Process for the preparation of new N-heterocyclic compounds The present invention relates to a process for the preparation of new N-heterocyclic compounds with valuable pharmacological properties.
It has been found that basically substituted iminodibenzyls (10,11-dihydro-5H-dibenzo [b, f] -azepines of the formula

where Y is hydrogen or a halogen atom, Z is hydrogen or a halogen atom, one of the symbols R1 and R2 is the methyl group and the other hydrogen, and R3 and R4 are low molecular weight alkyl radicals, which are also directly or via an oxygen atom or a low molecular weight "#,"

According to the invention, the compounds of the formula I are prepared by adding a compound of the formula

available. The reaction can be carried out in the presence or absence of an inert organic solvent, examples of which are benzene,
Toluene and xylene may be mentioned.
Compounds of the formula III which are suitable for the preparation of starting materials of the formula II are, in addition to the basic body iminodibenzyl, for example 3,7-dichloro-iminodibenzyl and the monosubstituted compounds 3-chloro-iminodibenzyl and 3-bromo-iminodibenzyl. Their production can, for. B.
by treating 3,5-diacetyl-iminodibenzyl with hydrazoic acid according to Schmitt, followed by partial hydrolysis of the acetamino compound to 3-amino-5-acetyl-iminodibenzyl, diazotization of the same, conversion of the diazonium halides with cupric halides according to Sandmeyer and hydrolysis to 3-halo-iminodibenzyl .
Basic alcohols of the formula IV are γ-dimethylamino-n-butanol, γ-diethylamino-n-butanol, γ-di-n-butylamino-n-butanol, γ-pyrrolidino-n-butanol, γ-piperidino-n-butanol , y-morpholino-n-butanol, - (4-methyl-piperazino) -n-butanol, y- (4-hydroxyethyl-piperazino) -n-butanol and y- (4-acetoxyethyl-piperazino) -n-butanol as well the y-dimethylamino-ss-methyl-propanol,
y-diethylamino-ss-methyl propanol, y-pyrrolidine-ss-methyl-propanol, y-piperidino-ss-methyl-propanol, y-morpholino-ss-methyl-propanol, y- (4-methyl-piperazino) ss- methyl-propanol, y- (4-hydroxyethyl-piperazino) s-methyl-propanol and y- (4-acetoxyethyl-piperazino) -ss-methyl-propanol. The former group of basic alcohols pick is z.
B., starting from 1,3-butylene glycol, by reacting it with acetyl chloride to give 3-chloro-n-butyl acetate and reacting the latter with a suitable secondary amine.
With inorganic or organic acids, such as hydrochloric acid, hydrobromic acid, sulfuric acid, phosphoric acid, methanesulfonic acid, maleic acid, fumaric acid, tartaric acid, benzoic acid, phthalic acid or 8-chlorotheophylls, the compounds of the formula I form salts, some of which are water-soluble.
In the following example, parts are parts by weight, unless otherwise noted; these are related to parts of volume as g to cm3. The temperatures are given in degrees Celsius. Example 26.5 parts of 5-chlorocarbonyl-iminodibenzyl (prepared from iminodibenzyl and phosgene) in 200 parts by volume of abs. Toluene are added dropwise to 35 parts of γ-dimethylamino-n-butanol within about one hour.
After the addition, the reaction mixture is refluxed for about 16 hours, whereupon it is cooled and set with water. The organic layer is separated, washed with water, and the basic components are extracted with 2N hydrochloric acid. The acidic solution is made alkaline with K.C03 solution and extracted with ether. The residue is the crude iminodibenzyl-5-carboxylic acid (y-dimethylamino-n-butyl) ester.
The reaction product obtained in this way is heated in an oil bath to 200-250 for about 6 hours until the evolution of CO 2 has ended. The residue is then distilled in a high vacuum, where the 5- (γ-dimethylamino-n-butyl) -iminodibenzyl at 165 / 0.02 Torr. transforms.
The hydrochloride prepared with ethanolic hydrochloric acid melts at 146-148 (from acetone).
In an analogous manner, the following is obtained: 5- (γ-diethylamino-n-butyl) -iminodibenzyl, b.p., o.os 196-200, hydrochloride mp. 165-166. 5- (y-piperidino-n-butyl) -3-chloro-iminodibenzyl, 5- (y-methyl-ethylamino-n-butyl) -iminodibenzyl, 5- (y-methyl-ethylamino-n-butyl) -3- chloro-iminodibenzyl, 5- (γ-morpholino-ss-methyl-propyl) -ininodibenzyl, bp.
