Die Herstellung der erfindungsgemäss zu verwen denden Ausgangsverbindungen kann erfolgen, indem man 6a-Methyl-llp,21-dioxy-4,17(20)-[cis]-pregna- dien-3-on (I, R = H), [Spero et a1., J.
Am. Chem. Soc. 78, 6213 (1956)] mit einem Di-nieder-alkyl-oxalat, vorzugsweise Methyl- oder Äthyloxylat, in Gegen wart eines Alkalimetall-Kondensationskatalysators in einem unter den Reaktionsbedingungen inerten Lösungsmittel, zu einem Alkalimetallenolat des 2- Alkoxyoxalyl-6a-methyl-11 @, 21-dioxy-4,17(20)-[cis]- pregnadien-3-ons (Il, M = Alkalimetall) umsetzt.
Diese Verbindung oder das entsprechende freie Enol (II, M = H) wird sodann mit Perchlorylfluorid (C10",F) zu 2-Fluor-2-alkoxyoxalyl-6.i-methyl-11,6',21- dioxy-4,17(20)-[cis]-pregnadien-3-on (11f) umgesetzt. Die Behandlung dieser Verbindung mit einer Base er gibt 2-Fluor-6a-methyl-11f,21-dioxy-4,17(20)-[cis]- pregnadien-3-on (IV).
Geht man von einem 21-Ester des 6a-Methyl- l 1 f,21-dioxy-4,17(20)-[cis]-pregna- dien-3-ons (I, R = Ac) und insbesondere dem Acetat aus, so erhält man die entsprechenden 2-Alkoxyoxalyl- verbindungen (II, R = Ac) und 2-Fluor-2-alkoxy- oxalylverbindungen (11I, R = Ac)
. Die Entfernung der 2-Alkoxyoxalylgruppe führt zu einer mindestens teilweisen Hydrolyse der 21-Estergruppe, so dass die 2-Fluor-2 1-oxyverbindung (IV, R = H) entsteht. Durch Veresterung dieser Verbindung, vorzugsweise mit Essigsäureanhydrid, wird die 21-Estergruppe wie der eingeführt (V, R = Ac).
Das erfindungsgemässe Verfahren umfasst sodann die oxydative Hydroxylierung dieser Verbindungen mit Osmiumtetroxyd und einem Oxydationsmittel, z. B. Wasserstoffsuperoxyd, einer Persäure, einem Alkylperoxyd, einem Aminoxydperoxyd, einem Aryliodoxyd, z. B. Phenyliodosoacetat, beispielsweise nach einer der im US-Patent Nrn. 2 769 825 und 2 769 823 oder bei Hogg. et a1., J.
Am. Chem. Soc. 77, 4436 (1955) beschriebenen Methode, unter Bildung eines 2-Fluor-6a-methyl-11f,17a,21-trioxy-4-pregnen- 3,20-dion-21-acylats, vorzugsweise des 21-Acetats (VI), welches mit einer Base zu 2-Fluor-6a-methyl- 11ss,17a,21-trioxy-4-pregnen-3,20-dion hydrolysiert oder mit einem Oxydationsmittel, wie beispielsweise Chromsäure, oder einem N-Halogenacylamid, z. B. N-Bromacetamid oder einem N-Halogenimid, z. B.
N-Chlorsuccinimid, in Pyridin oder dergleichen zum entsprechenden Ester des 2-Fluor-6:c-methyl-17a,21- dioxy-4-pregnen-3,11,20-trions oxydiert werden kann.
Die erfindungsgemäss erhältlichen 2-Fluorverbin- dungen besitzen hochgradige physiologische Eigen schaften sowie Aktivitätsspektren, welche von den jenigen der natürlichen Nebennierenrindenhormone wie Hydrocortison und Cortison oder anderer bekann ter Verbindungen ohne das zweiständige Fluor, ins besondere bezüglich ihres Mineral- und Wasserstoff- Wechsels verschieden sind. Diese Verbindungen weisen auch starke entzündungshemmende Wirksamkeit auf.
Sie erzeugen weniger Salz- und Wasserretention als die entsprechenden Analogen ohne die 2-Fluorgruppe, wodurch sie besonders wertvoll sind zur Behandlung von Entzündungserscheinungen im Zusammenhang mit chronischer, congestiver Herzschwäche, Leber- cirrhosis, Nierensyndromen sowie Eklampsie und Präeklampsie.
Die neuen 2-Fluorverbindungen sind in oralen, parenteralen oder topischen Zusammensetzungen an wendbar. Zur oralen Verabreichung kann das Ma terial in Form von Tabletten, beispielsweise unter Ver wendung von Polyäthylenglykol 4000 oder 6000 als Träger oder Laktose und/oder Saccharose als Verdün- ner verwendet werden. Zur örtlichen Anwendung eignen sie sich in Form von Salben, Lotionen, Geles, Cremen, Suppositorien, Stäbchen, wässrigen Suspen sionen usw.
Die Einverleibung eines Antibiotikums, insbeson dere Neomycinsulfat, in die pharmazeutische Zusam mensetzung bietet therapeutische Vorteile, da jeder der aktiven Bestandteile die guten Eigenschaften des anderen potenziert und ergänzt. Anstelle des oder zu sätzlich zu Neomycinsulfat können andere Antibiotika wie Novobiocin, Bacitracin, Circulin, Polymyxin-B- sulfat, Gramicidin, Streptomycinsulfat, Dihydro- streptomycinsulfat, Oxytetracyclin,
Chlortetracyclin, Tetracyclin, Chloramphenicol und die Sulfonamide zusammen mit den erfindungsgemäss hergestellten Steroiden für Präparate wie die obigen Salben verwen det werden.
<I>Präparat 1</I> 2 -Fluor- 6a- methyl-11 ss,21-dioxy-4,17(20)-[cis]- pregnadien-3-on.
Eine Lösung von 7,73 g 6a-Methyl-11ss,21-dioxy- 4, 17(20)-[cis]-pregnadien-3-on-21-acetat [Spero et a1., J. Am. Chem. Soc. 78, 6213 (1956)] in 100m1 tertiärem Butylalkohol wurde unter Rühren und Stickstoffatmosphäre auf 60 C erwärmt. Das Erwär men wurde sodann unterbrochen und 5,45 ml Äthyl- oxalat und 6,67 ml einer Methanollösung, welche 1,6 g Natriummethanolat enthielt, zugesetzt.
Nach halbstün digem Rühren wurden 100 ml Äther zugesetzt. Der entstandene gelbe Niederschlag wurde gesammelt, mit Äther gewaschen und getrocknet. Man erhielt 10,20 g.
Der Niederschlag wurde in 120 ml Methanol ge löst und die Lösung auf 0 C gekühlt. Dieser Lösung wurde unter Rühren und unter Stickstoffatmosphäre eine eisgekühlte Lösung von 3,34 g Perchlorylfluorid (CIO,F) in 100 ml Methanol zugesetzt, wobei die Reaktionstemperatur zwischen -5 und -15 C gehal ten wurde. Nach Zugabe allen Perchlorylfluorids wurde während einer weiteren halben Stunde gerührt und hierauf 7,35 ml 25'o/oiges Natriummethanolat zu gesetzt, das Rühren bei derselben Temperatur wäh rend weiteren 45 Minuten fortgesetzt und das Ge misch sodann unter vermindertem Druck konzentriert. Der Rückstand wurde mit einem Liter Eiswasser ver- mischt.
Der ausgefallene weisse Feststoff wurde gut mit Wasser verrieben und anschliessend im Vakuum- Exsiccator getrocknet. Es wurden 6,1g Material erhal ten, welche in Methylenchlorid gelöst und über eine 250-g-Säule Magnesiumsilikat (Florisil) geleitet wur den.
Die Säule wurde mit 125-ml-Portionen Lösungs mittel folgender Zusammensetzung und in folgender Reihenfolge entwickelt: 8 Portionen Hexankohlenwasserstoffe mit 7 % Aceton, 4 Portionen Hexankohlenwasserstoffe mit 1011/o Aceton, 5 Portionen Hexankohlenwasserstoffe mit 20% Aceton, und schliesslich 1 Portion Aceton.
Die Fraktionen 13 bis 17 eluierten 4,537 g 2-Fluor-6a-methyl-11ss,21- dioxy-4,17(20)-[cis]-pregnadien-3-on, F. 145 bis 155 C. Eine Umkristallisation aus Athylacetat hob den Schmelzpunkt auf 166 bis 168 C. Eine nochmals umkristallisierte Probe schmolz bei 188 bis 189 C und ergab folgende Analyse: Berechnet für C22H31F03: C 72,86; 1-18,62; F 5,24 gefunden: C 72,73; H 8,61; F 5,14. <I>Präparat 2</I> 2 -Fluor- 6a-methyl-l l ss,21-dioxy-4,17(20)-[cis]- pregnadien-3-on-21-acetat.
Eine Lösung von 3,338 g 2-Fluor-6a-methyl- llss,21-dioxy-4,17(20)-[cis]-pregnadien-3-on in etwa 20m1 Pyridin und 28m1 Essigsäureanhydrid wurde während etwa 16 Stunden auf Zimmertemperatur ge halten. Die Lösung wurde sodann in Wasser gegossen und führte zu einem amorphen Feststoff, welcher aus tertiärem Butylalkohol umkristallisiert 2-Fluor-6a- methyl -11 ss,21-dioxy-4,17(20)-[cis]-pregnadien-3-on- 21-acetat vom Schmelzpunkt 147 bis 156 C ergab.
<I>Beispiel</I> 2 -Fluor- 6a-methyl-11,8,17a,21-trioxy-4-pregnen- 3,20-dion-21-acetat.
Eine Lösung von 1,84 g 2-Fluor-6a-methyl- 11 ss,21-dioxy-4,17(20)-[cis]-pregnadien-3-on-21-acetat in 70 ml tertiärem Butylalkohol und 2,15 ml Pyridin wurde mit 2,77 g Osmiumtetroxyd in 0,94 ml ter tiärem Butylalkohol und 698 mg N-Methylmorpholin- oxydperoxyd (USP Nr. 2 769 823) in 6,05m1 ter tiärem Butylalkohol gemischt und während 16 Stun den bei Zimmertemperatur gerührt.
Ein Gramm Fil terhilfsmittel ( Magnesol >) und 17 ml einer 0,5 o/oigen wässrigen Natriumhydrosulfitlösung wurden sodann zugesetzt und das Gemisch filtriert, mit Wasser gewa schen und mit Äther extrahiert. Die Ätherlösung wurde mit Wasser, mit verdünnter Salzsäure und mit einer wässrigen Natriumchloridlösung gewaschen, über Natriumsulfat getrocknet und das Lösungsmittel ver dampft.
Das verbliebene Öl wurde in Methylenchlorid aufgelöst und über eine 130-g-Säule Magnesiumsilikat ( Florisil ) chromatographiert. Das gewünschte Pro- dukt wurde mit 15 und 20% Aceton in Hexanen ( Skellysolve B ) eluiert. Die derart gewonnenen 1,
305 g 2-Fluor-6a-methyl-llss,17a,21-trioxy-4-pre- gnen-3,20-dion-21-acetat waren amorph und konnten nicht kristallisiert werden. 2-Fluor-6a-methyl-11 ss, 17r-t,21-trioxy-4-pregnen-3,20-dion-21-acetat wies ein von<I>242</I> m,u. und a@L von 13 280 auf.
Gemäss dem oben beschriebenen Verfahren kön nen auch andere 21-Ester des 2-Fluor-6a-methyl-11ss, 21-dioxy-4,17(20)-[cis]-pregnadien-3-ons, beispiels weise die im letzten Absatz von Präparat 2 erwähnten, in die entsprechenden Ester eines 2-Fluor-6a-methyl- l 1ss,17a,21-trioxy-4-pregnen-3,20-dions umgewandelt werden.
Auf gleiche Weise wird 2-Fluor-6a-methyl- llss,21-dioxy-4,17(20)-[cisl-pregnadien-3-on zu 2- Fluor- 6a-methyl-11 ss,17a,21-trioxy-4-pregnen-3,20- dion umgesetzt.
Das so erhaltene Produkt kann wie folgt zum 2-Fluor- 6a-methyl-17a,21-dioxy-4-pregnen-3,11,20- trion-21-acetat oxydiert werden: Eine Lösung von 1 g Natriumbichromat-Dihydrat in 20 ml Essigsäure wurde auf Zimmertemperatur ge kühlt und anschliessend mit einer Lösung von 1,45 g rohem 2-Fluor-6a-methyl-11ss,17a,21-trioxy-4-pre- gnen-3,20-dion-21-acetat in 20m1 Essigsäure ver mischt. Das Gemisch wurde während 1 Stunde stehen gelassen und anschliessend mit Methylenchlorid extra hiert.
Die Extrakte wurden nacheinander mit wäss- rigem Natriumsulfit, mit wässrigem Natriumbicarbonat und mit Wasser gewaschen, über Natriumsulfat ge trocknet und das Lösungsmittel verdampft. Der Rück stand wurde aus Methanol umkristallisiert und ergab 0,4 g 2-Fluor-6a-methyl-17a,21-dioxy-4-pregnen-3, 11,20-trion-21-acetat, das bei 222 bis 225 C schmolz und bei 207 C sinterte, ein [aln von + 216 in Chloroform und folgende Analyse aufwies: Berechnet für C,4H31F06: C 66,34;<B>1-17,19</B> gefunden: C 66,94; H 7,24.
Hydrolyse des Acetats führt zum 2-Fluor-6a- methyl-11 ss,17a-21-trioxy-4-pregnen-3,20-dion: Eine Lösung von 438 mg 2-Fluor-6a-methyl-11ss, 17a,21-trioxy-4-pregnen-3,20-dion-21-acetat in 4 ml Methanol wurde durch Einleiten von Stickstoff vom Sauerstoff befreit. Eine Lösung von 404 ml Kalium- bicarbonat in 4 ml Wasser wurde auf ähnliche Weise von Sauerstoff befreit. Die beiden Lösungen wurden bei einer Temperatur zwischen 18 und 20 C unter Stickstoffatmosphäre gemischt.
Das Gemisch wurde sodann während 5 Stunden bei Zimmertemperatur gerührt, wobei es mit Stickstoff vor der Einwirkung von Sauerstoff geschützt wurde. Nach dieser Zeit wurde die Lösung mit Eisessig neutralisiert. Die neu trale Lösung wurde sodann durch Destillation bei Zimmertemperatur und unter vermindertem Druck konzentriert und anschliessend während 16 Stunden im Kühlschrank gekühlt. Das ausgefallene 2-Fluor-6a- methyl-11 ss,17a,21-trioxy-4-pregnen-3,20-dion wurde filtriert und getrocknet.
Das 2-Fluor-6a-methyl-17a,21-dioxy-4-pregnen- 3,11,20-trion-21-acetat kann analog hydrolysiert wer den.

305 g of 2-fluoro-6a-methyl-llss, 17a, 21-trioxy-4-pregnen-3,20-dione-21-acetate were amorphous and could not be crystallized. 2-fluoro-6a-methyl-11 ss, 17r-t, 21-trioxy-4-pregnen-3,20-dione-21-acetate had a grade of <I> 242 </I> m, u. and a @ L of 13,280.
According to the method described above, other 21-esters of 2-fluoro-6a-methyl-11ss, 21-dioxy-4,17 (20) - [cis] -pregnadien-3-ones, for example those in the last paragraph of preparation 2 mentioned, are converted into the corresponding esters of a 2-fluoro-6a-methyl- l 1ss, 17a, 21-trioxy-4-pregnen-3,20-dione.
In the same way, 2-fluoro-6a-methyl-11ss, 21-dioxy-4,17 (20) - [cisl-pregnadien-3-one becomes 2-fluoro-6a-methyl-11ss, 17a, 21-trioxy -4-pregnen-3,20- dione implemented.
The product obtained in this way can be oxidized to 2-fluoro-6a-methyl-17a, 21-dioxy-4-pregnen-3,11,20-trione-21-acetate as follows: A solution of 1 g of sodium dichromate dihydrate in 20 ml of acetic acid was cooled to room temperature and then with a solution of 1.45 g of crude 2-fluoro-6a-methyl-11ss, 17a, 21-trioxy-4-pregnen-3,20-dione-21-acetate in 20m1 acetic acid mixed. The mixture was left to stand for 1 hour and then extracted with methylene chloride.
The extracts were washed successively with aqueous sodium sulfite, with aqueous sodium bicarbonate and with water, dried over sodium sulfate and the solvent was evaporated. The residue was recrystallized from methanol and gave 0.4 g of 2-fluoro-6a-methyl-17a, 21-dioxy-4-pregnen-3, 11,20-trione-21-acetate, which melted at 222-225C and sintered at 207 C, had an [aln of +216 in chloroform and the following analysis: Calculated for C, 4H31F06: C, 66.34; found: C, 66.94; H 7.24.
Hydrolysis of the acetate leads to 2-fluoro-6a-methyl-11ss, 17a-21-trioxy-4-pregnen-3,20-dione: a solution of 438 mg of 2-fluoro-6a-methyl-11ss, 17a, 21 -trioxy-4-pregnen-3,20-dione-21-acetate in 4 ml of methanol was freed from oxygen by passing in nitrogen. A solution of 404 ml of potassium bicarbonate in 4 ml of water was deoxygenated in a similar manner. The two solutions were mixed at a temperature between 18 and 20 C under a nitrogen atmosphere.
The mixture was then stirred at room temperature for 5 hours while being protected from exposure to oxygen with nitrogen. After this time the solution was neutralized with glacial acetic acid. The neutral solution was then concentrated by distillation at room temperature and under reduced pressure and then cooled in the refrigerator for 16 hours. The precipitated 2-fluoro-6amethyl-11ss, 17a, 21-trioxy-4-pregnen-3,20-dione was filtered and dried.
The 2-fluoro-6a-methyl-17a, 21-dioxy-4-pregnen-3,11,20-trione-21-acetate can be hydrolyzed analogously to whoever.
