wirksamen Stoffe zuzusetzen. Erwärmen der Lösung auf etwa 60 bis 100 C C beschleunigt die Kohlenoxysulfidabspaltung. Die Aufarbeitung des Reaktionsgemisches kann durch Destillation, zweckmässig unter vermindertem Druck, erfolgen.
In den nachfolgenden Beispielen sind unter Teilen Gewichtsteile zu verstehen.
Beispiel I
Eine Mischung von 100 Teilen 2-Mercaptopyrimidin und 400 Teilen Dioxan wird zum Sieden unter Rückfluss erhitzt. Während 3 Stunden werden 100 Teile Phosgen eingeleitet. Dann lässt man das Gemisch unter Einleiten von Stickstoff erkalten und filtriert. Aus dem Filtrat wird das Lösungsmittel abdestilliert. Der Rückstand liefert bei der Destillation unter vermindertem Druck 76 Teile (74% der Theo rie) 2-Chlorpyrimidin; Kp.10 = 75 bis 76 ( > C C; Fp. 63 bis 660 C.

Beispiel 2
46 Teile 2-Mercapto-4-methylpyrimidin und 250 Teile Dioxan werden auf 1000 C erhitzt und 4 Stun den lang Phosgen eingeleitet. Dann wird wie in Beispiel 1 aufgearbeitet. Man erhält 35 Teile (74% der Theorie) 2-Chlor-4-methylpyrimidin ; Kp., = 75 bis 79O C ; Fp.47 bis 48 C.
Beispiel 3
In eine Lösung von 50 Teilen Phosgen in 320 Teilen Dioxan werden bei Raumtemperatur 50 Teile 2-Mercapto-4,6-dimethylpyrimidin eingetragen. Dann wird 1 Teil Dimethylformamid zugesetzt und das Gemisch unter Einleiten eines schwachen Phosgenstromes zum Sieden erhitzt (2 Stunden). Das Gemisch wird nach Beispiel 1 aufgearbeitet. Man erhält 43 Teile (84% der Theorie) 2-Chlor-4,6-dimethylpyrimidin; Kr. 12 = 97 97 bis 99" C; Fp. 29 bis 31"C.

Beispiel 4
In eine Mischung aus 2000 Teilen Chloroform, 668 Teilen 2-Mercaptobenzthiazol und 3 Teilen Dimethylformamid werden bei Oo C 400 Teile Phosgen eingeleitet. Das Reaktionsgemisch wird 1 Stunde bei 0 bis 10 C gerührt und anschliessend die Temperatur langsam unter Einleiten von Phosgen auf 600 C gesteigert. Unter weiterem Einleiten eines schwachen Phosgenstromes wird 8 Stunden unter Rückfluss gekocht. Das Chloroform wird danach unter Normaldruck abgedampft und der Rückstand im Vakuum destilliert. Es werden 548 Teile (81% der Theorie) 2-Chlorbenzthiazol vom Siedepunkt 141"C/30 mm erhalten.

Beispiel 5
Bei 0 C werden in eine Lösung von 99 Teilen Phosgen und 1 Teil Dimethylformamid in 500 Teilen Chloroform 151 Teile 2-Mercaptobenzoxazol eingetragen. Es wird 1 Stunde bei 0 bis lOoC erwärmt.
Nach achtstündigem Kochen unter Einleiten von Phosgen wird das Reaktionsgemisch durch Destillation aufgearbeitet. Man erhält in 83% iger Ausbeute 2-Chlorbenzoxazol vom Siedepunkt 1050 C/32 mm und Schmelzpunkt von 4,5 C.

Beispiel 6
In eine Lösung von 180 Teilen Phosgen und 400 Teilen Chloroform werden 60 Teile 2,5-Dimercapto- 1, 3, 4-thiadiazol bei 20 bis 25O C eingetragen. Das Gemisch wird 2 Stunden gerührt. Dann werden 2 Teile Dimethylformamid zugesetzt und weitere 48 Stunden bei Raumtemperatur gerührt. Nach Filtrieren wird das Lösungsmittel abdestilliert. Es werden als Rückstand 57 Teile 2,5-Dichlor-1,3,4-thiadiazol vom Fp. 66 bis 680 C erhalten. Die Ausbeute beträgt 92% der Theorie. Das Produkt kann, wenn erforderlich, durch Wasserdampfdestillation gereinigt werden.
Beispiel 7
In eine Lösung von 33 Teilen Phosgen in 200 Teilen Tetrahydrofuran werden 28 Teile 2-Mercaptopyrimidin eingetragen, wobei die Temperatur von 15 auf 23"C C steigt. Das Gemisch wird 5 Tage bei Raumtemperatur gerührt und dann abgesaugt. Aus dem Filtrat wird das Lösungsmittel abdestilliert und der Rückstand einer Wasserdampfdestillation unterworfen, aus deren Destillat 9 Teile 2-Chlorpyrimidin vom Schmelzpunkt 64 bis 65O C erhalten werden.

It is also known that they can be obtained from corresponding amino compounds by the action of nitrous acid in the presence of chlorine ions. These hydroxyl or amino compounds are not very easily accessible; In addition, the previous processes are cumbersome and give the halogen compounds only in moderate yields.
It has been found that chlorine-containing compounds of the 1,3, 4-thiadiazole, 1,3-benzothiazole, 1,3-benzoxazole and pyrimidine series whose chlorine atom or atoms is (are) adjacent to the ring nitrogen are obtained if the corresponding mercapto compounds are reacted with phosgene.

The reaction can be carried out at room temperature or at an elevated temperature, e.g. B. at the boiling point of the selected solvent.
Mercapto compound and phosgene are expediently used in approximately equivalent amounts; in general a slight excess of phosgene is used.
The mercapto compound initially forms a loose (easily cleavable) adduct with phosgene, which can be isolated in some cases, but its isolation is neither necessary nor advantageous. This intermediate product then breaks down into the desired chlorine compound, releasing carbon oxysulphide and hydrogen chloride. The disintegration of the intermediate product is expediently accelerated by catalysts and / or by heating; for example, N-substituted acid amides of lower carboxylic acids, such as dimethylformamide, N, N'-dimethylacetamide, or N-substituted cyclic carboxamides, such as N-methylpyrrolidone, are effective as catalysts. The underlying carboxylic acids generally contain 1 to 6 carbon atoms. These catalysts are generally added in an amount of about 0.2 to 0.5% by weight.

The compounds obtainable according to the invention are valuable intermediates, e.g. B. for the production of dyes.
To carry out the process, phosgene can be passed onto the mercapto compound and allowed to act; However, it is expedient to introduce the phosgene into a suspension or solution of the mercapto compound or to introduce it into the mercapto compound in a phosgene solution. If the phosgene is introduced into the solution or suspension of the mercapto compound, it is advantageous to carry out the reaction at the boiling point of the solvent in question. Since it is beneficial for the reaction to proceed to remove carbon oxysulphide formed from the reaction mixture, the pressureless version is preferred.
If lower N-alkylcarboxamides are added as a catalyst to accelerate the cleavage of carbon oxysulphide, these catalytically active substances can be added right from the start; In many cases, however, it is advantageous to first mix the reactants, that is to say the mercapto compound and the phosgene, if appropriate in a solvent, in the cold and only then to add the catalytically active substances. Heating the solution to about 60 to 100 ° C accelerates the carbon oxysulphide elimination. The reaction mixture can be worked up by distillation, advantageously under reduced pressure.

Example I.
A mixture of 100 parts of 2-mercaptopyrimidine and 400 parts of dioxane is heated to the boil under reflux. 100 parts of phosgene are passed in over the course of 3 hours. The mixture is then allowed to cool while passing in nitrogen and is filtered. The solvent is distilled off from the filtrate. The residue gives 76 parts (74% of theory) of 2-chloropyrimidine on distillation under reduced pressure; Bp 10 = 75 to 76 (> C C; mp 63 to 660 C.

Example 2
46 parts of 2-mercapto-4-methylpyrimidine and 250 parts of dioxane are heated to 1000 ° C. and phosgene is introduced for 4 hours. Then it is worked up as in Example 1. 35 parts (74% of theory) of 2-chloro-4-methylpyrimidine are obtained; B.p. = 75 to 790 C; Fp. 47 to 48 C.

50 parts of 2-mercapto-4,6-dimethylpyrimidine are introduced into a solution of 50 parts of phosgene in 320 parts of dioxane at room temperature. Then 1 part of dimethylformamide is added and the mixture is heated to boiling while passing in a gentle stream of phosgene (2 hours). Example 4
400 parts of phosgene are introduced at 0 ° C. into a mixture of 2000 parts of chloroform, 668 parts of 2-mercaptobenzothiazole and 3 parts of dimethylformamide. Example 5
At 0 C, 151 parts of 2-mercaptobenzoxazole are introduced into a solution of 99 parts of phosgene and 1 part of dimethylformamide in 500 parts of chloroform. It is heated for 1 hour at 0 to 10oC.
After boiling for eight hours while passing in phosgene, the reaction mixture is worked up by distillation. 2-chlorobenzoxazole with a boiling point of 1050 ° C./32 mm and a melting point of 4.5 ° C. is obtained in a yield of 83%.

Example 6
60 parts of 2,5-dimercapto-1,3,4-thiadiazole at 20.degree. To 250.degree. C. are introduced into a solution of 180 parts of phosgene and 400 parts of chloroform. The mixture is stirred for 2 hours. Then 2 parts of dimethylformamide are added and the mixture is stirred for a further 48 hours at room temperature. After filtering, the solvent is distilled off. 57 parts of 2,5-dichloro-1,3,4-thiadiazole with a melting point of 66 ° to 680 ° C. are obtained as residue. The yield is 92% of theory. If necessary, the product can be purified by steam distillation.
Example 7
28 parts of 2-mercaptopyrimidine are introduced into a solution of 33 parts of phosgene in 200 parts of tetrahydrofuran, the temperature rising from 15 to 23 "CC. The mixture is stirred at room temperature for 5 days and then filtered off with suction. The solvent is distilled off from the filtrate and the residue is subjected to steam distillation, from the distillate of which 9 parts of 2-chloropyrimidine with a melting point of 64 to 65 ° C. are obtained.
